RESUMO

The authors report a patient with coexistent double gammopathy, a Philadelphia chromosome-negative, bcr rearrangement-negative myeloproliferative disease resembling chronic myelocytic leukemia and a malignant lymphoma of B-cell origin. The double gammopathy consisted of IgM (kappa) and IgG (kappa). Peripheral blood, spleen, and marrow lymphocytes had primarily an IgG (kappa) isotype, whereas lymph node lymphocytes had predominantly an IgM (kappa) surface isotype. Increased numbers of marrow lymphocytes stained doubly for both IgM (kappa) and IgG (kappa). The results suggest that doubly isotypic as well as single isotypic lymphocytes contributed to the double gammopathy. Organ localization differed for lymphocytes with different antibody isotypes. This cluster of findings has not been described previously.

Water, perchloric acid extracts, and fractions of partially defatted whole mycelia of Coccidioides immitis contained delayed skin test activity when tested in C. immitis-infected guinea pits. Aqueous phenol extraction of these fractions resulted in partitioning of activity between aqueous-soluble and phenol-soluble fractions; activity was found to be water soluble after removal of phenol by extensive dialysis. Highest specific activity skin test antigen was invariably found in the phenol-soluble phase, water-soluble fraction. Material of equivalent activity could also be extracted directly from the defatted mycelia. Skin test active fractions contained glucose, mannose, 3-O-methylmannose, glucosamine, and amino acids.

Two human bladder cancer cell lines grew predictably in rats immunosuppressed with antilymphocyte serum. Intraperitoneal inoculation of tumor cell suspensions resulted in diffuse intraabdominal carcinomatosis with consequent host death after 10 to 20 days. Subcutaneous inoculation of tumor cell suspensions resulted in local tumors which grew exponentially for 20 to 30 days before eventual regression after 40 to 50 days; lung metastases developed in at least 13 per cent of the animals with subcutaneous tumors. The histologic appearance of the xenografted tumors closely resembled that of the original tumors. Subsequent in vitro culture of the xenografted tumors provided cell lines that were morphologically identical to the primary lines and that retained a human karyotype. It is proposed to employ this model of human bladder cancer to evaluate chemotherapeutic agents for possible use in the clinical disease.
