RESUMO

BACKGROUND: Based on social comparison theory, two experiments were conducted to explore the effects of depression and social comparison on adolescents, using the ultimatum game (UG). METHODS: Before the formal experiment began, a preliminary experiment tested the effectiveness of social comparison settings. This study used the UG paradigm to explore adolescents' social decision-making in the context of gain and loss through two experiments. These experiments were designed as a 2 (group: depressive mood group, normal mood group) × 2 (social comparison: upward, downward) × 3 (fairness level: fair 5:5, unfair 3:7, extremely unfair 1:9) three-factor hybrid study. RESULTS: (1) The fairer the proposal was, the higher the sense of fairness participants felt, and the higher their acceptance rate. (2) The acceptance rate of the participants for downward social comparison was significantly higher than that for upward social comparison, but there was no difference in fairness perception between the two social comparisons. (3) Under the context of gain, the acceptance rate of the depressive mood group was higher than that of the normal mood group, but there was no difference in the acceptance rate between the depressive mood group and the normal mood group under the loss context. Depressive mood participants had more feelings of unfairness in the contexts of both gain and loss. (4) The effects of depressive mood, social comparison and the fairness level of distribution on social decision-making interact. CONCLUSIONS: The interaction of social comparison, depressive mood and proposal type demonstrates that besides one's emotion, cognitive biases and social factors can also have an effect on social decision-making. These findings indicate that behavioral decision boosting may provide an avenue for appropriate interventions in helping to guide adolescents to make social decisions.

RESUMO

BACKGROUND: The type X collagen gene (Col10a1) is a signature gene of hypertrophic chondrocytes that are known as the main engine of long bone growth. Multiple transcription factors (TFs), including myocyte enhancer factor 2A (Mef2a), have previously been identified by in silico analysis as potential Col10al gene regulators. OBJECTIVES: In this study, we aimed to investigate the correlation between Mef2a and Col10a1 expression and the possible effects on chondrocyte proliferation and hypertrophic differentiation in vitro. METHODS: First, Mef2a expression in proliferating and hypertrophic chondrocytes were detected by quantitative real-time PCR (qRT-PCR) and Western blotting in two chondrocytic models, ATDC5 and MCT cells, as well as in mouse chondrocytes in situ. Transfection with Mef2a small interfering fragments or Mef2a overexpression plasmids in the above chondrocytic models were performed to determine how Mef2a knockdown or overexpression may influence Col10a1 expression. The binding between Mef2a and its putative binding site within the 150 bp Col10a1 cis-enhancer which was evaluated by the dual luciferase reporter assay. The effect of Mef2a on chondrocyte differentiation was determined by examining the chondrogenic marker gene expression by qRT-PCR and by alcian blue, alkaline phosphatase (ALP), and alizarin red staining of the ATDC5 cells stably knocked down by Mef2a. RESULTS: The expression of Mef2a in hypertrophic chondrocytes was significantly higher than that in proliferative chondrocytes in both chondrocytic models as well as in mouse chondrocytes in situ. Interference with Mef2a caused decreased Col10a1 expression, while overexpression of Mef2a upregulated Col10a1. The result of the dual luciferase reporter assay showed that Mef2a enhanced Col10a1 gene enhancer activity via its putative Mef2a binding site. For the staining of ATDC5 stable cell lines, although no significant differences were seen in ALP staining, significantly weaker alcian blue staining intensity was noticed in Mef2a knockdown stable cell lines compared to the control cells at day 21, while slightly weaker alizarin red staining was seen in the stable cell lines at days 14 and 21. Correspondingly, we detected decreased runt-related transcription factor 2 (Runx2), increased SRY-box transcription factor 9 (Sox9), as well as differential expression of other chondrogenic markers in ATDC5 stable cell lines compared with the controls. CONCLUSIONS: In conclusion, our results support that Mef2a upregulates Col10a1 expression possibly by interaction with its cis-enhancer. Altered levels of Mef2a affects the expression of chondrogenic marker genes, such as Runx2 and Sox9, but may only play an insignificant role during chondrocyte proliferation and maturation.

RESUMO

BACKGROUND: Network pharmacology is widely used in mechanistic studies of traditional Chinese medicines (TCMs). The present study aimed to predict the target and signaling pathway of Baihe Decoction in the intervention of coronary heart disease (CHD) based on a network pharmacology approach and molecular docking. METHODS: The active ingredients of Baihe Decoction were screened by the Traditional Chinese Medicine Systems Pharmacology (TCMSP), and their potential target genes and proteins in CHD were predicted. The targets were screened out using Online Mendelian Inheritance in Man and the Genecards database. Venn soft was used to obtain the common targets of drugs and diseases. The compound-target-disease network of Baihe Decoction in CHD was constructed in Cytoscape, and the functional protein interaction network was obtained through the STRING database. ClusterProfiler and Pathview were used to perform Gene Ontology function analysis and KEGG pathway enrichment analysis of the effective targets of Baihe Decoction in CHD. Finally, we used MOE software for molecular docking of the compounds to their targets. RESULTS: Fifteen active components of Baihe Decoction in CHD were screened, which corresponded to 145 targets in CHD, including 30 targets with strong correlations. The key targets included Jun, Aktl, MAPK1, RELA, IL6, CXCL8, EGFR, MAPK14, ESR1, and FOS, which were found to play important roles in the treatment of CHD. The results of molecular docking further illustrated the roles that the compounds with quercetin and ß-sitosterol play in the treatment of CHD through their interference with AKT1 and MAPK1 target proteins. CONCLUSIONS: This study has preliminarily revealed the mechanism of Baihe Decoction in the treatment of CHD. The components of TCM may intervene in the processes of CHD occurrence and development by regulating cardiomyocytes and antioxidative stress, and by participating in inflammation and immune response. Moreover, in the clinical syndrome differentiation of TCM, Baihe Decoction can be used as the main drug to treat CHD and angina pectoris due to qi stagnation and blood stasis caused by emotional discomfort.

RESUMO

Traditional seed pretreatment methods cause secondary pollution for the application of various chemicals. This study investigated the effect of dielectric barrier discharge (DBD) cold plasma on seedling growth. Effects of plasma-activated tap water (PATW) and plasma-activated seeds (PAS) were compared for germination rates, seedling height, dry weight, and chlorophyll content. Results show that compared with controls these growth parameters were all increased by more than 50%. The yields and contributions of hydrogen peroxide, nitrate, nitrite, and ammonium were quantified. Hydrogen peroxide and nitrate have an important role in seedling growth. By etching, the seed epidermis free radicals can reduce the apparent contact angle and increase the water absorption of the seeds. In addition to the low cost of PATW and PAS compared with commercial fertilizers, DBD does not involve any chemical addition. Thus, both PATW and PAS can be an alternative for improvement of agricultural production.

RESUMO

This study explores the influence of different social reference points and different comparison group sizes on risky decision-making. Participants were presented with a scenario describing an exam, and presented with the opportunity of making a risky decision in the context of different information provided about the performance of their peers. We found that behavior was influenced, not only by comparison with peers, but also by the size of the comparison group. Specifically, the larger the reference group, the more polarized the behavior it prompted. In situations describing social loss, participants were led to make riskier decisions after comparing themselves against larger groups, while in situations describing social gain, they become more risk averse. These results indicate that decision making is influenced both by social comparison and the number of people making up the social reference group.

RESUMO

The clinical thinking modes of "integrative disease and Syndrome" and "integrative macroscopical Syndrome-differentiation and microscopical Syndrome-differentiation" formed in the past half century were merely the primary train of thought and methodology of integrative traditional Chinese and Western medicine. In order to develop them to a higher level, stress should be put on the unity and holism of disordered organism and unity of function, structure and metabolism. It has long been ignored that a parallel and correlated relationship exists between the TCM Syndrome and pathological process of Western medicine, which is a common, set-formed and regular pathophysiological changes with certain development phase, presented in various diseases. Both have common features as that the same pathological process can be seen in different diseases and several pathological progresses can present in one disease, the same Syndrome also can be seen in different diseases and several Syndromes can present in a disease. Moreover, they have the compatible connotation and denotation. The author deduced that pathophysiological basis of main Syndrome is indeed the basic pathological process. Essence of Syndrome is the related pathological processes including abnormal changes in function, metabolism and morphological structure.
