PURPOSE: The present study aimed to explore the associations between the expression pattern of molecules in the Notch pathway in the cumulus cells of polycystic ovary syndrome (PCOS) patients and the quality of zygotes and embryos. METHODS: A total of 200 cumulus complexes surrounding mature oocytes were obtained from 40 patients with and without PCOS undergoing intracytoplasmic sperm injection (ICSI). The expressions of Notch-1, Notch-2, and Notch-3 genes were examined by Reverse Transcription Q-PCR assay. Moreover, immunocytochemistry was performed for the expressions of Jagged-1 and Jagged-2 proteins. The correlations between the Notch receptors and their ligand expressions and the qualities of the zygote and embryo were investigated. RESULTS: The expression levels of Notch-2, Notch-3, Jagged-1, and Jagged-2 were significantly lower in patients with PCOS than in normal women (p < 0.05), while Notch-1 showed no meaningful difference between the groups. A positive correlation was found between Notch-1 and embryo quality. Furthermore, only Notch-2 and Jagged-2 marginally correlated with zygote quality. CONCLUSION: The data of the present study indicated that evaluating the molecules in the Notch pathway in PCOS patients' cumulus cells provides a novel approach to predict the zygote and embryo quality. However, further studies on a larger population are needed to validate this finding.

BACKGROUND AND OBJECTIVES: Diabetic retinopathy (DR) is the leading cause of blindness worldwide, and therefore its early detection is important in order to reduce disease-related eye injuries. DR is diagnosed by inspecting fundus images. Since microaneurysms (MA) are one of the main symptoms of the disease, distinguishing this complication within the fundus images facilitates early DR detection. In this paper, an automatic analysis of retinal images using convolutional neural network (CNN) is presented. METHODS: Our method incorporates a novel technique utilizing a two-stage process with two online datasets which results in accurate detection while solving the imbalance data problem and decreasing training time in comparison with previous studies. We have implemented our proposed CNNs using the Keras library. RESULTS: In order to evaluate our proposed method, an experiment was conducted on two standard publicly available datasets, i.e., Retinopathy Online Challenge dataset and E-Ophtha-MA dataset. Our results demonstrated a promising sensitivity value of about 0.8 for an average of >6 false positives per image, which is competitive with state of the art approaches. CONCLUSION: Our method indicates significant improvement in MA-detection using retinal fundus images for monitoring diabetic retinopathy.

BACKGROUND: Menopause is a natural period in women's life and can be affected by several factors. The aim of this study was to identify the associated factors for age of natural menopause and among women with early and premature menopause based on a cohort study in Iran. METHODS: This population-based study was conducted on 894 post menopause women between 35 and 70 years old who participated in the Bandare-Kong Non-Communicable Diseases (BKNCD) Cohort Study, a part of Prospective Epidemiological Research Studies in Iran (PERSIAN) from March 2016 to February 2019. All women completed a standard self-reported questionnaire. Data were analyzed using chi-square test, independent t test, and ANOVA as well as a multivariable linear regression model. RESULTS: The mean age at natural menopause was 48.31 ± 6.34 years. After adjusting other variables, gravida, history of cardiac disease, socioeconomic status and residence status were predictive of age at menopause (P < 0.001). Among the premature menopause group, the mean age at menopause was significantly higher among women with diabetes compared to women without diabetes group (35.68 ± 2.92 vs. 33.82 ± 3.06; P = 0.043), while the mean age at menopause was significantly lower in women with infertility compared to women without infertility (29.13 ± 5.22 vs. 34.84 ± 2.826; P = 0.048). CONCLUSIONS: This study suggests that the predictors of menopausal age differed in women with premature menopause compared to overall menopause age. Prospective studies are needed to evaluation the effects of these factors on menopausal age.

OBJECTIVES: The effects of Anethum graveolens seed extract on fertility of male rats were investigated. METHODS: Male Wistar rats were divided into five groups according to the treatment they received during 42 days: control, low dose (0.5 g/kg) and high dose (5 g/kg) of aqueous extracts, and low dose (0.045 g/kg) and high dose (0.45 g/kg) of ethanol extracts of Anethum graveolens seed. Sperm count and motility and testosterone concentration were measured. Sections of the testes, epididymis, and seminal vesicles were stained with peroxidase-conjugated lectins of Ulex europaeus agglutinin, peanut agglutinin, Dolichos biflorus agglutinin, soy bean agglutinin and concanavalin A. The treated male rats were mated with females and the crown-rump lengths and weights of their newborn pups were measured. RESULTS: No significant differences in sperm count, sperm motility or testosterone concentration were observed in the experimental groups. However, female rats did not become pregnant after mating with rats given the high dose of the ethanol extract. The distribution of terminal sugars on the epithelial surface of the reproductive structures decreased in the experimental groups. CONCLUSION: Anethum graveolens extract decreased fertility rate by modifying some terminal sugars on the cell surface of male reproductive organs involved in sperm maturation, capacitation and oocyte recognition.

Recent studies on the analysis of the evolutionary relationships between the various taxa of the teleost fishes are based on the evaluation of the efficacy of different approaches, verifying the taxonomical congruence between the hypotheses inferred from molecular data and those from morphological data. The first objective of this study was to investigate the macro- and microscopic characterization of the body key scales for the sixteen Aphanius species, and to discuss their taxonomic significance. In addition, to reveal the phylogenetic origins of particular adaptations and to identify a possible cases of convergent evolution in scales of Aphanius species, we used a phylogenetic character mapping procedure that uses an independent data source, namely molecular data that is presumed to be functionally independent of the scale traits. Comparison of the resulted phylogenetic trees and scale data dendrogram implied that neither scale shape/scale surface morphology, nor scale surface microstructure can be used for the species identification and inferring phylogenetic relationships in the genus Aphanius. This conclusion confirms previous studies, indicating that the scale shape and ornamentations varies with habitat, age and size of the fish. We also concluded that the polygonal state in the scales of Aphanius species was a primitive shared character state.

RESUMO

The embryonic and early larval development of laboratory reared Zagros tooth-carp, Aphanius vladykovi Coad, 1988, are described and illustrated. Development and embryogenesis start with the external fertilization of sticky, transparent and spherical telolecithal/macrolecithal eggs with a mean diameter of 1.61± 0.12 mm and it continues with meroblastic/radial cleavage, blastulation/blastula formation, epibolic cell migration during gastrulation and organogenesis resulting in a newly hatched larvae of 5.23 ± 0.09 mm in length with attached yolk sac at about 164 hr (at 24 ± 1°C) after fertilization.

The lack of publicly available datasets of computed-tomography angiography (CTA) images for pulmonary embolism (PE) is a problem felt by physicians and researchers. Although a number of computer-aided detection (CAD) systems have been developed for PE diagnosis, their performance is often evaluated using private datasets. In this paper, we introduce a new public dataset called FUMPE (standing for Ferdowsi University of Mashhad's PE dataset) which consists of three-dimensional PE-CTA images of 35 different subjects with 8792 slices in total. For each benchmark image, two expert radiologists provided the ground-truth with the assistance of a semi-automated image processing software tool. FUMPE is a challenging benchmark for CAD methods because of the large number (i.e., 3438) of PE regions and, more especially, because of the location of most of them (i.e., 67%) in lung peripheral arteries. Moreover, due to the reporting of the Qanadli score for each PE-CTA image, FUMPE is the first public dataset which can be used for the analysis of mortality and morbidity risks associated with PE. We also report some complementary prognosis information for each subject.
