RESUMO

The structure of monkey (Chlorocebus aethiops) heparin-binding EGF-like growth factor (HB-EGF) gene has been investigated in this work in comparison with the known structure of human gene. It was shown that HB-EGF short form (SF-HB-EGF) specific exon 3a is mapped between exons 3 and 4 at distance 700 b.p. from exon 4. In a number of human and simian cell lines the main part of SF-HB-EGF mRNA does not contain HB-EGF mRNA specific exons 4 and 5. In comparison with HB-EGF mRNA in SF-HB-EGF mRNA P-form, but not L-form of is predominant, and this mRNA encodes a polypeptide with changed propeptide structure. Labeled SF-HB-EGF competes with HB-EGF and EGF for binding sites at A431 cell surface, which may be due to interaction with specific receptor. All the data suggest a specific role of SF-HB-EGF in cellular signalization.

RESUMO

A new mRNA coding for the heparin-binding EGF-like growth factor (HB-EGF) was found in Vero cells. The corresponding cDNA had C-156 in place of T, which resulted in a loss of the NheI site and a substitution of Leu-33 with Pro in the HB-EGF precursor. The known and new forms of the precursor were accordingly termed L and P. A possible conformational change in the corresponding propeptide region were assumed to affect processing of soluble secreted HB-EGF. The L and P mRNAs are differently expressed in various cell lines and have the identical 5'-untranslated sequences. Possibly, they are transcribed from one promoter and then alternatively spliced. Stimulation of resting Vero cells with tetraphorbol ester (TPA) substantially increased production of the L form, decreased production of the P form, and did not affect expression of the total HB-EGF mRNA. This was associated with an increase in binding of the diphtheria toxin, suggesting that the L HB-EGF precursor acts as its receptor.

RESUMO

In all secreted proteins related to the epidermal growth factor (EGF), EGF domains that occur in a mature factor are each encoded by two exons, and those that do not, by one exon. During splicing, additional exon 3a can be inserted between exons 3 and 4, which code for the EGF domain of the mature heparin-binding EGF-like growth factor (HB-EGF). The resulting mRNA codes for the short form of HB-EGF (SF HB-EGF), which retains the signal peptide, the propeptide, and the heparin-binding domain. However, its EGF domain lacks the C-terminal subdomain essential for the interaction with the EGF receptor (EGFR). Structural analysis suggested that SF HB-EGF is a secreted polypeptide that has high affinity for heparin, but weakly, if at all, interacts with EGFR. Data obtained in three different systems indicated that SF HB-EGF possesses a mitogenic activity but utilizes a signal transduction pathway other than that of HB-EGF.

RESUMO

All the structural genes of riboflavin biosynthesis are shown to be located on the 2.8 MD DNA fragment, using the collection of plasmids, carrying the Bacillus subtilis riboflavin operon fragments and Bacillus subtilis strains, containing various deletions of rib-operon for analysis. The proximal Bgl II site is shown to be located between promoter P1 and the first structural gene ribG. The distal Hind III site of fragment C is the left bound of the rib-operon.

RESUMO

The 210 degrees region of Bacillus subtilis DNA containing the rib operon and genes for the first (dapA) and last (lysA) steps of lysine biosynthesis was cloned. PstI fragments of B. subtilis m.m. 4.7 MD DNA containing the lys and the proximal part of rib operon were isolated from different B. subtilis strains (SB25 and SHgW) and shown to have the same restriction and genetic maps. The restriction mapping of EcoRI fragment of B. subtilis m.m. 6.3 MD DNA containing the rib operon has been carried out.

RESUMO

The possibility of expression of the Bacillus pumilus chloramphenicol acetyltransferase gene (cat) in Bacillus subtilis from the pR promoter of phage lambda has been investigated in this work. For this purpose, the plasmid pPL703 carrying the B. pumilus DNA segment with the cat gene lacking promoter has been combined with the plasmid pBM21 containing the pR promoter. The recombinant plasmid pEL1 is capable of providing the 60 mkg/ml chloramphenicol resistance in Bac. subtilis cells.

RESUMO

The insecticidal action of the new compound ethofenprox (trebon) in the form of 10 and 30% flow concentrate against house flies, cockroaches, bed bugs, rat fleas, mosquito imagines and larvae of Aedes and Culex genera was studied in the laboratory and in the field. The effective doses and the methods of spraying were established. 10% flow trebon is allowed by the USSR Ministry of Health to be useful in medical disinfection.

RESUMO

The comparative analysis of the low-molecular-weight heparin Clexane influence and the unfractionated heparin on the parameters of blood coagulation homeostasis in 70 patients with complicated forms of acute cholecystitis. The tendency for hypercoagulation is observed. Prophylactic use of Clexane in patients with complicated forms of acute cholecystitis, in comparison with unfractionated heparin, is accompanied by the decrease of coagulating potential of blood.

Blood-sucking mosquito fauna, season and circadian activity of predominant species, dynamics of hatching place formation in the villages and on the territories surrounding the Golodnaya and Dzhizakskaya steppes have been studied for many years and the materials have been presented. The data obtained serve as the basis for planning, prophylactic and eliminating measures.

Two residual insecticides (Icon 10% WP, ICI and Ficam W 80% WP, CAMCO) were in 1989 tested in field trials in Tajikistan, Azerbaijan and Uzbekistan against Anopheles superpictus, An. sacharovi, An. pulcherrimus, An. martinius, Culex pipiens, Aedes caspius, and Ae. vexans. In dosages recommended by the producing companies (0.025 g/m2 a. i. for Icon and 0.4 g/m2 for Ficam), the above preparations showed residual action during 2-3 months. Icon and Ficam did not cause irritability and repellency in mosquito, which did not migrate to unsprayed rooms of cattle sheds. In areas where the experimental trials were large-scale, a stable decrease in mosquito density was achieved. An insecticide action of ICON was also noted on house flies, cockroaches, bed bugs, fleas and spiders.

RESUMO

A high insecticidal activity of K-otrin (flow), a preparation containing 2.5% of deltametrin, towards various arthropods, was revealed in laboratory experiments and in field trials. The effective doses were determined: 0.01-0.1 g/m2. Residual effects of the insecticide on the treated surfaces persist for 3 months and longer. Toxicologic studies revealed the possibility of using K-otrin for medical disinfection. The preparation is permitted for use by the Ministry of Health.
