PURPOSE: Enterobacteriaceae carrying mcr-9, in particularly those also co-containing metallo-ß-lactamase (MBL) and TEM type ß-lactamase, present potential transmission risks and lack adequate clinical response methods, thereby posing a major threat to global public health. The aim of this study was to assess the antimicrobial efficacy of a combined ceftazidime/avibactam (CZA) and aztreonam (ATM) regimen against carbapenem-resistant Enterobacter cloacae complex (CRECC) co-producing mcr-9, MBL and TEM. METHODS: The in vitro antibacterial activity of CZA plus ATM was evaluated using a time-kill curve assay. Furthermore, the in vivo interaction between CZA plus ATM was confirmed using a Galleria mellonella (G. mellonella) infection model. RESULTS: All eight clinical strains of CRECC, co-carrying mcr-9, MBL and TEM, exhibited high resistance to CZA and ATM. In vitro time-kill curve analysis demonstrated that the combination therapy of CZA + ATM exerted significant bactericidal activity against mcr-9, MBL and TEM-co-producing Enterobacter cloacae complex (ECC) isolates with a 100% synergy rate observed in our study. Furthermore, in vivo survival assay using Galleria mellonella larvae infected with CRECC strains co-harboring mcr-9, MBL and TEM revealed that the CZA + ATM combination significantly improved the survival rate compared to the drug-treatment alone and untreated control groups. CONCLUSION: To our knowledge, this study represents the first report on the in vitro and in vivo antibacterial activity of CZA plus ATM against CRECC isolates co-harboring mcr-9, MBL and TEM. Our findings suggest that the combination regimen of CZA + ATM provides a valuable reference for clinicians to address the increasingly complex antibiotic resistance situation observed in clinical microorganisms.

Airborne fine particulate matter (PM2.5) can cause pulmonary inflammation and even fibrosis, however, the underlying molecular mechanisms of the pathogenesis of PM2.5 exposure have not been fully appreciated. In the present study, we explored the dynamics of glycolysis and modification of histone lactylation in macrophages induced by PM2.5-exposure in both in vivo and in vitro models. Male C57BL/6 J mice were anesthetized and administrated with PM2.5 by intratracheal instillation once every other day for 4 weeks. Mouse RAW264.7 macrophages and alveolar epithelial MLE-12 cells were treated with PM2.5 for 24 h. We found that PM2.5 significantly increased lactate dehydrogenase (LDH) activities and lactate contents, and up-regulated the mRNA expression of key glycolytic enzymes in the lungs and bronchoalveolar lavage fluids of mice. Moreover, PM2.5 increased the levels of histone lactylation in both PM2.5-exposed lungs and RAW264.7 cells. The pro-fibrotic cytokines secreted from PM2.5-treated RAW264.7 cells triggered epithelial-mesenchymal transition (EMT) in MLE-12 cells through activating transforming growth factor-ß (TGF-ß)/Smad2/3 and VEGFA/ERK pathways. In contrast, LDHA inhibitor (GNE-140) pretreatment effectively alleviated PM2.5-induced pulmonary inflammation and fibrosis via inhibiting glycolysis and subsequent modification of histone lactylation in mice. Thus, our findings suggest that PM2.5-induced glycolysis and subsequent modification of histone lactylation play critical role in the PM2.5-associated pulmonary fibrosis.

Owing to the extremely complex compositions and origins of waste-activated sludge (WAS), the multiple physiochemical properties of WAS have impacts on its dewaterability, and there is a complex interaction relationship among the multiple physiochemical properties, which makes it difficult to identify the controlling factors on WAS dewaterability. Accordingly, there is still no unified certainty in the appropriate ranges of physiochemical properties for the optimal dewaterability of sludge from different sources, resulting in a lack of clear theoretical basis for technical selection and optimization of sludge dewatering processes. The large consumption of conditioning chemicals and low process efficiency stand for the major deficiency of existing sludge conditioning technologies. This study proposed to use a non-linear, adaptive and self-organizing artificial neural network (ANN) model to integrate the multiple physiochemical properties of WAS affecting its dewaterability, and WAS dewatering performance under certain conditioning schemes could be predicated by ANN model with the multiple physiochemical properties and conditioning operation parameters as the input arguments. Thus, the laborious filtration experiments for screening conditioning chemicals could be replaced by the input adjustment of ANN model. Rooted mean squared error (RMSE) of 6.51 and coefficient of determination (R2) of 0.73 confirmed the satisfied stability and accuracy of established ANN model. Furthermore, the predictor-exclusive method revealed that the exclusion of polar interface free energy decreased most, which reflected the importance of surface hydrophilicity reduction in sludge dewaterability improvement. All the contributions presented here were believed to provide an intelligent insight to improve the experience operation status of WAS dewatering process.

In this paper, a new algorithm for extracting the laser fringe center is proposed. Based on a deep learning skeleton extraction network, the laser stripe center can be extracted quickly and accurately. Skeleton extraction is the process of reducing the shape image to its approximate central axis representation while maintaining the image's topological and geometric shape. Skeleton extraction is an important step in topological and geometric shape analysis. According to the characteristics of the wheelset laser curve dataset, a new skeleton extraction network, a hierarchical skeleton network (LuoNet), is proposed. The proposed architecture has three levels of the encoder-decoder network, and YE Module interconnection is designed between each level of the encoder and decoder network. In the wheelset laser curve dataset, the F1_score can reach 0.714. Compared with the traditional laser curve center extraction algorithm, the proposed LuoNet algorithm has the advantages of short running time, high accuracy, and stable extraction results.

An algorithm of laser curve segmentation for a train wheelset based on an encoder- decoder network is proposed. Aiming at the rich local features and simple semantic features of the train wheelset laser curve image, a neural network with shallow depth, high resolution, and good detail performance was designed. The proposed neural network makes full use of the dense connection mechanism and the upsampling module to enhance feature reuse and feature propagation. It can extract context semantic information at multiple scales with fewer parameters. Experimental results show that the encoder-decoder network has better performance than other neural networks in laser curve extraction of train wheelset. Based on the encoder-decoder neural network, mIOU, Recall, Accuracy, and F1_score of the laser curve dataset of the train wheelset, the score index reached 86.5%, 89.2%, 99.9%, and 85.0%, which can accurately extract the laser stripe of the train wheelset. Additionally, the encoder-decoder network can diminish the influence of noise on the extraction of laser fringes of a train wheelset to a certain extent. Therefore, it has good application in railway safety.

Regarding the ice periods of the Yellow River, it is difficult to obtain ice data information. To effectively grasp the ice evolution process in the ice periods of the typical reach of the Yellow River, a fixed-point air-coupled radar remote monitoring device is proposed in this paper. The device is mainly composed of an air-coupled radar ice thickness measurement sensor, radar water level measurement sensor, temperature measurement sensor, high-definition infrared night vision instrument, remote switch control, telemetry communication machine, solar and wind power supply, lightning protection, and slewing arm steel tower. The integrated monitoring device can monitor ice thickness, water level, air temperature, ice surface temperature, and other related parameters in real time. At present, devices have obtained the ice change process of fixed points in ice periods from 2020 to 2021. Through a comparison with manual data, the mean error of the monitoring results of the water level and ice thickness was approximately 1 cm. The device realizes the real-time monitoring of ice thickness and water level change in the whole cycle at the fixed position. Through video monitoring, it can take pictures and videos regularly and realize the connection between the visual river and monitoring data. The research results provide a new model and new technology for hydrological monitoring in the ice periods of the Yellow River, which has broad application prospects.

Silicon (Si), germanium (Ge), and gallium arsenide (GaAs) are familiar semiconductors that always act in the role of optical dielectrics. However, these semiconductors also have plasmonic behaviors in ultraviolet (UV) ranges due to the strong interband transitions or valence electrons. And few studies are aimed at investigating plasmonic properties in the semiconductor at the nanoscale. In this work, we discuss UV plasmonics and sensing properties in single and dimer Si, Ge, and GaAs triangular prism meta-antenna systems. The results show that obvious local surface plasmon resonances (LSPRs) can be realized in the proposed triangular prism meta-antennas, and the resonant wavelength, electromagnetic field distribution, surface charge distribution, and surface current density can be effectively tuned by structural and material parameters. In addition, we also find that the Si triangular prism meta-antenna shows more intense plasmonic responses in UV ranges than that in the Ge or GaAs triangular prism nanostructures. Especially, the phase difference between the triangular prism nanostructure and light source can effectively regulate the symbol and value of the surface charge. Moreover, the great enhancement of electric field can be seen in the dimer triangular prism meta-antennas when the distance of the gap is g<5 nm, especially g=1 nm. The most interesting result is that the maximum of refractive index sensitivity s and figure of merit (FoM) are greatly enlarged in dimer triangular prism meta-antennas. Particularly, the sensitivity can reach up to 215 nm/RIU in the dimer GaAs triangular prism meta-antennas, which is improved more than one order of magnitude. These research results may play important roles in applications of the photo detecting, plasmonic sensing and disinfecting in UV ranges.

Apple latent spherical virus (ALSV) is a latent virus with wide host range of plant species. In the present study, we prepared ALSV vectors expressing RNA silencing suppressors (RSSs) from eight plant viruses: P19 of carnation Italian ring spot virus (tombusvirus), 2b of peanut stunt virus (cucumovirus), NSs of tomato spotted wilt virus (tospovirus), HC-Pro of bean yellow mosaic virus (potyvirus), γb of barley stripe mosaic virus (hordeivirus), P15 of peanut clump virus (pecluvirus), P1 of rice yellow mottle virus (sobemovirus), or P21 of beet yellows virus (closterovirus). These vectors were inoculated to Nicotiana benthamiana to investigate the effects of RSSs on the virulence and accumulation of ALSV. Among the vectors, ALSV expressing NSs (ALSV-NSs) developed severe mosaic symptoms in newly developed leaves followed by plant death. Infection of ALSV-γb induced characteristic concentric ringspot symptoms on leaves, and plants infected with ALSV-HC-Pro showed mosaic and dwarf symptoms. Infection of the other five ALSV vectors did not show symptoms. ELISA and immunoblot assay indicated that virus titer increased in leaves infected with ALSV-NSs, γb, HC-Pro, or P19. RT-qPCR indicated that the amount of ALSV in plants infected with ALSV-NSs was increased by approximately 45 times compared with that of wtALSV without expression of any RSS. When ALSV-P19, NSs, or HC-Pro was inoculated to Cucumis sativus plants, none of these ALSV vectors induced symptoms, but accumulation of ALSV in plants infected with ALSV-NSs was increased, suggesting that functions of RSSs on virulence and accumulation of ALSV depend on host species.

BACKGROUND: Carbapenem-resistant hypervirulent Klebsiella pneumoniae strains have recently come into existence worldwide; however, researchers in northeast China are not aware of their clinical features and molecular characteristics. METHODS: Here, the molecular and virulent characteristics of 44 carbapenem-resistant K. pneumoniae (CRKP) isolates collected from January 2015 to December 2017 were studied. Multilocus sequence typing (MLST) and pulsed-field gel electrophoresis (PFGE) were carried out to define the clonal relatedness among the isolates. PCR and capsular serotyping of the virulence-associated genes, as well as biofilm formation and serum complement-mediated killing assays, were employed to determine the virulent potential. The genomic features and associated mobile genetic elements of JmsCRE57 were detected by whole genome sequencing. RESULTS: The only positive isolate was JmsCRE57, which belonged to the ST375 serotype K2 that expressed uge, mrkD, fimH, kpn, aerobactin and rmpA virulence-associated genes and showed strong biofilm formation and serum sensitivity. Sequencing results showed that the JmsCRE57 genome mainly consisted of a circular chromosome, three antimicrobial resistant plasmids and a virulent plasmid. The antimicrobial resistant plasmid expressing blaKPC-2, blaCTX-M-15, aph(3″)-Ib, aph(6)-Id, qnrB1, aac(3)-IIa, aac(6')-Ib-cr, blaOXA-1, blaTEM-1B, catB4, sul2, dfrA14 and blaSHV-99. The virulent plasmid belonged to the IncHI1B group, which is mainly composed of mucoid phenotype genes and siderophore-associated genes. The remaining CRKP strains that expressed uge, fimH, mrkD and kpn virulence-associated genes were not successfully typed. CONCLUSION: Our results provide new insights on the epidemiology of carbapenem-resistant K2 hypervirulent K. pneumoniae ST375 and CRKP ST76 strains in northeast China, which may help control their future outbreaks.

BACKGROUND: Enterobacter cloacae complex (ECC) is one of the most common extended-spectrum ß-lactamase and carbapenemase-producing pathogen that threatens millions of the elderly and vulnerable sick persons. The objective of this study was to perform the molecular characteristics of the carbapenem-resistant E. cloacae complex (CREC) emerged in Heilongjiang Province of China. METHODS: Six CREC strains were isolated from the patients with infectious diseases. The identities of ECC isolates were confirmed by sequencing the polymerase chain reaction (PCR) products of 16S rRNA gene. The characterization of the CREC isolates were analyzed by sequencing PCR products of the carbapenemase, ampC and fluoroquinolone resistance genes and performing multilocus sequence typing (MLST), pulsed-field gel electrophoresis (PFGE) and whole genome sequencing. RESULTS: All 6 isolates harbored multiple resistance genes. Of them, 5 carried metallo-ß-lactamases and one was blaKPC-2-positive. The levofloxacin and ciprofloxacin-resistant strains had substitutions of gyrA83, gyrA87, and parC80 in the quinolone-resistance determining regions. The MLST analyses revealed that 6 isolates belonged to five sequence types (ST520, ST528, ST1119, ST1120, and ST93) while the PFGE patterns of the isolates fallen into four clusters. The strain ST1120 was found to carry two separated plasmids that encode blaNDM-1 and blaIMP-4. CONCLUSIONS: Our study, for the first time, identified a CREC strain that co-produces blaNDM-1 and blaIMP-4 in the Northeast China. Our finding emphasizes an urgent need for more intensive surveillance and precaution measures to prevent the CERC spread.

The meta-analysis was conducted to evaluate the correlations between common genetic polymorphisms in the IFN-γ gene and susceptibility to breast cancer. The following electronic databases were searched without language restrictions: MEDLINE (1966 ~ 2013), the Cochrane Library Database (issue 12, 2013), EMBASE (1980 ~ 2013), CINAHL (1982 ~ 2013), Web of Science (1945 ~ 2013), and the Chinese Biomedical Database (CBM) (1982 ~ 2013). Meta-analysis was performed with the use of the STATA statistical software. Odds ratios (OR) with their 95 % confidence intervals (95 % CIs) were calculated. Nine clinical case-control studies met all the inclusion criteria and were included in this meta-analysis. A total of 1,182 breast cancer patients and 1,525 healthy controls were involved in this meta-analysis. Three functional polymorphisms were assessed, including rs2069705 C>T, rs2430561 T>A, and CA repeats 2/X. Our meta-analysis results indicated that IFN-γ genetic polymorphisms might be significantly associated with an increased risk of breast cancer (allele model: OR = 1.37, 95 % CI = 1.03 ~ 1.83, P = 0.031; dominant model: OR = 1.55, 95 % CI = 1.01 ~ 2.37, P = 0.046; homozygous model: OR = 2.23, 95 % CI = 1.30 ~ 3.82, P = 0.004; respectively), especially the rs2430561 T>A polymorphism. Subgroup analysis based on ethnicity suggested that genetic polymorphisms in the IFN-γ gene were closely correlated with increased breast cancer risk among Asians (allele model: OR = 1.21, 95 % CI = 1.02 ~ 1.58, P = 0.017; dominant model: OR = 3.44, 95 % CI = 2.07 ~ 5.71, P < 0.001; recessive model: OR = 1.58, 95 % CI = 1.06 ~ 2.37, P = 0.025; homozygous model: OR = 1.83, 95 % CI = 1.19 ~ 2.80, P = 0.006; respectively), but not among Caucasians (all P > 0.05). Our meta-analysis supported the hypothesis that IFN-γ genetic polymorphisms may contribute to an increased risk of breast cancer, especially the rs2430561 T>A polymorphism among Asians.

Citrus fruits are extensively cultivated fruits with high nutritional value. The identification of distinct ripeness stages in citrus fruits plays a crucial role in guiding the planning of harvesting paths for citrus-picking robots and facilitating yield estimations in orchards. However, challenges arise in the identification of citrus fruit ripeness due to the similarity in color between green unripe citrus fruits and tree leaves, leading to an omission in identification. Additionally, the resemblance between partially ripe, orange-green interspersed fruits and fully ripe fruits poses a risk of misidentification, further complicating the identification of citrus fruit ripeness. This study proposed the YOLO-CIT (You Only Look Once-Citrus) model and integrated an innovative R-LBP (Roughness-Local Binary Pattern) method to accurately identify citrus fruits at distinct ripeness stages. The R-LBP algorithm, an extension of the LBP algorithm, enhances the texture features of citrus fruits at distinct ripeness stages by calculating the coefficient of variation in grayscale values of pixels within a certain range in different directions around the target pixel. The C3 model embedded by the CBAM (Convolutional Block Attention Module) replaced the original backbone network of the YOLOv5s model to form the backbone of the YOLO-CIT model. Instead of traditional convolution, Ghostconv is utilized by the neck network of the YOLO-CIT model. The fruit segment of citrus in the original citrus images processed by the R-LBP algorithm is combined with the background segment of the citrus images after grayscale processing to construct synthetic images, which are subsequently added to the training dataset. The experiment showed that the R-LBP algorithm is capable of amplifying the texture features among citrus fruits at distinct ripeness stages. The YOLO-CIT model combined with the R-LBP algorithm has a Precision of 88.13%, a Recall of 93.16%, an F1 score of 90.89, a mAP@0.5 of 85.88%, and 6.1ms of average detection speed for citrus fruit ripeness identification in complex environments. The model demonstrates the capability to accurately and swiftly identify citrus fruits at distinct ripeness stages in real-world environments, effectively guiding the determination of picking targets and path planning for harvesting robots.

Carbapenem-resistant Enterobacterales (CRE) are some of the most important pathogens causing infections, which can be challenging to treat. We identified four blaIMP-carrying CRE isolates and collected clinical data. The transferability and stability of the plasmid were verified by conjugation, successive passaging, and plasmid elimination assays. The IncC blaIMP-4-carrying pIMP4-ECL42 plasmid was successfully transferred into the recipient strain, and the high expression of traD may have facilitated the conjugation transfer of the plasmid. Interestingly, the plasmid showed strong stability in clinical isolates. Whole-genome sequencing was performed on all isolates. We assessed the sequence similarity of blaIMP -harboring plasmid from our institution and compared it to plasmids for which sequence data are publicly available. We found that four blaIMP-carrying CRE belonged to four different sequence types. The checkerboard technique and time-kill assays were used to investigate the best antimicrobial therapies for blaIMP-carrying CRE. The time-kill assay showed that the imipenem of 1× minimum inhibitory concentration (MIC) alone had the bactericidal or bacteriostatic effect against IMP-producing strains at 4-12 h in vitro. Moreover, the combination of tigecycline (0.5/1/2 × MIC) and imipenem (0.5/1 × MIC) showed a bactericidal effect against the blaIMP-26-carrying CRECL60 strain.IMPORTANCECarbapenem-resistant Enterobacterales (CRE) are an urgent public health threat, and infections caused by these microorganisms are often associated with high mortality and limited treatment options. This study aimed to determine the clinical features, molecular characteristics, and plasmid transmissible mechanisms of blaIMP carriage as well as to provide a potential treatment option. Here, we demonstrated that conjugated transfer of the IncC blaIMP-4-carrying plasmid promotes plasmid stability, so inhibition of conjugated transfer and enhanced plasmid loss may be potential ways to suppress the persistence of this plasmid. The imipenem alone or tigecycline-imipenem combination showed a good bactericidal effect against IMP-producing strains. In particular, our study revealed that imipenem alone or tigecycline-imipenem combination may be a potential therapeutic option for patients who are infected with IMP-producing strains. Our study supports further trials of appropriate antibiotics to determine optimal treatment and emphasizes the importance of continued monitoring of IMP-producing strains in the future.

The COVID-19 pandemic challenged emergency management in cities worldwide. Many municipalities adopted restrictive, one-size-fits-all spatial regulations such as lockdowns without fully considering the inhabitants' daily activities and local economies. The existing epidemic regulations' unintended detrimental effects on socioeconomic sustainability necessitate a transition from the "lockdown" approach to more precise disease prevention. A spatially and temporally precise approach that balances epidemic prevention with the demands of daily activities and local economies is needed. Thus, the aim of this study was to propose a framework and key procedures for determining precise prevention regulations from the perspectives of the 15-minute city concept and spatiotemporal planning. Alternative regulations of lockdowns were determined by delineating 15-minute neighborhoods, identifying and reconfiguring facility supplies and activity demands in both normal and epidemic conditions, and performing cost-benefit analyses. Highly adaptable, spatially- and temporally-precise regulations can match the needs of different types of facilities. We demonstrated the process for determining precise prevention regulations in the case of the Jiulong 15-minute neighborhood in Beijing. Precise prevention regulations-which meet essential activity demands and are adaptable for different facility types, times, and neighborhoods-have implications for long-term urban planning and emergency management.

Short-chain fatty acids are important nutrients that regulate milk fat synthesis. They regulate milk synthesis via the sterol regulatory element binding protein 1 (SREBP1) pathway; however, the details are still unknown. Here, the regulation and mechanism of sodium acetate (SA) in milk fat synthesis in bovine mammary epithelial cells (BMECs) were assessed. BMECs were treated with SA supplementation (SA+) or without SA supplementation (SA-), and milk fat synthesis and activation of the SREBP1 pathway were increased (P = 0.0045; P = 0.0042) by SA+ and decreased (P = 0.0068; P = 0.0031) by SA-, respectively. Overexpression or inhibition of SREBP1 demonstrated that SA promoted milk fat synthesis (P = 0.0045) via the SREBP1 pathway. Overexpression or inhibition of TATA element modulatory factor 1 (TMF1) demonstrated that TMF1 suppressed activation of the SREBP1 pathway (P = 0.0001) and milk fat synthesis (P = 0.0022) activated by SA+. Overexpression or inhibition of TMF1 and SREBP1 showed that TMF1 suppressed milk fat synthesis (P = 0.0073) through the SREBP1 pathway. Coimmunoprecipitation analysis revealed that TMF1 interacted with SREBP1 in the cytoplasm and suppressed the nuclear localization of SREBP1 (P = 0.0066). The absence or presence of SA demonstrated that SA inhibited the expression of TMF1 (P = 0.0002) and the interaction between TMF1 and SREBP1 (P = 0.0001). Collectively, our research suggested that TMF1 was a new negative regulator of milk fat synthesis. In BMECs, SA promoted the SREBP1 pathway and milk fat synthesis by suppressing TMF1. This study enhances the current understanding of the regulation of milk fat synthesis and provides new scientific data for the regulation of milk fat synthesis.

Space-time prism is a fundamental concept in time geography that can model an individual's accessibility to resources under space-time constraints. A prism anchor is often defined by work, school, or home activity with a fixed location and schedule. Trips and other activities are relatively flexible and scheduled between prism anchors. This fixity-flexibility dichotomy may not capture the increasing complexity of human mobility behaviors or variations among individuals. Recent developments in location-aware technologies allow us to collect person-level mobility data with detailed space-time paths and contextual information. This article develops methods to extract prism anchors from these GPS-based survey data and examines whether home, work, and school activities can always be used to define prism anchors for everyone. To illustrate our methods, we use data collected in Minnesota and Beijing as two study cases. Results in both study cases suggest that not everyone has home, work, or school anchors, and people with the same socio-demographic background tend to have similar anchor types. By deriving home, work, and school anchors, we can better understand how a person's everyday schedules are governed by home, work, and school and refine person-based accessibility measures.

BACKGROUND: To compare the gait patterns between posterior cruciate retention and substitution in total knee arthroplasty (TKA). METHODS: Electronic databases including the PubMed, Embase, CINAHL, Web of Science, and Cochrane databases were searched to identify clinical trials investigating posterior cruciate retention versus substitution in TKA. The outcome measurements were the kinematic gait parameters (flexion at heel strike, maximum flexion during loading response, flexion range during loading, minimal flexion at terminal stance, maximal flexion at the swing, and total flexion during the gait cycle), Knee Society Score (KSS), knee flexion, knee extension, and walking speed. Statistical software Review Manager 5.4 and Stata 14.0 were used for data analysis. RESULTS: There were finally 9 studies included in this meta-analysis. The results did not reveal differences between posterior cruciate retention (CR) and posterior cruciate substitution (PS) groups in TKA, in terms of kinematic gait parameters, knee extension, walking speed, and KSS. However, the PS group had a significantly larger knee flexion angle than that in the CR group [weighted mean difference = - 3.20, 95% CI - 6.13 to - 0.28, P = 0.03]. CONCLUSION: Both the posterior cruciate retention and posterior cruciate substitution lead to obvious improvements in patient function and have their advantages in getting a good cup position. The PS design is significantly better on the knee flexion, while there are no statistical differences in kinematic gait parameters and outcome scores between them. This might indicate that surgeons do not necessarily need a PS design to substitute the posterior cruciate ligament during TKA.

Background: Previous studies have partly explored the role of B-cell CLL/lymphoma 7 protein family member B (BCL7B) in tumorigenesis and development. However, the prognosis and immunoregulatory value of BCL7B in pan-cancer patients remains unclear. Methods: Through The Cancer Genome Atlas (TCGA) and Genotype-Tissue Expression (GTEx) databases, the distinct expression of BCL7B gene in 33 tumors and adjacent normal tissues was analyzed. The Kaplan-Meier method (univariate Cox regression analysis and Kaplan-Meier curve) was used to identify the cancer types whose BCL7B gene expression was related to prognosis. The receiver operating characteristic (ROC) curve was used to elucidate the diagnosis value of BCL7B gene. Spearman's rank correlation coefficient was used to explore the relationship between BCL7B gene expression and immune cell infiltration, immune checkpoints, DNA methylation, DNA repair genes, immune-activating genes, immune-suppressing genes, immune subtypes, tumor mutation burden (TMB), and microsatellite instability (MSI). The Wilcoxon rank sum test and Kruskal-Wallis test were used to compare the expression of BCL7B gene in tumor tissues with different clinicopathological features. Gene set enrichment analysis (GSEA) was conducted to identify the tumor-related pathways in pan-cancer. The Human Protein Atlas (HPA) database was used to verify the BCL7B gene expression at the protein level. Results: High expression of BCL7B was associated with an inferior prognosis in glioblastoma multiforme (GBM), glioma (GBMLGG), kidney chromophobe (KICH), brain lower grade glioma (LGG), oral squamous cell carcinoma (OSCC), rectum adenocarcinoma (READ), and uveal melanoma (UVM). Low expression of BCL7B was associated with a poor prognosis in kidney renal clear cell carcinoma (KIRC), kidney renal papillary cell carcinoma (KIRP), skin cutaneous melanoma (SKCM), thyroid carcinoma (THCA), and sarcoma (SARC). The BCL7B gene expression had varying degrees of correlation with 24 immune cell subsets in 37 tumor environments such as adrenocortical carcinoma (ACC) and bladder urothelial carcinoma (BCLA). Spearman's rank correlation coefficient showed that BCL7B gene expression had different degrees of correlation with 47 immune checkpoints, 46 immune-activating genes, 24 immune-suppressing genes, 5 DNA repair genes, and DNA methylation, TMB, and MSI in 39 tumors. GSEA suggested that BCL7B was notably associated with cancer-related and immune-related pathways. Conclusion: In summary, BCL7B gene has a high diagnostic and prognostic value in pan-cancer and is related to the infiltration of 24 immune cell subsets in pan-cancer.

Coronavirus disease 2019 (COVID-19) caused by coronavirus-2 (SARS-CoV-2) infection has rapidly spread throughout the world and become a major threat to human beings. Cytokine storm is a major cause of death in severe patients. Abatacept can suppress cytokines used as antirheumatic drugs in clinical applications. This study analyzed the molecular mechanisms of abatacept treatment for COVID-19. Differentially expressed genes (DEGs) were identified by analyzing expression profiling of abatacept treatment for rheumatoid arthritis (RA) patients and SARS-CoV-2 infection patients. We found that 59 DEGs were upregulated in COVID-19 patients and downregulated following abatacept treatment. Gene set enrichment analysis (GSEA) and Gene Ontology (GO) analysis showed that immune and inflammatory responses were potential regulatory mechanisms. Moreover, we verified 8 targeting genes and identified 15 potential drug candidates for the treatment of COVID-19. Our study illustrated that abatacept could be a promising property for preventing severe COVID-19, and we predicted alternative potential drugs for the treatment of SARS-CoV-2 infection.

Improper sludge treatment will cause serious environmental problems, and sludge drying is the key to effective treatment. Almost all the existing sludge drying technologies use heating to overcome the great latent heat of moisture vaporization, which leads to high drying energy consumption. In this study, based on the particle high-speed self-rotation in the cyclone and micro-interface oscillations, the cyclone self-rotation drying (CSRD) technology was developed. It can realize drying of the dewatered landfill sludge (DLS) and the urban sewage dewatered sludge (UDSS) with mainly non-phase change. The obtained results reveal that at low carrier gas temperatures (< 100 °C) and very short residence time (< 15 s), the moisture content of the DLS decreased from 53% to 6.85%, and that of the UDSS decreased from 67% to 18.92%. Through calculation, the proportions of moisture non-phase change removal during the CSRD process touched 68.94% and 63.39%, respectively. Based on the experimental studies, we proposed an enlarged industrial application program (50 t/d) for the UDSS drying by employing the CSRD technology. The operating cost was 159.69 CNY/t H2O, showing prominent advantages. This study can provide guidelines for the practical application of CSRD technology and fill the scientific gap in the field of moisture non-phase change separation for sludge drying.
