RESUMO

Naturally occurring protein nanocages like ferritin are self-assembled from multiple subunits. Because of their unique cage-like structure and biocompatibility, there is a growing interest in their biomedical use. A multipurpose and straightforward engineering approach does not exist for using nanocages to make drug-delivery systems by encapsulating hydrophilic or hydrophobic drugs and developing vaccines by surface functionalization with a protein like an antigen. Here, a versatile engineering approach is described by mimicking the HIV-1 Gap polyprotein precursor. Various PREcursors of nanoCages (PREC) are designed and created by linking two ferritin subunits via a flexible linker peptide containing a protease cleavage site. These precursors can have additional proteins at their N-terminus, and their protease cleavage generates ferritin-like nanocages named protease-induced nanocages (PINCs). It is demonstrated that PINC formation allows concurrent surface decoration with a protein and hydrophilic or hydrophobic drug encapsulation up to fourfold more than the amount achieved using other methods. The PINCs/Drug complex is stable and efficiently kills cancer cells. This work provides insight into the precursors' design rules and the mechanism of PINCs formation. The engineering approach and mechanistic insight described here will facilitate nanocages' applications in drug delivery or as a platform for making multifunctional therapeutics like mosaic vaccines.

OBJECTIVE: To evaluate color, translucency parameter and optical properties (scattering (S), absorption (K) and transmittance (T)) of a multi-color polymer-infiltrated ceramic-network (PICN) material. METHODS: Samples of shades 1M1-HT, 1M2-HT, 2M2-HT, 3M2-HT, and 4M2-HT from VITA ENAMIC® multiColor (E-MC) High Translucent were fabricated (n = 3). CAD-CAM blocks were cut and polished to 1.00 ± 0.01 mm of thickness. Diffuse reflectance and color coordinates were measured against white and black backgrounds, using a calibrated spectroradiometer, CIE D65 illuminant and the CIE 45°/0° geometry. Color and translucency differences were evaluated using 50:50% perceptibility (PT and TPT) and 50:50% acceptability (AT and TAT) thresholds. S and K coefficients and T were calculated using Kubelka-Munk's equations. Data was statistically analyzed using Kruskal-Wallis, Mann-Whitney tests, and VAF coefficient. RESULT: Mean C* and b* values increased from incisal to cervical layers with statistically significant differences (p < 0.05). In general, ΔE00 between sequential layers were above PT for all shades. In addition, translucency parameter (TP) increased from cervical to incisal and ΔTP00 values were greater than TPT00 and lower than TAT00 between all sequential layers. Layers from all shades showed similar spectral behavior for S (97.4% ≤ VAF), K (85.0% ≤ VAF) coefficients and T (95.3% ≤ VAF). However, these values presented significant differences (p < 0.05) from cervical to incisal layers. SIGNIFICANCE: The gradient in color and translucency of this novel CAD-CAM multi-color PICN material can assist dental technicians and dentists to reach greater esthetics than the pre-existing CAD-CAM monolithicmaterials.

Within sub-Saharan Africa (SSA), although nematodes are viewed among the most important threats to crop production and food security, the presence of trained nematologists working within this discipline has traditionally been viewed as scarce. The few research studies concerning this subject address this topic from a country or sub-regional perspective and generally portray nematology as 'insufficient'. Over the past two decades, a few initiatives have been instrumental in building greater nematology expertise. For the first time a structured survey was undertaken, involving interviews with individuals from SSA that were (or currently are) involved in nematology training programmes, research, national extension services or in African universities. This paper provides evidence of the positive impact of various initiatives and shows an increase in the number of available nematology positions, together with high rates of graduates that return home to occupy qualified positions. Our findings will help researchers, policy makers and donors to identify areas requiring support to increase the promotion of nematology in SSA and to make an impact for end-users.

Existing cognitive diagnosis models conceptualize attribute mastery status discretely as either mastery or non-mastery. This study proposes a different conceptualization of attribute mastery as a probabilistic concept, i.e., the probability of mastering a specific attribute for a person, and developing a probabilistic-input, noisy conjunctive (PINC) model, in which the probability of mastering an attribute for a person is a parameter to be estimated from data. And a higher-order version of the PINC model is used to consider the associations among attributes. The results of simulation studies revealed a good parameter recovery for the new models using the Bayesian method. The Examination for the Certificate of Proficiency in English (ECPE) data set was analyzed to illustrate the implications and applications of the proposed models. The results indicated that PINC models had better model-data fit, smaller item parameter estimates, and more refined estimates of attribute mastery.
