Rhinoviruses (RV) are the major cause of chronic obstructive pulmonary disease and are associated with exacerbation development as well as community-acquired pneumonia in children, leading to substantial morbidity, mortality, and hospital admission. Here we have examined how changes at the amino terminal of the conserved VP4 epitope of different RV serotypes may affect pulmonary cytokine and chemokine responses and disease severity. Samples positive for rhinovirus were used for genetic characterization, followed by profiling gene expression of pulmonary Th1 and Th2 cytokines/chemokines by RT-PCR arrays. Genetic sequencing and homology 3D modeling revealed changes at the amino terminal of the conserved viral protein 4 (VP4) epitope in the RV-A101 serotype, especially serine at several positions that are important for interactive binding with the host immune cells. We found dysregulation of pulmonary gene expression of Th1- and Th2-related cytokines and chemokines in RV-A 101 and RV-C 8 pneumonia patients. These findings might contribute to a better understanding of RV immunity and the potential mechanisms underlying the pathogenesis of severe RV infections, but further functional studies are needed to confirm the causal relationship.

Cadmium (Cd) is getting worldwide attention due to its continuous accumulation in agricultural soils which is due to anthropogenic activities and finally Cd enters in food chain mainly through edible plants. Cadmium free food production on contaminated soils is great challenge which requires some innovative measures for crop production on such soils. The current study evaluated the efficiency of zinc oxide nanoparticles (ZnONPs) (0, 150 and 300 mg/kg) on the growth of wheat in texturally different soils including clay loam (CL), sandy clay loam (SCL), and sandy loam (SL) which were contaminated with were contaminated with 25 mg/kg of Cd before crop growth. Results depicted that doses of ZnONPs and soil textures significantly affected the biological yields, Zn and Cd uptake in wheat plants. The application of 300 mg/kg ZnONPs caused maximum increase in dry weights of shoot (66.6%), roots (58.5%), husk (137.8%) and grains (137.8%) in CL soil. The AB-DTPA extractable Zn was increased while Cd was decreased with doses of NPs depending upon soil textures. The maximum decrease in AB-DTPA extractable Cd was recorded in 300 mg/kg of ZnONPs treatment which was 58.7% in CL, 33.2% in SCL and 12.1% in SL soil as compared to respective controls. Minimum Cd concentrations in roots, shoots, husk and grain were found in 300 mg/kg ZnONPs amended CL soil which was 58%, 76.7%, 58%, and 82.6%, respectively. The minimum bioaccumulation factor (0.14), translocation index (2.46) and health risk index (0.05) was found in CL soil with the highest dose of NPs. The results concluded that use of ZnONPs significantly decreased Cd concentration while increased Zn concentrations in plants depending upon doses of NPs and soil textures.

The complement system, a network of highly-regulated proteins, represents a vital part of the innate immune response. Over-activation of the complement system plays an important role in inflammation, tissue damage, and infectious disease severity. The prevalence of MERS-CoV in Saudi Arabia remains significant and cases are still being reported. The role of complement in Middle East Respiratory Syndrome coronavirus (MERS-CoV) pathogenesis and complement-modulating treatment strategies has received limited attention, and studies involving MERS-CoV-infected patients have not been reported. This study offers the first insight into the pulmonary expression profile including seven complement proteins, complement regulatory factors, IL-8, and RANTES in MERS-CoV infected patients without underlying chronic medical conditions. Our results significantly indicate high expression levels of complement anaphylatoxins (C3a and C5a), IL-8, and RANTES in the lungs of MERS-CoV-infected patients. The upregulation of lung complement anaphylatoxins, C5a, and C3a was positively correlated with IL-8, RANTES, and the fatality rate. Our results also showed upregulation of the positive regulatory complement factor P, suggesting positive regulation of the complement during MERS-CoV infection. High levels of lung C5a, C3a, factor P, IL-8, and RANTES may contribute to the immunopathology, disease severity, ARDS development, and a higher fatality rate in MERS-CoV-infected patients. These findings highlight the potential prognostic utility of C5a, C3a, IL-8, and RANTES as biomarkers for MERS-CoV disease severity and mortality. To further explore the prediction of functional partners (proteins) of highly expressed proteins (C5a, C3a, factor P, IL-8, and RANTES), the computational protein-protein interaction (PPI) network was constructed, and six proteins (hub nodes) were identified.

BACKGROUND: In COVID-19 patients, undetected co-infections may have severe clinical implications associated with increased hospitalization, varied treatment approaches and mortality. Therefore, we investigated the implications of viral and bacterial co-infection in COVID-19 clinical outcomes. METHODS: Nasopharyngeal samples were obtained from 48 COVID-19 patients (29% ICU and 71% non-ICU) and screened for the presence of 24 respiratory pathogens using six multiplex PCR panels. RESULTS: We found evidence of co-infection in 34 COVID-19 patients (71%). Influenza A H1N1 (n = 17), Chlamydia pneumoniae (n = 13) and human adenovirus (n = 10) were the most commonly detected pathogens. Viral co-infection was associated with increased ICU admission (r = 0.1) and higher mortality (OR 1.78, CI = 0.38-8.28) compared to bacterial co-infections (OR 0.44, CI = 0.08-2.45). Two thirds of COVID-19 critically ill patients who died, had a co-infection; and Influenza A H1N1 was the only pathogen for which a direct relationship with mortality was seen (r = 0.2). CONCLUSIONS: Our study highlights the importance of screening for co-infecting viruses in COVID-19 patients, that could be the leading cause of disease severity and death. Given the high prevalence of Influenza co-infection in our study, increased coverage of flu vaccination is encouraged to mitigate the transmission of influenza virus during the on-going COVID-19 pandemic and reduce the risk of severe outcome and mortality.

Abscisic acid (ABA) priming is known to enhance plant growth and survival under salinity. However, the mechanisms mediating this long-term acclimatization to salt stress are still obscure. Specifically, the long-term transcriptional changes and their effects on ion relations were never investigated. This motivated us to study the long-term (8 days) effect of one-time 24 h root priming treatment with 10 µM ABA on transcription levels of relevant regulated key genes, osmotically relevant metabolites, and ionic concentrations in Vicia faba grown under 50 mM NaCl salinity. The novelty of this study is that we could demonstrate long-term effects of a one-time ABA application. ABA-priming was found to prevent the salt-induced decline in root and shoot dry matter, improved photosynthesis, and inhibited terminal wilting of plants. It substantially increased the mRNA level of AAPK and 14-3-3 ABA inducible kinases and ion transporters (PM H+ -ATPase, VFK1, KUP7, SOS1, and CLC1). These ABA-induced transcriptional changes went along with altered tissue ion patterns. Primed plants accumulated less Na+ and Cl- but more K+ , Ca2+ , Zn2+ , Fe2+ , Mn2+ , NO3 - , and SO4 2- . Priming changed the composition pattern of organic osmolytes under salinity, with glucose and fructose being dominant in unprimed, whereas sucrose was dominant in the primed plants. We conclude that one-time ABA priming mitigates salt stress in Vicia faba by persistently changing transcription patterns of key genes, stabilizing the ionic and osmotic balance, and improving photosynthesis and growth.

Antigenic drift of the hemagglutinin (HA) and neuraminidase (NA) proteins of the influenza virus cause a decrease in vaccine efficacy. Since the information about the evolution of these viruses in Saudi is deficient so we investigated the genetic diversity of circulating H1N1 viruses. Nasopharyngeal aspirates/swabs collected from 149 patients hospitalized with flu-like symptoms during 2014 and 2015 were analyzed. Viral RNA extraction was followed by a reverse transcription-polymerase chain reaction and genetic sequencing. We analyzed complete gene sequences of HA and NA from 80 positive isolates. Phylogenetic analysis of HA and NA genes of 80 isolates showed similar topologies and co-circulation of clades 6b. Genetic diversity was observed among circulating viruses belonging to clade 6B.1A. The amino acid residues in the HA epitope domain were under purifying selection. Amino acid changes at key antigenic sites, such as position S101N, S179N (antigenic site-Sa), I233T (antigenic site-Sb) in the head domain might have resulted in antigenic drift and emergence of variant viruses. For NA protein, 36% isolates showed the presence of amino acid changes such as V13I (n = 29), I314M (n = 29) and 12% had I34V (n = 10). However, H257Y mutation responsible for resistance to neuraminidase inhibitors was missing. The presence of amino acid changes at key antigenic sites and their topologies with structural mapping of residues under purifying selection highlights the importance of antigenic drift and warrants further characterization of recently circulating viruses in view of vaccine effectiveness. The co-circulation of several clades and the predominance of clade 6B.1 suggest multiple introductions in Saudi.

MERS-CoV, a highly pathogenic virus in humans, is associated with high morbidity and case fatality. Inflammatory responses have a significant impact on MERS-CoV pathogenesis and disease outcome. However, CD4+ T-cell induced immune responses during acute MERS-CoV infection are barely detectable, with potent inhibition of effector T cells and downregulation of antigen presentation. The local pulmonary immune response, particularly the Th1 and Th2-related immune response during acute severe MERS-CoV infection is not fully understood. In this study, we offer the first insights into the pulmonary gene expression profile of Th1 and Th2-related cytokines/chemokines (Th1 & Th2 responses) during acute MERS-CoV infection using RT2 Profiler PCR Arrays. We also quantified the expression level of primary inflammatory cytokines/chemokines. Our results showed a downregulation of Th2, inadequate (partial) Th1 immune response and high expression levels of inflammatory cytokines IL-1α and IL-1ß and the neutrophil chemoattractant chemokine IL-8 (CXCL8) in the lower respiratory tract of MERS-CoV infected patients. Moreover, we identified a high viral load in all included patients. We also observed a correlation between inflammatory cytokines, Th1, and Th2 downregulation and the case fatality rate. Th1 and Th2 response downregulation, high expression of inflammatory cytokines, and high viral load may contribute to lung inflammation, severe infection, the evolution of pneumonia and ARDS, and a higher case fatality rate. Further study of the molecular mechanisms underlying the Th1 and Th2 regulatory pathways will be vital for active vaccine development and the identification of novel therapeutic strategies.

Phytoremediation of heavy metal contaminated soils represents a promising technique and salt-tolerant hyperaccumulators for multiple metals are the need of time. Therefore, phytoremediation potential of four salt-tolerant grass species [Dhab (Desmostachya bipinnata), Kallar (Leptochloa fusca), Para (Brachiaria mutica) and Sporobolus (Sporobolus arabicus Boiss)] was evaluated for cadmium (Cd) and lead (Pb) in a hydroponic study. The plants were harvested after a growth period of 3 months in a nutrient solution containing different levels of Cd (0, 5, and 25 mg L-1) and Pb (0, 25, and 125 mg L-1). Results indicated that Dhab grass showed the highest root and shoot dry matter yield followed by Para, Kallar and Sporobolus grass irrespective of metal or its level under which they were grown. All the grass species showed considerable Cd-accumulating potential with an accumulation of >150 mg kg-1of shoot dry matter at a higher level of Cd-contamination (25 mg L-1). While in case of shoot Pb-accumulation only Para grass performed well and accumulated Pb >1000 mg kg-1 of shoot dry matter at the higher level of Pb-contamination (125 mg L-1). Moreover, Para and Dhab grasses performed better for shoot Cd-uptake, while only Para grass showed promising shoot Pb uptake potential. In conclusion, these grass species could be penitentially used for phytoremediation of salt-affected Cd and Pb contaminated soils.

BACKGROUND: The negative effect of soil-applied phosphorus (P) on zinc (Zn) uptake by plants and its concentration in food grains could be a possible reason for low dietary intake of Zn. Likewise, owing to its critical role in plant tolerance to other abiotic stresses, potassium (K) was thought to retrieve P-induced decrease in grain Zn concentration of wheat. To test the above hypothesis, the effect of K application (50 mg kg-1 soil) on Zn concentration in shoot/grains and its shoot-to-grain translocation was studied in wheat (Triticum aestivum L. cv. Galaxy-2013) under low and optimal supply of both P (10 and 50 mg kg-1 ) and Zn (0 and 5 mg kg-1 ). RESULTS: The response of growth parameters and grain yield to optimal Zn, P and K applications indicated that all nutrients were limiting plant growth at low levels. Irrespective of P level, Zn application at optimal rate increased Zn concentration and uptake by straw and grains of wheat. Contrarily, optimal P application decreased Zn concentration but increased Zn uptake by wheat straw. More specifically, combined application of Zn and P at optimal levels decreased Zn concentration in grains from 43 to 32 mg kg-1 compared with optimal Zn application alone. Potassium application to optimal P- and Zn-supplied plants increased remobilization of pre-anthesis straw Zn store to grains by 50% and decreased Zn concentration in straw. Consequently, K application along with optimal Zn and P supply to plants completely retrieved P-induced loss in grain Zn concentration and also increased grain Zn uptake from 891 to 1249 µg per pot without significantly affecting grain yield. CONCLUSION: The K-induced increase in grain Zn concentration is attributed to K-driven higher post-anthesis Zn uptake and remobilization of pre-anthesis straw Zn store to grains. © 2018 Society of Chemical Industry.

Nickel (Ni) toxicity in agricultural crops is a widespread problem while little is known about the role of biochar (BC) and other organic amendments like farm manure (FM) from cattle farm and compost (Cmp) on its alleviation. A greenhouse experiment was conducted to evaluate the effects of BC, Cmp and FM on physiological and biochemical characteristics of maize (Zea mays L.) under Ni stress. Maize was grown in Ni spiked soil without and with two rates of the amendments (equivalent to 1% and 2% organic carbon, OC) applied separately to the soil. After harvest, plant height, root length, dry weight, chlorophyll contents, gas exchange characteristics and trace elements in plants were determined. In addition, post-harvest soil characteristics like pHs, ECe and bioavailable Ni were also determined. Compared to the control, all of the amendments increased plant height, root length, shoot and root dry weight with the maximum increase in all parameters by FM (2% OC) treatment. Similarly, total chlorophyll contents and gas exchange characteristics significantly increased with the application of amendments being maximum with FM (2% OC) application. Amendments significantly increased copper, zinc, manganese and iron concentrations and decreased Ni concentrations in the plants. The highest reduction in shoot Ni concentration was recorded with FM (2% OC) followed by BC (2% OC) being 73.2% and 61.1% lower compared to the control, respectively. The maximum increase in soil pH and decrease in AB-DTPA extractable Ni was recorded with BC (2% OC) followed by FM (2% OC). It is concluded that FM (2% OC) was the most effective in reducing Ni toxicity to plants by reducing Ni uptake while BC (2% OC) was the most effective in decreasing bioavailable Ni in the soil through increasing soil pH. However, long-term field studies are needed to evaluate the effects of these amendments in reducing Ni toxicity in plants.

Little is known about the effect of elemental sulfur on lead uptake and its toxicity in wheat. A pot experiment was conducted with the purpose to examine the impact of sulfur on improving Pb solubility in soil, and uptake and accumulation in wheat plants. The effect of three levels of lead (0, 50, and 100 mg/kg soil) and sulfur (0, 150, and 300 mmol/kg soil) was tested in all possible combinations. Root dry matter, straw, and grain yields, and the photosynthetic and transpiration rates decreased significantly with increase in the concentration of Pb in the soil. However, sulfur fertilization in the presence of Pb improved the photosynthetic and transpiration rates and consequently increased the straw and grain yields of wheat. It also enhanced Pb accumulation in roots, its translocation from roots to shoot, and accumulation in grain. S and Zn contents of different plant parts were also enhanced. Thus, by mitigating the toxic effect of Pb and improving wheat growth, sulfur enhances Pb accumulation by the aboveground plant parts and hence the phytoextraction capacity of wheat. However, total accumulation of Pb shows that wheat plant cannot be considered as a suitable candidate for phytoremediation.

BACKGROUND: Low phosphorus (P) efficiency from existing granular fertilisers necessitates searching for efficient alternatives to improve wheat productivity in calcareous soil. RESULTS: Multi-location trials have shown that phosphoric acid (PA) produced 16% higher wheat grain over commercial P fertilisers, i.e. diammonium phosphate (DAP) and triple superphosphate (TSP). Methods of P application significantly influenced grain yield and the efficiency of methods was observed in the order: PA placement below seed > PA, DAP or TSP fertigation > DAP or TSP broadcast. The sub-surface application of PA produced highest grain yields (mean of all rates), i.e. 4669, 4158 and 3910 kg ha(-1) in Bagh, Bhalwal and Shahpur soil series, respectively. Phosphoric acid at 66 kg P2 O5 ha(-1) was found more effective in increasing gain yield over that of control. Trend in grain P uptake was found similar to that observed for grain yield. Maximum P uptake by grain was recorded at the highest P rate and the lowest at zero P. The significant increase in P uptake with P rates was generally related to the increase in yield rather than its concentration in grain. Phosphorus agronomic efficiency (PAE) and phosphorus recovery efficiency (PRE) were found higher at lower P rate (44 kg P2 O5 ha(-1) ) and decreased with P application. However, PA applied by the either method resulted in higher PAE and PRE compared to DAP and TSP. CONCLUSION: Phosphoric acid is suggested as an efficient alternative to commercial granular P fertilisers for wheat production in alkaline calcareous soils. © 2015 Society of Chemical Industry.

BACKGROUND: Cadmium concentration in food grains could be minimised through application of beneficial plant nutrients such as silicon. Therefore, the impact of silicon application on immobilisation of Cd in soil and its concentration in low and high shoot-Cd (LSCd and HSCd, respectively) cultivars of wheat were evaluated in a pot experiment. Selected LSCd cultivars (Iqbal-2000 and Lasani-2008) and HSCd cultivars (Inqlab-91 and Sehar-2006) were grown on artificially Cd contaminated soil at 10 mg Cd kg(-1) . Three levels of Si (50, 100 and 150 mg kg(-1) soil), applied as calcium silicate (CaSiO3 ), were tested. RESULTS: None of the wheat cultivars showed any symptoms of toxicity or growth retardation against applied Cd stress. Silicon applied to Cd-treated plants did not improve root and shoot dry matter; however, it increased grain yield significantly at the highest rate of application (150 mg kg(-1) soil). Similarly, Si application at 150 mg kg(-1) decreased plant available soil Cd without affecting soil pH. Silicon application not only caused a linear decrease in Cd contents of shoots and grains but also decreased its translocation from roots to shoots and grains. Decrease in shoot Cd concentration was higher in HSCd than LSCd cultivars whereas the reverse was true for Cd concentration in grains. CONCLUSION: Si addition decreased Cd concentration in wheat cultivars by causing a decrease in both plant-available soil Cd and its translocation from roots to shoots. Application of Si at 150 mg kg(-1) proved to be an effective level of Si that could significantly lower Cd concentration in wheat grains.

Human cardioviruses or Saffold viruses (SAFVs) of the family Picornaviridae are newly emerging viruses whose genetic and phenotypic diversity are poorly understood. We report here the full genome sequence of 11 SAFV genotypes from Pakistan and Afghanistan, along with a re-evaluation of their genetic diversity and recombination. We detected 88 SAFV from stool samples of 943 acute flaccid paralysis cases using reverse transcriptase-PCR targeting the 5' untranslated region (UTR). Further characterization based on complete VP1 analysis revealed 71 SAFVs belonging to 11 genotypes, including three previously unidentified genotypes. SAFV showed high genetic diversity and recombination based on phylogenetic, pairwise distance distributions and recombination mapping analyses performed herein. Phylogenies based on non-structural and UTRs were highly incongruent indicating frequent recombination events among SAFVs. We improved the SAFV genotyping classification criteria by determining new VP1 thresholds based on the principles used for the classification of enteroviruses. For genotype assignment, we propose a threshold of 23 and 10â% divergence for VP1 nucleotide and amino acid sequences, respectively. Other members of the species Theilovirus, such as Thera virus and Theiler's murine encephalomyelitis virus, are difficult to classify in the same species as SAFV, because they are genetically distinct from SAFV, with 41-56â% aa pairwise distances. The new genetic information obtained in this study will improve our understanding of the evolution and classification of SAFV.

Since 2005, a large poliomyelitis outbreak associated with type 2 circulating vaccine-derived poliovirus (cVDPV2) has occurred in northern Nigeria, where immunization coverage with trivalent oral poliovirus vaccine (tOPV) has been low. Phylogenetic analysis of P1/capsid region sequences of isolates from each of the 403 cases reported in 2005 to 2011 resolved the outbreak into 23 independent type 2 vaccine-derived poliovirus (VDPV2) emergences, at least 7 of which established circulating lineage groups. Virus from one emergence (lineage group 2005-8; 361 isolates) was estimated to have circulated for over 6 years. The population of the major cVDPV2 lineage group expanded rapidly in early 2009, fell sharply after two tOPV rounds in mid-2009, and gradually expanded again through 2011. The two major determinants of attenuation of the Sabin 2 oral poliovirus vaccine strain (A481 in the 5'-untranslated region [5'-UTR] and VP1-Ile143) had been replaced in all VDPV2 isolates; most A481 5'-UTR replacements occurred by recombination with other enteroviruses. cVDPV2 isolates representing different lineage groups had biological properties indistinguishable from those of wild polioviruses, including efficient growth in neuron-derived HEK293 cells, the capacity to cause paralytic disease in both humans and PVR-Tg21 transgenic mice, loss of the temperature-sensitive phenotype, and the capacity for sustained person-to-person transmission. We estimate from the poliomyelitis case count and the paralytic case-to-infection ratio for type 2 wild poliovirus infections that â¼700,000 cVDPV2 infections have occurred during the outbreak. The detection of multiple concurrent cVDPV2 outbreaks in northern Nigeria highlights the risks of cVDPV emergence accompanying tOPV use at low rates of coverage in developing countries.

In calcareous soils, phosphorus (P) retention and immobilization take place due to precipitation and adsorption. Since soil pH is considered a major soil variable affecting the P sorption, an acidic P fertilizer could result in low P adsorption compared to alkaline one. Therefore, P adsorption from DAP and phosphoric acid (PA) required to produce desired soil solution P concentration was estimated using Freundlich sorption isotherms. Two soils from Faisalabad and T. T. Singh districts were spiked with 0, 10, and 20 % CaCO3 for 15 days. Freundlich adsorption isotherms (P = aC(b/a)) were constructed, and theoretical doses of PA and DAP to develop a desired soil solution P level (i.e., 0.20 mg L(-1)) were calculated. It was observed that P adsorption in soil increased with CaCO3. Moreover, at all the levels of CaCO3, P adsorption from PA was lower compared to that from DAP in both the soils. Consequently, lesser quantity of PA was required to produce desired solution P, 0.2 mg L(-1), compared to DAP. However, extrapolating the developed relationship between soil CaCO3 contents and quantity of fertilizer to other similar textured soils needs confirmation.

Owing to a competitive interaction, zinc (Zn) contained in highly Cu-contained wastewater was hypothesized to mitigate Cu toxicity-induced negative effects on the growth and quality of lettuce. Thus, growth, metal accumulation and biochemical responses of lettuce irrigated with simulated wastewater (SW, control), Cu-contaminated SW (CuSW, 20 mg Cu L-1), Zn-contaminated SW (ZnSW, 100 mg Zn L-1) and both Cu- and Zn-contaminated SW (CuZnSW, 20 mg Cu and 100 mg Zn L-1) were evaluated. Results revealed that irrigation with CuSW negatively affected growth (dry matters, root length and plant height) and quality (low mineral concentrations) of lettuce, which were associated with higher Cu uptake. Irrigation with Zn + Cu-contaminated SW retrieved Cu toxicity and improved root and shoot dry matters and root length by 13.5%, 46% and 19%, respectively compared to that with alone Cu-contaminated SW. Moreover, CuZnSW improved lettuce leaf quality compared to CuSW and increased concentrations of Mg (30%), P (15%), Ca (41%), Mn (24%) and Fe (23%). Moreover, compared to CuSW, CuZnSW improved flavonoids (54%), total polyphenolic compounds (1.8-fold), polyphenolic acids (77%) and antiradical activities (16.6%). Most importantly, Zn addition boosted up lettuce Cu tolerance index by 18% under Cu-contaminated SW treatment. Pearson's correlation analysis among various growth and mineral parameters demonstrated that shoot Zn concentration was positively related to elemental concentrations, phytochemical contents and antioxidant activity under Cu-contaminated environment. Thus, it is concluded that Zn supplementation retrieves negative effects of Cu toxicity to lettuce grown with Cu-contaminated wastewater.

Introduction: Low soil fertility and high fertilizer costs are constraints to wheat production, which may be resolved with integrating fertilizer phosphorus (P) and farm-yard manure (FYM). Study objectives were to evaluate P source impacts on soil, P efficiency, and wheat growth in a calcareous soil. Methods: Treatments included P fertilizer (0, 17, 26, or 39 kg P ha-1) and/or FYM (0 or 10 T ha-1) in a: 1) incubation experiment and 2) wheat (Triticum aestivum spp.) field experiment. Results and Discussion: Soil organic matter increased (30-72%) linearly for both fertilizer and FYM, whereas pH decreased (0.1-0.3 units) with fertilizer only. Addition of fertilizer and FYM increased plant available P (AB-DTPA extractable soil P) an average of 0.5 mg P kg-1 soil week-1 with incubation. The initial increase was 1-9 mg P kg-1, with further increase after 84 d of ~3-17 mg P kg-1. There was also a significant increase of available P in the soil supporting plants in the field study, although the magnitude of the increase was only 2 mg kg-1 at most for the highest fertilizer rate + FYM. Grain (66 to 119%) and straw (25-65%) yield increased significantly, peaking at 26 kg P ha-1 + FYM. The P Absorption Efficiency (PAE), P Balance (PB), and P Uptake (PU) increased linearly with P rate, with the highest levels at the highest P rate. The P Use Efficiency (PUE) was highest at the lowest rates of P, with general decreases with increasing P, although not consistently. Principal component analysis revealed that 94.34 % of the total variance was accounted for with PC1 (84.04 %) and PC2 (10.33 %), with grain straw yield significantly correlated to SOM, PU, and PAE. Regression analysis showed highly significant correlation of PB with P-input (R2= 0.99), plant available P (R2= 0.85), and PU (R2= 0.80). The combination of FYM at the rate of 10 T ha-1 and fertilizer P at 26 kg P ha-1 was found as the optimum dose that significantly increased yield. It is concluded that FYM concoction with fertilizer-P not only improved SOM and residual soil P, but also enhanced wheat yields with reasonable P efficiency.

Oilseed rape (Brassica napus L.) is one of the most important oilseed crops. It has relatively high boron (B) requirements for growth. In this study, a hydroponic experiment was performed to determine the critical B requirement and B distribution in B. napus. The plants were grown for four weeks at a range of B levels (from 0.25 to 1000 µM) supplied in a nutrient solution. The results showed significant differences in the root and shoot dry matter and B accumulation in these tissues among the supplied B levels. Severe visible symptoms of B deficiency were observed on the leaves at levels lower than 1 µM B and toxicity at 1000 µM B in the nutrient solution. The maximum shoot and root dry matter were recorded at 25 µM B in the nutrient solution. The plants supplied with the lowest and the highest B levels produced 35% and 37% less shoot dry matter than those supplied with 25 µM B, while the corresponding decreases in the root dry matter were 48% and 36%, respectively. The critical concentration of B, which is the lowest concentration at which plants produce 90% of the maximum shoot dry matter, was proven to be 1 µM B for oilseed rape. At this level of external B supply, the B concentration in the shoot was 26.9 mg kg-1 DM. It was found that with the increase in B levels in the nutrient solution, the relative distribution of B between the roots and the shoots shifted in favor of the shoots.

Viral metagenomics focused on particle-protected nucleic acids was used on the stools of South Asian children with nonpolio acute flaccid paralysis (AFP). We identified sequences distantly related to Seneca Valley virus and cardioviruses that were then used as genetic footholds to characterize multiple viral species within a previously unreported genus of the Picornaviridae family. The picornaviruses were detected in the stools of >40% of AFP and healthy Pakistani children. A genetically diverse and highly prevalent enteric viral infection, characteristics similar to the Enterovirus genus, was therefore identified substantially expanding the genetic diversity of the RNA viral flora commonly found in children.
