Bladder cancer is a malignancy characterized by significant heterogeneity. RNA methylation has received an increasing amount of attention in recent years. RNA data were collected from the GEO database, and cell subsets were classified according to specific cell markers. Epithelial, immunological, and fibroblast cells were clustered individually to explore the tumor heterogeneity. To distinguish between malignant and benign cells, the InferCNV R package was employed. The monocle2 R package was used for pseudotime analysis. The Decouple R package was used for transcription factor analysis of each cell subgroup, and PROGENy was used to predict the activity of pathways related to tumors. The target lncRNA was screened for model construction. In addition, the qPCR experiment was used to detect the transcription level of lncRNA. Epithelial cells, fibroblasts, and T cells significantly differ in tumor and normal tissues. The lncRNAs related to m6A/m5C/m1A were intersected to construct the model. Finally, six model lncRNAs (PSMB8-AS1, THUMPD3-AS1, U47924.27, XXbac-B135H6.15, MIR99AHG, and C14orf132) were screened. High-risk individuals were shown to have a better prognosis. qPCR experiments showed that the model lncRNA was differentially expressed between normal and tumor cells. Immunotherapy will be more effective in treating individuals with lower risk than those with higher risk using 4 candidate drugs. The prognostic m6A/m5C/m1A-related lncRNA model was constructed for evaluating the clinical outcomes of bladder cancer patients and guiding clinical medication.

INTRODUCTION: Renal cell carcinoma (RCC) is a grave malignancy that poses a significant global health burden with over 400,000 new cases annually. Disulfidptosis, a newly discovered programmed cell death process, is linked to the actin cytoskeleton, which plays a vital role in maintaining cell shape and survival. The role of disulfidptosis is poorly depicted in the clear cell histologic variant of RCC (ccRCC). METHODS: Three sets of ccRCC cohorts, ICGC_RECA-EU (n = 91), GSE76207 (n = 32) and TCGA-KIRC (n = 607), were included in our study, the batch effect of which was removed using the "combat" function. Correlation was calculated using the "rcorr" function of the "Hmisc" package for Pearson analysis, which was visualized using the "pheatmap" package. Principal component analysis was performed by the "vegan" package, visualized using the "scatterplot3d" package. Long non-coding RNAs (lncRNAs) associated with disulfidptosis were screened out using least absolute shrinkage and selection operator (LASSO) and COX analysis. Tumor mutation, immune landscaping and immunotherapy prediction were performed for further characterization of two risk groups. RESULTS: A total of 1822 disulfidptosis-related lncRNAs was selected, among which 308 lncRNAs were found to be significantly associated with the clinical outcome of ccRCC patients. We retained 11 disulfidptosis-related lncRNAs, namely, AP000439.3, RP11-417E7.1, RP11-119D9.1, LINC01510, SNHG3, AC156455.1, RP11-291B21.2, EMX2OS, AC093850.2, HAGLR and RP11-389C8.2, through LASSO and COX analysis for prognosis model construction, which displayed satisfactory accuracy (area under the curve, AUC, values all above 0.6 in multiple cohorts) in stratification of ccRCC prognosis. A nomogram model was constructed by integrating clinical factors with risk score, which further enhanced the prediction efficacy (AUC values all above 0.7 in multiple cohorts). We found that patients of male gender, higher clinical stages and advanced pathological T stage were inclined to have higher risk score values. Dactinomycin_1911, Vinblastine_1004, Daporinad_1248 and Vinorelbine_2048 were identified as promising candidate drugs for treating ccRCC patients of higher risk score value. Moreover, patients of higher risk value were prone to be resistant to immunotherapy. CONCLUSION: We developed a prognosis predicting model based on 11 selected disulfidptosis-related lncRNAs, the efficacy of which was verified in different cohorts. Furthermore, we delineated an intricate portrait of tumor mutation, immune topography and pharmacosensitivity evaluations within disparate risk stratifications.

INTRODUCTION: Prostate cancer is a common cancer among male population. The aberrant expression of histone modifiers has been identified as a potential driving force in numerous cancer types. However, the mechanism of histone modifiers in the development of prostate cancer remains unknown. METHODS: Expression profiles and clinical data were obtained from GSE70769, GSE46602, and GSE67980. Seruat R package was utilized to calculate the gene set enrichment of the histone modification pathway and obtain the Histone score. Least absolute shrinkage and selection operator (LASSO) and Cox regression analyses were employed to identify marker genes with prognostic value. Kaplan-Meier survival analysis was conducted to assess the efficacy of the prognostic model. In addition, microenvironment cell populations counter (MCPcounter), single-sample gene set enrichment analysis (ssGSEA), and xCell algorithms were employed for immune infiltration analysis. Drug sensitivity prediction was performed using oncoPredict R package. RESULTS: We screened differentially expressed genes (DEGs) between Histone-high score (Histone-H) and Histone-low score (Histone-L) groups, which were enriched in RNA splicing and DNA-binding transcription factor binding pathways. We retained four prognostic marker genes, including TACC3, YWHAH, TAF1C and TTLL5. The risk model showed significant efficacy in stratification of the prognosis of prostate cancer patients in both internal and external cohorts (p < .0001 and p = .032, respectively). In addition, prognostic gene YWHAH was infiltrated in abundance of fibroblasts and highly correlated with Entinostat_1593 drug sensitivity score and the value of risk score. CONCLUSION: We innovatively developed a histone modification-related prognostic model with high prognostic potency and identified YWHAH as possible diagnostic and therapeutic biomarkers for prostate cancer. It provides novel insights to address prostate cancer and enhance clinical outcomes, thereby opening up a new avenue for customized treatment alternatives.

INTRODUCTION: Clear cell renal cell carcinoma (ccRCC) is the most prevalent and aggressive subtype of renal cell carcinoma, originating from renal tubular epithelial cells in the kidney. Hypoxia proves to be a feature commonly observed in solid tumors, leading to increased resistance to treatment and tumor progression. METHODS: scRNA-seq data were procured from GSE159115 data set. We utilized UMAP and NMF algorithm for clustering and dimensionality reduction. The FindAllMarkers function was used to compare various groups and identify potential hypoxia marker genes. A series of in vitro experiments, including CFA, flow cytometry targeting cell cycle, CCK-8, and EDU, was applied to investigate how ANGPTL4 regulated the ccRCC progression. Two cell lines of ccRCC cells, 786-O and Caki, were used for si-ANGPTL4 transfection. RESULTS: We annotated a total of a total of 6 cell clusters, namely ccRCC malignant cells, T cells, endothelial cells, myeloid cells, smooth muscle cells, and B cells. We observed higher levels of hypoxia-score in the ccRCC malignant cells, while lowest hypoxia-score in T and B cells. We detected multiple hypoxia-related subclusters of TME cells in ccRCC, among which S100A4 CD8+ T cells and nonhypoxia CD8+ T cells were found with a marked elevation of T cell inhibitory gene score. We identified that ANGPTL4+ endothelial cells might function as an integrative role in tumor angiogenesis. Multiple TME subclusters showed high potency in stratification of the prognosis of ccRCC patients. Moreover, by a series of in vitro experiment, we found ANGPTL4 regulated the ccRCC cell proliferation, probably through ERK/P38 pathway. CONCLUSION: We discerned multiple hypoxia-related subclusters of TME cells in ccRCC, which displayed distinct functional features and great potency in predicting prognosis of ccRCC patients. We identified the role of ANGPTL4 in regulating ccRCC proliferation via ERK/p38 pathway.

Clear-cell renal cell carcinoma (ccRCC) appears as the most common type of kidney cancer, the carcinogenesis of which has not been fully elucidated. Tumor heterogeneity plays a crucial role in cancer progression, which could be largely deciphered by the implement of scRNA-seq. The bulk and single-cell RNA expression profile is obtained from TCGA and study conducted by Young et al. We utilized UMAP, TSNE, and clustering algorithm Louvain for dimensionality reduction and FindAllMarkers function for determining the DEGs. Monocle2 was utilized to perform pseudo-time series analysis. SCENIC was implemented for transcription factor analysis of each cell subgroup. A series of WB, CFA, CCK-8, and EDU analysis was utilized for the validation of the role of MT2A in ccRCC carcinogenesis. We observed higher infiltration of T/NK and B cells in tumorous tissues, indicating the role of immune cells in ccRCC carcinogenesis. Transcription factor analysis revealed the activation of EOMES and ETS1 in CD8 + T cells, while CAFs were divided into myo-CAFs and i-CAFs, with i-CAFs showing distinct enrichment of ATF3, JUND, JUNB, EGR1, and XBP1. Through cell trajectory analysis, we discerned three distinct stages of cellular evolution, where State2 symbolizes normal renal tubular cells that underwent transitions into State1 and State3 as the CNV score ascended. Functional enrichment examination revealed an amplification of interferon gamma and inflammatory response pathways within tumor cells. The consensus clustering algorithm yielded two molecular subtypes, with cluster 2 being associated with advanced tumor stages and an abundance of infiltrated immune cells. We identified 17 prognostic genes through Cox and LASSO regression models and used them to construct a prognostic model, the efficacy of which was verified in multiple cohorts. Furthermore, we investigated the role of MT2A, one of our hub genes, in ccRCC carcinogenesis, and found it to regulate proliferation and migration of malignant cells. We depicted a detailed single-cell landscape of ccRCC, with special focus on CAFs, endothelial cells, and renal tubular cells. A prognostic model of high stability and accuracy was constructed based on the DEGs. MT2A was found to be actively implicated in ccRCC carcinogenesis, regulating proliferation and migration of the malignant cells.

BACKGROUND: Most plant breeding and agricultural practices are based on selecting genotypes for yield. However, this is inadequate to screen crop varieties for specific attributes, such as drought tolerance. In this study, we quantified the response of oat (Avena sativa L.) plant physiological and morphological traits to drought stress and selected some key traits to establish a genotype by yield*trait (GYT)-based method for ranking 30 oat genotypes. The effectiveness of this method was also evaluated under drought conditions. RESULTS: Water-deficit treatment significantly reduced leaf chlorophyll, root morphological traits, groat yield and associated components, such as mean grain weight. We observed that the genotypes 'JUSTICE' and 'BOLINA' had the smallest and largest yield loss, respectively, after exposure to drought stress, but showed opposite trends in the biomass allocation of roots and grains. This indicated that drought tolerance was highly dependent on the distribution of photoassimilates. Our results also illustrated that the GYT method is a trade-off approach and more effective in selecting oat ideotypes under drought conditions than the yield-related index method because it combines yield, yield stability, and related agronomic traits in the calculation process. CONCLUSION: Drought-tolerant genotypes had more biomass allocated to roots and grains with higher chlorophyll content and better root structure, e.g. longer root lengths than drought-sensitive lines. By integrating yield and yield-related traits, the GYT approach is more practical than traditional single-trait selection methods when assessing drought tolerance. © 2023 His Majesty the King in Right of Canada. BACKGROUND: Modern wheat cultivars have been developed having distinct advantages in many aspects under drought stress, such as plasticity in biomass allocation and root system architecture. A better understanding of the biomass allocation mechanisms that enable modern wheat to achieve higher yields and yield-based water use efficiency (WUEg ) is essential for implementing best management strategies and identifying phenotypic traits for cultivar improvement. We systematically investigated the biomass allocation, morphological and physiological characteristics of three ploidy wheat genotypes under 80% and 50% field water-holding capacity (FC) conditions. Some crucial traits were also assessed in a complementary field experiment. RESULTS: The diploid and tetraploid genotypes were found to allocate more biomass to the root system, especially roots in the topsoil under drought stress. Our data illustrated that lower WUEg and yield of these old genotypes were due to excessive investment in the root system, which was associated with severely restricted canopy development. Modern hexaploid genotypes were found to allocate smaller biomass to roots and larger biomass to shoots. This not only ensured the necessary water uptake, but also allowed the plant to distribute more assimilates and limited water to the shoots. Therefore, the hexaploid genotypes have evolved a stable plant canopy structure to optimize WUEg and grain yield. CONCLUSION: This study demonstrated that the biomass shift from below ground to above ground or a more balanced root:shoot ratio tended to optimize water use and yield of the modern cultivars. BACKGROUND: The newly developed sugarcorn is conceived for dual-purpose use as a potential biofuel feedstock and a high-energy silage crop. Its agronomic traits are, however, not fully appraised under the umbrella of nitrogen (N) management and with canopy reflectance indicator. A 3-year field study was conducted to examine the responses of silage biomass, stalk sugar concentration, sugar and juice yields to various N applications; and determine the quantitative relationships between canopy reflectance, expressed as the normalized difference vegetation index (NDVI), and stalk sucrose or other sugar measures in a dual-purpose sugarcorn (cv. 'CO384xC103'), in comparison with a commercial leafy silage-specific hybrid (cv. 'Pride A5892G3 EDF'). RESULTS: The moderate N rate, 125 kg ha-1 , produced similar stalk sucrose, silage and grain yields, compared to the high rate (250 kg N ha-1 ), regardless of application methods. The NDVI signatures measured at the V8-V10 stage exhibited significant (P < 0.01) and exponential relationships with stalk sucrose concentrations, sucrose and juice yields at the R3 stage, and with silage yield at approximately 65% whole-plant moisture, the optimum silage-harvest window. CONCLUSION: The results indicate that the moderate N rate, 125 kg ha-1 , which is recommended for conventional grain corn production in the region, was likely close to the economic optimum N rate for leafy silage-specific and sugarcorn. Canopy reflectance, measured at the early growth stages, can be used as a potential indicator of sugar and silage production, and this quantitative relationship necessitates further evaluation with more genotypes and under wide environmental conditions. © 2019 Her Majesty the Queen in Right of Canada Journal of The Science of Food and Agriculture © 2019 Society of Chemical Industry.

BACKGROUND: The eco-physiological mechanism of wheat yield loss resulting from chilling stress is a fundamental scientific issue. However, previous studies have focused on hexaploid wheats, and few studies on the morphological and physiological plasticity of wheat plants. Six different wheat genotypes were tested under chilling stress to investigate the physio-morphological parameters as well as the loss of grain yield in growth chambers. RESULTS: Chilling stress resulted in significant loss in grain yield in all genotypes. Under chilling stress, diploid wheats generated zero harvest, and tetraploid genotypes also suffered from a pronounced loss in grain yield, compared with the control group. In contrast, hexaploid genotypes acquired relatively high maintenance rate of grain yield among three species. CONCLUSIONS: Diploid and tetraploid wheat genotypes maintained relatively large leaf area and high photosynthetic rates, but they were subjected to significant declines in vascular bundle number and productive tillers as a consequence of the inhibition by sink growth under chilling stress. The hexaploid wheats were found to have relatively low leaf area and photosynthetic rates. These genotypes also stored more soluble carbohydrates and exhibited stronger sink enhancement, ensuring the translocation and redistribution of assimilates. Our findings provided a new theoretical understanding of yield stabilization in the domestication process of wheat genotypes under chilling stress. © 2017 Her Majesty the Queen in Right of Canada. BACKGROUND: The hypothesis that positive and negative interactions account for adaptive strategies was tested in a controlled study with two oat (Avena sativa) genotypes: 'Manotick' with erect leaves and 'Oa1316-1' with prostrate leaves. An increasing competition pattern was designed by varying the number of seeds planted in each container and the space between containers, thus creating different planting density regimes (i.e. alternative and solid treatments). RESULTS: Total biomass of individual plants tended to decrease exponentially with increasing density in both genotypes. Under high density stress, Manotick allocated more biomass to the roots and produced 50% more tillers, leading to more non-productive tillers and lower harvest index in the alternative than in the solid treatment. In contrast, Oa1316-1 allocated more biomass to panicles and stems, and less to the roots, with fewer tillers. CONCLUSIONS: With increasing density and strengthening intraspecific competition, Manotick reduced aboveground biomass allocation, leading to lower yield, while Oa1316-1 decreased allocation to the roots, but increased allocation to the panicles under an increasingly competitive environment. These adjustments were mechanically derived from negative and positive interactions, ensuring greater yield in the prostrate type. Our findings provided a novel rationale for a planting strategy based on plant type selections.

BACKGROUND: Optical sensing is a potential tool to estimate plant N status, but soil water deficits may interefere with forming a clear relationship. A greenhouse study was conducted with oat plants treated with three water regimes and four N levels to determine whether optical sensing could be used to estimate leaf N and relative water content (RWC). RESULTS: Leaf N was strongly correlated with reflectance at 550 nm and at around 705 nm, and N treatments caused a red-edge peak shift to lower wavelength. The ratio of the first derivative reflectance at 741-696 nm (FDRE) was identified to be a good estimator of leaf N at jointing (R(2) = 0.90) and heading (R(2) = 0.86) stages across water treatments. Leaf N also had a stronger association with the red-edge position (REP) at both stages (R(2) = 0.83 and 0.78), or with the ratio R4 (R760/R550) at jointing (R(2) = 0.88), than with chlorophyll meter (SPAD) readings. Under water stress, the predictive accuracy of leaf N increased with these reflectance indices, but decreased using SPAD readings. CONCLUSION: The results indicate that specific reflectance indices of FDRE, REP and R4 may be used for a rapid and non-destructive estimation of oat plant N status over a range of water regimes.

It is crucial to clarify how the iron nanostructure activates plant growth, particularly in combination with arbuscular mycorrhizal fungi (AMF). We first identified 1.0 g·kg-1 of nanoscale zerovalent iron (nZVI) as appropriate dosage to maximize maize growth by 12.7-19.7% in non-AMF and 18.9-26.4% in AMF, respectively. Yet, excessive nZVI at 2.0 g·kg-1 exerted inhibitory effects while FeSO4 showed slight effects (p > 0.05). Under an appropriate dose, a nano core-shell structure was formed and the transfer and diffusion of electrons between PS II and PS I were facilitated, significantly promoting the reduction of ferricyanide and NADP (p < 0.05). SEM images showed that excessive nZVI particles can form stacked layers on the surface of roots and hyphae, inhibiting water and nutrient uptake. TEM observations showed that excessive nanoparticles can penetrate into root cortical cells, disrupt cellular homeostasis, and substantially elevate Fe content in roots (p < 0.05). This exacerbated membrane lipid peroxidation and osmotic regulation, accordingly restricting photosynthetic capacity and AMF colonization. Yet, appropriate nZVI can be adhered to a mycelium surface, forming a uniform nanofilm structure. The strength of the mycelium network was evidently enhanced, under an increased root colonization rate and an extramatrical hyphal length (p < 0.05). Enhanced mycorrhizal infection was tightly associated with higher gas exchange and Rubisco and Rubisco enzyme activities. This enabled more photosynthetic carbon to input into AMF symbiont. There existed a positive feedback loop connecting downward transfer of photosynthate and upward transport of water/nutrients. FeSO4 only slightly affected mycorrhizal development. Thus, it was the Fe nanostructure but not its inorganic salt state that primed AMF symbionts for better growth.

Plastic fragments are widely found in the soil profile of terrestrial ecosystems, forming plastic footprint and posing increasing threat to soil functionality and carbon (C) footprint. It is unclear how plastic footprint affects C cycling, and in particularly permanent C sequestration. Integrated field observations (including 13C labelling) were made using polyethylene and polylactic acid plastic fragments (low-, medium- and high-concentrations as intensifying footprint) landfilling in soil, to track C flow along soil-plant-atmosphere continuum (SPAC). The result indicated that increased plastic fragments substantially reduced photosynthetic C assimilation (p < 0.05), regardless of fragment degradability. Besides reducing C sink strength, relative intensity of C emission increased significantly, displaying elevated C source. Moreover, root C fixation declined significantly from 21.95 to 19.2 mg m-2, and simultaneously root length density, root weight density, specific root length and root diameter and surface area were clearly reduced. Similar trends were observed in the two types of plastic fragments (p > 0.05). Particularly, soil aggregate stability was significantly lowered as affected by plastic fragments, which accelerated the decomposition rate of newly sequestered C (p < 0.05). More importantly, net C rhizodeposition declined averagely from 39.77 to 29.41 mg m-2, which directly led to significant decline of permanent C sequestration in soil. Therefore, increasing plastic footprint considerably worsened C footprint regardless of polythene and biodegradable fragments. The findings unveiled the serious effects of plastic residues on permanent C sequestration across SPAC, implying that current C assessment methods clearly overlook plastic footprint and their global impact effects.

Background: The relationship between the C-reactive protein (CRP) and prognosis in prostate cancer (PCa) has been widely discussed over the past few years but remains controversial. Material and methods: In our meta-analysis, we searched 16 reliable studies in the PubMed, Embase, and Cochrane Library databases. Otherwise, we have successfully registered on the INPLASY. We also performed random- and fixed-effects models to evaluate the hazard ratio (HR) and 95% confidence interval (CI), respectively. Result: The result of our meta-analysis shows that elevated CRP levels were related to worse overall survival (OS) (HR = 1.752, 95% CI = 1.304-2.355, p = 0.000), cancer-specific survival (CSS) (HR = 1.663, 95% CI = 1.064-2.6, p = 0.026), and progression-free survival (PFS) (HR = 1.663, 95% CI = 1.064-2.6, p = 0.026) of PCa patients. There was significant heterogeneity, so we performed a subgroup analysis according to the staging of the disease and found the same result. Furthermore, the heterogeneity was also reduced, and no statistical significance. Conclusion: Our study shows that the level of CRP could reflect the prognosis of prostate cancer patients. We find that PCa patients with high levels of CRP often have worse OS, CSS, and PFS, although the stages of the patients' disease are different. More studies are needed to verify this idea.

Non-hydraulic root signals (nHRS) are affirmed as a unique positive response to soil drying, and play a crucial role in regulating water use efficiency and yield formation in dryland wheat production. Strigolactones (SLs) can enhance plant drought adaptability. However, the question of whether strigolactones enhance grain yield and water use efficiency by regulating nHRS and antioxidant defense systems in dryland wheat remains unanswered. In this study, pot experiments were conducted to investigate the effects of strigolactones on nHRS, antioxidant defense system, and grain yield and water use efficiency in dryland wheat. The results showed that external application of SLs increased drought-induced abscisic acid (ABA) accumulation and activated an earlier trigger of nHRS at 73.4% field capacity (FC), compared to 68.5% FC in the control group (CK). This phenomenon was mechanically associated with the physiological mediation of SLs. The application of SLs significantly enhanced the activities of leaf antioxidant enzymes, reduced ROS production, and mitigated oxidative damage to lipid membrane. Additionally, root biomass, root length density, and root to shoot ratio were increased under strigolactone treatment. Furthermore, exogenous application of SLs significantly increased grain yield by 34.9% under moderate drought stress. Water use efficiency was also increased by 21.5% and 33.3% under moderate and severe drought conditions respectively, compared to the control group (CK). The results suggested that the application of strigolactones triggered earlier drought-sensing mechanism and improved the antioxidant defense ability, thus enhancing grain yield and water use efficiency in dryland wheat production.

G protein-coupled receptors (GPCRs) are a class of receptors on cell membranes that regulate various biological processes in cells, such as cell proliferation, differentiation, migration, apoptosis, and metabolism, by interacting with G proteins. However, the role of G protein-coupled receptors in predicting the prognosis of renal clear cell carcinoma is still unknown. The transcriptome data and clinical profiles of renal clear cell carcinoma patients, were downloaded from TCGA databases, and the validation group data were downloaded from number GSE167573, including 63 tumor samples and 14 normal samples. Single-cell RNA sequencing data were downloaded from the GEO database, No. GSE152938 and selected samples were used for GSEA enrichment analysis, WGCNA subgroup analysis, single-cell data analysis, and mutation analysis to explore the role of G protein-coupled receptor-related genes in the diagnosis and prognosis of renal clear cell carcinoma and to verify their reliability with cellular experiments. Preliminary studies observed a lower growth activity during the vegetative stage with higher growth attributes at the pod-filling stage among the high nitrogen (N) utilisation efficiency (NUtE) oilseed rape (Brassica napus L.) genotypes, compared with the low NUtE genotypes. Therefore, we hypothesised that there would exist a critical growth stage when distinctive phenotypic traits are exhibited to regulate yield formation and NUE. A field experiment and a hydroponic culture were conducted to characterise the differences in shoot and root physiological indicators of the high and low NUtE oilseed rape genotypes at seedling, bud, bolting, flowering and pod-filling stages. We found that flowering was the critical period when the reverse growth habit occurred between high and low NUtE genotypes. The high NUtE genotypes displayed larger values of root traits, stronger N uptake kinetics parameters, higher activity of leaf glutamine synthetase (GS) and glutamate synthetase (GOGAT), larger SPAD values and net photosynthetic rate, ultimately leading to higher seed yield and NUE. Our results indicate that flowering is the critical growth stage to distinguish the high from low NUtE oilseed rape genotypes, and plant breeders may focus on selecting root and shoot phenotypic traits from flowering stage onwards to achieve both high yields and NUE for oilseed rape genotypes.

Real time PCR assays were developed to detect and quantify the transgene DNA of a commercially released Bacillus thuringiensis (Bt) corn (Zea mays L.) hybrid (DKC42-23), which was derived from the event MON863 and also carried a neomycin phosphotransferase gene (the nptII gene). We applied the real time PCR assays to investigate the persistence of the transgene DNA in a field trial grown with DKC42-23 over 3 years, in combination with bacterial natural transformation. The results showed that under continuous cultivation of DKC42-23, its transgene DNA was detectable in the field plots all year around. Meanwhile, when soil DNA extracts from DKC42-23 plots were used as donor in bacterial natural transformation, successful recovery of kanamycin resistant (Km(R)) transformants indicated that the nptII gene carried by DKC42-23 could be taken up and integrated into naturally competent Pseudomonas stutzeri pMR7 cells, leading to the restoration of the antibiotic resistance of P. stutzeri pMR7. However, after the cultivation of a soybean line in the same plots for the subsequent growing season, the presence of transgene DNA of DKC42-23 was reduced to undetectable levels at the end of that growing season. Therefore, existing corn-soybean crop rotation practices reduce the availability of transgene DNA in soil and thus minimize the risks that might be attributable to horizontal gene transfer. The real time PCR assays are useful for investigating the persistence of transgene DNA derived from the MON863 event in soil environments.

Ecologically relevant low UV-B is reported to alter reactive oxygen species metabolism and anti-oxidative systems through an up-regulation of enzymes of the phenylpropanoid pathway. However, little is known about low UV-B-induced changes in carotenoid profile and their impacts on light harvesting and photoprotection of photosystem II (PSII) in plants. We investigated carotenoids profile, chlorophyll pigments, phenolics, photosynthetic efficiency and growth in Arabidopsis thaliana (Col-0) plants grown under photosynthetically active radiation (PAR), PAR+ ultraviolet (UV)-A and PAR+UV-A+B regimes for 10 days in order to assess plant acclimation to low UV-B radiation. A chlorophyll fluorescence assay was used to examine UV-B tolerance in plants further exposed to acute high UV-B for 4 and 6 h following a 10-day growth under different PAR and UV regimes. We found that both PAR+ UV-A and PAR+UV-A+B regimes had no negative effect on quantum efficiency, electron transport rate, rosette diameter, relative growth rate and shoot dry weight of plants. Chronic PAR+ UV-A regime considerably (P < 0.05) increased violaxanthin (26 %) and neoxanthin (92 %) content in plants. Plant exposure to chronic PAR+UV-A+B significantly (P < 0.05) increased violaxanthin (48 %), neoxanthin (63 %), lutein (33 %), 9-cis ß-carotene (28 %), total ß-carotene (29 %) and total phenolics (108 %). The maximum photochemical efficiency (Fv/Fm) in leaves was found to be positively correlated with total phenolics (rho = 0.81 and rho = 0.91, P < 0.05 for 4 and 6 h, respectively) and non-photochemical quenching (qN) (rho = 0.81 and rho = 0.84, P < 0.05 for 4 and 6 h, respectively) in plants exposed to acute high UV-B for 4 and 6 h following a 10-day growth under chronic PAR+UV-A+B. There was also a significant positive correlation (rho = 0.93, P < 0.01) between qN and lutein content in the plants exposed to acute high UV-B stress for 4 h following plant exposure to chronic PAR+UV-A+B. The findings from our study indicate that plants grown under chronic PAR+UV-A+B displayed higher photoprotection of PSII against acute high UV-B stress than those grown under PAR and PAR+ UV-A regimes. An induction of phenolics and lutein-mediated development of qN were involved in the photoprotection of PSII against UV-B-induced oxidative stress.
