RESUMO

The Dlk1-Dio3 domain is important for normal embryonic growth and development. The heart is the earliest developing and functioning organ of the embryo. In this study, we constructed a transcriptional termination model by inserting termination sequences and clarified that the lack of long non-coding RNA (lncRNA) expression in the Dlk1-Dio3 domain caused the death of maternal insertion mutant (MKI) and homozygous mutant (HOMO) mice starting from E13.5. Parental insertion mutants (PKI) can be born and grow normally. Macroscopically, dying MKI and HOMO embryos showed phenomena such as embryonic edema and reduced heart rate. Hematoxylin and eosin (H.E.) staining showed thinning of the myocardium in MKI and HOMO embryos. In situ hybridization (IHC) and quantitative reverse-transcription polymerase chain reaction (qRT-PCR) showed downregulation of lncGtl2, Rian, and Mirg expression in MKI and HOMO hearts. The results of single-cell RNA sequencing (scRNA-Seq) analysis indicated that the lack of lncRNA expression in the Dlk1-Dio3 domain led to reduced proliferation of epicardial cells and may be an important cause of cardiac dysplasia. In conclusion, this study demonstrates that Dlk1-Dio3 domain lncRNAs play an integral role in ventricular development.

A pair of metal-organic frameworks (MOFs) of JXNU-15 (formulated as [Co6(µ3-OH)6(BTB)2(BPY)3]n, BTB3- = benzene-1,3,5-tribenzoate and BPY = 4,4'-bipyridine) and its fluorinated JXNU-15(F) ([Co6(µ3-OH)6(SFBTB)2(BPY)3]n) based on the fluorous 1,3,5-tri(3,5-bifluoro-4-carboxyphenyl)benzene (SFBTB3-) ligands were presented. The detailed comparisons of the acetylene/carbon dioxide (C2H2/CO2) separation abilities between the isostructural JXNU-15(F) and JXNU-15 were presented. In comparison with the parent JXNU-15, the higher C2H2 uptake, larger adsorption selectivity of the C2H2/CO2 (50/50) mixture, and enhanced C2H2/CO2 separation performance endow JXNU-15(F) with highly efficient C2H2/CO2 separation performance, which is demonstrated by singe-component gas adsorptions and dynamic gas mixture breakthrough experiments. The fluorine substituents exert the crucial effects on the enhanced C2H2/CO2 separation ability of JXNU-15(F) and play the dominant role in the C2H2-framework interactions, as uncovered by computational simulations. This work illustrates a powerful fluorine substitution strategy for boosting C2H2/CO2 separation ability for MOFs.

An immobilized metal affinity (IMAC) adsorbent was prepared for selective enrichment of adenine type CKs, via grafting polymer chain pendant with iminodiacetic acid (IDA) from polydopamine (PDA)/polyethyleneimine (PEI)-coated magnetic graphene oxide (magGO) via surface-initiated-atom transfer radical polymerization (SI-ATRP). The prepared IMAC sorbent exhibited remarkable adsorption performances and good selectivity for adenine-type CKs and was utilized as a sorbent of magnetic solid-phase extraction (MSPE) for effective enrichment of four adenine-type CKs in bean sprouts. Under the optimized extraction conditions, an analytical method for four adenine type CKs in bean sprouts was established by combining the MSPE combined with ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). The recoveries of the analytes were between 80.4 ± 1.9% and 114.6 ± 1.5% (n = 3). The limits of detection (LODs) range from 0.63 to 2.30 pg⋅mL-1. The relative standard deviations of intra-day and inter-day were less than 12.6%. The established method was successfully applied to the selective extraction and sensitive detection of trace adenine-type CKs in plant samples.

Here, a hexanuclear Co6(µ3-OH)6 cluster-based metal-organic framework (MOF), [Co6(µ3-OH)6(BTB)2(bpy)3]n (JXNU-15) (bpy = 4,4'-bipyridine), with the 1,3,5-tri(4-carboxyphenyl)benzene (BTB3-) ligand was synthesized for the challenging propadiene/propylene separation. The combination of a large pore volume and a suitable pore environment boosts the significantly high propadiene (C3H4) uptake (311 cm3 g-1 at 298 K and 100 kPa) for JXNU-15. An amine-functionalized MOF of JXNU-15(NH2) was further obtained with the 1,3,5-tri(4-carboxyphenyl)benzene analogue of 3,3″-diamino-5'-(3-amino-4-carboxyphenyl)-[1,1':3',1″-terphenyl]-4,4″-dicarboxylic ligand. The comparative studies of propadiene/propylene(C3H4/C3H6) separation performance between isostructural JXNU-15 and JXNU-15(NH2) are provided. JXNU-15(NH2) exhibits an impressive C3H4 capacity at low pressures with 69.1 cm3 g-1 at 10 kPa, which is twice that of JXNU-15 under the same conditions. Moreover, the separation selectivity of JXNU-15(NH2) is 1.3-fold higher as compared to JXNU-15. JXNU-15(NH2) with enhanced C3H4/C3H6 separation performance was elegantly illustrated by gas separation experiments and theoretical simulations. This work presents an amine-functionalized strategy for the enhancement of the C3H4/C3H6 separation performance of MOF.

Two series of novel 4-phenoxypyridine derivatives containing imidazole-4-carboxamide and 4-methyl-5-oxo-4,5-dihydro-1,2,4-triazole-3-carboxamide moieties were synthesized and evaluated for their in vitro inhibitory activities against c-Met kinase and antiproliferative activities against MKN-45, A549 and H460 cancer cell lines. The results indicated that most of the compounds showed moderate to good antitumor activities. The most promising compound T14 (with c-Met IC50 value of 0.012 µM) showed remarkable antiproliferative activities against MKN-45, A549 and H460 cell lines with IC50 values of 0.64 µM, 1.92 µM and 2.68 µM, respectively. Their preliminary structure-activity relationships (SARs) studies indicate that imidazole-4-carboxamide was more preferred as linker part, and electron-withdrawing groups (especially halogen groups) on the terminal phenyl rings were beneficial for improving the antitumor activities.

On the basis of N-(3-amino-4-methoxyphenyl)acrylamide scaffold, a series of novel compounds containing 3-substitutional-1-methyl-1H-indole, 2-substitutional pyrrole or thiophene moieties were synthesized and their in vitro antiproliferation activities against A549 and H1975 cell lines were evaluated. The results indicated that most of the compounds showed moderate to excellent antitumor activities. Especially, compounds 9a (A549 IC50 = 1.96 µM, H1975 IC50 = 0.095 µM), 17i (A549 IC50 = 4.17 µM, H1975 IC50 = 0.052 µM), 17j (A549 IC50 = 1.67 µM, H1975 IC50 = 0.061 µM) exhibited comparable antitumor activities and selectivity ratios compared to the positive control osimertinib (A549 IC50 = 2.91 µM, H1975 IC50 = 0.064 µM). In vitro inhibitory activities against EGFR kinases containing different mutations were also tested. Compound 17i showed remarkable inhibitory activity (with IC50 value of 1.7 nM) to EGFRL858R/T790M kinase and selectivity (22-folds compared to EGFRWT kinase). Furthermore, acridine orange/ethidium bromide (AO/EB) staining assay, cell apoptosis assay, cell cycle distribution assay and wound-healing assay of the compounds 9a and 17i were performed on H1975 cell line. The results showed dose-dependent activities of the induction of apoptosis, G0/G1-phase arrestation and inhibition of migration, which were similar to the positive control osimertinib. Additionally, molecular docking analysis was performed to seek the possible binding mode between the selected compounds (9a, 17i-17j) and EGFRL858R/T790M kinase. The results demonstrated that compound 17i is a promising candidate and worth further study.

N-doped carbon quantum dots (N-CDs) with polyaminobenzene hydrazine as precursor were prepared by solvothermal method for the monitoring of pH fluctuation in HeLa cells via fluorescence imaging. The N-CDs show two emission wavelengths at 582 and 640 nm under different pH with two excitation wavelengths. The fluorescence intensity at 640 nm (λex = 520 nm) and the ratio of F582/F640 (λex = 470 nm) linearly increase with pH in the range of 2.4 ~ 3.6 (R2 = 992) and 5.6 ~ 7.6 (R2 = 0.987), respectively. The sensor exhibits high sensitivity and reversibility and anti-interference capability, thus enabling sensing pH change in intracellular environment in real time, as demonstrated by successful monitoring of intracellular pH fluctuation during H2O2 stimulation in HeLa cells.

Poly(ionic liquid)-modified stationary phases can have multiple interactions with solutes. However, in most stationary phases, separation selectivity is adjusted by changing the poly(ionic liquid) anions. In this work, two poly(ionic liquid)-modified silica stationary phases were prepared by introducing the cyano or tetrazolyl group on the pendant imidazolium cation on the polymer chains. Various analytes were selected to investigate their mechanism of retention in the stationary phases using different mobile phases. Two poly(ionic liquid)-modified stationary phases can provide various interactions toward solutes. Compared to the cyano-functionalized poly(ionic liquid) stationary phase, the tetrazolyl-functionalized poly(ionic liquid) stationary phase provides additional cation-exchange and π-π interactions, resulting in different separation selectivity toward analytes. Finally, applicability of the developed stationary phases was demonstrated by the efficient separation of nonsteroidal anti-inflammatory drugs.

Graphene oxide has received extensive attention because of its unique properties and potential applications. In this study, magnetic nitrogen-doped graphene was prepared by one-step hydrothermal reaction using urea as the dopant and reductant, and ferroferric oxide nanoparticles were in situ deposited on the surface of the nanohybrids. The magnetic nitrogen-doped graphene was characterized using various physical and chemical methods. It was used as a new adsorbent for the magnetic solid-phase extraction of four nonsteroidal anti-inflammatory drugs from the river water. The parameters influencing the extraction efficiency were optimized in detail. Under optimal conditions, this method provided a wide linear range (5-200 ng/mL). The limits of detection were in the range of 1.07-5.10 ng/mL. The recoveries varied from 81.2 to 121.5% with relative standard deviations of less than 10.8%. Overall, we can conclude that the proposed method offers an efficient pretreatment and enrichment and can be successfully applied for the extraction and determination of nonsteroidal anti-inflammatory drugs in complex matrices.

An immobilized metal affinity (IMA) adsorbent was prepared by grafting bottlebrush polymer pendant with iminodiacetic acid (IDA) from the surface of polydopamine (PDA)-coated magnetic graphene oxide (magGO), via surface-initiated atom transfer radical polymerization (SI-ATRP). Poly(hydroxyethyl methacrylate) (PHEMA) was grafted firstly from the PDA-coated magGO as the backbone, and then poly(glycidyl methacrylate) was grafted from the PHEMA chains via the second SI-ATRP to afford the bottlebrush polymer-grafted magGO Thereafter, IDA was anchored on the nanocomposites to produce the IMA adsorbent after chelating copper ions. The adsorbent was characterized by various physical and physicochemical methods. Its adsorption properties were evaluated by using histidine-rich proteins (bovine hemoglobin, BHb) and other proteins (lysozyme and cytochrome-C). The results show that its maximum adsorption capacity to BHb was 378.6 mg g-1, and the adsorption equilibrium can be quickly reached within 1 h. The adsorbent has excellent reproducibility and reusability. It has been applied to selectively purify hemoglobin from human whole blood, indicating its potential in practical applications. In recent years, anaplastic lymphoma kinase (ALK) rearrangement-positive anaplastic large cell lymphoma (ALCL) has rising morbidity and mortality. Unfortunately, no ALK inhibitor has been approved by the FDA for single treatment of ALK rearrangement-positive ALCL. In this study, we investigated the antitumor effect of ZYY, a novel ALK inhibitor, showing a strong growth inhibitory effect on Karpas299 cells in vitro and in vivo. Specifically, ZYY significantly reduced the mRNA and protein expression of ALK and its downstream signaling proteins in Karpas299 cells. Furthermore, ZYY induced G1 phase arrest and promoted apoptosis in Karpas299 cells. Furthermore, we demonstrated that ZYY-induced apoptosis was mainly related to the mitochondria-dependent endogenous pathway. In vitro studies further showed that ZYY induced autophagy in Karpas299 cells, along with increased levels of the autophagy-related proteins, including LC3II and Beclin-1. Moreover, knockdown Beclin-1 and application of autophagy inhibitor chloroquine potentiated ZYY-induced cytotoxicity and apoptosis in vitro, indicating that cytoprotective autophagy might be triggered by ZYY in Karpas299 cells. Taken together, the novel ALK inhibitor ZYY has tremendous potential for treating human ALCL, and a combination of autophagy and ALK inhibition could effectively elicit potent antitumor effects.

Finely tuning the photodegradation behavior of the layer-by-layer (LbL) film from the view of controlling the chemical structure of the film-building polymer is still a challenge in related fields. To meet this requirement, a photodegradable polymer (P1) is rationally designed for assembling a visible-light-degradable multilayer film with polystyrene sulfonate (PSS). Compared with similar photopolymers (P2 and P3), this asymmetric picolinium-containing polymer can significantly enhance the degradation rate of as-prepared LbL films; under the same degradation condition, the degradation rate of (P1/PSS)10 is 3 and 6.6 times that of (P2/PSS)10 and (P3/PSS)10, respectively. Moreover, near-infrared light (NIR) is available for triggering the degradation of this film with the assistance of upconversion nanoparticles of YbTm@Lu. The cell cytotoxicity and cell proliferation experiments reveal that P1 is nontoxic and favorable for cell proliferation at concentrations of up to 500 µg mL-1 . As for (PSS/P1)10 films, the ratio of cell number of these two samples ((PSS/P1)10 modified: photodegraded) increases dramatically and reaches about 1.67:1 after 72 h incubation. On the basis of these results, it is anticipated that P1 and this LbL film is an exceptional candidate for visible-light/NIR degradable materials in materials and biological science, medicine, and optics.

In this work, core-shell structured magnetic mesoporous carbon nanospheres were fabricated from the carbonization of metal-polyphenol coordination polymer-coated Fe3 O4 nanoparticles. The preparation method is simple, fast, versatile, and easy to scale up. Magnetic mesoporous carbon nanospheres exhibit a high specific surface area, high superparamagnetism, and high adsorption efficiencies for phthalates. Four phthalates were extracted from aqueous solutions by using magnetic mesoporous carbon nanospheres via magnetic solid phase extraction. Subsequent analysis was performed by using high-performance liquid chromatography with ultraviolet detection. The analytical method has good linearity in the concentration range of 1-200 ng/mL for diethyl phthalate, diisobutyl phthalate, and dicyclohexyl phthalate, and 3-200 ng/mL for dipropyl phthalate. The limits of detection were in the range of 0.10-0.62 ng/mL. Compared with previous methods, this method has a lower detection limit, wider linearity range, and faster adsorption and desorption rates. The results indicate that magnetic mesoporous carbon nanospheres are suitable for the enrichment of hydrophobic substances from aqueous solutions.

Polymeric ionic liquids (PILs) with 1-vinyl-3-ethylimidazolium cations and two different anions of Br- and PF6- were assembled onto the surface of graphene (G) nanosheets. The derived two composites, i.e., PIL(Br)-G and PIL(PF6)-G, were further efficiently immobilized onto the surface of silica nanoparticles via self-assembly technique. The obtained two PIL-G/SiO2 nanocomposites exhibited diverse adsorption performances toward proteins through adjusting the anions of PILs. Electrostatic attractions between proteins and the nanocomposites dominated protein adsorption, while the presence of PF6- anions weakened electrostatic interactions and deteriorated the selective adsorption of target protein, i.e., bovine serum albumin (BSA) in this case. Specifically, PIL(Br)-G/SiO2 nanocomposite displayed high selectivity toward BSA and a high adsorption efficiency of ca. 98% was achieved for 100 mg L-1 BSA in a Britton-Robinson (B-R) buffer at pH 5. HPLC analysis demonstrated the selectivity of PIL(Br)-G/SiO2 nanocomposite toward BSA in the presence of abundant hemoglobin and cytochrome c. The practical applicability was verified by performing selective isolation of human serum albumin (HSA) from human whole blood. The authors describe the preparation of copper-doped magnetic microspheres (Cu-Fe3O4) by a solvothermal method. Due to their good magnetic property and high affinity for compounds containing an imidazole moiety (containing N-H), they are excellent adsorbents for such compounds as tested by eighteen compounds. Specifically, a method has been developed for magnetic solid-phase extraction (MSPE) of theophylline (TP) from plasma. The method enables selective enrichment of TP over many potential interferents that can occur in plasma. Following elution with alkaline methanol, TP was quantified by HPLC-UV at a detection wavelength of 272 nm. Under the optimized conditions, a linear response is found for the 0.02 to 20 µg·mL-1 concentration range, and the limit of detection is as low as 3 ng·mL-1. Recoveries from spiked samples range from 91.2 to 100.4%, and the repeatabilities are between 2.9 and 12% (for n = 6). The method was successfully applied to the determination of TP in rabbit and rat plasma. Poly(2-naphthyl acrylate) was first grafted onto silica-coated magnetic nanoparticles by surface-initiated atom transfer radical polymerization to prepare a reversed-phase magnetic adsorbent. The resulting polymer brush displays enhanced extraction efficiency by offering active sites on the surfaces of adsorbent. It was applied to the preconcentration of the non-steroidal antiinflammatory drugs (NSAIDs) indomethacin (InDo) and diclofenac (DIC). These drugs interact with the sorbent through hydrophobic and π-interactions, and via electrostatic attraction. By coupling the magnetic solid-phase extraction with HPLC, a method for analysis of InDo and DIC in the environmental water samples was established. The limits of detection range from 0.62 to 0.64 ng·mL-1, and the relative standard deviations for intra-and inter-day analyses of spiked water samples are <11.9%, and relative recoveries are between 62.1 and 96.7%. Graphical abstract A reversed-phase magnetic adsorbent was prepared by grafting poly(2-naphthyl acrylate) brush on the surface of silica coated magnetic nanoparticles. Due to the two conjugated aromatic rings of the monomer, the polymer brush can effectively extract non-steroidal anti-inflammatory drugs through strong π- and hydrophobic interactions.

The authors report on the preparation of composites made from graphene oxide (GO) and zeolitic imidazolate framework-8 (ZIF-GO), with various fractions of GO. GO acts as the template and as a modulator for the surface properties of the composites. It also improves the selective adsorption of specific proteins, i.e. hemoglobin (Hb) in this case. The adsorption capacity for Hb is as high as 436 mg g-1 when using a composite containing 20% of GO as sorbent, and 95% of specific activity is maintained for the Hb recovered. The sorbent is applied to selectively isolate Hb from human whole blood. Graphical abstract Graphene oxide-zeolitic imidazolate framework-8 composites (ZIF-GO) with varying mass ratios of GO were prepared in order to tune surface properties and to improve the adsorption selectivity toward hemoglobin.

Two series of novel 5,7-diarylimidazo[1,2-a]pyridine-8-carbonitrile derivatives (3a-3q and 7a-7n) were designed by modification of CA-4 pharmacophore to develop colchicine targeted antitubulin agents. All compounds were efficiently synthesized and evaluated for their cytotoxicity against five selected cancer cell lines (HT-29, H460, A549, MKN-45 and SMMC-7721) which got an insight in structure and activity relationships (SARs). Several molecules (7e, 7f, 7h-7j and 7m) were disclosed to exhibit promising antiproliferative activity with IC50 values in double-digit nanomolar degree. Optimization toward these compounds led to the discovery of a promising lead 7e, which showed noteworthy potency with IC50 value ranging from 0.01 to 3.2µM superior to CA-4 and Crolibulin. Importantly, immunofluorescence staining and colchcine competitive binding assay revealed that microtubule dynamics was disrupted by 7e by binding at the colchicine site of tubulin. Moreover, molecular docking studies suggested the binding of this mimic at colchcine-binding site is similar to Crolibulin, as was in conformity with the observed SARs for these compounds.

In case of organic frameworks (COFs) as adsorbents in the pretreatment of complex food matrices, challenges such as poor dispersion and non-specific adsorption of interfering macromolecules like proteins are often encountered. To address this issue, this work prepared a three-dimensional covalent organic framework (3D-COF) with a novel bcu topology based on polyhedral oligomeric silsesquioxane (POSS). Subsequently, gluconolactone (GDL) was modified onto the external surface of the material via the reaction with the exposed reactive residues. The resulting POSS-COF@GDL adsorbent has an enhanced hydrophilicity in the external surface, thereby significantly improves the dispersion of materials in aqueous solution and reduces the adsorption ability toward protein. Whereas, the inner of material retains hydrophobic pores that exhibit high adsorption efficiency to small hydrophobic molecules. Compared with the traditional pretreatment methods, POSS-COF@GDL can directly extract bisphenols (BPs) in milk samples without any additional treatment. The established sample pretreatment method is coupled with high-performance liquid chromatography-ultraviolet detection (HPLC-UV), resulting in recoveries of 71.8 to 93.6%, intra- and inter-day relative standard deviations (RSDs) of <8.3%, and limits of detection (LODs) of 0.042-0.16 ng∙mL-1 for four BPs.

Circulating tumor cell (CTC) in the blood is the main cause of cancer metastasis for death in cancer patients. It is extremely important for cancer diagnosis at an early stage and treatment to simultaneously detect and kill the CTCs. In this work, a new hybridized nanolayer, namely gold nanoparticle/gold nanorods@ Polydopamine (AuNPs/AuNRs@PDA), was coated on the Ω-shaped fiber optics (Ω-FO) for localized surface plasmonic resonance (LSPR) to perform tumor cell sensing and photothermal treatment (PTT). The PDA nanolayer was formed on a bare fiber optic through the self-polymerization of dopamine under mild conditions. The AuNRs and AuNPs were absorbed on the surface of the PDA nanolayer to form a hybridized nanolayer. The hybridized nanolayer-modified Ω-FO LSPR exhibited a high refractive index sensitivity (RIS) of 37.59 (a.u/RIU) and photothermal conversion efficiency. After being modified with the recognition element of aptamer, the Ω-FO LSPR was used to develop a sensitive and specifical tumor cell sensing. Under the irradiation of near-infrared light (NIR) laser, the Ω-FO LSPR can kill the captured tumor cells with the apoptotic/necrotic rate of 62.6 % and low side-effect for the nontarget cells. The FO LSPR sensor realized the dual functions of CTC sensing and PTT, which provided a new idea for the early diagnosis and treatment of cancer.
