RESUMO

A new porphyrin-based sensing platform in hexadecyl trimethyl ammonium bromide (CTAB) microemulsion is developed for highly sensitive detection of theophylline. In this sensing system, the zinc-porphyrin-cinnamic acid conjugate (Zn-TPPCA) works as fluorescence probe while theophylline can decrease fluorescence intensity of the probe. Further studies indicate the linear relationship between the fluorescence quenching value and the concentration of theophylline within a given range. And the introduction of CTAB microemulsion can greatly enhance sensibility and stability of this detecting system and facilitate the detection of theophylline. On the basis above, a highly sensitive sensing platform for theophylline is created with a low limit of detection (LOD) of 0.0083 µg mL-1 under the optimal detection conditions. And further application of this method in determination of commercially available theophylline preparation shows excellent results. Subsequent studies on quenching mechanism indicate that static quenching appears between Zn-TPPCA and theophylline. Therefore, this work provides not only a highly sensitive method for determination of theophylline but also further evidence for creation of biosensors for drugs with porphyrin derivatives.

In this work, 3-Phenyl-N-[4-(10,15,20-triphenyl-porphyrin-5-yl)-phenyl]- acrylamide (TPPCA) was synthesized for the determination of metronidazole (MTZ). It was found that the type of fluorescence quenching was static quenching determined by Stern-Volmer plot and UV absorption spectroscopy, and thermodynamic related parameters were also obtained. Furthermore, the corresponding measurement conditions: the acidity of the system, the type of surfactant, the concentration of TPPCA and the sequence of reagent addition were optimized. Under the optimal experimental conditions, the linear range of MTZ was determined to be 0.01-0.20 µg mL-1, and the limit of detection (LOD) was 0.004 µg mL-1. Importantly, this report provides a simple, fast, and sensitive probe for the determination of MTZ in pharmaceutical practice.

A series of novel porphyrin-cinnamic acid (porphyrin/CA) conjugates (4a-4c) have been synthesized by condensation of 5-(4-Hydroxyphenyl)-10,15,20-triphenyl- porphyrin (1) with different substituted cinnamic acids (CAs) through an alkyl linker due to the biological activities of CAs and the application of porphyrins in photodynamic therapy (PDT) of cancer. Their related zinc (II) complexes (5a-5c) were also prepared. These novel compounds have been fully characterized by 1H NMR, Infrared (IR), Mass spectra (Ms) and Elemental analysis. The photophysical properties of these target molecules were studied by absorption and Fluorescence spectroscopy. In solution, the effect of pH, ionic strength in acid media and concentration on the aggregation behaviors of 4a has also been investigated by UV-Vis spectra. The broadened and red shifted Soret band indicated the formation of J-aggregates when the pH value was up to 2.0 in THF-aqueous solution. Furthermore, the higher ionic strength of 0.3 M NaCl in acid media resulted in the generation of J-aggregates in THF-aqueous solution. And the significant blue shift of Soret band also demonstrated the formation of H-aggregates of 4a at 1.1 × 10-4 M in THF.

Red blood cells (RBCs) possess intact cyto-architectures while haemoglobin (Hb) is a cell-free, homogeneous solution. Both RBCs and Hb are generalized oxygen carriers. In this paper, kinetic studies on oxygen-releasing of high concentration of Hb and RBCs under various conditions were carried out regarding Hb and RBCs as fluids. Among them, Hb under specific conditions was seen as the simplest Hb-based oxygen carrier (HBOC), Also, factors affecting the oxygen releasing of Hb and RBCs, including osmotic pressure, viscosity and allosteric agent, have been well studied. Analysis of the results from the measurement above showed that kinetics of oxygen releasing of either pure Hb or the simplest HBOCs was obviously different from that of RBCs. The oxygen-releasing time of Hb was shorter and the oxygen-releasing rates of Hb were quicker than those of RBCs under various conditions. Therefore, as fluids, only by changing the milieus it exists in, Hb could not achieve the expected oxygen-releasing effect on the microcirculation so well as RBCs do in the same system, irrespective of the interaction between the fluids and blood vessels. Furthermore, kinetic properties of HBOCs must be considered and matched with those of RBCs in the study of HBOCs.
