RESUMO

In females, the pathophysiological mechanism of poor ovarian response (POR) is not fully understood. Considering the expression level of p62 was significantly reduced in the granulosa cells (GCs) of POR patients, this study focused on identifying the role of the selective autophagy receptor p62 in conducting the effect of follicle-stimulating hormone (FSH) on antral follicles (AFs) formation in female mice. The results showed that p62 in GCs was FSH responsive and that its level increased to a peak and then decreased time-dependently either in ovaries or in GCs after gonadotropin induction in vivo. GC-specific deletion of p62 resulted in subfertility, a significantly reduced number of AFs and irregular estrous cycles, which were same as pathophysiological symptom of POR. By conducting mass spectrum analysis, we found the ubiquitination of proteins was decreased, and autophagic flux was blocked in GCs. Specifically, the level of nonubiquitinated Wilms tumor 1 homolog (WT1), a transcription factor and negative controller of GC differentiation, increased steadily. Co-IP results showed that p62 deletion increased the level of ubiquitin-specific peptidase 5 (USP5), which blocked the ubiquitination of WT1. Furthermore, a joint analysis of RNA-seq and the spatial transcriptome sequencing data showed the expression of steroid metabolic genes and FSH receptors pivotal for GCs differentiation decreased unanimously. Accordingly, the accumulation of WT1 in GCs deficient of p62 decreased steroid hormone levels and reduced FSH responsiveness, while the availability of p62 in GCs simultaneously ensured the degradation of WT1 through the ubiquitin‒proteasome system and autophagolysosomal system. Therefore, p62 in GCs participates in GC differentiation and AF formation in FSH induction by dynamically controlling the degradation of WT1. The findings of the study contributes to further study the pathology of POR.

A large number of oocytes in the perinatal ovary in rodents get lost for unknown reasons. The granulosa cell-oocyte mutual communication is pivotal for directing formation of the primordial follicle; however, little is known if paracrine factors participate in modulating programmed oocyte death perinatally. We report here that pregranulosa cell-derived fibroblast growth factor 23 (FGF23) functioned in preventing oocyte apoptosis in the perinatal mouse ovary. Our results showed that FGF23 was exclusively expressed in pregranulosa cells, while fibroblast growth factor receptors (FGFRs) were specifically expressed in the oocytes in perinatal ovaries. FGFR1 was one of the representative receptors in mediating FGF23 signaling during the formation of the primordial follicle. In cultured ovaries, the number of live oocytes declines significantly, accompanied by the activation of the p38 mitogen-activated protein kinase signaling pathway, under the condition of FGFR1 disruption by specific inhibitors of FGFR1 or silencing of Fgf23. As a result, oocyte apoptosis increased and eventually led to a decrease in the number of germ cells in perinatal ovaries following the treatments. In the perinatal mouse ovary, pregranulosa cell-derived FGF23 binds to FGFR1 and activates at least the p38 mitogen-activated protein kinase signaling pathway, thereby regulating the level of apoptosis during primordial follicle formation. This study reemphasizes the importance of granulosa cell-oocyte mutual communication in modulating primordial follicle formation and supporting oocyte survival under physiological conditions.

For heterojunction system, the lack of stable interfacial driving force and definite charge transfer channel makes the charge separation and transfer efficiency unsatisfactory. The photoreaction mechanism occurring at the interface also receives less attention. Herein, a 2D/2D Z-scheme junction BiOBr@NiFe-LDH with large-area contact featured by abundant interfacial hydrogen bonds and a strong interfacial electric field (IEF) is synthesized, and in-situ photoinduced metallic species assisting charge transfer mechanism is demonstrated. The hydrogen bonds between O atoms from BiOBr and H atoms from NiFe-LDH induce a significant interfacial charge redistribution, establishing a robust IEF. Notably, during photocatalytic reaction, Bi0 and Ni0 are in-situ isolated from BiOBr and NiFe-LDH in heterojunction, which separately act as electron transport mediator and electron trap to further accelerate charge transfer efficiency up to 71.2%. Theoretical calculations further demonstrate that the existence of Bi0 strengthens the IEF. Therefore, high-speed spatial charge separation is realized in Bi0/BiOBr@Ni0/NiFe-LDH, leading to a prominent photocatalytic activity with a tetracycline removal ratio of 88.3% within 7 minutes under visible-light irradiation and the presence of persulfate, far exceeding majority of photocatalysts reported previously. This study provides valid insights for designing hydrogen bonding heterojunction systems, and advances mechanistic understanding on in-situ photoreaction at interfaces.

Single-cell western blotting (scWB) is a prevalent technique for high-resolution protein analysis on low-abundance cell samples. However, the extensive signal loss during repeated antibody stripping precludes multiplex protein detection. Herein, we introduce Fluorescent-quenching Aptamer-based Single-cell Western Blotting (FAS-WB) for multiplex protein detection at single-cell resolution. The minimal size of aptamer probes allows rapid in-gel penetration, diffusion, and elution. Meanwhile, the fluorophore-tagged aptamers, coordinated with complementary quenching strands, avoid the massive signal loss conventionally caused by antibody stripping during repeated staining. Such a strategy also facilitates multiplex protein analysis with a limited number of fluorescent tags. We demonstrated FAS-WB for co-imaging four biomarker proteins (EpCAM, PTK7, HER2, CA125) at single-cell resolution with lower signal loss and enhanced signal-to-noise ratio compared to conventional antibody-based scWB. Being more time-saving (less than 25 min per cycle) and economical (1/1000 cost of conventional antibody probes), FAS-WB offers a highly efficient platform for profiling multiplex proteins at single-cell resolution.

Workpiece repositioning error has always been a key factor affecting manufacturing accuracy. The issues become more sensitive when machining microstructures with special morphologies, where the declination error caused by the repositioning may lead to microstructural defects. To solve this practical problem, in this paper, we report the design of a fixture that can detect the plane angular displacement error between the workpiece and the tool, namely the Rotation Correction Fixture (RCF). The fiducial marker referred to as polar microstructure is proposed and placed on the RCF edge. Angular displacement measurement is realized by observing the microstructural changes. Simultaneously, a Full-scale Rotation Detection (FRD) method is proposed to obtain the full-scale and high-precision angular displacement, including coarse extraction based on Fourier transform and fine extraction based on the Fast and Robust Feature-based Positioning method. Template matching is employed to eliminate the phase ambiguity in the Fourier transform. The results show that the proposed method can realize the calibration of the workpiece declination with a standard deviation error of 250.24 seconds, which meets the needs of workpiece precision positioning well.

A 32×32 100 GHz silicon photonic integrated arrayed waveguide grating router (AWGR) is experimentally demonstrated for dense wavelength division multiplexing (DWDM) applications. The dimension of the AWGR is 2.57 mm×1.09 mm with a core size of 1.31 mm×0.64 mm. It exhibits 6.07 dB maximum channel loss non-uniformity with -1.66 dB best-case insertion loss and average channel crosstalk of -15.74 dB. In addition, in the case of 25 Gb/s signals, the device successfully realizes high-speed data routing. The AWG router provides clear optical eye diagrams and low power penalty at bit-error-rates of 10-9.

We propose a compact, ultrabroadband and temperature-insensitive adiabatic directional coupler based on rib silicon waveguide-enabling arbitrary splitting ratios. Simulation results show that the device can achieve arbitrary splitting ratios from 1400 to 1600 nm, equal to 50%:50%, 60%:40%, 70%:30%, 80%:20%, and 90%:10% for the fundamental transverse electric mode. The designed device has an excess loss of less than 0.19 dB on the operational waveband. Furthermore, the proposed device shows a great robustness to fabrication imperfection, with a waveguide width deviation of 50 nm and ambient temperature change from 0°C to 200°C. These properties make the design a potential candidate for ultrahigh-density photonic integration chips.

A single-shot imaging system with multiple frames has been developed, which can record sequential multiple frames by delaying multiple optical images with fiber bundles and then capturing the images with a single intensified camera. The observed optical object is imaged through four lenses onto the end faces of four sets of fiber bundles. These fiber bundles with different lengths can provide different delays for delivering optical images, which determine the inter-frame separation times. The optical images exported from the fiber bundles are captured with a single intensified CMOS camera simultaneously. This imaging system has been applied for investigating the dynamic x-ray spot of the rod-pinch diode via a combination of scintillators, which are used to convert x-ray images to optical images. Four sequential x-ray images in a single shot have been obtained, which show the dynamic development of the rod-pinch x-ray spot. The results experimentally reveal the dynamics of the electrons flow bombarding the rod, which roughly agrees with the theoretical modeling of the rod-pinch diode.

3D doping structure has significant advantages in modulation efficiency and loss compared with 2D modulator doping profiles. However, to the best of our knowledge, previous work on 3D simulation methods for interdigitated doping designs applied simplified models, which prohibited complex 3D doping. In this work, innovative omni junctions, based on the effective 3D Monte-Carlo method, are believed to be the first proposed for high-performance modulators. Simulation results show that the modulation efficiency reaches 0.88 V·cm, while the loss is only 16 dB/cm, with capacitance below 0.42 pF/mm. This work provides a modulator design with superior modulation efficiency and serviceability for high-speed datacom.

The coupled process of thiosulfate-driven denitrification (NO3-→NO2-) and Anammox (TDDA) was a promising process for the treatment of wastewater containing NH4+-N and NO3--N. However, the high concentration of SO42- production limited its application, which needs to be alleviated by an economical and effective way to promote the application of TDDA process. In this study, TDDA process was started in a relatively short time by stepwise replacing nitrite with nitrate and operated continuously for 146 days. Results presented that the average total nitrogen removal efficiency of 82.18% can be acquired at a high loading rate of 1.98 kg N/(m3·d) with maximum nitrogen removal efficiency up to 87.04%. It was observed that the increase of S/N ratio improved the denitrification efficiency and slightly inhibit the Anammox process. Batch tests showed that Sulfammox process appeared in TDDA process under certain conditions, further contributing 2.59% nitrogen removal and 10.46% sulfur removal (14.42 mg/L NH4+-N and 37.68 mg/L SO42--S were removed). This finding was mainly attributed to the reduction of sulfate in TDDA system to elemental S0 or HS-, which subsequently was used as an electron donor to realize the recycling of sulfate (SO42--S) pollutants and promote the sulfur-nitrogen (S-N) cycle. High-throughput analysis displayed that Anammox bacteria (Candidatus_Kuenenia), Sulfur-oxidizing bacteria (Thiobacillus) with relatively high abundance of 5.37%, 7.74%, respectively, guaranteeing the excellent nitrogen and sulfate removal performance in the reactor. The enrichment of phyla Chloroflexi (31.79%), Proteobacteria (31.82%), class Ignavibacteriales (10.55%), genus Planctomycetes (13.57%) further verified the exitence of Sulfammox process in the TDDA reactor. This study provides a new perspective for the practical application of TDDA in terms of reducing the production of high concentration SO42- and saving operational cost and strengthening deeply nitrogen removal.

The construction of a guide star catalog is crucial for a star sensor to achieve accurate star map recognition and attitude determination. At present, the methods of a guide star catalog for a large field of view (FOV) star sensor have been relatively mature. However, for a small-FOV star sensor, there are still certain problems, such as a large storage capacity of a guide star catalog, uneven distribution of stars, and easy occurrence of voids. To address these problems, we propose a construction method of a small-FOV star sensor guide star catalog based on the combination of the improved spherical spiral method (ISSM) and extreme learning machine (ELM) named the ISSM-ELM. First, a spiral reference point is used as an optical axis pointing of the star sensor, and the guide stars are preliminarily screened based on the star-diagonal distance between the star and the reference point, and the star-density and magnitude characteristics of the guide star. Then the ELM is used to supplement the guide star empty sky area to construct an integrity guide star catalog. The experimental results demonstrate that the proposed method can reduce the storage capacity of the guide star catalog, and improve its uniformity, integrity, and average brightness.

Inferior contact interface and low charge transfer efficiency seriously restrict the performance of heterojunctions. Herein, chemically bonded α-Fe2 O3 /Bi4 MO8 Cl (M=Nb, Ta) dot-on-plate Z-scheme junctions with strong internal electric field are crafted by an in situ growth route. Experimental and theoretical results demonstrate that the internal electric field provides a powerful driving force for vectorial migration of photocharges between Bi4 MO8 Cl and α-Fe2 O3 , and the interfacial Fe-O bond not only serves as an atomic-level charge flow highway but also lowers the charge transfer energy barrier, thereby accelerating Z-scheme charge transfer and realizing effective spatial charge separation. Impressively, α-Fe2 O3 /Bi4 MO8 Cl manifests a significantly improved photocatalytic activity for selective oxidation of aromatic alcohols into aldehydes (Con. ≥92 %, Sel. ≥96 %), with a performance improvement of one to two orders of magnitude. This work presents atomic-level insight into interfacial charge flow steering.

Developing efficient piezocatalytic systems for two-electron water splitting (TEWS) with producing H2 and H2 O2 shows great promise to meet the industrial demand. Herein, Ag single atoms (SAs) and clusters are co-anchored on carbon nitride (AgSA+C -CN) to serve as the multifunctional sites for efficient TEWS. The Ag SAs enhance the in-plane piezoelectric polarization of CN that is intimately modulated by the atomic coordination induced charge redistribution, and Ag clusters afford strong interfacial electric field to remarkably promote the out-of-plane migration of piezoelectrons from CN. Moreover, AgSA+C -CN yields a larger piezoresistive effect that elevates carrier mobility under strain. Consequently, a superior H2 and H2 O2 evolution rate of 7.90 mmol g-1 h-1 and 5.84 mmol g-1 h-1 is delivered by AgSA+C -CN, respectively, far exceeding that of the previously reported piezocatalysts. This work not only presents the SAs decoration as an available polarization enhancement strategy, but also sheds light on the superiority of multi-sites engineering in piezocatalysis.

The Ebbinghaus illusion (EI) is an optical illusion of relative size perception that reflects the contextual integration ability in the visual modality. The current study investigated the genetic basis of two subtypes of EI, EI overestimation, and EI underestimation in humans, using quantitative genomic analyses. A total of 2825 Chinese adults were tested on their magnitudes of EI overestimation and underestimation using the method of adjustment, a standard psychophysical protocol. Heritability estimation based on common single nucleotide polymorphisms (SNPs) revealed a moderate heritability (34.3%) of EI overestimation but a nonsignificant heritability of EI underestimation. A meta-analysis of two phases (phase 1: n = 1986, phase 2: n = 839) of genome-wide association study (GWAS) discovered 1969 and 58 SNPs reaching genome-wide significance for EI overestimation and EI underestimation, respectively. Among these SNPs, 55 linkage-disequilibrium-independent SNPs were associated with EI overestimation in phase 1 with genome-wide significance and their associations could be confirmed in phase 2 cohort. Gene-based analyses found seven genes to be associated with EI overestimation at the genome-wide level, two from meta-analysis, and five from classical two-stage analysis. Overall, this study provided consistent evidence for a substantial genetic basis of the Ebbinghaus illusion.

The arbitrary ratio power splitter is widely used in photonic integrated circuits (PICs), for signal monitoring, power equalization, signal feedback, and so on. Here we designed a fabrication-tolerant, compact, broadband, and low-loss arbitrary ratio power splitter. The proposed arbitrary ratio power splitter was realized with an adiabatically tapered silicon rib waveguide with 70 nm shallow etches and an Si3N4 waveguide. The fabrication analysis confirmed that both of them are robust to fabrication errors. 3D finite-difference time-domain simulations show a very low excess loss (less than 0.02 dB for Si3N4 waveguide and 0.05 dB for Si rib waveguide), and a broadband operating wavelength range (100 nm). Good fabrication tolerance and standard critical dimensions make the arbitrary ratio power splitter compatible with the standard fabrication process of commercial silicon photonic foundries.

A silicon-on-insulator polarization diversity scheme is proposed. Based on an asymmetrical evanescent coupler, a broadband and compact polarization splitter-rotator comprising mode conversion tapers and mode sorting asymmetric Y junctions is optimized with silicon dioxide upper cladding and a silicon nitride waveguide. The simulation results show mode conversion loss is less than 0.2 dB, and the extinction ratio is lower than -17dB in the wavelength range of 1.48µm to 1.67µm.

Previous studies have evaluated the relationship between prenatal air pollution exposure and low birth weight, but the results are inconsistent. The purpose of this meta-analysis is to quantitatively analyze the relationship between maternal air pollutant exposure and low birth weight (LBW). PubMed and Web of Science databases were searched to obtain the studies on the relationship between the prenatal exposure of air pollutants and LBW that published as of June 2020. The pooled effects of air pollutant exposure and LBW were calculated using random-effect model (for studies with significant heterogeneity) or fixed-effect model (for studies without significant heterogeneity). Totally, 54 studies were included in this meta-analysis. The pooled effect of PM2.5, PM10, NO2, CO, SO2, and O3 exposure on LBW were 1.081 (95% CI: 1.043, 1.120), 1.053 (95% CI: 1.030, 1.076), 1.030 (95% CI: 1.008, 1.053), 1.007 (95% CI: 1.001, 1.014), 1.125 (95% CI: 1.017, 1.244), and 1.045 (95% CI: 1.005, 1.086), respectively. NO2 (per 10 ppb increase) and CO (per 100 ppb increase) exposure in the first trimester were positively correlated with LBW, of which the pooled effect was 1.022 (95% CI: 1.009, 1. 035) and 1.008 (95% CI: 1.004, 1.012), respectively. PM2.5 (per 10 µg/m3 increase) exposure in the third trimester significantly affected the LBW, of which the pooled effect was 1.053 (95% CI: 1.010, 1.097). In addition, PM10 (per 10 µg/m3 increase) exposure in the second trimester also significantly affected the LBW, with the pooled effect of 1.011 (95% CI: 1.005, 1.017). Prenatal exposure of the major air pollutants during the entire pregnancy could increase the risk of LBW, while the susceptible window of the pollutants varied.

Visual cognition in humans has traditionally been studied with cognitive behavioral methods and brain imaging, but much less with genetic methods. Perceptual rivalry, an important phenomenon in visual cognition, is the spontaneous perceptual alternation that occurs between two distinct interpretations of a physically constant visual stimulus (e.g., binocular rivalry stimuli) or a perceptually ambiguous stimulus (e.g., the Necker cube). The switching rate varies dramatically across individuals and can be voluntarily modulated by observers. Here, we adopted a genomic approach to systematically investigate the genetics underlying binocular rivalry, Necker cube rivalry and voluntary modulation of Necker cube rivalry in young Chinese adults (Homo sapiens, 81% female, 20 ± 1 years old) at multiple levels, including common single nucleotide polymorphism (SNP)-based heritability estimation, SNP-based genome-wide association study (GWAS), gene-based analysis, and pathway analysis. We performed a pilot GWAS in 2441 individuals and replicated it in an independent cohort of 943 individuals. Common SNP-based heritability was estimated to be 25% for spontaneous perceptual rivalry. SNPs rs184765639 and rs75595941 were associated with voluntary modulation, and imaging data suggested genotypic difference of rs184765639 in the surface area of the left caudal-middle frontal cortex. Additionally, converging evidence from multilevel analyses associated genes such as PRMT1 with perceptual switching rate, and MIR1178 with voluntary modulation strength. In summary, this study discovered specific genetic contributions to perceptual rivalry and its voluntary modulation in human beings. These findings may promote our understanding of psychiatric disorders, as perceptual rivalry is a potential psychiatric biomarker.SIGNIFICANCE STATEMENT Perceptual rivalry is an important visual phenomenon in which our perception of a physically constant visual input spontaneously switches between two different states. There are individual variations in perceptual switching rate and voluntary modulation strength. Our genomic analyses reveal several loci associated with these two kinds of variation. Because perceptual rivalry is thought to be relevant to and potentially an endophenotype for psychiatric disorders, these results may help understand not only visual cognition, but also psychiatric disorders.

The six basic emotions (disgust, anger, fear, happiness, sadness, and surprise) have long been considered discrete categories that serve as the primary units of the emotion system. Yet recent evidence indicated underlying connections among them. Here we tested the underlying relationships among the six basic emotions using a perceptual learning procedure. This technique has the potential of causally changing participants' emotion detection ability. We found that training on detecting a facial expression improved the performance not only on the trained expression but also on other expressions. Such a transfer effect was consistently demonstrated between disgust and anger detection as well as between fear and surprise detection in two experiments (Experiment 1A, n = 70; Experiment 1B, n = 42). Notably, training on any of the six emotions could improve happiness detection, while sadness detection could only be improved by training on sadness itself, suggesting the uniqueness of happiness and sadness. In an emotion recognition test using a large sample of Chinese participants (n = 1748), the confusion between disgust and anger as well as between fear and surprise was further confirmed. Taken together, our study demonstrates that the "basic" emotions share some common psychological components, which might be the more basic units of the emotion system.
