RESUMO

We studied the effect of gonadotropin-releasing hormone agonist surfagon (2 µg/kg, once, intraperitoneally) on anxious behavior of adult gonadectomized and non-gonadectomized male rats. It was shown that surfagon significantly increased anxiety of both gonadectomized and non-gonadectomized rats in the open-field test and in elevated plus maze.

RESUMO

The effect of serotoninotropic substances with positive and negative effects on serum concentrations of thyrotropic hormone, total triiodothyronine, and total thyroxin was studied in old male rats. Chronic treatment with L-tryptophan and citalopram reduced serum level of total thyroxin in old intact animals and serum concentration of thyrotropic hormone in old thyrectomied rats. DL-p-chlorophenylalanine increased the serum concentrations of total thyroxin in old intact rats and in thyroidectomied animals treated with triiodothyronine. Chronic methysergide therapy was associated with reduction of serum concentration of thyrotropic hormone in old intact and old thyroidectomied rats and of total triiodothyronine in old intact animals.

RESUMO

Copper deficiency in adult rats was induced by addition of silver chloride to the feed. The concentrations of silver, copper, iron, and zinc and relative activity of genes for copper transporting proteins and copper enzymes were measured in the cortex, cerebellum, hippocampus, amygdala, pituitary gland, and hypothalamus. Silver was accumulated only in the hypothalamic-pituitary system. These changes were accompanied by a decrease in the concentration of copper and increase in the contents of iron and zinc. Activity of genes for copper transport enzymes (high-affinity copper transporter; and two copper transport ATPases, ATP7A and ATP7B) and copper enzymes that were formed in the intracellular secretory pathway did not decrease in the brain of rats with copper deficiency. Relative activity of genes for intracellular copper enzymes (Cu(2+)/Zn(2+) superoxide dismutase and subunit IV of cytochrome c oxidase), concentration of immunoreactive polypeptides of superoxide dismutase, and enzymatic activity of superoxide dismutase remained unchanged under these conditions.

Fibrillogenesis was induced in rats by injection of a fragment of neurotoxic protein, beta-amyloid protein precursor, into the cerebral ventricle. Copper, iron, and zinc concentrations and relative activities of genes of copper-transporting protein and extracellular and intracellular cuproenzymes were evaluated in different brain compartments of these animals. Copper and zinc concentrations decreased significantly in different compartments of the brain of rats with experimental fibrillogenesis, while iron content did not change. According to the data of RT-PCR analysis, activities of genes of copper-transporting protein and extracellular coenzyme decreased. The expression of intracellular cuproenzyme genes and the content of SOD1 protein did not change, SOD1 activity in the cytosol decreased, and active SOD1 was detected in the mitochondrial intermembrane space. The relationship between fibrillogenesis and copper metabolism is discussed.

RESUMO

The efficacy of a combined therapy with reminyl and ovestin in low doses used for pharmacocorrection of cognitive disorders in women after total ovariectomy has been studied. The results show a pronounced efficacy of the proposed combined therapy as compared to the standard hormonal replacement therapy in women with hypoestrogenic syndrome, which is confirmed by a significant decrease in the expression of mental impairments evaluated using the BCRS, CCSE, and MMSE methods.

The activity of selective serotonin re-uptake inhibitor cypramil (cipramil, citalopram) has been studied in young male rats with thyroid hormone dysbalance induced by thyroidectomy. Thyroidectomy increased the level of depressed behavior in the Porsolt forced swim test and enhanced the expression of emotional behavior in the open-field test. The replacement treatment of thyroidectomized rats with triiodothyronine (T3) produced an antidepressant and anxiolytic effects. The chronic administration of cypramil also produced an antidepressant action in the Porsolt test, the drug effect being more pronounced in the case of a combined treatment with cypramil and T3 (synergism). Cypramil reduced the horizontal motor activity and the oprientation-research activity in the open-field test, but these drug effects were less pronounced in the case of joint administration with T3.

The activity of L-tryptophan was studied on young male rats in depressive state caused by thyreoid hormone dysbalance. The immobilization time in the Porsolt forced swim test in thyreoidectomized rats was significantly increased in comparison to the intact control. The replacement therapy with triiodothyronin (T3) reduced the level of depressed behavior to the initial value. Chronic administration of L-tryptophan significantly decreased the total immobilization time in all the test groups, irrespective of the hormonal state. However, the effect of L-tryptophan was more pronounced in T3-treated rats.
