BACKGROUND: The Atlantic cod is a prolific species in the Atlantic, despite its inconsistent specific antibody response. It presents a peculiar case within vertebrate immunology due to its distinct immune system, characterized by the absence of MHCII antigen presentation pathway, required for T cell-dependent antibody responses. Thorough characterisation of immunoglobulin loci and analysis of the antibody repertoire is necessary to further our understanding of the Atlantic cod's immune response on a molecular level. RESULTS: A comprehensive search of the cod genome (gadmor3.0) identified the complete set of IgH genes organized into three sequential translocons on chromosome 2, while IgL genes were located on chromosomes 2 and 5. The Atlantic cod displayed a moderate germline V gene diversity, comprising four V gene families for both IgH and IgL, each with distinct chromosomal locations and organizational structures. 5'RACE sequencing revealed a diverse range of heavy chain CDR3 sequences and relatively limited CDR3 diversity in light chains. The analysis highlighted a differential impact of V-gene germline CDR3 length on receptor CDR3 length between heavy and light chains, underlining different recombination processes. CONCLUSIONS: This study reveals that the Atlantic cod, despite its inconsistent antibody response, maintains a level of immunoglobulin diversity comparable to other fish species. The findings suggest that the extensive recent duplications of kappa light chain genes do not result in increased repertoire diversity. This research provides a comprehensive view of the Atlantic cod's immunoglobulin gene organization and repertoire, necessary for future studies of antibody responses at the molecular level.

In this work, the Mn, Co, Ce co-doped corn cob biochar (MCCBC) as catalytic particle electrodes in a three-dimensional heterogeneous electro-Fenton-like (3D-HEFL) system for the efficient degradation of coking wastewater was investigated. Various characterization methods such as SEM, EDS, XRD, XPS and electrochemical analysis were employed for the prepared materials. The results showed that the MCCBC particle electrodes had excellent electrochemical degradation performances of COD in coking wastewater, and the COD removal and degradation rates of the 3D/HEFL system were 85.35% and 0.0563 min-1 respectively. RSM optimized conditions revealed higher COD removal rate at 89.23% after 31.6 min of electrolysis. The efficient degradability and wide adaptability of the 3D/HEFL system were due to its beneficial coupling mechanism, including the synergistic effect between the system factors (3D and HEFL) as well as the synergistic interactions between the ROS (dominated by •OH and supplemented by O2•-) in the system. Moreover, the COD removal rate of MCCBC could still remain at 81.41% after 5 cycles with a lower ion leaching and a specific energy consumption of 11.28 kWh kg-1 COD. The superior performance of MCCBC, as catalytic particle electrodes showed a great potential for engineering applications for the advanced treatment of coking wastewater.

A Value of Information (VOI) analysis can play a key role in decision-making for adopting new approach methodologies (NAMs). We applied EPA's recently developed VOI framework to the Threshold of Toxicological Concern (TTC). Obtaining/deriving a TTC value for use as a toxicity reference value (TRV) for substances with limited toxicity data was shown to provide equivalent or greater health protection, immense return on investment (ROI), greater net benefit, and substantially lower costs of delay (CoD) compared with TRVs derived from either traditional human health assessment (THHA) chronic toxicity testing in lab animals or the 5-day in vivo EPA Transcriptomic Assessment Product (ETAP). For all nine exposure scenarios examined, the TTC was more economical terms of CoD and ROI than the ETAP or the THHA; expected net benefit was similar for the TTC and ETAP with both of these more economical than the THHA The TTC ROI was immensely greater (5,000,000-fold on average) than the ROI for THHA and the ETAP ROI (100,000-fold on average). These results support the use of the TTC for substances within its domain of applicability to waive requiring certain in vivo tests, or at a minimum, as an initial screening step before conducting either the ETAP or THHA in vivo studies.

With the aim to upcycle fish side-streams, enzymatic hydrolysis is often applied to produce protein hydrolysates with bioactive properties or just as a protein source for food and feed. However, the production of hydrolysates generates a side-stream. For underutilized fish and fish backbone this side-stream will contain fish bones and make it rich in minerals. The aim of this study was to assess the relative bioaccessibility (using the standardized in vitro model INFOGEST 2.0) of minerals in a dietary supplement compared to bone powder generated after enzymatic hydrolysis of three different fish side-streams: undersized whole hake, cod and salmon backbones consisting of insoluble protein and bones. Differences in the bioaccessibility of protein between the powders were also investigated. The enzyme hydrolysis was carried out using different enzymes and hydrolysis conditions for the different fish side-streams. The content and bioaccessibility of protein and the minerals phosphorus (P), calcium (Ca), potassium (K) and magnesium (Mg) were measured to evaluate the potential of the powder as an ingredient in, e.g., dietary supplements. The bone powders contained bioaccessible proteins and minerals. Thus, new side-streams generated from enzymatic hydrolysis can have possible applications in the food sector due to bioaccessible proteins and minerals.

Malnutrition is one of the major factors of bone and cartilage disorders. Pacific cod (Gadus macrocephalus) processing waste is a cheap and highly promising source of bioactive substances, including collagen-derived peptides and amino acids, for bone and cartilage structure stabilization. The addition of these substances to a functional drink is one of the ways to achieve their fast intestinal absorption. Collagen hydrolysate was obtained via enzymatic hydrolysis, ultrafiltration, freeze-drying, and grinding to powder. The lyophilized hydrolysate was a light gray powder with high protein content (>90%), including collagen (about 85% of total protein) and a complete set of essential and non-essential amino acids. The hydrolysate had no observed adverse effect on human mesenchymal stem cell morphology, viability, or proliferation. The hydrolysate was applicable as a protein food supply or a structure-forming food component due to the presence of collagen fiber fragments. An isotonic fitness drink (osmolality 298.1 ± 2.1 mOsm/L) containing hydrolysate and vitamin C as a cofactor in collagen biosynthesis was prepared. The addition of the hydrolysate did not adversely affect its organoleptic parameters. The production of such functional foods and drinks is one of the beneficial ways of fish processing waste utilization.

During recent years, there has been a renewed interest in establishing farming of Atlantic cod (Gadus morhua) in Norway. However, a fatal abdominal disorder compromises animal welfare and causes economic losses. A similar problem was present during a previous attempt to establish Atlantic cod farming more than a decade ago. In this paper, we provide the first in-depth description of this intestinal disorder, which is correctly denoted 'strangulating obstruction'. In affected fish, part of the intestine is permanently entrapped (incarcerated) under fibrous strands in the mesentery. The entrapment interferes with blood flow and physically blocks the intestine, causing a strangulating obstruction with severe venous congestion and ischemia of the intestinal wall. Furthermore, comparison of macroscopical and histological anatomy of farmed and wild Atlantic cod is presented and risk factors associated with the anatomical differences are discussed.

The degradation of typical dye wastewater is a focus of research in the printing and dyeing industry. In this study, a combined micro-electrolysis and microbial treatment method was established to treat refractory dye wastewater, and the pivotal factors in the microbial treatment were optimized. In the series and coupled modes, the removal rates of chroma reached 98.75% and 92.50%, and the removal rates of chemical oxygen demand (COD) reached 96.17% and 82.29%, respectively. The high-throughput sequencing results showed that the microbial communities in the microbial system varied at different treatment stages. From the culture stage to the domestication stage, the dominant phylum was Proteobacteria; however, the community abundance of microorganisms decreased. A combination of micro-electrolysis and biological methods can alter the characteristics of the microbial community, increase the number of dominant phyla, and increase the abundance of microorganisms. The degradation effect of the series mode and the overall strengthening effect of micro-electrolysis on the microorganisms were better than those of the coupled mode. In actual wastewater, the maximum removal rates of chroma, COD, total nitrogen (TN), ammonia nitrogen (NH3-N), and total phosphorus (TP) are 97.50%, 98.90%, 94.35%, 93.95%, and 91.17%, respectively. Three-dimensional fluorescence spectrum analysis showed that microbial processes could significantly degrade fluorescent components in wastewater, and methanogenic active enzymes in anaerobic processes could continue to react. The combined process can realize the efficient treatment of toxic dye wastewater by reducing the toxicity of wastewater and efficiently degrading organic matter, which has important guiding significance for the treatment of refractory dye wastewater.

A portable and integrated electrochemical detection system has been constructed for on-site and real-time detection of chemical oxygen demand (COD). The system mainly consists of four parts: (i) sensing electrode with a copper-cobalt bimetallic oxide (CuCoOx)-modified screen-printed electrode; (ii) an integrated electrochemical detector for the conversion, amplification, and transmission of weak signals; (iii) a smartphone installed with a self-developed Android application (APP) for issuing commands, receiving, and displaying detection results; and (iv) a 3D-printed microfluidic cell for the continuous input of water samples. Benefiting from the superior catalytic capability of CuCoOx, the developed system shows a high detection sensitivity with 0.335 µA/(mg/L) and a low detection limit of 5.957 mg/L for COD determination and possessing high anti-interference ability to chloride ions. Moreover, this system presents good consistency with the traditional dichromate method in COD detection of actual water samples. Due to the advantages of cost effectiveness, portability, and point-of-care testing, the system shows great potential for water quality monitoring, especially in resource-limited remote areas.

A continuous stirred tank bioreactor (CSTB) with cell recycling combined with ceramic membrane technology and inoculated with Rhodococcus opacus PD630 was employed to treat petroleum refinery wastewater for simultaneous chemical oxygen demand (COD) removal and lipid production from the retentate obtained during wastewater treatment. In the present study, the COD removal efficiency (CODRE) (%) and lipid concentration (g/L) were predicted using two artificial intelligence models, i.e., an artificial neural network (ANN) and a neuro-fuzzy neural network (NF-NN) with a network topology of 6-25-2 being the best for NF-NN. The results revealed the superiority of NF-NN over ANN in terms of determination coefficient (R2), root mean square error (RMSE), and mean absolute percentage error (MAPE). Three learning algorithms were tested with NF-NN; among them, the Bayesian regularization backpropagation (BR-BP) outperformed others. The sensitivity analysis revealed that, if solid retention time and biomass concentrations were maintained between 35 and 75 h and 3.0 g/L and 3.5 g/L, respectively, high CODRE (93%) and lipid concentration (2.8 g/L) could be obtained consistently.

Basking sharks (Cetorhinus maximus) seasonally aggregate in coastal surface waters of the North Atlantic, providing opportunities for visual observation. While putative courtship displays have been observed, actual copulation has not been documented. Here we examine video collected by an unmanned aerial vehicle ("drone") of novel behavioral interactions between basking sharks in Cape Cod Bay, Massachusetts in May 2021. The behaviors, including close following and tight concentric circling, are consistent with pre-copulatory behavior observed in other shark species. These observations provide new insights into the pre-copulatory behavior of basking sharks.

Despite decades of active fisheries management, many stocks of Atlantic cod in its southern range are in a depleted state and mortality estimates remain high. Recovery of these stocks, as defined by management areas, could be confounded by cod distributions shifting outside of these areas. Here, we assess data from internationally coordinated trawl surveys to investigate the distribution of three cod stocks in the Celtic Seas ecoregion, Irish Sea, Celtic Sea, and West of Scotland, from 1985 to 2021. We mapped cod densities, analyzed trends in mean weighted depth and bottom temperature, and calculated the center of gravity and equivalent area of the stocks. The distribution of the West of Scotland stock shifted north and east, spilling into the North Sea, while the Irish Sea and Celtic Sea stocks shifted west. Each stock showed decreasing trends in equivalent area, but there were no clear trends in the average depth occupied by the fish. There was no apparent relationship between temperature and the distribution of cod, as bottom temperature varied little from 1993 to 2021. Although Irish Sea cod showed a shift into warmer water, this was due to changes in survey distribution. The shift in distribution of the West of Scotland cod stock towards the North Sea whilst impairing local recovery provides further justification for the recent definition of its incorporation into a larger stock unit that includes the northwest of the North Sea. The Irish Sea and Celtic Sea cod stocks are neither shifting northwards, nor into deeper waters, but remained within current boundaries. This suggests that recent temperature conditions did not affect their distribution, but this may change as temperatures increase towards the limit for reproduction.

Polar cod (Boreogadus saida) is an endemic key species of the Arctic Ocean ecosystem. The ecology of this forage fish is well studied in Arctic shelf habitats where a large part of its population lives. However, knowledge about its ecology in the central Arctic Ocean (CAO), including its use of the sea-ice habitat, is hitherto very limited. To increase this knowledge, samples were collected at the under-ice surface during several expeditions to the CAO between 2012 and 2020, including the Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) expedition. The diet of immature B. saida and the taxonomic composition of their potential prey were analysed, showing that both sympagic and pelagic species were important prey items. Stomach contents included expected prey such as copepods and amphipods. Surprisingly, more rarely observed prey such as appendicularians, chaetognaths, and euphausiids were also found to be important. Comparisons of the fish stomach contents with prey distribution data suggests opportunistic feeding. However, relative prey density and catchability are important factors that determine which type of prey is ingested. Prey that ensures limited energy expenditure on hunting and feeding is often found in the stomach contents even though it is not the dominant species present in the environment. To investigate the importance of prey quality and quantity for the growth of B. saida in this area, we measured energy content of dominant prey species and used a bioenergetic model to quantify the effect of variations in diet on growth rate potential. The modeling results suggest that diet variability was largely explained by stomach fullness and, to a lesser degree, the energetic content of the prey. Our results suggest that under climate change, immature B. saida may be at least equally sensitive to a loss in the number of efficiently hunted prey than to a reduction in the prey's energy content. Consequences for the growth and survival of B. saida will not depend on prey presence alone, but also on prey catchability, digestibility, and energy content.

Egg quality in fishes is commonly determined by fertilisation success and cleavage patterns as a phenotypic outcome of underlying regulatory mechanisms. Although these phenotypic estimators of egg quality are useful in farming conditions, these "good quality" egg batches do not always translate to good larval growth and survival. The identification of genes involved in embryonic development may help find links between genetic factors of maternal origin and egg quality. Herein, the relative expression of seven stage-specific developmental genes of Atlantic cod was analysed using quantitative PCR to understand the function during embryogenesis and its relationship with egg quality. Genes ccnb2 and pvalb1 showed significant differential expression between developmental stages and significant upregulation from blastula and somite stages, respectively. The comparison of spawning batches showed that the relative gene expression of genes ccnb2, acta, tnnt3 and pvalb1 was significantly higher from the middle of the spawning season where phenotypic quality estimators establish the best egg quality. Moreover, a positive significant correlation was observed between quality estimators based on egg morphology and the genetic expression of genes acta and acta1 during somitogenesis. This study suggests that the combination of quality estimators, genetics and batch timing could help optimise reproductive protocols for commercial stocks of Atlantic cod.

In the present study, the technical feasibility of an electrocoagulation-treatment wetland continuous flow system, for the removal of organic matter from landfill leachate (LL), was evaluated. The response surface methodology (MSR) was used to assess the individual and combined effects of the applied potential and distance between electrodes, on the removal efficiency and optimization of the electrocoagulation process. The hybrid treatment wetland system consisted of a vertical flow system coupled to a horizontal subsurface flow system, both planted with Canna indica. For a chemical oxygen demand (COD) concentration - without pretreatment of 5142.8 ± 2.5 mg L-1, the removal percentage for the electrocoagulation system was 79.4 ± 0.16%, under the optimal working conditions (Potential: 20 V; Distance: 2.0 cm). The COD removal efficiency in the treatment wetland with Canna indica showed a dependence with the hydraulic retention time, reaching 59.2 ± 0.2 % over 15 days. The overall efficiency of the system was about 91.5 ± 0.02 % removal of COD. In addition, a decrease in the biochemical oxygen demand (94.8 ± 0.14%) and total suspended solids (88.2 ± 0.22%), also related to the contamination levels of the LL, were obtained. This study, for the first time, shows that the coupling of electrocoagulation together with a treatment wetland system is a good alternative for the removal of organic contaminants present in LL.

Oil refineries produce annually large quantities of oily sludge and non-biodegradable wastewater during petroleum refining that require adequate management to minimize its environmental impact. The fraction solid of the oily sludge accounts for 25 wt% and without treatment for their valorization. This work is focused on the valorization of these solid particles through their transformation into porous materials with enhanced properties and with potential application in the catalytic wet air oxidation (CWAO) of a non-biodegradable spent caustic refinery wastewater. Hence, dealing with the valorization and treatment of both refinery wastes in a circular approach aligned with the petrol refinery transformations by 2050. The obtained oily sludge carbonaceous materials showed improved surface area (260-762 m2/g) and a high Fe content. The good catalytic performance of these materials in CWAO processes has been attributed to the simultaneous presence of surface basic sites and iron species. Those materials with higher content of Fe and basic sites yielded the highest degradation of organic compounds present in the spent caustic refinery wastewater. In particular, the best-performing material ACT-NP 1.1 (non-preoxidated and thermically treated with 1:1 mass ratio KOH:solid) showed a chemical oxygen demand (COD) removal of 60 % after 3 h of reaction and with a higher degradation rate than that achieved with thermal oxidation without catalyst (WAO) and that using an iron-free commercial activated carbon. Moreover, the biodegradability of the treated wastewater increased up to 80% (from ca. 31% initially of the untreated effluent). Finally, this material was reused up to three catalytic cycles without losing metal species and keeping the catalytic performance.

This study has carried out a systematic review of 36 scientific papers (reporting 63 case studies) published in the last 15 years about the treatment of industrial, agri-food and municipal wastewater in lagoons. A concentration of studies from a few countries (Italy, Algeria and Iran) and about municipal wastewater (70% of papers) was revealed by the bibliographic analysis. Aeration was supplied in more than 50% of case studies; the storage capacity of lagoons (adopted as a measure of size) was extremely variable (over seven orders of magnitude), while their depth was generally lower than a few metres. The efficiency of lagoon treatments at removing COD was in a wide range (25-98%). Very few studies analysed the energy intensity of treatments in lagoons. The meta-analysis applied to a further selection of 10 papers with 29 case studies revealed significant differences in pH and dissolved oxygen concentration, due to aeration or type of treated wastewater. Treatment efficiency was higher in aerated lagoons compared to non-aerated systems, and did not depend on the type of treated wastewater. Based on the analysis of the reviewed papers, an urgent research need on this topic arises, mainly due to the oldness of most analysed studies. Practical suggestions are given to optimise the depuration performances of lagoons: (i) application of intermittent and night aeration; (ii) reduced air flow rates; (iii) adaptation of microbial biomass to high contents of inhibiting compounds in wastewater; (iv) construction of baffles to keep the planned hydraulic retention time avoiding short-circuit; (v) integration of lagoons with other treatments (e.g., constructed wetlands); (vi) ferti-irrigation of crops with lagoon effluents rather than disposal into water bodies.

Supercritical water gasification technology provides a favorable technology to achieve pollution elimination and resource utilization of phenolic wastewater. In this study, the reaction mechanism of phenolic wastewater supercritical water gasification was investigated using a combination of experimental and computational methods. Five reaction channels were identified to elucidate the underlying pathway of phenol decomposition. Importantly, the rate-determining step was found to be the dearomatization reaction. By integrating computational and experimental analyses, it was found that phenol decomposition via the path with the lowest energy barrier generates cyclopentadiene, featuring a dearomatization barrier of 70.97 kcal/mol. Additionally, supercritical water plays a catalytic role in the dearomatization process by facilitating proton transfer. Based on the obtained reaction pathway, alkali salts (Na2CO3 and K2CO3) are employed as a catalyst to diminish the energy barrier of the rate-determining step to 40.00 kcal/mol and 37.14 kcal/mol. Alkali salts catalysis significantly improved carbon conversion and pollutant removal from phenolic wastewater, increasing CGE from 58.44% to 93.55% and COD removal efficiency from 94.11% to 99.79%. Overall, this study provides a comprehensive understanding of the decomposition mechanism of phenolic wastewater in supercritical water.

It is very important to choose a suitable method and catalyst to treat coking wastewater. In this study, Fe-Ce-Al/MMT catalysts with different Fe/Ce molar ratios were prepared, characterized by XRD, SEM, and N2 adsorption/desorption, and treated with coking wastewater. The results showed that the optimal Fe-Ce-Al/MMT catalyst with a molar ratio of Fe/Ce of 7/3 has larger interlayer spacing, specific surface area, and pore volume. Based on the composition analysis of real coking wastewater and the study of phenol simulated wastewater, the response surface test of the best catalyst for real coking wastewater was carried out, and the results are as follows: initial pH 3.46, H2O2 dosage 19.02 mL/L, Fe2+ dosage 5475.39 mL/L, reaction temperature 60 °C, and reaction time 248.14 min. Under these conditions, the COD removal rate was 86.23%.

The effect of coagulant dosage in a chemically enhanced primary treatment (CEPT) on the performance of a conventional wastewater treatment plant (WWTP) has been investigated. Lab-scale experiments simulations were carried out in order to evaluate the effect of coagulant addition on the primary settling performance. In these experiments, FeCl3 was used as coagulant. Later, the WWTP was theoretically simulated using a commercial software (WEST®) to evaluate the effect of coagulation/flocculation on the global system, based on the results obtained at lab-scale. According to these results, the CEPT modifies the organic matter balance in the WWTP, decreasing the contribution of readily (SS) and slowly (XS) biodegradable fractions of COD to the aerobic biological process up to 27.3% and 80.8%, respectively, for a dosage of FeCl3 of 24 mg L-1. Consequently, total suspended solids in the aerobic reactor and the secondary purged sludge decreased up to 33% and 13%, respectively. However, the influence on effluent quality was negligible. On the contrary, suspended solids concentration in the sludge to be treated by anaerobic digestion increased, mainly regarding the Ss and Xs fractions, which caused an 8.1% increase in biogas production potential, with approximately 60% of CH4 concentration.

The present work deals with the optimization of basic fuchsin dye removal from an aqueous solution using the ultraviolet UV/H2O2 process. Response Surface Modeling (RSM) based on Box-Behnken experimental design (BBD) was applied as a tool for the optimization of operating conditions such as initial dye concentration (10-50 ppm), hydrogen peroxide dosage (H2O2) (10-20 mM/L) and irradiation time (60-180 min), at pH = 7.4 under ultra-violet irradiation (254 nm and 25 W intensity). Chemical oxygen demand (COD abatement) was used as a response variable. The Box-Behnken Design can be employed to develop a mathematical model for predicting UV/H2O2 performance for COD abatement. COD abatement is sensitive to the concentration of hydrogen peroxide and irradiation time. Statistical analyses indicate a high correlation between observed and predicted values (R2 > 0.98). In the BBD predictions, the optimal conditions in the UV/H2O2 process for removing 99.3% of COD were found to be low levels of pollutant concentration (10 ppm), a high concentration of hydrogen peroxide dosage (20 mM/L), and an irradiation time of 80 min.
