RESUMO

Bisphenol AF (BPAF), as one of structural analogs of BPA, has been increasingly used in recent years. However, limited studies have suggested its adverse effects similar to or higher than BPA. In order to explore the general toxicity and genotoxicity of subacute exposure to BPAF, the novel 28-day multi-endpoint (Pig-a assay + micronucleus [MN] test + comet assay) genotoxicity evaluation platform was applied. Male rats were randomly distributed into seven main experimental groups and four satellite groups. The main experimental groups included BPAF-treated groups (0.5, 5, and 50 µg/kg·bw/d), BPA group (10 µg/kg·bw/d), two solvent control groups (PBS and 0.1% ethanol/99.9% oil), and one positive control group (N-ethyl-N-nitrosourea, 40 mg/kg bw). The satellite groups included BPAF high-dose recovery group (BPAF-HR), oil recovery group (oil-R), ENU recovery group (ENU-R), and PBS recovery group (PBS-R). All groups received the agents orally via gavage for 28 consecutive days, and satellite groups were given a recovery period of 35 days. Among all histopathologically examined organs, testis and epididymis damage was noticed, which was further manifested as blood-testis barrier (BTB) junction protein (Connexin 43 and Occludin) destruction. BPAF can induce micronucleus production and DNA damage, but the genotoxic injury can be repaired after the recovery period. The expression of DNA repair gene OGG1 was downregulated by BPAF. To summarize, under the design of this experiment, male reproductive toxicity of BPAF was noticed, which is similar to that of BPA, but its ability to induce micronucleus production may be stronger than that of BPA.

The mycotoxin altertoxin I (ATX-I) is one of secondary metabolites produced by Alternaria fungi and is frequently detected as food and feed contaminants. Little is known about the genotoxicity of the ATX-I. In order to evaluate potential genotoxicity and general toxicity of ATX-I, the novel 28-day multiendpoint (Pig-a assay + micronucleus [MN] test + comet assay) genotoxicity platform was applied. Male Sprague-Dawley (SD) rats were randomized to five groups (six rats per group), that is, a positive control group (N-ethyl-N-nitrosourea [ENU], 40 mg/kg.bw/d), two solvent control groups (PBS and corn oil), and two ATX-I-treated groups (low-dose group [1.10 µg/kg.bw/d] and high-dose group [5.51 µg/kg.bw/d]). Treatments were administered by oral gavage to male SD rats for 28 consecutive days. Histopathological damages in the liver, kidney, and spleen were observed, but without significant changes in hematological and serum biochemical parameters. Genotoxic endpoints indicated that ATX-I could cause DNA damage. To summarize, in a relatively low-dose range, ATX-I may not have direct genotoxicity in vivo but could induce liver, kidney, and spleen damage.

The three-dimensional trajectory data of vehicles have important practical meaning for traffic behavior analysis. To solve the problems of narrow visual angle in single-camera scenes and lack of continuous trajectories in 3D space by current cross-camera trajectory extraction methods, we propose an algorithm of vehicle spatial distribution and 3D trajectory extraction in this paper. First, a panoramic image of a road with spatial information is generated based on camera calibration, which is used to convert cross-camera perspectives into 3D physical space. Then, we choose YOLOv4 to obtain 2D bounding boxes of vehicles in cross-camera scenes. Based on the above information, 3D bounding boxes around vehicles are built with geometric constraints which are used to obtain projection centroids of vehicles. Finally, by calculating the spatial distribution of projection centroids in the panoramic image, 3D trajectories of vehicles are extracted. The experimental results indicate that our algorithm can effectively complete vehicle spatial distribution and 3D trajectory extraction in various traffic scenes, which outperforms other comparison algorithms.

This study investigated the concentration of cephalosporin, the resistant levels of lactose fermentative opportunistic pathogenic Enterobacteriaceae bacteria (LFOPEB) against seven antibiotics and one cephalosporin-resistant gene in cephalosporin wastewater (CPWW) treatment plant and its discharge receiving river. Although large numbers of bacteria have been removed during the CPWW treatment process, the antibiotic resistant rates of the isolates to ß-lactam antibiotics significantly increased (p = 0.032) after treatment, while the percentage of resistant LFOPEB to non-ß-lactam antibiotics did not change dramatically. Furthermore, the discharge of the effluent of CPWW treatment plant (CPWWeff) led to an obvious increase in the percentages of ß-lactam antibiotic-resistant LFOPEB and relative abundance of the blaTEM-2 gene in the downstream receiving river (RWdown) in comparison with those in the upstream receiving river (RWup). The antibiotic resistant phenotypes of isolates in the influent of CPWW treatment plant (CPWWin), CPWWeff and RWdown appeared to be seriously affected by the cephalosporin residues, which suggested that main antibiotic resistance phenotypes in antibiotic contaminated water were closely associated with its antibiotic composition. Therefore, CPWW treatment process has been proved to result in selective growth of ARB and proliferation of ARG. Besides, CPWWeff was also proved to be an important supplier of ARB and ARG to the receiving river.

The objective of this study was to investigate the degradation of nonbiodegradable organic pollutants in biologically cephalosporin pharmaceutical wastewater using different electrodes such as non-nano-scale electrode (traditional coated), nano-scale (nano-coated) electrode, and perforated electrode after biotreatment. The traditional coated electrode plate, nano-coated electrode plate, and two different perforated titanium dioxide (TiO2) electrode plates with an average pore size of 10 µm and 20 µm were chosen as the anode. The results demonstrated that traditional coated electrode, nano-scale electrode, and perforated electrode could effectively remove nonbiodegradable organic pollutants from pharmaceutical wastewater. The perforated electrode with an average pore size of 10 µm exhibited the best degradation effect with a 90 % decrease in the chemical oxygen demand (COD) (COD content reduced from 320 mg L(-1) to 32 mg L(-1)). During catalytic degradation, the electrical conductivity of pharmaceutical wastewater increased and the pH increased and finally reached equilibrium. It was also found that the perforated TiO2 electrode produced relatively large amounts of dissolved oxygen during the catalytic oxidation process, reaching above 4 mg L(-1), whereas the nano-coated electrode produced little dissolved oxygen. The biotoxicities of all wastewater samples increased firstly then decreased slightly during the electrical catalytic oxidation, but the final biotoxicities were all higher than initial ones.

In order to reduce the energy consumption of human daily movement without providing additional power, we considered the biomechanical behavior of the knee during external impedance interactions. Based on the theory of human sports biomechanics, combined with the requirements of human-machine coupling motion consistency and coordination, an unpowered exoskeleton-assisted device for the knee joint is proposed in this paper. The effectiveness of this assisted device was verified using gait experiments and distributed plantar pressure tests with three modes: "not wearing exoskeleton" (No exo.), "wearing exoskeleton with assistance " (Exo. On), and "wearing exoskeleton without assistance" (Exo. Off). The experimental results indicate that (1) This device can effectively enhance the function of the knee, increasing the range of knee movement by 3.72% (p < 0.001). (2) In the early stages of the lower limb swing, this device reduces the activity of muscles in relation to the knee flexion, such as the rectus femoris, vastus lateralis, and soleus muscles. (3) For the first time, it was found that the movement length of the plantar pressure center was reduced by 6.57% (p = 0.027). This basic principle can be applied to assist the in-depth development of wearable devices.

Furan is a widespread endogenous contaminant in heat-processed foods that can accumulate rapidly in the food chain and has been widely detected in foods, such as wheat, bread, coffee, canned meat products, and baby food. Dietary exposure to this chemical may bring health risk. Furan is classified as a possible category 2B human carcinogen by the International Agency for Research on Cancer, with the liver as its primary target organ. Hepatic fibrosis is the most important nontumoral harmful effect of furan and also an important event in the carcinogenesis of furan. Although the specific mechanism of furan-induced liver fibrosis is still unclear, it may involve oxidative stress and genetic toxicity, in which the activation of cytochrome P450 2E1 (CYP2E1) may be the key event. Thus, we conducted a study using an integrating multi-endpoint genotoxicity platform in 120-day in vivo subchronic toxicity test in rats. Results showed that the rats with activated CYP2E1 exhibited DNA double-strand breaks in D4, gene mutations in D60, and increased expression of reactive oxygen species and nuclear factor erythroid 2-related factor 2 in D120. Necrosis, apoptosis, hepatic stellate cell activation, and fibrosis also occurred in the liver, suggesting that furan can independently affect liver fibrosis through oxidative stress and genotoxicity pathways. Point of Departure (PoD) was obtained by benchmark-dose (BMD) method to establish health-based guidance values. The human equivalent dose of PoD derived from BMDL05 was 2.26 µg/kg bw/d. The findings laid a foundation for the safety evaluation and risk assessment of furan and provided data for the further construction and improvement of the adverse outcome pathway network in liver fibrosis.

Human body enhancement is an interesting branch of robotics. It focuses on wearable robots in order to improve the performance of human body, reduce energy consumption and delay fatigue, as well as increase body speed. Robot-assisted equipment, such as wearable exoskeletons, are wearable robot systems that integrate human intelligence and robot power. After careful design and adaptation, the human body has energy-saving sports, but it is an arduous task for the exoskeleton to achieve considerable reduction in metabolic rate. Therefore, it is necessary to understand the biomechanics of human sports, the body, and its weaknesses. In this study, a lower limb exoskeleton was classified according to the power source, and the working principle, design idea, wearing mode, material and performance of different types of lower limb exoskeletons were compared and analyzed. The study shows that the unpowered exoskeleton robot has inherent advantages in endurance, mass, volume, and cost, which is a new development direction of robot exoskeletons. This paper not only summarizes the existing research but also points out its shortcomings through the comparative analysis of different lower limb wearable exoskeletons. Furthermore, improvement measures suitable for practical application have been provided.

Alternariol (AOH) is one of the toxins of Alternaria, and it has been widely detected in a variety of foods. It has been reported to be cytotoxic, dermally toxic, genotoxic, and potentially carcinogenic in vitro. However, in vivo toxicity data are lacking. This study used a novel in vivo 28-day multi-endpoint (Pig-a assay + micronucleus test + comet assay) genotoxicity evaluation system to evaluate the general toxicity and genotoxicity of AOH. A total of 42 male Sprague-Dawley rats were randomly distributed into three AOH-treated groups (5.51, 10.03, and 22.05 µg/kg bw), one AOH high-dose recovery group (AOH-HR, 22.05 µg/kg bw), one positive control group (N-ethyl-N-nitrosourea, 40 mg/kg bw), and two vehicle control groups (corn oil and PBS). Treatments were administered by oral gavage for 28 consecutive days. Histopathological lesions were observed in the liver, kidney, and spleen in all AOH-treated groups. No statistical difference was found in each genotoxicity index within 28 days in the AOH-treated groups compared with those in the corn oil group. On day 42, in the AOH-HR group, the rate of Pig-a mutant phenotype reticulocytes (RETCD59-) significantly increased. On day 56, both RETCD59- and the rate of Pig-a mutant phenotype erythrocytes (RBCCD59-) were significantly reduced. These findings indicated that AOH might cumulatively induce genetic mutations.

Alternariol monomethyl ether (AME), a typical Alternaria toxin, has often been detected in grains. We have measured the general toxicity and genotoxicity of AME with a 28-day multi-endpoint (Pig-a assay + in vivo micronucleus [MN] test + comet assay) platform. Male Sprague-Dawley rats were administered AME (1.84, 3.67, or 7.35 µg/kg body weight/day), N-Ethyl-N-nitrosourea (40 mg/kg body weight/day), or corn oil by gavage for 28 consecutive days. Another group (AME-high-dose + recovery) was maintained for a further 14 days after the end of the AME administration. Hematology and serum biochemistry results suggested that AME might compromise the immune system. The histopathology results indicated that AME can cause liver (inflammatory cell infiltration, steatosis, and edema), kidney (renal glomerular atrophy), and spleen (white pulp atrophy) damage. The genotoxicity results showed that AME can induce gene mutations, chromosome breakage, and DNA damage, but the effects were diminished after the recovery period. According to point-of-departure analysis (BMDL10), the risk to the population of exposure to AME cannot be ignored and further assessment is needed.

Opening the blood brain barrier (BBB) under imaging guidance may be useful for the treatment of many brain disorders. Rapidly applied magnetic fields have the potential to generate electric fields in brain tissue that, if properly timed, may enable safe and effective BBB opening. By tuning magnetic pulses generated by a novel electropermanent magnet (EPM) array, we demonstrate the opening of tight junctions in a BBB model culture in vitro, and show that induced monophasic electrical pulses are more effective than biphasic ones. We confirmed, with in vivo contrast-enhanced MRI, that the BBB can be opened with monophasic pulses. As electropermanent magnets have demonstrated efficacy at tuning B0 fields for magnetic resonance imaging studies, our results suggest the possibility of implementing an EPM-based hybrid theragnostic device that could both image the brain and enhance drug transport across the BBB in a single sitting.

On the basis of analyzing the movement law of 3D circular braided yarn, the three-cell model of 3D five-direction circular braiding composite material is established. By analyzing the node position relationship in various cell models, the calculation formulas of braiding angle, cell volume, fiber volume and fiber volume content in various cell models are obtained. It is found that there are four different braiding angles in four internal cells, and the braiding angles in internal cells gradually increase from inside to outside. The braiding angles of upper and lower surface cells are approximately equal. With the increase of the length of the knuckles, the braiding angles of each cell decrease, and the braiding angles of the four inner cells decrease greatly, while the braiding angles of upper and lower surfaces decrease slightly. The results of parametric analysis showed that with the increase of the length of the knuckles and the inner diameter of cells, the mass of cells increased proportionally, while the total fiber volume content of cells decreased. With the increase of braiding yarn number and axial yarn number, the unit cell mass decreases in direct proportion, and the unit cell total fiber volume content increases. Through the research results of this paper, the geometrical characteristics of the cell model under different braided parameters can be obtained, which greatly improves the analysis efficiency.

Deoxynivalenol (DON), zearalenone (ZEN), and fumonisin B1 (FB1), as the main mycotoxins contaminating rice, often coexist in food. Thus, we have measured the genotoxicity of the three rice fungal contaminants, singly and in different combinations, with a 28-day multi-endpoint (Pig-a assay + in vivo micronucleus [MN] test + comet assay) genotoxicity platform. Male Sprague-Dawley rats received the agents orally via gavage for 28 consecutive days, before performing the abovementioned tests. Results indicated that low dose of a single mycotoxin did not show significant genotoxicity. However, some of these mycotoxins in combination induced significant genotoxicity in the peripheral blood and tissues, at sacrifice. In the peripheral blood, the binary combination of DON and FB1 significantly induced MN. In the liver, ZEN might aggravate the DNA-damaging effects of DON and FB1. Therefore, the genotoxicity of sub-chronic exposure to mycotoxins in combination cannot be ignored.

Small soft robotic systems are being explored for myriad applications in medicine. Specifically, magnetically actuated microrobots capable of remote manipulation hold significant potential for the targeted delivery of therapeutics and biologicals. Much of previous efforts on microrobotics have been dedicated to locomotion in aqueous environments and hard surfaces. However, our human bodies are made of dense biological tissues, requiring researchers to develop new microrobotics that can locomote atop tissue surfaces. Tumbling microrobots are a sub-category of these devices capable of walking on surfaces guided by rotating magnetic fields. Using microrobots to deliver payloads to specific regions of sensitive tissues is a primary goal of medical microrobots. Central nervous system (CNS) tissues are a prime candidate given their delicate structure and highly region-specific function. Here we demonstrate surface walking of soft alginate capsules capable of moving on top of a rat cortex and mouse spinal cord ex vivo, demonstrating multi-location small molecule delivery to up to six different locations on each type of tissue with high spatial specificity. The softness of alginate gel prevents injuries that may arise from friction with CNS tissues during millirobot locomotion. Development of this technology may be useful in clinical and preclinical applications such as drug delivery, neural stimulation, and diagnostic imaging.

Hyperuricemia, as a critical risk factor for various adverse clinical outcomes, shows a trend of increasing prevalence among young-aged population. Dietary adjuvant therapy by function foods, such as tart cherry, is promising. Thus, effects of tart cherry powder specialized in hyperuricemia were explored via establishing a hyperuricemia model in Sprague Dawley rats by cotreatment with oteracil potassium and adenine. The results indicated that low dose of tart cherry powder (0.17 g/kg·bw) showed effects on hyperuricemia by slightly decreasing serum uric acid and improving kidney injury, whereas high dose of tart cherry powder (0.50 g/kg·bw) could merely alleviate kidney injury. Meanwhile, adenosine deaminase activity rather than xanthine oxidase activity was affected at low dose, which reveals low dose of tarty cherry powder may be beneficial to hyperuricemia through reduction of ADA activity, and its reported potentials on antioxidation or anti-inflammation provide clues for further study.

Nuclear quadrupole resonance (NQR) is commonly used to characterize solid materials containing quadrupolar nuclei. For example, NQR is a promising technique for detecting plastic explosives and other forbidden substances as well as for authenticating pharmaceutical products. Spatially-resolved NQR measurements are of particular interest for enabling automated sample positioning, evaluation of sample heterogeneity, and chemometric authentication of objects. This paper proposes a rapid "single-shot" method for spatially-resolved NQR with the potential to benefit such applications. The proposed method takes advantage of the fact that certain NQR relaxation rates are field-dependent: the observed field dependence is used to convert relaxation time distributions measured in a static field gradient (estimated via Laplace inversion of time-domain data) into spatial distributions. The method was validated using 35Cl and 37Cl NQR of sodium chlorate and other compounds. Effective spatial resolution was also improved by using machine learning (ML) to classify the measured spatial distributions. In particular, experimental results demonstrate accurate ML-based classification of 3D-printed objects containing arbitrary binary distributions of sodium chlorate. Such distributions can thus be used as NQR-based "embedded barcodes" for authenticating high-value objects.

An investigation to study the distribution and persistence of cephalosporins in the cephalosporin producing wastewater was carried out in this paper. The target cephalosporins included ceftriaxone (CRO), cefalexin (CEF), cefotaxime (CTX), cefazolin (CZO), cefuroxime (CXM), cefoxitin (CFX) and cefradine (CF). A rapid and reliable detection method for cephalosporins was established based on solid phase extraction and ultra-performance liquid chromatography - tandem mass spectrometry. In the cephalosporin producing wastewater effluent (CPWWeff), the limit of quantification for the targets ranged from 27.5ng/L to 131.8ng/L, and the recoveries for all of the analytes ranged from 73% to 102%. The mean concentrations of the seven cephalosporins were 12.85-141.55µg/L and 0.05-24.38µg/L in cephalosporin producing wastewater influent and effluent, respectively. Although high removal efficiencies were achieved for the cephalosporins (78.8-99.7%), up to 1.9kg of cephalosporins was discharged per day from the investigated C-WWTP. The degradation processes of CRO, CEF, CZO and CXM followed first-order kinetics in CPWWeff under all of the testing conditions. The degradation rates of tested cephalosporins were accelerated by high temperature and light. Persistence of CXM was the highest among the four tested cephalosporins in CPWWeff.

A novel sonoelectrochemical catalytic oxidation-driven process using a nanocoated electrode to treat methylene blue (MB) wastewater was developed. The nano-scale (nanocoated) electrode generated more hydroxyl radicals than non-nano-scale (non-nanocoated) electrodes did. However, hydroxyl radicals were easily adsorbed by the nanomaterial and thus were not able to enter the solution. Supersonic waves were found to enhance the mass-transfer effect on the nanocoated electrode surface, resulting in rapid diffusion of the generated hydroxyl radicals into the solution. In solution, the hydroxyl radicals then reacted with organic pollutants in the presence of ultrasonic waves. The effect of the nanocoated electrode on the MB wastewater treatment process was enhanced by ultrasound when compared to the non-nanocoated electrode used under the same conditions. The synergy of the nanocoated electrode and ultrasonic waves towards MB degradation was then studied. The optimum operating conditions resulted in a 92% removal efficiency for TOC and consisted of a current of 600 mA, an ultrasound frequency of 45 kHz, and a supersonic power of 250 W. The mechanism of ultrasound enhancement of the nanocoated electrode activity with respect to MB treatment is discussed. The reaction intermediates of the sonoelectrochemical catalytic oxidation process were monitored, and degradation pathways were proposed. The sonoelectrochemical catalytic oxidation-driven process using nanocoated electrodes was found to be a very efficient method for the treatment of non-biodegradable wastewater.

The toxicity of pharmaceutical wastewaters has recently been the focus of the public in China. This study aimed to evaluate the conventional pollution parameters and toxicities of different raw and treated pharmaceutical wastewaters to algae Scenedesmus obliquus and bacteria Vibrio fischeri. Wastewater samples were collected from 16 pharmaceutical wastewater treatment plants in China. The results of the conventional parameters analysis indicated that the total suspended solids, chemical oxygen demand (COD), ammonia (NH3-N), and total phosphorus (TP) were largely removed after treatment. Pharmaceutical effluents were mainly polluted with organics and phosphorus as indicated by the average COD (388 mg/L) and TP (3.16 mg/L) concentrations. The toxicity test results indicated that the influent samples were toxic to both test species. Although the toxicities could be remarkably reduced after treatment, 10 out of the 16 effluent samples exceeded the acute toxicity discharge limit of the Chinese national standards. Spearman rank correlation coefficients indicated a significantly positive correlation between the toxicity values of S. obliquus and V. fischeri. Compared with S. obliquus, V. fischeri detected more pharmaceutical effluent samples with toxicities. Meanwhile, the toxicity indicators were significantly and positively correlated with the COD and NH3-N concentrations based on a Spearman rank correlation analysis.

Acute toxicity of wastewater from 5 nodes of technological process in the pharmaceutical factory sewage treatment station was studied by luminescent bacteria tests. The EC50, TUa and LID of the wastewater in underground regulating tanks was 3.44%, 29 and 625, respectively, indicating the water was extremely/highly toxic; for the wastewater in surface regulating tanks, the EC50, TUa and LID was 2.46%, 41 and 244, respectively, also extremely/highly toxic; for the wastewater in middle sediment tanks, the EC50 > 100% and LID was 10, which was moderately toxic; for the wastewater in secondary sediment tanks and the final effluents, the EC50 was above 100% and LID was 1, with no observed toxicity. The results indicated that the existing treatment process effectively reduced the acute toxicity of the pharmaceutical wastewater to luminescent bacteria, the effluents showed no observed toxicity to luminescent bacteria, which was lower than the relative effluent limits of pharmaceutical wastewater. The wastewater in lower concentration did not inhibit the luminosity, but enhanced the luminosity.
