RESUMO

Flexible engineering of the complex shapes of the surface nanoscale axial photonics (SNAP) bottle microresonators (SBMs) is challenging for future nanophotonic technology applications. Here, we experimentally propose a powerful approach for the one-step fabrication of SBMs with simultaneous negative and positive radius variations, exhibiting a distinctive "bump-well-bump" profile. It is executed by utilizing two focused and symmetrical CO2 laser beams exposed on the fiber surface for only several hundred milliseconds. The spectral characteristics of different eigenmodes are analyzed, providing deep insights into the complex physical processes during the CO2 laser exposure. The shapes of the SBMs can be flexibly adjusted by the exposure time, laser power, and applied pre-strains. As a proof of this technique, the developed approach enables the efficient production of a bat SBM, ensuring a uniform field amplitude of the bat mode over the length exceeding 120 µm with 7% deviation. Our proposed technique provides a powerful technique for the efficient fabrication of SBMs with predetermined shapes, laying the groundwork for its applications on microscale optical signal processing, quantum computing, and so on.

RESUMO

A simple generic method for enhancing extracellular protein yields in engineered bacteria is still lacking. Here, we demonstrated that phage-encoded holin can be used to export proteins to the extracellular medium in both Gram-negative Escherichia coli and -positive Lactococcus lactis. When a putative holin gene LLNZ_RS10380 annotated in the genome of L. lactis NZ9000 (hol380) was recombinantly expressed in E. coli BL21(DE3), the Hol380 oligomerized up to hexamer in the cytoplasmic membrane, yielding membrane pore to allow the passage of cytosolic ß-galatosidase (116 kDa), whose extracellular production reached 54.59 U/µl, accounting for 76.37% of the total activity. However, the overexpressed Hol380 could not release cytosolic proteins across the membrane in L. lactis NZ9000, but increased the secretory production of staphylococcal nuclease to 2.55-fold and fimbrial adhesin FaeG to 2.40-fold compared with those guided by signal peptide Usp45 alone. By using a combination of proteomics and transcriptional level analysis, we found that overexpression of the Hol380 raised the accumulation of Ffh and YidC involved in the signal recognition particle pathway in L. lactis, suggesting an alternative road participating in protein secretion. This study proposed a new approach by expressing holin in bacterial cell factories to export target proteins of economic or medical interest.

Dissecting the genetic basis of yield traits in hybrid populations and identifying the candidate genes are important for molecular crop breeding. In this study, a BC1F3:4 population, the line per se (LPS) population, was constructed by using elite inbred lines Zheng58 and PH4CV as the parental lines. The population was genotyped with 55,000 SNPs and testcrossed to Chang7-2 and PH6WC (two testers) to construct two testcross (TC) populations. The three populations were evaluated for hundred kernel weight (HKW) and yield per plant (YPP) in multiple environments. Marker-trait association analysis (MTA) identified 24 to 151 significant SNPs in the three populations. Comparison of the significant SNPs identified common and specific quantitative trait locus/loci (QTL) in the LPS and TC populations. Genetic feature analysis of these significant SNPs proved that these SNPs were associated with the tested traits and could be used to predict trait performance of both LPS and TC populations. RNA-seq analysis was performed using maize hybrid varieties and their parental lines, and differentially expressed genes (DEGs) between hybrid varieties and parental lines were identified. Comparison of the chromosome positions of DEGs with those of significant SNPs detected in the TC population identified potential candidate genes that might be related to hybrid performance. Combining RNA-seq analysis and MTA results identified candidate genes for hybrid performance, providing information that could be useful for maize hybrid breeding.

A novel coumarin-thiourea conjugate was synthesized facilely. It served as a fluorescent turn-on chemosensor for selective detection of Hg2+ ion over other common competitive metal ions including Li+, Na+, K+, Ag+, Cu2+, Fe2+, Zn2+, Co2+, Ni2+, Mn2+, Sr2+, Ca2+, Mg2+, Al3+, Cr3+ and Fe3+ ions based on the Hg2+-promoted desulfurization and cyclization reactions. Addition of Hg2+ ion to the sensor solution in 2:8 EtOH/H2O induced a hypsochromic shift of the UV-Vis absorption band from 360 nm to 340 nm accompanying distinct enhancement in the absorption intensity while addition of other metal ions failed to bring about substantial change in the absorption spectra. Addition of Hg2+ to the sensor solution also caused marked increase in the fluorescence emission intensity and most common competitive metal ions did not interfere with the selective sensing of Hg2+ ion by the sensor. The detection limit of Hg2+ ion by the probe was calculated to be 1.46 × 10-7 M and the probe could be utilized for selective detection of Hg2+ ion by fluorescence turn-on mode over a broad pH range of 1-11.

The black layer (BL) is traditionally used as an indicator for kernel harvesting in maize, as it turns visibly dark when the kernel reaches physiological maturity. However, the molecular roles of BL in kernel development have not been fully elucidated. In this work, microscopy images showed that BL began to appear at a growth stage earlier than 10 days after pollination (DAP), and its color gradually deepened to become dark as the development period progressed. Scanning electron microscopy observations revealed that BL is a tissue structure composed of several layers of cells that are gradually squeezed and compressed during kernel development. Laser-capture microdissection (LCM) was used to sample BL and its neighboring inner tissue, basal endosperm transfer layer (BETL), and outer tissue, inner epidermis (IEP), from 20 DAP of kernels. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry profiling (MALDI-TOF MS profiling) detected 41, 104, and 120 proteins from LCM-sampled BL, BETL, and IEP, respectively. Gene ontology (GO) analysis indicated that the 41 BL proteins were primarily involved in the response to stress and stimuli. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis found that the BL proteins were enriched in several defense pathways, such as the ascorbate and aldarate metabolic pathways. Among the 41 BL proteins, six were BL-specific proteins that were only detected from BL. Annotations of five BL-specific proteins were related to stress responses. During kernel development, transcriptional expression of most BL proteins showed an increase, followed by a decrease, and reached a maximum zero to 20 DAP. These results suggest a role for BL in stress responses for protecting filial tissue against threats from maternal sides, which helps to elucidate the biological functions of BL.

A new spiropyran derivative acts as colorimetric and fluorescent sensor for metal ions. Addition of Cu2+, Al3+, Cr3+ and Ce3+ to the sensor solution brings about marked increase in the absorption intensities while addition of other metal ions (Na+, K+, Li+, Sr2+, Hg2+, Zn2+, Co2+, Ni2+ and Ag+) fails to cause substantial change in the UV-Vis spectra. The sensor can be used for detection of Hg2+, Cu2+, Ce3+, Cr3+ and Al3+ ions by the naked eye and by fluorescence enhancement. The detection limit of Cu2+ and Hg2+ ions by the sensor is estimated to be 10 and 14 µM, respectively. Job's plot analysis on the sensor-Cu2+ complexation reveals the formation of a 2:1 stoichiometric complex between the sensor and Cu2+ ion.

The exploration of polysaccharides from microorganisms is of great importance. In this study, a new type of exopolysaccharide excreted by Fusarium merismoides A6 (FM-EPS) was isolated, and the extraction conditions were optimized using a response surface methodology (RSM). The extraction temperature at 0 °C, a precipitation time of 7.83 h, and an ethanol precipitation concentration of 77.64% were predicted and proved to be the best extraction conditions with the maximum extraction yield of 0.74 g/mL. Then, two fractions of F. merismoides A6 exopolysaccharides (FM-EPS1 and FM-EPS2) were obtained through DEAE Sepharose fast flow column chromatography. As indicated by monosaccharide composition analysis, both fractions mainly consisted of mannose, glucose, galactose, and ribose, with an average molecular weight of 5.14 × 104 and 6.50 × 104 g/mol, respectively. FT-IR and NMR spectroscopy indicated the FM-EPSs had both α- and ß-glycosidic bonds. Moreover, the determination of antioxidant and antiproliferative activities in vitro proved that FM-EPSs had good antioxidant activities and antiproliferation activities. FM-EPS1 showed stronger antioxidant activities than FM-EPS2. FM-EPS2 showed antiproliferation activities on HeLa and HepG2 cells, while FM-EPS1 had no obvious antiproliferative activity. Therefore, FM-EPSs could be explored as potential antioxidant and anticancer agent applied in food, feed, nutraceutical, pharmaceutical, cosmetics, and chemical industries.

A new near-infrared (NIR) probe based on a coumarinyl ligand (CL) was designed and synthesized. The probe CL can be used for simultaneous fluorescent turn-on and colorimetric detection of Hg2+ and Ag+ in ethanol/water medium. Colorless solution of probe CL changed to light yellow or dark yellow after addition of Hg2+ or Ag+ ions. Meanwhile the maximum absorption band shifted from 379 nm to 404 nm and the intensity increased enormously (for Hg2+) or moderately (for Ag+). Probe CL displayed an extraordinarily large Stokes shift of 316 nm and addition of Hg2+ or Ag+ to probe CL induced enhancement in the intensity of fluorescence emission at 695 nm by 15 or 8 fold. The detection limit of CL for Hg2+ and Ag+ ions is 0.83 and 8.8 µM, respectively. The applicable pH for sensing Hg2+ by probe CL is in a broad range of 2-12. Application of probe CL for in vitro U87MG cell imaging to detect Hg2+ ions was confirmed.

Genomic prediction (GP) across different populations and environments should be enhanced to increase the efficiency of crop breeding. In this study, four populations were constructed and genotyped with DNA chips containing 55,000 SNPs. These populations were testcrossed to a common tester, generating four hybrid populations. Yields of the four hybrid populations were evaluated in three environments. We demonstrated by using real data that the prediction accuracies of GP across structured hybrid populations were lower than those of within-population GP. Including relatives of the validation population in the training population could increase the prediction accuracies of GP across structured hybrid populations drastically. G × E models (including main and genotype-by-environment effect) had better performance than single environment (within environment) and across environment (including only main effect) GP models in the structured hybrid population, especially in the environment where yields had higher heritability. GP by implementing G × E models in two cross-validation schemes indicated that, to increase the prediction accuracy of a new hybrid line, it would be better to field-test the hybrid line in at least one environment. Our results would be helpful for designing training population and planning field testing in hybrid breeding.

A fluorescent and colorimetric probe L1 based on a simple coumarin derivative for detection of Hg2+ and F- ions was developed. Upon addition of Hg2+ and F- ions, L1 underwent desulfurization and desilylation, respectively, to induce marked increase in the fluorescence intensity and sharp color change from light yellow to dark purple and light brown, respectively. Probe L1 could be used for sensing and for quantitative measurement of Hg2+ and F- ions by both UV-vis and fluorescence spectra. The bifunctional probe exhibited a high selectivity over other competitive cations and anions and could be used in both organic and aqueous media over a wide pH range.

Extraction polysaccharide from microorganism is a research hotspot. In this work, a new type of water-soluble exopolysaccharides (EPS) was isolated from Bacillus licheniformis. Firstly, response surface methodology (RSM), based on a three-level, three-factor, was used to determine optimum conditions for EPS extraction. And RSM analysis indicated optimum condition was at the temperature of 8 °C for 10.44 h with ethanol at a concentration of 79.22% (v/v), the maximum yield of EPS was 3.07 g/mL. Secondly, EPS were seperated using DEAE-Sepharose Fast Flow column chromatography and acquired two polysaccharide fractions, BL-P1 and BL-P2. BL-P1 had larger molecular weight than BL-P2 from structural analyses, because of higher content of mannose, ribose, glucuronic acid, galactose, arabinose and fructose in BL-P2. Moreover, the characterization of BL-P1 and BL-P2 was investigated with Fourier transform infrared spectroscopy (FT-IR) and nuclear magnetic resonance (NMR) spectroscopy, the results indicated that EPS was mainly composed of→3)-α-d-Galp-(1→, →3,5)-α-l-Araf-(1→, →3)-ß-d-Glcp-(1→, ß-d-Glcp-(1 → and→4)-ß-l-Fucp-(1 → 4)-ß-d-Xylp-(1 → 4)-α-l-Rhap (1 → 3) -ß-d-Manp-(4 → residues. In vitro antioxidant activity assay, EPS exhibited potent quenching capacities on hydroxyl and 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals in a dose-dependent manner. Furthermore, BL-P2 had higher activity than BL-P1 in inhibiting α-amylase and α-glucosidase, which would have potential to be applied in nutraceutical and pharmaceutical industries.

The popularity of genomic selection (GS) has increased owing to its prospects in commercial breeding. It is necessary to enhance GS to increase its efficiency. In this study, a maize BC1F3:4 population, consisting of 481 families, was evaluated for days to anthesis in four environments, and genotyped with DNA chips including 55,000 single nucleotide polymorphisms (SNPs). This population was used to investigate whether GS could be enhanced by borrowing information from the genetic basis and genotype-by-environment (G × E) interaction. The results showed that: 1) fitting the top four large-effect SNPs as fixed effects could increase prediction accuracy, including three minor-effect SNPs explaining less than 10% phenotypic variance; 2) the increase of prediction accuracy when fitting large-effect SNPs as fixed effects was related to the decrease of genetic variance; 3) generally, the GS model fitting large-effect SNPs as fixed effects and G × E component enhanced GS. Therefore, we propose fitting large-effect markers as fixed effects and G × E effect for crop breeding projects in order to obtain accurately predicted phenotypic data and conduct efficient selection of desired plants.

A new fluorescent chemosensor based on calix[4]arene bearing four iminoquinoline subunits on the upper rim was conveniently synthesized, which showed a remarkable enhanced fluorescent intensity in the presence of Cu(2+) ion and a high selectivity toward Cu(2+) ion over a wide range of tested metal ions in acetonitrile.

A new approach to the synthesis of meta-substituted enantiopure inherently chiral calix[4]arenes by introducing L-Boc-proline as dual functions of the chiral auxiliary has been described. Moreover, the absolute configurations of the enantiomers were determined by CD spectra, X-ray crystallographic analysis, and the chemical derivative method.
