After the Fukushima nuclear power plant accident in 2011, many types of survey meters were used, including Geiger-Müller (GM) survey meters, which have long been used to measure ß-rays. Recently, however, a novel radiation survey meter that uses a plastic-scintillation sensor has been developed. Although manufacturers' catalog data are available for these survey meters, there have been no user reports on performance. In addition, the performance of commercial plastic-scintillation survey meters has not been evaluated. In this study, we experimentally compared the performance of a plastic-scintillation survey meter with that of a GM survey meter. The results show that the two instruments performed very similarly in most respects. The GM survey meter exhibited count losses when the radiation count rate was high, whereas the plastic-scintillation survey meter remained accurate under such circumstances, with almost no count loss at high radiation rates. For measurements at background rates (i.e., low counting rates), the counting rates of the plastic-scintillation and GM survey meters were similar. Therefore, an advantage of plastic-scintillation survey meters is that they are less affected by count loss than GM survey meters. We conclude that the plastic-scintillation survey meter is a useful ß-ray measuring/monitoring instrument.

Given the new recommendations for occupational eye lens doses, various lead glasses have been used to reduce irradiation of interventional radiologists. However, the protection afforded by lead glasses over prescription glasses (thus over-glasses-type eyewear) has not been considered in detail. We used a phantom to compare the protective effects of such eyewear and regular eyewear of 0.07 mm lead-equivalent thickness. The shielding rates behind the eyewear and on the surface of the left eye of an anthropomorphic phantom were calculated. The left eye of the phantom was irradiated at various angles and the shielding effects were evaluated. We measured the radiation dose to the left side of the phantom using RPLDs attached to the left eye and to the surface/back of the left eyewear. Over-glasses-type eyewear afforded good protection against x-rays from the left and below; the average shielding rates on the surface of the left eye ranged from 0.70-0.72. In clinical settings, scattered radiation is incident on physicians' eyes from the left and below, and through any gap in lead glasses. Over-glasses-type eyewear afforded better protection than regular eyewear of the same lead-equivalent thickness at the irradiation angles of concern in clinical settings. Although clinical evaluation is needed, we suggest over-glasses-type Pb eyewear even for physicians who do not wear prescription glasses.

We demonstrate ultralinear and ultrawideband frequency-modulated continuous-wave (FMCW) signal generation using an optical parametric wideband frequency modulation (OPWBFM) method. The OPWBFM method optically expands the bandwidths of FMCW signals beyond the electrical bandwidths of optical modulators via a cascaded four-wave mixing (FWM) process. Compared to the conventional direct modulation approach, the OPWBFM method simultaneously achieves high linearity and a short measurement time of the frequency sweep. On the other hand, it is also known that the OPWBFM method expands the phase noise of idlers as well as their bandwidths if an input conjugate pair has different phase noise. To avoid this phase noise expansion, it is crucial to synchronize the phase of an input complex conjugate pair of an FMCW signal using an optical frequency comb. For demonstration, we successfully generated an ultralinear 140-GHz FMCW signal by using the OPWBFM method. Moreover, we employ a frequency comb in the conjugate pair generation process, leading to the mitigation of phase noise expansion. By using a 140-GHz FMCW signal, we achieve a range resolution of ∼1 mm through fiber-based distance measurement. The results show the feasibility of an ultralinear and ultrawideband FMCW system with a sufficiently short measurement time.

In 2011, the International Commission on Radiological Protection (ICRP) recommended a significant reduction in the lens-equivalent radiation dose limit, thus from an average of 150 to 20 mSv/year over 5 years. In recent years, the occupational dose has been rising with the increased sophistication of interventional radiology (IVR); management of IVR staff radiation doses has become more important, making real-time radiation monitoring of such staff desirable. Recently, the i3 real-time occupational exposure monitoring system (based on RaySafeTM) has replaced the conventional i2 system. Here, we compared the i2 and i3 systems in terms of sensitivity (batch uniformity), tube-voltage dependency, dose linearity, dose-rate dependency, and angle dependency. The sensitivity difference (batch uniformity) was approximately 5%, and the tube-voltage dependency was <±20% between 50 and 110 kV. Dose linearity was good (R2 = 1.00); a slight dose-rate dependency (~20%) was evident at very high dose rates (250 mGy/h). The i3 dosimeter showed better performance for the lower radiation detection limit compared with the i2 system. The horizontal and vertical angle dependencies of i3 were superior to those of i2. Thus, i3 sensitivity was higher over a wider angle range compared with i2, aiding the measurement of scattered radiation. Unlike the i2 sensor, the influence of backscattered radiation (i.e., radiation from an angle of 180°) was negligible. Therefore, the i3 system may be more appropriate in areas affected by backscatter. In the future, i3 will facilitate real-time dosimetry and dose management during IVR and other applications.

The total capacity of optical submarine cable systems as a global communication infrastructure must be continuously enlarged. Multi-core fibers (MCFs) have been studied as methods to maximize the total cable capacity under electrical power and cable space limitations. In particular, standard cladding MCFs, which are expected to have high productivity and mechanical reliability, are attractive for early deployment in submarine cable systems. In this paper, we demonstrate high-capacity trans-Pacific class transmission using standard cladding uncoupled 4-core fibers, achieving a transmission capacity of 55.94 Tbit/s over 12,040 km. In addition, based on the results of this and our previous coupled MCF transmission experiments, we summarize the characteristics of coupled and uncoupled MCFs applied to optical submarine cable systems.

RESUMO

An IF-over-fiber (IFoF)-based analog transport technology for mobile fronthaul applications has recently attracted significant attention. However, most previous studies have employed discrete optical components. For the analog transport technology to be a more cost-effective and power-efficient solution, it is necessary to utilize existing integrated optical transceivers. In this paper, we demonstrate IFoF transmission using a commercial off-the-shelf transmitter optical sub-assembly (TOSA). Although the TOSA was developed for a digital system employing non-return-to-zero (NRZ) signals, we show that it is also possible for the TOSA to support high-capacity analog transmission. As a demonstration, by using the TOSA, we could successfully transmit 64- and 256-ary quadrature-amplitude-modulated (64/256QAM) orthogonal-frequency-division-multiplexed (OFDM) signals with net bit rates of 54.74 and 36.49 Gbps per wavelength, respectively. Since the TOSA has four wavelength channels, the total capacities are 218.94 and 145.98 Gbps, respectively. To the best of our knowledge, these rates are the highest among all the demonstrations using analog transport technology.

RESUMO

Wideband signal generation using frequency/phase modulation (FM/PM) is the key fundamental function for various applications such as radar and analog communication systems. It is well known that analog FM/PM communication systems can dramatically improve signal quality by spectral expansion. In classical communication theories, the Armstrong indirect method is one of the most popular methods for bandwidth expansion of FM signals. In the Armstrong method, a narrowband signal is converted to a broadband signal with the help of a nonlinear frequency multiplier. In this paper, we propose a photonic Armstrong method enabled by direct detection. By utilizing the nonlinearity caused by direct detection, we can increase bandwidth so that it is double that of the original signal in the photonic Armstrong method. Also, it completely eliminates signal-to-signal beat interference (SSBI) at the same time. We conducted an experiment to verify the concept and confirmed these advantages experimentally.

Since the Fukushima Daiichi Nuclear Power Plant (FDNPP) accident, we have established an archive system of livestock and wild animals from the surrounding ex-evacuation zone. Wildlife within the alert zone have been exposed to low-dose-rate (LDR) radiation for a long continuous time. In this study, we analysed the morphological characteristics of the testes and in vitro fertilization (IVF) capacity of cryopreserved sperm of racoons from the ex-evacuation zone of the FDNPP accident. The radioactivity of caesium-137 (137 Cs) was measured by gamma-ray spectrometry, and the measured radioactivity concentration was 300-6,630 Bq/kg in the Fukushima raccoons. Notably, normal spermatogenesis was observed in the seminiferous tubules of the testes, with the germinal epithelium composed of a spermatogenic cell lineage with no evident ultrastructural alterations; freeze-thawing sperm penetration ability was confirmed using the interspecific zona pellucida-free mouse oocytes IVF assays. This study revealed that the chronic and LDR radiation exposure associated with the FDNPP accident had no adverse effect on the reproductive characteristics and functions of male raccoons.

Cellular agriculture is an emerging scientific discipline that leverages the existing principles behind stem cell biology, tissue engineering, and animal sciences to create agricultural products from cells in vitro. Cultivated meat, also known as clean meat or cultured meat, is a prominent subfield of cellular agriculture that possesses promising potential to alleviate the negative externalities associated with conventional meat production by producing meat in vitro instead of from slaughter. A core consideration when producing cultivated meat is cell sourcing. Specifically, developing livestock cell sources that possess the necessary proliferative capacity and differentiation potential for cultivated meat production is a key technical component that must be optimized to enable scale-up for commercial production of cultivated meat. There are several possible approaches to develop cell sources for cultivated meat production, each possessing certain advantages and disadvantages. This review will discuss the current cell sources used for cultivated meat production and remaining challenges that need to be overcome to achieve scale-up of cultivated meat for commercial production. We will also discuss cell-focused considerations in other components of the cultivated meat production workflow, namely, culture medium composition, bioreactor expansion, and biomaterial tissue scaffolding.

For weakly coupled mode-division multiplexed (MDM) transmission systems, we design and implement optical coherent receiver prototypes with real-time multiple-input multiple-output (MIMO) digital signal processing to equalize two degenerate linearly polarized modes with dual polarization. Using field programmable gate array circuits, we implement real-value 8 × 2 MIMO adaptive equalization with externally separated phase compensators based on the least mean square algorithm, which enables not only training equalization but also fast carrier-phase tracking. With the optical coherent MIMO receiver prototype, we demonstrate real-time weakly coupled 10 × MDM wavelength-division multiplexed dual-polarization quadrature phase shift keying transmission over 48-km few-mode fibers. This report shows a record number of multiplexed spatial modes, namely, 10 modes with dual polarization, in real-time MDM transmission experiments.

BACKGROUND/AIMS: Lacerations of the oral mucosa and fractures of alveolar processes commonly occur in traumatic dental injuries (TDIs). Impaired wound healing and tissue regeneration have severe consequences on the quality of life. Bone marrow mesenchymal stem cells (BMMSCs) possess the ability of self-renewal and multipotential differentiation. Treatment with low-level sodium fluoride (NaF) has emerged as a promising approach to enhance wound repair. The aim of this study was to assess the effects of low-level NaF on soft tissue healing and on the proliferation, migration and extracellular matrix synthesis of BMMSCs. MATERIAL AND METHODS: BMMSCs derived from mice were treated with 50 µM, 500 µM, or 5 mM NaF for 12, 24, and 48 hours, and cell proliferation was assessed by the MTS assay. Cell motility was detected at 12 and 24 hours by a wound healing assay, and osteoblastic differentiation for 21 days by 1% Alizarin Red S staining in 50 µM NaF-treated BMMSCs. Gene expression of Runx2 and Osteocalcin was evaluated by quantitative real-time PCR. An experimental rat skin wound model was employed, and levels of c-Myc, Ki67, fibronectin, and vimentin were assessed by immunohistochemistry. RESULTS: There was a significant induction in the proliferation and migration of BMMSCs treated with 50 µM NaF. The expression of Ki67 and c-Myc protein was increased in tissues treated with 50 µM NaF, and the expression of fibronectin and vimentin in the 50 µM NaF-treated tissues was stimulated. Alizarin Red staining revealed enhanced mineralization in 50 µM NaF-treated BMMSCs with increased expression of Runx2 and Osteocalcin, indicating their upregulated osteogenic differentiation. CONCLUSION: Low-level NaF could promote soft tissue healing and hard tissue regeneration.

MicroRNA (miRNA) is a non-coding RNA involved in regulating both cancer gene promotion and suppression. We investigated the role of miRNA in inducing radiation resistance in cancer cell lines using clinically relevant radioresistant (CRR) cells. Analysis using miRNA arrays and qPCR revealed that miR-7-5p is highly expressed in all examined CRR cells. Additionally, CRR cells lose their radioresistance when daily irradiation is interrupted for over 6 months. MiR-7-5p expression is reduced in these cells, and treating CRR cells with a miR-7-5p inhibitor leads to a loss of resistance to irradiation. Conversely, overexpression of miR-7-5p in CRR cells using a miR-7-5p mimic shows further resistance to radiation. Overexpression of miR-7-5p in parent cells also results in resistance to radiation. These results indicate that miR-7-5p may control radioresistance in various cancer cells at the clinically relevant dose of irradiation. Furthermore, miR-7-5p downregulates mitoferrin and reduces Fe2+, which influences ferroptosis. Our findings have great potential not only for examining radiation resistance prior to treatment but also for providing new therapeutic agents for treatment-resistant cancers.

Recent advances in bioengineering have enabled cell culture systems that more closely mimic the native cellular environment. Here, we demonstrated that human induced pluripotent stem cell (iPSC)-derived myogenic progenitors formed highly-aligned myotubes and contracted when seeded on two-dimensional micropatterned platforms. The differentiated cells showed clear nuclear alignment and formed elongated myotubes dependent on the width of the micropatterned lanes. Topographical cues from micropatterning and physiological substrate stiffness improved the formation of well-aligned and multinucleated myotubes similar to myofibers. These aligned myotubes exhibited spontaneous contractions specifically along the long axis of the pattern. Notably, the micropatterned platforms developed bundle-like myotubes using patient-derived iPSCs with a background of Pompe disease (glycogen storage disease type II) and even enhanced the disease phenotype as shown through the specific pathology of abnormal lysosome accumulations. A highly-aligned formation of matured myotubes holds great potential in further understanding the process of human muscle development, as well as advancing in vitro pharmacological studies for skeletal muscle diseases.

Human induced-pluripotent stem cells (iPSCs) are a promising resource for propagation of myogenic progenitors. Our group recently reported a unique protocol for the derivation of myogenic progenitors directly (without genetic modification) from human pluripotent cells using free-floating spherical culture. Here we expand our previous efforts and attempt to determine how differentiation duration, culture surface coatings, and nutrient supplements in the medium influence progenitor differentiation and formation of skeletal myotubes containing sarcomeric structures. A long differentiation period (over 6 weeks) promoted the differentiation of iPSC-derived myogenic progenitors and subsequent myotube formation. These iPSC-derived myotubes contained representative sarcomeric structures, consisting of organized myosin and actin filaments, and could spontaneously contract. We also found that a bioengineering approach using three-dimensional (3D) artificial muscle constructs could facilitate the formation of elongated myotubes. Lastly, we determined how culture surface coating matrices and different supplements would influence terminal differentiation. While both Matrigel and laminin coatings showed comparable effects on muscle differentiation, B27 serum-free supplement in the differentiation medium significantly enhanced myogenesis compared to horse serum. Our findings support the possibility to create an in vitro model of contractile sarcomeric myofibrils for disease modeling and drug screening to study neuromuscular diseases.

Ultra-dense spatial-division multiplexing (SDM) is achieved by mode multiplexed technique with multiple cores in a single fiber, namely few-mode multi-core fiber. Using a 9.8-km six-mode nineteen-core fiber, we demonstrate an ultra-dense SDM transmission of 16-channels wavelength-division-multiplexed (WDM) dual-polarization quadrature phase shift keying signals, achieving a record spatial multiplicity of 114. With the help of ultra-dense Super-Nyquist WDM techniques in the 4.5-THz bandwidth of the full C-band, we demonstrate 2.05 Pbit/s transmission over 9.8-km six-mode nineteen-core fibers. In this experiment, the highest aggregate spectral efficiency of 456 bit/s/Hz is achieved.

BACKGROUND: It is often difficult to differentiate between the depressive states seen in late-life depression and late-onset Alzheimer' disease (AD) in the clinical setting. METHODS: Thirty-four outpatients were recruited, all fulfilling the criteria of aged 65 years or above, scores of 14 or more on the Hamilton depression rating scale (HAM-D), and 26 or less on the Mini-Mental State Examination (MMSE). At the initial visit, they were administered the Neurobehavioral Cognitive Status Examination (COGNISTAT). At 1 month, a diagnosis of either senile depression (n = 24) or Alzheimer' disease (n = 10) was made. RESULTS: The COGNISTAT revealed that the late-life depression group showed significantly higher scores in orientation and comprehension subtests compared with the AD group. At the study endpoint (6 months after treatment), MMSE detected significant improvements in the late-life depression group (n = 15), but no changes in the late-onset AD group (n = 7). Scores for memory, similarities, and judgment on the second COGNISTAT were significantly improved in the depressed group, whereas calculation scores deteriorated significantly in the AD group. CONCLUSION: The COGNISTAT could prove useful in differentiating late-life depression from late-onset AD, despite similar scores on MMSE.

OBJECTIVE: Treatment-resistant depression is a challenging problem in the clinical setting. Tipepidine has been used as a non-narcotic antitussive in Japan since 1959. METHODS: We administered tipepidine to 11 patients with treatment-resistant depression. Tipepidine was given for 8 weeks as an augmentation. RESULTS: Tipepidine significantly improved depression scores on the Hamilton Rating Scale for depression. Add-on treatment with tipepidine significantly improved scores on the trail making test and Rey auditory verbal learning test. However, no changes were observed in blood concentrations of stress-related hormones (adrenocorticotropic hormone, cortisol, dehydroepiandrosterone sulphate) with tipepidine augmentation. CONCLUSION: Tipepidine might be a potential therapeutic drug for treatment-resistant depression.

Standard fractionated radiotherapy for the treatment of cancer consists of daily irradiation of 2-Gy X-rays, 5 days a week for 5-8 weeks. To understand the characteristics of radioresistant cancer cells and to develop more effective radiotherapy, we established a series of novel, clinically relevant radioresistant (CRR) cells that continue to proliferate with 2-Gy X-ray exposure every 24 h for more than 30 days in vitro. We studied three human and one murine cell line, and their CRR derivatives. Guanine nucleotide-binding protein 1 (GBP1) gene expression was higher in all CRR cells than their corresponding parental cells. GBP1 knockdown by siRNA cancelled radioresistance of CRR cells in vitro and in xenotransplanted tumor tissues in nude mice. The clinical relevance of GBP1 was immunohistochemically assessed in 45 cases of head and neck cancer tissues. Patients with GBP1-positive cancer tended to show poorer response to radiotherapy. We recently reported that low dose long-term fractionated radiation concentrates cancer stem cells (CSCs). Immunofluorescence staining of GBP1 was stronger in CRR cells than in corresponding parental cells. The frequency of Oct4-positive CSCs was higher in CRR cells than in parental cells, however, was not as common as GBP1-positive cells. GBP1-positive cells were radioresistant, but radioresistant cells were not necessarily CSCs. We concluded that GBP1 overexpression is necessary for the radioresistant phenotype in CRR cells, and that targeting GBP1-positive cancer cells is a more efficient method in conquering cancer than targeting CSCs.

Differentiated embryo chondrocyte 2 (DEC2/Sharp-1/Bhlhe41), a basic helix-loop-helix (bHLH) transcription factor, has been shown to regulate the transcription of target genes by binding to their E-box elements. We identified a possible DEC2-response element (consensus E-box: CACGTG) in the promoter region of Twist1. Forced expression of DEC2 significantly repressed Twist1 promoter activity under normoxia and under hypoxia as assessed by a luciferase reporter assay. In addition, over-expression of DEC2 repressed Twist1 mRNA expression assessed by quantitative real-time PCR. Site-directed mutagenesis studies showed that mutagenesis of the consensus E-box sequence eliminated the ability of DEC2 to reduce the Twist1 promoter activity. Chromatin immunoprecipitation (ChIP) assays confirmed that the DEC2-mediated repression is primarily achieved by binding to the E-box in the Twist1 promoter. Knockdown of DEC2 by siRNA significantly attenuated the repression of Twist1 expression. DEC2 and Twist1 exhibit inversed protein expression patterns during development of mouse tongue embryo tissue. Given the fact that DEC2 protein is emerging as an important regulator in a vast array of cellular events, including cell differentiation, maturation of lymphocytes and the molecular clock, our study elucidates an important mechanism by which DEC2 regulates cellular function by modulating the expression of Twist1.

Space-division multiplexing with uncoupled multi-core fiber is a promising technology to drastically increase a fiber capacity in optical communication systems. Trans-oceanic class ultra-long-haul transmission was successfully achieved by using 7-core MCF with suppressed inter-core crosstalk. By using a combination of MCF and spectral efficient modulation format, the fiber capacity could be increased from 28.8 Tbit/s to 140 Tbit/s and a capacity-distance product exceeding 1 Exabit/s·km was obtained in 7 cores x 201 λ x 100 Gbit/s transmission over 7326 km. These results indicate that the MCF transmission will be one of promising candidates for future ultra-high capacity optical communication systems.
