RESUMO

STUDY HYPOTHESIS: Cellular aging of the egg following ovulation, also known as post-ovulatory aging, is associated with aberrant cortical mechanics and actomyosin cytoskeleton functions. STUDY FINDING: Post-ovulatory aging is associated with dysfunction of non-muscle myosin-II, and pharmacologically induced myosin-II dysfunction produces some of the same deficiencies observed in aged eggs. WHAT IS KNOWN ALREADY: Reproductive success is reduced with delayed fertilization and when copulation or insemination occurs at increased times after ovulation. Post-ovulatory aged eggs have several abnormalities in the plasma membrane and cortex, including reduced egg membrane receptivity to sperm, aberrant sperm-induced cortical remodeling and formation of fertilization cones at the site of sperm entry, and reduced ability to establish a membrane block to prevent polyspermic fertilization. STUDY DESIGN, SAMPLES/MATERIALS, METHODS: Ovulated mouse eggs were collected at 21-22 h post-human chorionic gonadotrophin (hCG) (aged eggs) or at 13-14 h post-hCG (young eggs), or young eggs were treated with the myosin light chain kinase (MLCK) inhibitor ML-7, to test the hypothesis that disruption of myosin-II function could mimic some of the effects of post-ovulatory aging. Eggs were subjected to various analyses. Cytoskeletal proteins in eggs and parthenogenesis were assessed using fluorescence microscopy, with further analysis of cytoskeletal proteins in immunoblotting experiments. Cortical tension was measured through micropipette aspiration assays. Egg membrane receptivity to sperm was assessed in in vitro fertilization (IVF) assays. Membrane topography was examined by low-vacuum scanning electron microscopy (SEM). MAIN RESULTS AND THE ROLE OF CHANCE: Aged eggs have decreased levels and abnormal localizations of phosphorylated myosin-II regulatory light chain (pMRLC; P = 0.0062). Cortical tension, which is mediated in part by myosin-II, is reduced in aged mouse eggs when compared with young eggs, by ∼40% in the cortical region where the metaphase II spindle is sequestered and by ∼50% in the domain to which sperm bind and fuse (P < 0.0001). Aging-associated parthenogenesis is partly rescued by treating eggs with a zinc ionophore (P = 0.003), as is parthenogenesis induced by inhibition of mitogen-activated kinase (MAPK) 3/1 [also known as extracellular signal-regulated kinase (ERK)1/2] or MLCK. Inhibition of MLCK with ML-7 also results in effects that mimic those of post-ovulatory aging: fertilized ML-7-treated eggs show both impaired fertilization and increased extents of polyspermy, and ML-7-treated young eggs have several membrane abnormalities that are shared by post-ovulatory aged eggs. LIMITATIONS, REASONS FOR CAUTION: These studies were done with mouse oocytes, and it remains to be fully determined how these findings from mouse oocytes would compare with other species. For studies using methods not amenable to analysis of large sample sizes and data are limited to what images one can capture (e.g. SEM), data should be interpreted conservatively. WIDER IMPLICATIONS OF THE FINDINGS: These data provide insights into causes of reproductive failures at later post-copulatory times. LARGE SCALE DATA: Not applicable. Vertebrate eggs are arrested at metaphase of meiosis II, a state classically known as cytostatic factor arrest. Maintenance of this arrest until the time of fertilization and then fertilization-induced exit from metaphase II are crucial for reproductive success. Another key aspect of this meiotic arrest and exit is regulation of the metaphase II spindle, which must be appropriately localized adjacent to the egg cortex during metaphase II and then progress into successful asymmetric cytokinesis to produce the second polar body. This study examined the mitogen-activated protein kinases MAPK3 and MAPK1 (also known as ERK1/2) as regulators of these two related aspects of mammalian egg biology, specifically testing whether this MAPK pathway affected myosin-II function and whether myosin-II perturbation would produce some of the same effects as MAPK pathway perturbation. Inhibition of the MEK1/2-MAPK pathway with U0126 leads to reduced levels of phosphorylated myosin-regulatory light chain (pMRLC) and causes a reduction in cortical tension, effects that are mimicked by treatment with the myosin light chain kinase (MLCK) inhibitor ML-7. These data indicate that one mechanism by which the MAPK pathway acts in eggs is by affecting myosin-II function. We further show that MAPK or MLCK inhibition induces loss of normal cortical spindle localization or parthenogenetic egg activation. This parthenogenesis is dependent on cytosolic and extracellular calcium and can be rescued by hyperloading eggs with zinc, suggesting that these effects of inhibition of MLCK or the MAPK pathway are linked with dysregulation of ion homeostasis.

Changes occurring as the prophase I oocyte matures to metaphase II are critical for the acquisition of competence for normal egg activation and early embryogenesis. A prophase I oocyte cannot respond to a fertilizing sperm as a metaphase II egg does, including the ability to prevent polyspermic fertilization. Studies here demonstrate that the competence for the membrane block to polyspermy is deficient in prophase I mouse oocytes. In vitro fertilization experiments using identical insemination conditions result in monospermy in 87% of zona pellucida (ZP)-free metaphase II eggs, while 92% of ZP-free prophase I oocytes have four or more fused sperm. The membrane block is associated with a postfertilization reduction in the capacity to support sperm binding, but this reduction in sperm-binding capacity is both less robust and slower to develop in fertilized prophase I oocytes. Fertilization of oocytes is dependent on the tetraspanin CD9, but little to no release of CD9 from the oocyte membrane is detected, suggesting that release of CD9-containing vesicles is not essential for fertilization. The deficiency in membrane block establishment in prophase I oocytes correlates with abnormalities in two postfertilization cytoskeletal changes: sperm-induced cortical remodeling that results in fertilization cone formation and a postfertilization increase in effective cortical tension. These data indicate that cortical maturation is a component of cytoplasmic maturation during the oocyte-to-egg transition and that the egg cortex has to be appropriately primed and tuned to be responsive to a fertilizing sperm.

This study aimed to determine the minimum anesthetic concentration (MAC) and dose-related cardiovascular effects of isoflurane during controlled ventilation in cinereous vultures (Aegypius monachus). The MAC was determined for 10 cinereous vultures as the midpoint between the end-tidal isoflurane concentration that allows gross purposeful movement and that which prevents the movement in response to clamping a pedal digit. Immediately after the MAC was determined, the cardiovascular effects of isoflurane at 1.0, 1.5, and 2.0 times the MAC were investigated in seven of the 10 birds. The MAC of isoflurane for 10 cinereous vultures during controlled ventilation was 1.06 +/- 0.07% (mean +/- SD). When the isoflurane concentration was increased to 1.5 and 2.0 times the MAC, there was significant dose-dependent decrease in the arterial blood pressure. However, the heart rate did not change over a range of 1.0 to 2.0 times the MAC.

The toxicity of solutions containing nonionic surfactants Tween 80, Brij 35 and/or phenanthrene to Pseudomonas putida ATCC 17484 was investigated. The fraction of direct contact between micellar-phase phenanthrene and bacterial cell surface was estimated by using the toxicity data and a mathematical model. The mathematical model was used to calculate phenanthrene concentration in the micellar phase and aqueous pseudophase separately. The first-order death rate constant increased from 0.088+/-0.016 to 0.25+/-0.067 h(-1) when the phenanthrene concentration was increased from 0 to 5.17 x 10(-6)M (equals water solubility). The intrinsic toxicity of surfactant was higher in Brij 35 than in Tween 80. When phenanthrene concentration was increased to 9.7 x 10(-5)M in surfactant solutions, the death rate constant increased to 1.8 +/- 0.024 and 0.41 +/- 0.088 h(-1) for 8.4 x 10(-4)M Brij 35 and 7.6 x 10(-4)M Tween 80. The direct-contact fraction was 0.083 and 0.044 for Brij 35 and Tween 80, respectively, under these conditions using exponential model. The toxicity increased with increasing phenanthrene concentration at a fixed surfactant concentration. The toxicity decreased with increasing the surfactant concentration at a fixed phenanthrene concentration due to decreased contact of bacteria with phenanthrene present in the interior of surfactant micelles.

This study investigated the effects of ovariohysterectomy on reactivity of German Shepherd dogs. Fourteen healthy dogs ranging in age from 5 to 10 months were assigned to an ovariohysterectomy or a sexually intact group. Their behaviours were digitally video recorded 4-5 months after treatment and analysed for treatment effects on reactivity. Responses to the approach of an unfamiliar human leading an unknown dog were assigned the following reactivity scores: severe reactivity, 3; moderate reactivity, 2; defensive or mild reactivity, 1; attentive or no reactivity, 0. Median reactivity scores in response to the approach of an unfamiliar human walking with an unknown dog were calculated for each observation period. Dogs in the ovariohysterectomized group showed more reactivity, and median reactivity scores were higher in the ovariohysterectomy group compared with those of the sexually intact group. Ovariohysterectomy of 5-10 month old German Shepherd bitches specifically, and perhaps bitches of any breed generally, may induce an increase in reactivity. Practitioners may benefit from recognizing that a range of behavioural changes may occur post-ovariohysterectomy.

This study investigated the use of sprayed intraperitoneal bupivacaine to relieve postoperative pain behavior and biochemical stress response after laparoscopic ovariohysterectomy (LOVH) in dogs. Sixteen sexually intact female dogs were randomly assigned to two groups. The sprayed intraperitoneal bupivacaine (SIB) group received 4.4 mg/kg of sprayed intraperitoneal bupivacaine diluted to 0.25% with an equivalent volume of saline after pneumoperitoneum. The control group received 1.76 mL/kg of saline in a similar fashion. Both groups received preoperative periportal 5% bupivacaine (1 mL) before incision. Postoperative pain was measured using the short form of the Glasgow composite measures pain scale (CMPS-SF, 0-24). Serum cortisol and glucose concentrations were measured preoperatively and 0.5, 1, 2, 4, 6, 12, and 24h postoperatively. The SIB group had significantly lower CMPS-SF compared to the control group 1, 2, 4, 6, and 12h after the operation. Cortisol concentrations were significantly increased from preoperative concentrations in the control group at 0.5, 1, 2, and 4h post operation and at 0.5 and 1h post operation in the SIB group. No significant differences were seen in serum glucose within each group. This report suggests that the use of sprayed intraperitoneal bupivacaine can be used as part of a multimodal approach for pain management after LOVH in dogs.

To investigate how socialization can affect the types and characteristics of vocalization produced by cats, feral cats (n=25) and house cats (n=13) were used as subjects, allowing a comparison between cats socialized to people and non-socialized cats. To record vocalization and assess the cats' responses to behavioural stimuli, five test situations were used: approach by a familiar caretaker, by a threatening stranger, by a large doll, by a stranger with a dog and by a stranger with a cat. Feral cats showed extremely aggressive and defensive behaviour in most test situations, and produced higher call rates than those of house cats in the test situations, which could be attributed to less socialization to other animals and to more sensitivity to fearful situations. Differences were observed in the acoustic parameters of feral cats in comparison to those of house cats. The feral cat produced significantly higher frequency in fundamental frequency, peak frequency, 1st quartile frequency, 3rd quartile frequency of growls and hisses in agonistic test situations. In contrast to the growls and hisses, in meow, all acoustic parameters like fundamental frequency, first formant, peak frequency, 1st quartile frequency, and 3rd quartile frequency of house cats were of significantly higher frequency than those of feral cats. Also, house cats produced calls of significantly shorter in duration than feral cats in agonistic test situations. These results support the conclusion that a lack of socialization may affect usage of types of vocalizations, and the vocal characteristics, so that the proper socialization of cat may be essential to be a suitable companion house cat.

Cell division is inherently mechanical, with cell mechanics being a critical determinant governing the cell shape changes that accompany progression through the cell cycle. The mechanical properties of symmetrically dividing mitotic cells have been well characterized, whereas the contribution of cellular mechanics to the strikingly asymmetric divisions of female meiosis is very poorly understood. Progression of the mammalian oocyte through meiosis involves remodeling of the cortex and proper orientation of the meiotic spindle, and thus we hypothesized that cortical tension and stiffness would change through meiotic maturation and fertilization to facilitate and/or direct cellular remodeling. This work shows that tension in mouse oocytes drops about sixfold during meiotic maturation from prophase I to metaphase II and then increases ∼1.6-fold upon fertilization. The metaphase II egg is polarized, with tension differing ∼2.5-fold between the cortex over the meiotic spindle and the opposite cortex, suggesting that meiotic maturation is accompanied by assembly of a cortical domain with stiffer mechanics as part of the process to achieve asymmetric cytokinesis. We further demonstrate that actin, myosin-II, and the ERM (Ezrin/Radixin/Moesin) family of proteins are enriched in complementary cortical domains and mediate cellular mechanics in mammalian eggs. Manipulation of actin, myosin-II, and ERM function alters tension levels and also is associated with dramatic spindle abnormalities with completion of meiosis II after fertilization. Thus, myosin-II and ERM proteins modulate mechanical properties in oocytes, contributing to cell polarity and to completion of meiosis.

This study aimed to compare the behavioural reactivity of Jindo dogs with two different coat colours. Fawn (16 males, 15 females; mean age+/-S.D.=7.2+/-2.1 years) and white (10 males, 10 females; mean age+/-S.D.=6.9+/-2.1 years) Jindo dogs were exposed to a set of behavioural tests. All of the dogs were videotaped during the testing period to allow further analysis. The intensity of social, aggressive, fearful, and submissive reactivity and the frequency of urination as a scent-marking behaviour were scored on a scale running from 0 to 4 points. For each dog, each variable was defined as the average of the scores of nine behaviour tests. Then, the behavioural reactivities of Jindo dogs of each coat colour were compared. The results suggested that Jindo dogs of fawn coat colour exhibited a significantly lower intensity of fearful and submissive reactivity than those of white coat colour. In addition, fawn Jindo dogs produced scent-marking behaviour significantly more frequently. The results of the present study may provide useful information for scientific researchers, potential owners and breeders of Jindo dogs.
