RESUMO

Aquaporins (AQPs), also known as water channels, appear to be particularly promising in maintaining male reproductive potential. Therefore, this study aimed to determine the presence of classical AQPs in the bovine (Bos taurus) reproductive system and analyze changes in their expression with age using immunohistochemistry and Western blotting. Of the six classical AQPs, AQP0, AQP1, AQP4, AQP5 and AQP6 were detected, while AQP2 was absent. In the testis, AQP0 was visible in Leydig cells in selected animals, while AQP1 was found in myoid cells surrounding the seminiferous tubules of mature individuals. This characteristic expression patterns of AQP0, limited only to certain bulls, is difficult to explain unequivocally. It is possible that AQP0 expression in cattle is subject to individual variability or changes in response to specific physiological conditions. In the caput and corpus epididymis, AQP0 showed weak expression in epithelial cells of immature animals and stronger expression in basal and principal cells of reproductive bulls. In all animals, AQP1 was present on the apical surface of epithelial cells in the initial segment of the caput epididymis. AQP4, AQP5 and AQP6 were identified in principal and basal cells along the entire epididymis of reproductive bulls. The abundance of AQP4 and AQP6 increased from the caput to the cauda epididymis with the growth and development of the animals. In all males, AQP4, AQP5 and AQP6 were observed in epithelial cells of the vas deferens, and their expression in this section increased with age. In conclusion, the abundance and distribution of the classical AQPs in various cell types and parts of the male reproductive system indicate their crucial role in maintaining water homeostasis, which is essential for normal reproductive function in cattle.

The increasing incidence of male infertility in humans and animals creates the need to search for new factors that significantly affect the course of reproductive processes. Therefore, the aim of this study was to determine the temporospatial expression of aquaglyceroporins (AQP3, AQP7 and AQP9) in the bovine (Bos taurus) reproductive system using immunohistochemistry and Western blotting. The study also included morphological analysis and identification of GATA-4. In brief, in immature individuals, AQP3 and AQP7 were found in gonocytes. In reproductive bulls, AQP3 was observed in spermatocytes and spermatogonia, while AQP7 was visible in all germ cells and the Sertoli cells. AQP7 and AQP9 were detected in the Leydig cells. Along the entire epididymis of reproductive bulls, aquaglyceroporins were visible, among others, in basal cells (AQP3 and AQP7), in epididymal sperm (AQP7) and in the stereocilia of the principal cells (AQP9). In males of all ages, aquaglyceroporins were identified in the principal and basal cells of the vas deferens. An increase in the expression of AQP3 in the testis and cauda epididymis and a decrease in the abundance of AQP7 in the vas deferens with age were found. In conclusion, age-related changes in the expression and/or distribution patterns of AQP3, AQP7 and AQP9 indicate the involvement of these proteins in the normal development and course of male reproductive processes in cattle.

The purpose of this study was to analyze the ultrastructure of the testes of sexually immature calves and reproductive bulls of the Polish Holstein-Friesian Black-and-White breed. Utilizing TEM, this study identified three distinct stages of seminiferous tubule development in calves, characterized by varying shapes, distributions, and arrangements of individual cells. In immature animals, early developing spermatocytes, prespermatogonia, and pre-Sertoli cells were observed within the seminiferous tubules. In sexually mature bulls, all cells of the spermatogenic series were observed, situated on a thin, multilayered basal lamina, which forms characteristic undulations. An abundant smooth endoplasmic reticulum was observed in the cytoplasm of spermatogonia in both groups of animals, forming characteristic membranous swirls. In adult bulls, spermatogonia maintain contact with each other through numerous cytoplasmic bridges and cell connections, forming small spaces with visible microvilli between them. The ultrastructural analysis facilitated the identification of morphological changes occurring during the maturation of pre-Sertoli cells, transitioning from a large euchromatic nucleus to a nucleus in which the formation of characteristic vesicles and tubules could be observed. It should also be emphasized that two types of Sertoli cells, namely dark and light electron-dense cells, can be found in cattle. These cells differ from each other, indicating that they may perform different functions. The widespread recognition of the presence of two types of Sertoli cells in cattle will undoubtedly contribute to a better understanding of the processes occurring within the testes and provide a basis for further research in this area.

Aquaporins located in the male reproductive system seem to play a key role in the proper course of many processes occurring within it, thus maintaining a high reproductive potential.

PURPOSE: Chronic pancreatitis (CP) is associated with serious complications and reduced quality of life. Kidney failure is a frequent complication of acute pancreatitis (AP), however limited information is available regarding the impact of CP on this condition. In the kidney, 9 aquaporins (AQPs) are expressed to maintain body water homeostasis and concentrate urine. The purpose of this study was to morphologically assess and analyze the location and expression of AQP2, AQP3 and AQP4 and determine whether CP affects renal structure and expression of AQPs in collecting duct (CD) principal cells. MATERIALS/METHODS: CP was induced in domestic pigs through intramuscular injections of cerulein (1 â€‹µg/kg â€‹bw/day for 6 days; n â€‹= â€‹5); pigs without CP (n â€‹= â€‹5) were used as a control group. Kidney samples were collected 6 weeks after the last injection and subjected to histological examination. Expression of AQPs was determined by immunohistochemistry and Western blot. RESULTS: The kidneys of animals with CP exhibited moderate changes, including glomerular enlargement, increased collagen percentage, numerous stromal erythrorrhages and inflammatory infiltrations compared to control group. Although the total abundance of AQP2 in the CD decreased in pigs after cerulein administration, the difference was not statistically significant. Expression of AQP3 and AQP4 was limited to the basolateral membrane of the CD cells. AQP4 abundance remained relatively stable in both groups, while AQP3 expression increased nearly three-fold in pigs with CP. CONCLUSION: This study identified morphological alterations and a statistically significant increase in the expression of renal AQP3 when pigs developed CP.

The experiment was carried out on 10 clinically healthy Polish-Friesian var. Black-and-White cow calves, during the first seven days of postnatal life. The results indicate that renal removal of potassium depends primarily on the quantity reabsorbed in the tubules, whereas clearance of the electrolyte, due to stable levels in the blood plasma, depends on the amount excreted in the urine. With stable tubular reabsorption of potassium, a relatively unchanging amount of excreted potassium was observed in the urine. However, reduced tubular reabsorption caused a significant increase in excretion and clearance of the electrolyte. Changes in the amount of filtered potassium play a minor role in the regulation of excretion. Small changes in the blood plasma potassium concentration observed primarily resulted from changes in glomerular filtration rate and tubular reabsorption, since the concentration of electrolyte in the blood after birth remained within the physiological range. The results ofthis study suggest that neonate calf kidneys are sufficiently prepared to regulate kalemia. Atrial natriuretic peptide is not directly involved in the regulation of tubular reabsorption of potassium in calves in the first week of life, although it is highly likely that the peptide is involved in the excretion of potassium in the urine in calves during the first seven days of life.

The experiment was carried out on 13 male Polish Black and White dairy calves of 75% share of the Holstein-Friesian (HF) breed during the second week of life. The animals were divided into two groups. One group (n=7) was fed mother's milk and the second (n=6) milk replacer. The dynamics of changes in concentration of selected blood biochemical components connected with nitrogen metabolism (plasma total protein, albumin, urea, endogenous creatinine) and with mineral metabolism (sodium, chloride, calcium, magnesium, zinc, copper and plasma osmotic pressure) were analyzed in both groups. The results show that the type of ingested food influences the concentration of indicators reflecting nitrogen metabolism. Changes of these parameters in calves fed milk replacer are possibly connected with advantageous catabolic changes. Stable concentrations of main extracellular fluid electrolytes and blood plasma osmotic pressure were found in both groups of calves. Constant blood plasma calcium, magnesium, zinc and copper concentrations observed during this study might also indicate the relative maturity of mechanisms maintaining water and electrolyte balance. Nevertheless, it seemsjustifiable to monitor the copper concentrations in plasma of young calves.

Improving the methods utilized to facilitate reproduction is associated with a constant need to search for new factors that not only significantly affect reproductive processes, but also create new possibilities when assessing male reproductive potential. Aquaporins (AQPs) belong to a family of small (28-30 kDa) proteins that facilitate the transport of water and other small molecules. There have been 13 AQPs (AQP0-AQP12) discovered in mammals, and these proteins are present in a wide range of cell types. Almost all AQPs, except AQP6 and AQP12 are present in the male reproductive organs and sperm of mammals and birds. Increasing evidence suggests that these proteins are involved in a number of processes responsible for the optimal functioning of the male reproductive system. This review presents the current state of knowledge regarding the abundance and distribution of AQPs in the male reproductive organs and sperm of various livestock and poultry species, including buffalo, cattle, sheep, horses, pigs, turkeys and goose. Furthermore, the possible physiological and pathophysiological significance of AQPs in male reproduction, as well as hormonal regulation of quantities are discussed. It can be concluded from the studies analyzed in this paper that abundance patterns of AQPs may be considered in the future as specific and universal biomarkers of male fertility and infertility in animal husbandry.

Cancer cell lines are widely used as in vitro models to elucidate biological processes in cancer, and as a tool to evaluate anticancer agents. In fact, the use of an appropriate cancer cell line in cancer research is crucial for investigating new, potential factors involved in carcinogenesis. One of them is aquaporin-3 (AQP3), which is a small, hydrophobic, integral membrane protein with a predominant role in water and glycerol transport. Recently, altered expression of AQP3 has been reported in many types of cancer. Increasing evidence strongly suggests that AQP3 plays a key role in cancer cell proliferation, migration and invasion. In this study, we performed an insightful characteristic of AQP3 location and its protein expression in MCF-7 human breast adenocarcinoma and HepG2 hepatocellular carcinoma cell lines in the context of cancer biology using immunocytochemistry, immunofluorescence and Western blot analyses. AQP3 was found to be located in the cell membrane and cytoplasm of MCF-7 cells, and in the cytoplasm and nuclear membrane of HepG2 cells. Immunoblotting of proteins derived from both cell lines revealed a clear band with a molecular weight of approx. 30 kDa representing an unglycosylated form of AQP3. However, the expression of this protein was higher in MCF-7 than in HepG2. Concluding, our results clearly indicated variability in both the expression levels and subcellular location of the AQP3 protein in MCF-7 and HepG2 cell lines. This leads to the possibility that the expression patterns and subcellular location of AQP3 in the tested cancer cell lines are tissue-of-origin specific, and may be related to the aggressiveness of cancer cells and their mobility.

BACKGROUND: To this day, the effect of multi-drug immunosuppressive protocols on renal expression of AQPs is unknown. This study aimed to determine the influence of rapamycin-based multi-drug immunosuppressive regimens on the expression of aquaporins (AQPs) 1, 2, 3, and 4 in the rat kidney. METHODS: For 6 months, 24 male Wistar rats were administered immunosuppressants, according to the three-drug protocols used in patients after organ transplantation. The rats were divided into four groups: the control group, the TRP group (tacrolimus, rapamycin, prednisone), the CRP group (cyclosporine A, rapamycin, prednisone), and the MRP group (mycophenolate mofetil, rapamycin, prednisone). Selected red cell indices and total calcium were measured in the blood of rats and quantitative analysis of AQP1, AQP2, AQP3 and AQP4 immunoexpression in the kidneys were performed. RESULTS: In the TRP and CRP groups, a mild increase of mean corpuscular hemoglobin concentration, hematocrit and total calcium were observed. Moreover, decreased expression of AQP1-4 was found in all experimental groups, with the highest decrease in the CRP group. CONCLUSIONS: The long-term immunosuppressive treatment using multi-drug protocols decreased AQP1-4 expressions in renal tubules, possibly leading to impaired urine-concentrating ability in rat.

Westernized diet is characterized by a high content of saturated fatty acids (SFA) and a low level of omega-3 polyunsaturated fatty acids (PUFA), often accompanied by an imbalance in the omega-6/omega-3 PUFA ratio. Since increased intake of SFA and n-6 PUFA is considered as a cardiovascular disease risk factor, this study was conducted to determine whether a three-month dietary supplementation of high-fat diets (HFDs) with saturated fatty acids and a significant proportion of various n-6 and n-3 PUFA ratios would affect the architecture and protein expression patterns of the murine heart. Therefore, three HFD (n = 6) feeding groups: rich in SFA, dominated by PUFA with the n-6/n-3-14:1, and n-6/n-3-5:1, ratios were compared to animals fed standard mouse chow. For this purpose, we performed two-dimensional electrophoresis with MALDI-ToF mass spectrometry-based identification of differentially expressed cardiac proteins, and a histological examination of cardiac morphology. The results indicated that mice fed with all HFDs developed signs of hypertrophy and cardiac fibrosis. Animals fed SFA-rich HFD manifested the most severe cardiac hypertrophy and fibrosis lesions, whereas less pronounced changes were observed in the group of animals that ingested the highest amount of omega-3 FA. In general, all HFDs, regardless of FA composition, evoked a comparable pattern of cardiac protein changes and affected the following biological processes: lipid metabolism and FA ß-oxidation, glycolysis, TCA cycle, respiratory chain, myocardium contractility, oxidative stress and PUFA eicosanoid metabolism. However, it should be noted that three proteins, namely IDH3A, LDHB, and AK1, were affected differently by various FA contents. High expression of these myocardial proteins found in the group of animals fed a HFD with the highest n-3 PUFA content could be closely related to the observed development of hypertrophy.

The aim of these studies was to analyse and compare changes in selected parameters of renal function in terms of water-electrolyte balance regulation in single- and twin-pregnant goats. Clearance analyses were carried out on 16 pregnant White Improved goats (8 in single and 8 in twin gestation). Blood plasma and urine samples were analysed for the concentration of inulin, endogenous creatinine, sodium, potassium, and chlorides. It has been demonstrated that glomerular filtration rate (GFR) in the goat kidney does not change significantly during gestation. GFR recorded from the 1st week until the 20th week of gestation in twin-pregnant goats was only slightly higher compared to those observed in single-pregnant does. Blood plasma concentrations of major electrolytes, i.e. sodium, potassium and chloride ions, did not differ significantly in pregnant and non-pregnant goats, and remained within the reference values. From the very beginning of gestation, the single-pregnant goats showed increased renal potassium clearance; however, the level of sodium clearance remained stable. On the other hand, sodium clearance increased from the 2nd month of gestation in the twin-pregnant goats, while the load of excreted potassium did not change. These changes had probably resulted from varied levels of aldosterone and progesterone and their mutual proportions differing between the groups.

The converting enzyme catalyzes the conversion of angiotensin I to angiotensin II. Ang II is the key component of the renin-angiotensin-aldosterone (RAA) system, regulating water-electrolyte balance in newborn calves. Captopril is an inhibitor of the angiotensin-converting enzyme. The aim of this study was to examine the effect of captopril-induced reduction of convertase activity on copper and iron concentrations of blood plasma in calves. The experiment was carried out on 10 Holstein-Friesian female calves, during the first week of life. Copper and iron concentrations in blood plasma were examined before and after captopril administration (0.5, 1, 2, 4, and 6 hours after giving the inhibitor) on subsequent days of the experimental period. The results demonstrated that the copper concentration of blood plasma increased with age. On the seventh day, the copper concentration stabilised at the level observed in adult cattle. Measured before captopril administration, the iron concentration in blood plasma changed: the highest iron concentration was observed on the first day of life, which was followed by a decrease on the third day, and thereafter an increase on the seventh day. These changes may significantly influence the neonatal adaptation of newborn calves, particularly hemopoiesis efficiency. Captopril did not cause statistically significant changes in plasma copper concentration in calves. However, the reduction of angiotensin convertase activity induced by captopril administration resulted in a drop of plasma iron concentration, observed already within 1-2 hours after administration of the inhibitor, and especially within two days post partum. The results indicate that an efficient mechanism maintaining a constant concentration of selected minerals may involve changes in the reabsorption of these minerals from the system fluids to tissues.

Urine is a biological diagnostic material suitable not only for the analysis of kidney and urinary tract functions but also the function of other tissues and organs. The urine proteome of adult mammals differs from the urine proteome of neonatal ones. The establishment of urinary protein maps of healthy newborn calves is important for diagnosing and monitoring the progression of various diseases. The experiment was carried out on a Polish-Friesian var. of Black-and-White male calves in the sixth day of postnatal life. The two proteomics approaches used for separation and identification of urinary proteins were: 2-DE with MALDI-TOF-TOF-MS/MS and 1-DE with LC-MS/MS. This resulted in the identification of 692 urinary proteins. The majority of them were classified as extracellular proteins (40.32%), as well as proteins involved in regulation of major cellular processes (31.07%). We have observed the presence of unique proteins associated with embryonic (ameloblastin, alpha-fetoprotein, Delta-like protein, embryo-specific fibronectin 1 transcript variant, Indian hedgehog homolog) and kidney development (angiotensin-converting enzyme, angiotensinogen, aquaporin-1, calbindin, glypican 3, nidogen 1, pro-cathepsin H). Additionally, proteins involved in the renal regulation of water and electrolyte balance (angiotensinogen, angiotensin-converting enzyme, aquaporin-1, ezrin, uromodulin) were detected. Presented in the current study 1-D and 2-D urinary proteomic maps are the basis for the identification and detection of prognostic biomarkers important for defining a calf's health status.

The aim of this study was the clearance assessment of renal function in single- and twin-pregnant goats and a comparison of selected parameters associated with calcium-phosphate management in terms of litter size. Clearance studies were carried out on 16 pregnant Polish White Improved goats (8 single pregnancies and 8 twin pregnancies). It was demonstrated that the kidneys of pregnant goats regulate the calcium-phosphate balance to a great extent. In spite of observed differences in parameters of renal functions, plasma calcium and inorganic phosphorus concentrations were comparable with normal reference values in further weeks of pregnancy. Significant differences in renal function between single and twin pregnancies were not observed. Gut absorption and/or bone tissue metabolism seem important for the maintenance of appropriate calcium-phosphate status.

To date, 13 aquaporin isoforms (AQPs) have been discovered in mammals, of which as many as 9 are located in epithelial cells lining the individual sections of the nephron and collecting tubules. Detailed analysis of the location and expression of AQPs in the kidneys of laboratory animals and humans allowed to define the key role of these proteins in renal excretion of water and other small molecules. Unfortunately, despite the significant advances in knowledge in this area, still little is known about this subject in livestock, including cattle. Therefore, the aim of the study was to determine the expression and AQPs location in the nephron segment in the bovine kidney by immunohistochemistry and Western blot. The distribution of a total of 8 aquaporins was determined as a result of the conducted experiments. The results obtained in the present study clearly indicate that aquaporins in cattle are involved in the renal regulation of water excretion and maintenance of proper acid-base balance. Undoubtedly, changes in the distribution and expression of AQPs in bovine kidneys may be the cause of water balance disorders and disruption of the normal body fluid composition. Kidney diseases in cattle are poorly described in veterinary medicine. Knowledge of cellular location and expression of all AQPs in the bovine kidney under normal physiological condition allows a deeper understanding of the renal regulation of body homeostasis. It creates new perspective for diagnosis and pharmacotherapy in cattle in the future.

The aim of this study was to determine the influence of different combinations of immunosuppressive drugs on the morphology, ultrastructure, and expression of proliferating cell nuclear antigen and cytoskeleton proteins in the rat dorsolateral prostate. The studies were conducted on 48 male Wistar rats. The animals were divided into eight groups: a control group and seven experimental groups. For 6 months, the animals in the experimental groups were administered a combination of drugs including rapamycin (Rapa), cyclosporin A, tacrolimus (Tac), mycophenolate mofetil, and prednisone (Pred), according to the standard three-drug regimens for immunosuppressive therapy used in clinical practice. An evaluation of the morphology and ultrastructure was conducted, and a quantitative evaluation of the expression of proliferating cell nuclear antigen and desmin- and cytokeratin-positive cells with weak, moderate, and strong expression was performed. The combination of Rapa, Tac, and Pred caused the smallest morphological and ultrastructural changes in the rat prostate cells. In the case of rats whose treatment was switched to Rapa monotherapy, a decreased percentage of proliferating cells of both the glandular epithelium and the stroma was found. Decreases in body weight and changes in the expression of cytokeratin and desmin were observed in all the experimental rats. The combination of Rapa, Tac, and Pred would seem to be the most beneficial for patients who do not suffer from prostate diseases. Our results justify the use of inhibitors of the mammalian target of Rapa in the treatment of patients with prostate cancer. The changes in the expression of cytoskeleton proteins may be the result of direct adverse effects of the immunosuppressive drugs, which are studied in this article, on the structure and organization of intermediate filament proteins.
