RESUMO

Different biomarkers are used to evaluate the severity of atopic dermatitis (AD); however, it remains difficult to determine the severity of localized skin lesions. MIF plays an essential role in the pathophysiology of skin inflammation. To establish whether the MIF level in the stratum corneum (SC) serves as a marker of the severity of AD lesions, we examined the SC MIF (scMIF) levels in AD patients. The SC of the cheek, neck and upper arm skin was collected using tape stripping, and the scMIF levels were measured. Consequently, the scMIF levels were found to be significantly higher in the involved skin lesions than the uninvolved areas within the same patient. Moreover, the scMIF levels were significantly correlated with the severity of local skin lesions. These findings suggest that the scMIF level can be used as an effective marker for evaluating the local severity of AD.

RESUMO

The effect of OH-groups on the surface of a Ni catalyst for low-temperature (473 K) steam reforming of methane in an electric field (EF) was investigated. Ni-doped YSZ (Zr0.65Y0.05Ni0.3O2) was chosen as a highly active catalyst for this purpose. The effects on catalyst activity of adding hydrogen and steam in the pre-treatment were assessed with and without EF. When an EF was applied, activity increased irrespective of the electronic state of Ni, whereas the metallic Ni state was necessary for activity without EF. Furthermore, the highest activity with EF was observed for the pre-treatment with a mixture of H2 and H2O. Investigation of the superiority using XPS measurements showed an increase in the amount of Ni(OH)2, OH groups and H2O near the surface after the activity test, which are regarded as the reaction sites with EF. This finding suggests that a pre-treatment with steam increases the surface OH groups and Ni(OH)2 on the Ni catalyst, and enhances surface proton conduction, thereby improving the activity.

RESUMO

When used in closed-type plant factories, light-emitting diode (LED) illumination systems have the particular advantages of low heat emission and high luminous efficiency. The effects of illumination quality and intensity on the growth and morphogenesis of many plant species have been examined, but improvements are needed to optimize the illumination systems for better plant products with lower resource investments. In particular, new strategies are needed to reduce the wastage of plant products related to leaf senescence, and to better control the ingredients and appearance of leafy vegetables. Although the quality of light is often altered to change the characteristics of plant products, the transcriptional status underlying the physiological responses of plants to light has not been established. Herein, we performed a comprehensive gene expression analysis using RNA-sequencing to determine how red, blue, and red/blue LEDs and fluorescent light sources affect transcriptome involved in the leaf aging of leaf lettuce. The RNA-sequencing profiling revealed clear differences in the transcriptome between young and old leaves. Red LED light caused large variation between the two age classes, while a pure or mixed blue LED light spectrum induced fewer transcriptome differences between young and old leaves. Collectively, the expression levels of genes that showed homology with those of other model organisms provide a detailed physiological overview, incorporating such characteristics as the senescence, nutrient deficiency, and anthocyanin synthesis of the leaf lettuce plants. Our findings suggest that transcriptome profiles of leaf lettuce grown under different light sources provide helpful information to achieve better growth conditions for marketable and efficient green-vegetable production, with improved wastage control and efficient nutrient inputs.

RESUMO

INTRODUCTION: Myositis ossificans is a benign, self-limiting, tumor-like lesion that usually affects the elbow and thigh; occurrence in the hand is uncommon. We report a rare case of a patient with myositis ossificans in the thenar region. CASE PRESENTATION: A 15-year-old Japanese girl presented to our hospital with a 2-month history of a painful mass in the right thenar region without previous trauma. The clinical and radiological examination findings suggested an osteoblastic malignancy. A diagnosis of myositis ossificans was made on the basis of an incisional biopsy. Despite the location of the lesion in the thenar region, a normal functional outcome was achieved after marginal resection of the mature lesion. CONCLUSIONS: Clinicians should consider myositis ossificans as a possible diagnosis for a soft tissue mass in the hand, thereby avoiding unnecessarily aggressive therapy.

RESUMO

INTRODUCTION: Patients with rheumatoid arthritis (RA) treated with abatacept (ABT) are at increased risk for vaccine-preventable infections. The aim of the present study is to evaluate the humoral response to 23-valent pneumococcal polysaccharide (PPSV23) vaccination in RA patients receiving ABT. METHODS: The immunogenicity study was nested within a randomized, double-blind placebo-controlled study, designed to evaluate the efficacy of the PPSV23. PPSV23 was given to 111 RA patients, who were classified into three groups: RA control (n = 35), methotrexate (MTX) alone (n = 55), and ABT (n = 21). Before and 4-6 weeks after vaccination, we measured the patients' concentrations of antibodies against pneumococcal serotypes 6B and 23F using an enzyme-linked immunosorbent assay and determined their antibody functionality using a multiplexed opsonophagocytic killing assay, reported as the opsonization index (OI). RESULTS: The pneumococcal serotype-specific IgG concentrations and OIs were both significantly increased in all treatment groups in response to PPSV23 vaccination. In the ABT group, the IgG responses for the 6B serotype were lower compared with those in the MTX alone or control groups, whereas the OI responses were similar to those in the other two groups. In a subgroup analysis, the pneumococcal serotype-specific IgG responses were significantly lower in both serotypes (6B and 23F) in the ABT/MTX group; however, the OI responses in the ABT group were not different from the control group. There was no association between the pneumococcal serotype-specific IgG and OI responses for the 6B serotype in patients receiving ABT in contrast to the control or MTX alone patients. No severe adverse effects were observed in any of the treatment groups. CONCLUSIONS: OI responses indicate antibody functionality rather than simply their amount, so the similarity of these measurements between all three groups suggests that RA patients receiving ABT still benefit from receiving the PPSV23 vaccination, even though they produce less IgG in response to it. The results suggest an influence of ABT on the humoral response to PPSV23 vaccination under MTX treatment; however, preserved opsonin responses are expected in RA patients treated with ABT plus MTX. RESUMO

Vaccination against Streptococcus pneumoniae is recommended for rheumatoid arthritis (RA) patients receiving immunosuppressive treatments. The objective of this study was to evaluate the humoral response to 23-valent pneumococcal polysaccharide vaccination (PPSV23) in RA patients receiving methotrexate (MTX) alone or in combination with a tumor necrosis factor inhibitor, golimumab (GOM).PPSV23 was given to 114 RA patients, who were classified into three groups: RA control (n = 35), MTX alone (n = 55), and GOM + MTX (n = 24). Before and 4 to 6 weeks after vaccination, concentrations of antibodies against pneumococcal serotypes 6B and 23F were measured using an enzyme-linked immunosorbent assay and antibody functionality was determined using a multiplexed opsonophagocytic killing assay, reported as the opsonization index (OI).The IgG concentrations and OIs were both significantly increased in all treatment groups in response to PPSV23 vaccination. In the GOM + MTX group, the IgG responses were lower than those in the MTX alone or control groups, whereas the OI responses were similar to those in the other 2 groups. Furthermore, discrepancies between the IgG and OI responses were found in GOM + MTX group. No severe adverse effect was observed in any treatment groups.OI responses indicate that antibody functionality rather than antibody quantity is important. The similarity of these measurements between all 3 groups suggests that RA patients receiving MTX + GOM still benefit from receiving the PPSV23 vaccination, even though they produce less IgG in response to it. These results can help clinicians to better schedule and evaluate pneumococcal vaccination for RA patients.

RESUMO

The current report presents the case of a 41-year-old male exhibiting a giant cell tumor of the tendon sheath (GCT-TS) arising from the patellar tendon sheath. Plain radiography and magnetic resonance imaging revealed a well-localized mass that wrapped around the patellar tendon, and extended from the subcutis into the infrapatellar fat pad and tibia. Following histopathological determination of the diagnosis, a piecemeal resection was performed. Nodular-type GCT-TS occurs less frequently in large joints compared with the small joints of the fingers and toes. The current report presents the unique features of a case of GCT-TS extending around the patellar tendon, and invading into the knee joint and proximal tibia bone.

RESUMO

INTRODUCTION: It is difficult to make a definite diagnosis of a cracked tooth solely based on an inspection within the root canal, especially in case of microcracks. At present, there seems to be no established method to detect dentinal microcracks in roots; therefore, the current detection techniques need to be improved. Vibrothermography (VibroIR) helps to detect microcracks by the friction heat generated from ultrasonic vibration. The purpose of this study was to establish a novel method using VibroIR to detect dentinal microcracks. METHODS: The root canals of 20 roots with cracks and control roots were prepared after removing the tooth crowns. A tapered indenter was inserted into the root canal and pressed until a microcrack was created under an optical microscope. Using VibroIR, the detection trials for dentinal microcracks were performed with an ultrasonic vibration power ranging from 0.43 to 1.48 W at an angle of 0°, 30°, 45°, 60°, and 90° between the ultrasonic vibration point and the microcrack line. After the detection test, the microcrack width was measured with an optical microscope. RESULTS: Frictional heat was detected in the microcracks with thermography at 0.89 to 1.48 W and at an ultrasonic vibration point angle less than 60° from the crack line for 10 seconds. Microcracks with a width of 4 to 35.5 µm were detected with this method. CONCLUSIONS: VibroIR may be an effective method for the diagnosis of root dentinal microcracks.
