RESUMO

Pyrethroids are synthetic organic insecticides. Deltamethrin, as one of the pyrethroids, has high insecticidal activity against pests and parasites and is less toxic to mammals, and is widely used in cities and urban areas worldwide. After entering the natural environment, deltamethrin circulates between solid, liquid and gas phases and enters organisms through the food chain, posing significant health risks. Increasing evidence has shown that deltamethrin has varying degrees of toxicity to a variety of organisms. This review summarized worldwide studies of deltamethrin residues in different media and found that deltamethrin is widely detected in a range of environments (including soil, water, sediment, and air) and organisms. In addition, the metabolism of deltamethrin, including metabolites and enzymes, was discussed. This review shed the mechanism of toxicity of deltamethrin and its metabolites, including neurotoxicity, immunotoxicity, endocrine disruption toxicity, reproductive toxicity, hepatorenal toxicity. This review is aim to provide reference for the ecological security and human health risk assessment of deltamethrin.

RESUMO

The yak (Bos grunniens) is a species adapted to the hypoxic environment in the plateau area. The heart is a hypoxia-sensitive organ involved in this adaptation. Herein, we used single-cell RNA-seq technology and clustering to determine the presence of 11 cell populations in the yak heart. We analyzed gene expression differences and expression patterns in each cell subpopulation at different altitudes. The cells related to altitude changes are mainly smooth muscle cells and vascular endothelial cells. Of the four transcription factors (TFs, MEF2B, FOXP4, ARID5A, and HES4) found in smooth muscle cells, only MEF2B was specifically expressed in vascular smooth muscle cells. Three key TFs (HNF1B, DMRTA1, and ARNTL2) were also found in the cardiomyocyte module. Compared with data extracted from low-altitude yak, we observed that the high altitude yak has enhanced contraction and relaxation of vascular smooth muscle cells and an increased metabolic level of cardiomyocytes. These may be strategies for the yak to adapt to high-altitude hypoxia environments.

RESUMO

HmsB, a temperature-dependent sRNA, promotes biofilm formation by Yersinia pestis, but whether its own expression is regulated by other regulators is still poorly understood. RovM is a global regulator that activates biofilm formation but represses the virulence of Y. pestis. In this work, the results of primer extension, quantitative real-time PCR (qRT-PCR), and LacZ fusion demonstrated that RovM was able to activate hmsB expression. However, the results of electrophoretic mobility shift assay (EMSA) showed that His-RovM did not bind to the upstream DNA region of hmsB. Thus, RovM may exert its regulatory action on hmsB expression in an indirect manner. The data presented here enriched the content of the regulatory circuits that control gene expression in Y. pestis.

RESUMO

Background and Aim: Leptin (LEP) is an autocrine and paracrine factor produced by the fat pad and acinar epithelial cells of the breast. This study aimed to investigate the effects of LEP on yak mammary epithelial cells (YMECs) and the expression of STAT3. In addition, we evaluated the possible effects of prolactin (PRL) on the function of LEP. Materials and Methods: The YMECs were treated with 0, 50, 100, 200, 400, and 800 ng/mL LEP for 48 h in the absence of PRL and the presence of 500 ng/mL PRL. The growth activity of YMECs was measured using the cell counting kit-8 assay. The changes in the lactation signaling pathway-related factor STAT3 were detected at the mRNA, protein, and protein phosphorylation levels using the reverse transcriptase-quantitative polymerase chain reaction and Western blotting. To explore whether LEP affects the activation of STAT3 through JAK2/JAK3 in YMECs, the JAK2/3 signaling pathway inhibitor AG490 was used at a fixed concentration of LEP. Results: Each concentration of LEP significantly promoted the expression of STAT3 mRNA (p < 0.05) in YMECs in the presence of PRL. In the absence of PRL, all concentrations of LEP were found to inhibit the expression of the STAT3 protein (p < 0.05). The expression of the STAT3 protein in YMECs was found to first increase followed by a decrease with an increase in the concentration of LEP. In addition, the phosphorylation level of STAT3 increased in all groups, except the 100 ng/mL concentration group. The STAT3 phosphorylation trend and protein expression were different, such that the level of protein phosphorylation was higher than that of the STAT3 protein (p < 0.05). The addition of AG490 reduced the expression of the STAT3 mRNA, STAT3 protein, and STAT3 phosphorylation in the LEP and LEP + PRL groups. Conclusion: Altogether, the results indicated that different concentrations of LEP exerted varying effects on the growth of YMECs and the expression of STAT3, and the activity of STAT3 was primarily activated by JAK2. The addition of LEP can effectively inhibit the downregulation of the JAK2/STAT3 signal pathway by AG490, mitigate its inhibitory effect on the proliferation of YMECs, and reduce apoptosis. We believe that these findings will provide a theoretical and experimental basis for future research in this field.

RESUMO

Background: Femoral neck fracture (FNF) is a commonly encountered injury in orthopedic practice, and many studies have been conducted in this field. However, no bibliometric studies regarding the global research trend concerning FNF have been performed. This study aims to analyze the knowledge framework, research hotspots, and theme trends in the field of FNF research. Methods: The scientific outputs related to FNF from 1994 to 2021 were retrieved from the Web of Science Core Collection. Three bibliometric tools were used for this study. The main analyses include publication and citation counts, contributions of countries, institutions, authors, funding agencies and journals, and clustering of keywords. Results: In total, 3,553 articles were identified. The annual publication counts of FNF showed an ascending tendency as a whole. The United States has the most prominent contributions, with the most number of publications and the highest H-index. Karolinska Institutet devoted the most in this domain. Professors Bhandari M, Schemitsch EH, Frihagen F, Parker MJ, and Rogmark C were the core authors in this field. The most productive journal was Injury International Journal of the Care of the Injured. Keywords were divided into four clusters: epidemiology and mortality, fracture prevention, internal-fixation and risk factors, and hip replacement. A trend of balanced and diversified development existed in these clusters. Keywords with the ongoing bursts, including "outcome," "reoperation," "complication," "revision," "displaced intracapsular," "fracture," and "adult," are considered as the research hotspots in the future and deserve more attention. Conclusions: The management of FNF in young patients is drawing more attention from orthopedic surgeons, and it is expected that these research topics may continue to be the research hotspots and focus in the near future.

RESUMO

Forms distribution and ecotoxicity of sulfonamides in root-soil interface of maize were studied by chemical analysis and luminescent bacteria toxicity assay. The results indicated that the water soluble residues were the main form in rhizosperic soils, and the organic solvent extractable residues were the main form in far-rhizosperic soils. The three forms of sulfonamides showed relatively lower toxicity to luminescent bacteria with the relative inhibition rates less than 30%. The toxicity of water soluble residues was lower than that of the organic solvent extractable residues, and the bound residues showed the lowest toxicity to luminescent bacteria. The contents and relative inhibition rates of water soluble residues in rhizosperic soils were higher than those in far-rhizosperic soils, while the contents and relative inhibition rates of organic solvent extractable and bound residues in rhizosperic soils were lower than those in far-rhizosperic soils.

RESUMO

Soil and vegetable samples were collected from 13 different livestock farms of different sizes in Xinxiang of China, and the residues of three sulfonamides including sulfadiazine, sulfamonomethoxine, and sulfamethoxazole were analyzed by HPLC with a fluorimetric detector, The results indicated that the total concentration ranges of the three sulfonamides in soil and vegetable were 7.60-176.26 µg · kg⁻¹ and ND-32, 70 µg · kg⁻¹, respectively. The mean concentrations were 70.73 µg · kg⁻¹ and 7.08 µg · kg⁻¹ for soil and vegetables. The residue levels in soil were all lower than the ecotoxic effect trigger value (100 µg · kg⁻¹) set by the Veterinary Medicine International Coordination Commission, indicating the low risk for organisms in soil. The concentrations of three sulfonamides varied significantly in different kinds of vegetables and were all lower than the acceptable daily intake values [50 µg · (kg ·d)⁻¹] set by Joint FAO/WHO Expert CommIttee on Food Additives. But we cannot neglect the potential ecotoxicity and resistance for human via food chain.
