RESUMO

A11 dopaminergic neurons regulate somatosensory transduction by projecting from the diencephalon to the spinal cord, but the function of this descending projection in itch remained elusive. Here, we report that dopaminergic projection neurons from the A11 nucleus to the spinal dorsal horn (dopaminergicA11-SDH ) are activated by pruritogens. Inhibition of these neurons alleviates itch-induced scratching behaviors. Furthermore, chemogenetic inhibition of spinal dopamine receptor D1-expressing (DRD1+ ) neurons decreases acute or chronic itch-induced scratching. Mechanistically, spinal DRD1+ neurons are excitatory and mostly co-localize with gastrin-releasing peptide (GRP), an endogenous neuropeptide for itch. In addition, DRD1+ neurons form synapses with GRP receptor-expressing (GRPR+ ) neurons and activate these neurons via AMPA receptor (AMPAR). Finally, spontaneous itch and enhanced acute itch induced by activating spinal DRD1+ neurons are relieved by antagonists against AMPAR and GRPR. Thus, the descending dopaminergic pathway facilitates spinal itch transmission via activating DRD1+ neurons and releasing glutamate and GRP, which directly augments GRPR signaling. Interruption of this descending pathway may be used to treat chronic itch.

Recent studies have shown that information from peripherally presented images is present in the human foveal retinotopic cortex, presumably because of feedback signals. We investigated this potential feedback signal by presenting noise in fovea at different object-noise stimulus onset asynchronies (SOAs), whereas subjects performed a discrimination task on peripheral objects. Results revealed a selective impairment of performance when foveal noise was presented at 250-ms SOA, but only for tasks that required comparing objects' spatial details, suggesting a task- and stimulus-dependent foveal processing mechanism. Critically, the temporal window of foveal processing was shifted when mental rotation was required for the peripheral objects, indicating that the foveal retinotopic processing is not automatically engaged at a fixed time following peripheral stimulation; rather, it occurs at a stage when detailed information is required. Moreover, fMRI measurements using multivoxel pattern analysis showed that both image and object category-relevant information of peripheral objects was represented in the foveal cortex. Taken together, our results support the hypothesis of a temporally flexible feedback signal to the foveal retinotopic cortex when discriminating objects in the visual periphery.

Human occipital-temporal cortex features several areas sensitive to faces, presumably forming the biological substrate for face perception. To date, there are piecemeal insights regarding the functional organization of these regions. They have come, however, from studies that are far from homogeneous with regard to the regions involved, the experimental design, and the data analysis approach. In order to provide an overall view of the functional organization of the face-sensitive areas, it is necessary to conduct a comprehensive study that taps into the pivotal functional properties of all the face-sensitive areas, within the context of the same experimental design, and uses multiple data analysis approaches. In this study, we identified the most robustly activated face-sensitive areas in bilateral occipital-temporal cortices (i.e., AFP, aFFA, pFFA, OFA, pcSTS, pSTS) and systemically compared their regionally averaged activation and multivoxel activation patterns to 96 images from 16 object categories, including faces and non-faces. This condition-rich and single-image analysis approach critically samples the functional properties of a brain region, allowing us to test how two basic functional properties, namely face-category selectivity and face-exemplar sensitivity are distributed among these regions. Moreover, by examining the correlational structure of neural responses to the 96 images, we characterize their interactions in the greater face-processing network. We found that (1) r-pFFA showed the highest face-category selectivity, followed by l-pFFA, bilateral aFFA and OFA, and then bilateral pcSTS. In contrast, bilateral AFP and pSTS showed low face-category selectivity; (2) l-aFFA, l-pcSTS and bilateral AFP showed evidence of face-exemplar sensitivity; (3) r-OFA showed high overall response similarities with bilateral LOC and r-pFFA, suggesting it might be a transitional stage between general and face-selective information processing; (4) r-aFFA showed high face-selective response similarity with r-pFFA and r-OFA, indicating it was specifically involved in processing face information. Results also reveal two properties of these face sensitive regions across the two hemispheres: (1) the averaged left intra-hemispheric response similarity for the images was lower than the averaged right intra-hemispheric and the inter-hemispheric response similarity, implying convergence of face processing towards the right hemisphere, and (2) the response similarities between homologous regions in the two hemispheres decreased as information processing proceeded from the early, more posterior, processing stage (OFA), indicating an increasing degree of hemispheric specialization and right hemisphere bias for face information processing. This study contributes to an emerging picture of how faces are processed within the occipital and temporal cortex.

Practice improves our perceptual ability. However, the neural mechanisms underlying this experience-dependent plasticity in adult brain remain unclear. Here, we studied the long-term neural correlates of motion perceptual learning. Subjects' behavioral performance and BOLD signals were tracked before, immediately after, and 2 weeks after practicing a motion direction discrimination task in noise over six daily sessions. Parallel to the specificity and persistency of the behavioral learning effect, we found that training sharpened the cortical tuning in MT, and enhanced the connectivity strength from MT to the intraparietal sulcus (IPS, a motion decision-making area). In addition, the decoding accuracy for the trained motion direction was improved in IPS 2 weeks after training. The dual changes in the sensory and the high-level cortical areas suggest that learning refines the neural representation of the trained stimulus and facilitates the information transmission in the decision process. Our findings are consistent with the functional specialization in the visual cortex, and provide empirical evidence to the reweighting theory of perceptual learning at a large spatial scale. Two types of encoding tasks have been employed in previous research to investigate the beneficial effect of unitisation on familiarity-based associative recognition (unitised familiarity effect), namely the compound task and the interactive imagery task. Here we show how these two tasks could differentially engage subsequent recollection-based associative recognition and consequently lead to the turn-on or turn-off of the unitised familiarity effect. In the compound task, participants studied unrelated word pairs as newly learned compounds. In the interactive imagery task, participants studied the same word pairs as interactive images. An associative recognition task was used in combination with the Remember/Know procedure to measure recollection-based and familiarity-based associative recognition. The results showed that the unitised familiarity effect was present in the compound task but was absent in the interactive imagery task. A comparison of the compound and the interactive imagery task revealed a dramatic increase in recollection-based associative recognition for the interactive imagery task. These results suggest that unitisation could benefit familiarity-based associative recognition; however, this effect will be eliminated when the memory trace formed is easily accessed by strong recollection without the need for a familiarity assessment.

As a traditional bone implant material, titanium (Ti) and its alloys have the disadvantages of lack of biological activity and susceptibility to stress shielding effect. Adipose stem cells (ADSCs) and exosomes were combined with the scaffold material in the current work to effectively create a hydroxyapatite (HA) coated porous titanium alloy scaffold that can load ADSCs and release exosomes over time. The composite made up for the drawbacks of traditional titanium alloy materials with higher mechanical characteristics and a quicker rate of osseointegration. Exosomes (Exos) are capable of promoting the development of ADSCs in porous titanium alloy scaffolds with HA coating, based on experimental findings from in vitro and in vivo research. Additionally, compared to pure Ti implants, the HA scaffolds loaded with adipose stem cell exosomes demonstrated improved bone regeneration capability and bone integration ability. It offers a theoretical foundation for the combined use of stem cell treatment and bone tissue engineering, as well as a design concept for the creation and use of novel clinical bone defect repair materials.

In recent years, the rate of implant failure has been increasing. Microbial infection was the primary cause, and the main stages included bacterial adhesion, biofilm formation, and severe inhibition of implant osseointegration. Various biomaterials and their preparation methods have emerged to produce specific implants with antimicrobial or bactericidal properties to reduce implant infection caused by bacterial adhesion and effectively promote bone and implant integration. In this study, we reviewed the research progress of bone integration promotion and antibacterial action of superhydrophilic surfaces based on titanium alloys. First, the adverse reactions caused by bacterial adhesion to the implant surface, including infection and bone integration deficiency, are briefly introduced. Several commonly used antibacterial methods of titanium alloys are introduced. Secondly, we discuss the antibacterial properties of superhydrophilic surfaces based on ultraviolet photo-functionalization and plasma treatment, in contrast to the antibacterial principle of superhydrophobic surface morphology. Thirdly, the osteogenic effects of superhydrophilic surfaces are described, according to the processes of osseointegration: osteogenic immunity, angiogenesis, and osteogenic related cells. Finally, we discuss the challenges and prospects for the development of this superhydrophilic surface in clinical applications, as well as the prominent strategies and directions for future research.

It is widely acknowledged that holistic processing is a key characteristic of face perception. Although holistic processing implies the automatic integration of face parts, it is unclear whether such processing requires the awareness of face parts. Here, we investigated the interactions between visible face parts and face parts rendered invisible using continuous flash suppression (CFS). In the first experiment with the upper half-face visible and the lower half-face invisible, the results showed that perceived face identity was influenced by the invisible lower half-face, suggesting that integration occurs between the visible and invisible face parts, a variant of the "composite face effect". In the second experiment, we investigated the influence of visible face parts on the processing of invisible face parts, as measured by the time it took for the invisible parts to break out from CFS. The results showed a visible-to-invisible facilitation effect, that the aligned invisible face parts broke through CFS faster than when the visible and invisible face parts were misaligned. Visible eyes had a stronger influence on the invisible nose/mouth than the other way around. Such facilitation of processing from visible to invisible parts was also found when Chinese characters were used as stimuli. These results show that information integration occurs across the consciousness boundary.

Although face processing has been studied extensively, the dynamics of how face-selective cortical areas are engaged remains unclear. Here, we uncovered the timing of activation in core face-selective regions using functional Magnetic Resonance Imaging and Magnetoencephalography in humans. Processing of normal faces started in the posterior occipital areas and then proceeded to anterior regions. This bottom-up processing sequence was also observed even when internal facial features were misarranged. However, processing of two-tone Mooney faces lacking explicit prototypical facial features engaged top-down projection from the right posterior fusiform face area to right occipital face area. Further, face-specific responses elicited by contextual cues alone emerged simultaneously in the right ventral face-selective regions, suggesting parallel contextual facilitation. Together, our findings chronicle the precise timing of bottom-up, top-down, as well as context-facilitated processing sequences in the occipital-temporal face network, highlighting the importance of the top-down operations especially when faced with incomplete or ambiguous input.

OBJECTIVE: Currently, it is unclear whether a radical-by-radical (RBR) reading strategy exists in Chinese pure alexia, compared to the letter-by-letter (LBL) reading strategy in alphabetic languages. In this study, we focus on exploring the reading features of a Chinese pure alexic patient during the process of language recovery. METHOD: We investigated this issue using a series of neuropsychological tests at 4 days, 11 days, and 8 months after admission of a Chinese pure alexic patient. RESULTS: The results showed that most words could not be read aloud initially by the patient. One week later, 2 clinical features were noted: RBR reading strategy and integration impairment. Finally, there were statistically significant differences in the reaction time between the single characters and compound characters, which indicated that a radical number effect may exist. CONCLUSION: Combined with the results of previous studies, the current case study indicates that there is evidence suggesting that the RBR reading strategy in Chinese pure alexia is of limited benefit, a result that appears fundamentally different from the LBL reading strategy.
