Exposure to high level of arsenic (As) through the ingestion of contaminated soil, dust and food plants can pose health risk to humans. This study investigates the total arsenic (As), arsenobetaine (AsB), monomethylarsenate (MMA), dimethylarsenate (DMA), arsenite (As3+) and arsenate (As5+) concentrations in poultry feed, manure, agricultural soils and food plants collected from Khyber Pakhtunkhwa Province, Pakistan. The total mean As concentrations in the edible parts of food plants ranged from 0.096 mg kg-1 to 1.25 mg kg-1 with percentile (P) values (P25-0.039, P50-0.0765, P75-0.165 1 mg kg-1 to P25-0.95, P50-1.23, P75-1.6 1 mg kg-1) and exceeded the food safety limit (0.1 mg kg-1) of Food & Agriculture Organization (FAO) and World Health Organization (WHO) in all plant species except Pisum sativum (pea) and Mentha arvensis (mint). The risk to human health was assessed through the average daily intake (ADI), hazards quotient (HQ), health risk index (HRI) and lifetime cancer risk (LTCR). The highest average daily intake of As via the ingestion of Malva neglecta (mallow, a leafy plant) was observed for adults and children. The ADI for adults and children (2.36 × 10-4 mg kg-1 day-1 and 6.33 × 10-4 mg kg-1 day-1) was about 13% and 5%, respectively, of the Bench Mark Dose Limit (BMDL0.5) of 3.00 × 10-3 mg kg-1 day-1 set by WHO. The HRI was 3 times more in the children (2.1) than the adults (0.79), posing non-cancer health risks (health risk index > 1) for children. The LTCR values were slightly higher (1.53 × 10-4) relative to USEPA and WHO limits (1 × 10-6 to 1 × 10-4) for children whereas a minimal cancer risk was observed for adults via consumption of selected food plants. The results showed that poultry manure can contaminate food plants that may lead to cancer and non-cancer risks in agricultural areas, Pakistan. Thus, it is important to minimize As concentration in poultry feed to safeguard human health and environment from adverse effects.

Cognitive decline in dementia is associated with deficiency of the cholinergic system. In this study, five mono-carbonyl curcumin analogs were synthesized, and on the basis of their promising in vitro anticholinesterase activities, they were further investigated for in vivo neuroprotective and memory enhancing effects in scopolamine-induced amnesia using elevated plus maze (EPM) and novel object recognition (NOR) behavioral mice models. The effects of the synthesized compounds on the cholinergic system involvement in the brain hippocampus and their binding mode in the active site of cholinesterases were also determined. Compound h2 (p < 0.001) and h3 (p < 0.001) significantly inhibited the cholinesterases and reversed the effects of scopolamine by significantly reducing TLT (p < 0.001) in EPM, while (p < 0.001) increased the time exploring the novel object. The % discrimination index (DI) was significantly increased (p < 0.001) in the novel object recognition test. The mechanism of cholinesterase inhibition was further validated through molecular docking study using MOE software. The results obtained from the in vitro, in vivo and ex vivo studies showed that the synthesized curcumin analogs exhibited significantly higher memory-enhancing potential, and h3 could be an effective neuroprotective agent. However, more study is suggested to explore its exact mechanism of action.

The antibiotics released by human and animals end up in the environmental sinks like soil and water to cause contamination and induce resistance in the microflora. The knowledge of fate and behavior of antibiotics in diverse geographical, climatological, and physicochemical is limited. Therefore, present study investigated the spatial distribution of antibiotics and their relationship with various factors and the source-sink relationship between soil and poultry manure. This was achieved by employing spatially constrained hierarchical clustering, global and local spatial autocorrelation, and spatial regression techniques. Most of the antibiotics co-occurred in both soil and poultry manure matrices, however antibiotic concentration in soil (1.20 µg kg-1 < antibiotics ≤21.38 µg kg-1) was lower than that in the poultry manure (7.05 µg kg-1< antibiotics ≤60.2 µg kg-1). Majority of the antibiotics showed spatial independence in both poultry manure and soil, except for sulfadiazine, sulfanilamide and sulfapyridine with Moran's I > - 0.111. Local indicator of spatial association indicated localized spatial clustering and outlier behavior of antibiotics. The underlying reasons for spatial heterogeneity of antibiotics resolved by spatial regression models indicated elevation, S%, C%, pH and mean annual temperature as the major factors. The influence of antibiotic concentration in poultry manure as a source was significant but marginal compared to the other predictors of spatial heterogeneity.

The study aimed to assess the physicochemical parameters (pH, electrical conductivity (EC), total dissolve solid (TDS), oxidation reduction potential (ORP), Temperature) and potential toxic metals (PTMs), including Ni, Mn, Cr, Cu, Cd, Pb, Co, Fe and Zn in the groundwater of Lower Dir, Pakistan. Furthermore, the pollution sources and spatial distribution pattern of PTMs were also investigated via principal component analysis (PCA) and geographic information system (GIS) application to understand the changing behaviors of PTMs in groundwater. The average concentrations of physicochemical parameters such as pH, EC, TDS, ORP and Temperature were 7.1, 418⯵S/cm, 251â¯mg/L, 193â¯mV and 25.7 âC, while the concentrations of PTMs; Ni, Mn, Cr, Cu, Cd, Pb, Co, Fe and Zn were 0.25, 0.34, 0.09, 0.29, 0.10, 0.08, 0.10, 0.83 and 0.25 â¯mg/L, respectively. Among the selected metals, Mn, Cr, Cd, Pb, Co and Fe were exceeded the WHO guidelines and their percentage contribution were 43%, 57%, 45%, 70%, 70% and 62%, respectively. The increasing order of PTMs were; Pbâ¯>â¯Coâ¯>â¯Feâ¯>â¯Crâ¯>â¯Cdâ¯>â¯Mnâ¯>â¯Cuâ¯>â¯Niâ¯>â¯Zn in the study area. PCA represented three significant factors, which explained 76% variability in the groundwater. Whereas, clustering analysis (CA) grouped groundwater into three distinct clusters less polluted (C1), moderate polluted (C2) and highly polluted (C3). Human health risk assessment was carried out to check the suitability of groundwater for drinking and domestic uses. The HQ and HRIs values of Cd were >1, suggested that the groundwater sources are unfit for drinking and domestic purposes and may be caused potential health risk after long term ingestion.
