RESUMO

A simple and time-saving strategy was developed for the amplification of electrochemiluminescence (ECL) by dropping the noncoplanar tetraphenylethylene (TPE) solution on the surface of gold electrode. The self-assembled TPE aggregates exhibited inherent porous structures, endowing them with high specific surface area and oxygen adsorption capability. Therefore, the fabricated porous structures could lead to a 50-fold increase in the ECL signal of luminol in neutral aqueous solution, in comparison to that on the bare electrode. In contrast, the aggregates of the two typical coplanar polycyclic aromatic hydrocarbons (PAHs), perylene and pyrene, gave a weaker ECL enhancement, owing to their disc-like molecular structure and densely packed layers under aggregated conditions. The proposed ECL system has been successfully applied for the detection of hydrogen peroxide (H2O2) in the linear range of 0.25-1000 µM with a detection limit (S/N = 3) of 0.1 µM. Our findings provide inspiration for revealing the role of inherent molecular structure in the aggregate configuration, and they provide attractive perspectives for the usage of noncoplanar molecules in analytical applications.

RESUMO

The ratio of reduced and oxidized glutathione (GSH/GSSG ratio) is a greater first indication of disease risk than the total concentration of GSH. However, the interferences from thiolated biomolecules, especially cysteine (Cys), make the accurate detection of GSH/GSSG ratio a technical problem. In this work, we successfully used a mixture of quantum dots (QDs) and ZnAl-LDH nanosheets to fabricate a high electrochemiluminescence resonance energy transfer (ERET) efficiency sensor for GSH from the disturbances of amino acids, especially Cys and GSSG. The mechanisms of high ERET efficiency and selectivity were well investigated with spectroscopy analysis and theoretical calculation. The results showed that the interaction force between ZnAl-LDH nanosheets and molecules proved a long-range-ordered space and selective transmission for molecules. On the basis of these interesting phenomena, we successfully measured the GSH/GSSG ratio in whole blood and serum samples.

Introduction: Carboxylesterases (CXEs) and glutathione S-transferases (GSTs) can terminate olfactory signals during chemosensation by rapid degradation of odorants in the vicinity of receptors. The tea grey geometrid, Ectropis grisescens (Lepidoptera, Geometridae), one of the most devastating insect herbivores of tea plants in China, relies heavily on plant volatiles to locate the host plants as well as the oviposition sites. However, CXEs and GSTs involved in signal termination and odorant clearance in E. grisescens remains unknown. Methods: In this study, identification and spatial expression profiles of CXEs and GSTs in this major tea pest were investigated by transcriptomics and qRT-PCR, respectively. Results: As a result, we identified 28 CXEs and 16 GSTs from female and male antennal transcriptomes. Phylogenetic analyses clustered these candidates into several clades, among which antennal CXEs, mitochondrial and cytosolic CXEs, and delta group GSTs contained genes commonly associated with odorants degradation. Spatial expression profiles showed that most CXEs (26) were expressed in antennae. In comparison, putative GSTs exhibited a diverse expression pattern across different tissues, with one GST expressed specifically in the male antennae. Disscussion: These combined results suggest that 12 CXEs (EgriCXE1, 2, 4, 6, 8, 18, 20-22, 24, 26, and 29) and 5 GSTs (EgriGST1 and EgriGST delta group) provide a major source of candidate genes for odorants degradation in E. grisescens.

Stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is one of the most destructive diseases of wheat and has been largely managed using demethylation inhibitor (DMI) fungicide triadimefon in China. To determine the sensitivity of Pst, a Chinese Pst isolate and its sexually produced progeny isolates were tested with triadimefon using the detached leaf method. The half maximal effective concentration (EC50) values varied greatly among the progeny isolates, ranging from 0.06 mg L-1 to 7.89 mg L-1. Twenty-six of the 56 tested progeny isolates were less sensitive to triadimefon than the parental isolate. A single-nucleotide mutation at the 401 position resulting in an amino acid change from tyrosine (Y) to phenylalanine (F) in the 134th codon (Y134F) of the cytochrome P450 sterol 14a-demethylase enzyme (CYP51), the target gene of DMI fungicide, was identified in the parental isolate. The 87 tested progeny isolates segregated into 19 homozygous wild type (AA), 40 heterozygous (AT), and 28 homozygous mutant (TT) genotypes, fitting a 1:2:1 ratio (χ2 = 2.43; P = 0.30). The mutant isolates had higher EC50 values than the wild type isolates. Significant differences in logEC50 were found between the mutant isolates and the wild type isolates (P = 2.2e-16). However, homozygous and heterozygous mutant isolates were not significantly different (P = 0.21), indicating dominant mutation. Twenty-two progeny isolates were used to inoculate a susceptible wheat variety, and latency period and lesion growth were recorded to compare wild type and mutant isolates for the pathogenicity fitness components. A moderate but significant negative correlation was detected between lesion growth and sensitivity to triadimefon (r = -0.53; P = 0.01). No significant variation in lesion growth was found between homozygous and heterozygous mutant isolates (P = 0.83). In the case of latency period and triadimefon sensitivity, no significant correlation was found (P = 0.17). These results are useful for understanding reduced sensitivity in the pathogen population and improving stripe rust management.
