PURPOSE: The aim of the present study was to examine the acute and chronic effects of wild blueberry supplementation on mood, executive function, and serum biomarkers of neuroplasticity, inflammation, and oxidative stress in emerging adults with moderate-to-severe depressive symptoms. METHODS: In this double-blind trial, 60 emerging adults (Mage = 20.0 years, 32% male) with self-reported depressive symptoms were randomly assigned to receive a single blueberry drink (acute phase), followed by 6 weeks of daily blueberry supplementation (chronic phase), or a matched placebo drink. The primary outcome was Beck Depression Inventory-II (BDI-II) scores at 6-week follow-up. Further measures included momentary affect (PANAS-X) and accuracy on an executive function task. The data were analyzed using ANCOVAs adjusted for baseline values, sex, and habitual fruit and vegetable intake. Estimated marginal means were calculated to compare the treatment arms. RESULTS: The blueberry drink significantly improved positive affect (p = 0.026) and executive function (p = 0.025) at 2 h post-ingestion, with change scores being positively correlated in the blueberry group (r = 0.424, p = 0.017). However, after six weeks of supplementation the reduction in BDI-II scores was greater in the placebo group by 5.8 points (95% CI: 0.8-10.7, p = 0.023). Generalized anxiety and anhedonia also decreased significantly more in the placebo group. No significant differences were found for any of the biomarkers. CONCLUSIONS: Six weeks of wild blueberry supplementation were inferior to placebo in reducing depressive symptoms. Nevertheless, the correlated improvements in positive affect and executive function after a single dose of blueberries point to a beneficial, albeit transient, psychological effect. These contrasting results suggest a biphasic, hormetic-like response that warrants further investigation. TRIAL REGISTRATION: NCT04647019, dated 30 November, 2020.

BACKGROUND: As the sector of the population over 65y increases, cognitive decline and dementia become a public health issue. Interventions to improve brain health and thus, quality of life for older adults are needed. OBJECTIVE: It was hypothesized that those consuming a flavonoid-rich, lyophilized wild blueberry powder would evidence improvements in cognitive performance as measured behaviorally and electrophysiologically compared to those consuming a placebo powder across a 6-month intervention period. DESIGN: In a double-blind, randomized placebo-controlled trial, participants experiencing cognitive issues as determined by scores on the Montreal Cognitive Assessment (MoCA) were randomized to consume either wild blueberry (n = 44) or placebo (n = 42) powder daily for 6 months. Participants who were not experiencing any cognitive issues were included as a reference group (n = 45). Participants were tested at baseline and outcome on the Cambridge Neurological Test Automated Battery (CANTAB) and in an electrophysiological paradigm known as event-related potentials (ERP). RESULTS: Tests of specific cognitive abilities using the CANTAB showed speed of processing not only improved in the blueberry intervention group relative to the placebo group across the 6-month intervention, but blueberries also restored speed of processing to the level of the reference group. The ERP results also showed that, relative to those consuming placebo, speed of processing improved for those in the blueberry group; this improvement was most prominent in those 75-80y. CONCLUSIONS: Consumption of wild blueberries for six months improves cognitive aging sequelae by improving the speed of information processing in older adults.Trial registration: ClinicalTrials.gov identifier: NCT01515098.

Research background: The presence of Yersinia enterocolitica on raw food products raises the concern of yersiniosis as most of the berries are consumed raw. This is a challenging issue from the food safety aspect since it could increase the occurrence of foodborne diseases among humans. Thus, it is crucial to implement an effective sanitation before the packaging. Experimental approach: This study aims to synthesize and characterize thymol-loaded polyvinyl alcohol (Thy/PVA) nanoparticles as a sanitizer for postharvest treatment of blueberries. Thy/PVA nanoparticles were characterized by spectroscopic and microscopic approaches, prior to the analyses of antimicrobial properties. Results and conclusions: The diameter size of the nanoparticles was on average 84.7 nm, with a surface charge of -11.73 mV. Based on Fourier transform infrared (FTIR) measurement, the Thy/PVA nanoparticles notably shifted to the frequency of 3275.70, 2869.66, 1651.02 and 1090.52 cm-1. A rapid burst was observed in the first hour of release study, and 74.9 % thymol was released from the PVA nanoparticles. The largest inhibition zone was displayed by methicillin-resistant Staphylococcus aureus (MRSA), followed by Y. enterocolitica and Salmonella typhi. However, amongst these bacteria, the inhibition and killing of Y. enterocolitica required a lower concentration of Thy/PVA nanoparticles. The treatment successfully reduced the bacterial load of Y. enterocolitica on blueberries by 100 %. Novelty and scientific contribution: Thymol is a plant-based chemical without reported adverse effects to humans. In this study, by using the nanotechnology method of encapsulation with PVA, we improved the stability and physicochemical properties of thymol. This nanoparticle-based sanitizer could potentially promote the postharvest microbiological safety of raw berries, which may become an alternative practice of food safety.

Currently, dietary supplements of plant origin based on raw materials containing anthocyanins are widely used. These compounds belong to the class of flavonoids and are glycosides of the flavylic cation. The properties of anthocyanins are associated with their hypolipidemic, hypoglycemic and antioxidant activity. The total content of anthocyanins should be taken into account when developing recipes for dietary supplements. The qualitative composition of individual anthocyanins is an important criterion for the authenticity of this type of product. The purpose of the research was to study anthocyanin content and composition in some dietary supplements that have undergone the procedure of state registration. Material and methods. 34 samples of dietary supplements based on raw materials containing anthocyanins were analyzed. Determination of the total content of anthocyanin pigments was carried out by differential spectrophotometry. The qualitative composition of individual anthocyanins (anthocyanin profile) was determined by reverse-phase HPLC with photometric detection at 510 nm. The peaks of individual compounds were identified by comparing the chromatogram of the sample with experimental and literary data on the order of elution of the most common anthocyanins. Results. The total content of anthocyanins in the analyzed samples varied in a wide range - from 0.013 to 20.8 mg per serving. The study of the anthocyanin profile showed compliance with the declared composition except two samples: in the first, acai extract was used instead of blueberry extract, in the second, black currant extract instead of acai extract. Despite the presence of anthocyanins in the vast majority of dietary supplements studied, only 33% of them can be positioned as sources of anthocyanins. Conclusion. The use of purified extracts with a high content of anthocyanins could solve the problem of low concentration of bioactive compounds in dietary supplements. The conducted research confirms the need for careful monitoring of the content of anthocyanin pigments in products.

Osteoporosis-related bone fragility fractures are a major public health concern. Given the potential for adverse side effects of pharmacological treatment, many have sought alternative treatments, including dietary changes. Based on recent evidence that polyphenol-rich foods, like blueberries, increase calcium absorption and bone mineral density (BMD), we hypothesized that blueberry polyphenols would improve bone biomechanical properties. To test this, 5-month-old ovariectomized Sprague-Dawley rats (n = 10/gp) were orally gavaged for 90 days with either a purified extract of blueberry polyphenols (0-1000 mg total polyphenols/kg bw/day) or lyophilized blueberries (50 mg total polyphenols/kg bw/day). Upon completion of the dosing regimen, right femur, right tibia, and L1-L4 vertebrae were harvested and assessed for bone mineral density (BMD), with femurs being further analyzed for biomechanical properties via three-point bending. There were no differences in BMD at any of the sites analyzed. For bone mechanical properties, the only statistically significant difference was the high dose group having greater ultimate stress than the medium dose, although in the absence of differences in other measures of bone mechanical properties, we concluded that this result, while statistically significant, had little biological significance. Our results indicate that blueberry polyphenols had little impact on BMD or bone mechanical properties in an animal model of estrogen deficiency-induced bone loss.

BACKGROUND: Blueberry is an attractive fruit due to its blue or purple-colored pigments with effective anti-cancer and anti-inflammatory properties. Accordingly, the demand for blueberry cultivation is rapidly increasing. However, various cultivars from different regions are cultivated without characterizing it. METHODS AND RESULTS: We present a method to characterize crops using the SAPD meter. For characterizing each cultivar, the photosynthetic potential of each cultivar, which was measured by SAPD meter three times a day at three heights of a crop, was compared under the same conditions. CONCLUSION: A difference in photosynthetic potential even in the same SHB-type cultivars was found. Therefore, applying cultivation methods suitable to the characteristics of each cultivar is necessary. At the same time, it would be meaningful for researchers and breeders to use this method for genome-wide association study in order to develop molecular markers for increasing photosynthesis efficiency.

Blueberries (Vaccinium spp.) have been considered to be a superfood because of their health benefits. Stem blight or dieback of blueberry has been frequently observed in commercial plantations, with incidences between 15 and 30% being observed in China. The causal agents of blueberry stem blight and dieback were surveyed at four commercial blueberry plantations in the Shandong, Fujian, Guizhou, and Yunnan Provinces of China during 2017 and 2019. Twenty-eight isolates of Neofusicoccum were obtained from 75 diseased and dead stem samples. Two taxa were identified. Of these, one novel species, Neofusicoccum vaccinii, was identified based on morphological characteristics and DNA sequence analysis of the concatenated internal transcribed spacer, the translation elongation factor-1α gene, and the beta-tubulin gene. Koch's postulates tests indicated that N. vaccinii was pathogenic toward blueberry. N. parvum is the other species isolated in this study. The optimal temperature for mycelial growth was 30°C for both N. vaccinii and N. parvum. N. vaccinii, however, was more virulent than N. parvum in this study at temperatures ranging from 25 to 30°C. Coinoculation of N. vaccinii and N. parvum did not lead to increased disease severity. On the contrary, the aggressiveness of N. vaccinii was suppressed by the presence of N. parvum at 25 to 35°C.

BACKGROUND: Gestational diabetes mellitus (GDM) is a growing public health concern and maternal obesity and poor dietary intakes could be implicated. Dietary polyphenols and fiber mitigate the risk of diabetes and its complications, but little is known about their efficacy in preventing GDM. OBJECTIVES: We examined the effects of whole blueberry and soluble fiber supplementation on primary outcomes of cardiometabolic profiles in women at high risk of developing GDM. METHODS: Women (n = 34; mean ± SD age: 27 ± 5 y; BMI: 35.5 ± 4.0 kg/m2; previous history of GDM ∼56%; Hispanic ∼79%) were recruited in early pregnancy (<20 weeks of gestation) and randomly assigned to 1 of the following 2 groups for 18 wk: intervention (280 g whole blueberries and 12 g soluble fiber per day) and standard prenatal care (control). Both groups received nutrition education and maintained 24-h food recalls throughout the study. Data on anthropometrics, blood pressure, and blood samples for biochemical analyses were collected at baseline (<20 weeks), midpoint (24-28 weeks), and end (32-36 weeks) of gestation. Diagnosis of GDM was based on a 2-step glucose challenge test (GCT). Data were analyzed using a mixed-model ANOVA. RESULTS: Maternal weight gain was significantly lower in the dietary intervention than in the control group at the end of the trial (mean ± SD: 6.8 ± 3.2 kg compared with 12.0 ± 4.1 kg, P = 0.001). C-reactive protein was also lower in the intervention than in the control group (baseline: 6.1 ± 4.0 compared with 6.8 ± 7.2 mg/L; midpoint: 6.1 ± 3.7 compared with 7.5 ± 7.3 mg/L; end: 5.5 ± 2.2 compared with 9.5 ± 6.6 mg/L, respectively, P = 0.002). Blood glucose based on GCT was lower in the intervention than in the control (100 ± 33 mg/dL compared with 131 ± 40 mg/dL, P < 0.05). Conventional lipids (total, LDL, and HDL cholesterol and triglycerides) did not differ between groups over time. No differences were noted in infant birth weight. CONCLUSIONS: Whole blueberry and soluble fiber supplementation may prevent excess gestational weight gain and improve glycemic control and inflammation in women with obesity.This trial was registered at clinicaltrials.gov as NCT03467503.

PURPOSE: Acute intervention with wild blueberry (WBB) has previously revealed positive cognitive and mood effects in typically developing children; however, it is unclear whether effects persist after daily supplementation. In addition, no data have been published exploring the metabolite profiles of children following berry consumption, to our knowledge. A study of this kind could provide insight into a mechanism of action for the cognitive and mood improvements observed previously in children. The aim of this pilot study was to assess cognitive performance and urinary metabolite concentrations in healthy 7-10-year-old children across a 4 week daily WBB drink intervention. METHODS: This pilot study examined the effects of daily WBB consumption for 4 weeks (766 mg total polyphenols; 253 mg anthocyanins; equivalent to 240 g fresh blueberries per day) on cognition and mood in 15 healthy 7-10-year-old children. Polyphenol metabolites were measured in 24 h urine before and after the 4 week intervention. RESULTS: Chronic WBB-related benefits were seen on cognitively demanding trials on the modified attention network task, a task measuring executive functioning. Specifically, the WBB group maintained significantly higher accuracy on incongruent trials (96%; SE 0.03) compared with placebo participants (85%; SE 0.03; p = 0.038) after the 4 week intervention, suggesting WBB was of most benefit on the more difficult aspects of the task. No significant WBB-related effects were observed on the auditory verbal learning task or the child's version of the positive and negative affect schedule. Urinary metabolite analyses indicated significant increases in different metabolites in WBB and placebo groups after 4 week consumption. CONCLUSION: The research demonstrates 24 h WBB bioavailability in a child cohort for the first time with increases in urinary hippuric acid excretion during 2 week daily WBB consumption. This study highlights the importance of conducting a larger study in children investigating the mechanism of action behind cognitive effects using bioavailability data.

Invasive blueberry species endanger the sensitive environment of wetlands and protection laws call for management measures. Therefore, methods are needed to identify blueberry bushes, locate them, and characterise their distribution and properties with a minimum of disturbance. UAVs (Unmanned Aerial Vehicles) and image analysis have become important tools for classification and detection approaches. In this study, techniques, such as GIS (Geographical Information Systems) and deep learning, were combined in order to detect invasive blueberry species in wetland environments. Images that were collected by UAV were used to produce orthomosaics, which were analysed to produce maps of blueberry location, distribution, and spread in each study site, as well as bush height and area information. Deep learning networks were used with transfer learning and unfrozen weights in order to automatically detect blueberry bushes reaching True Positive Values (TPV) of 93.83% and an Overall Accuracy (OA) of 98.83%. A refinement of the result masks reached a Dice of 0.624. This study provides an efficient and effective methodology to study wetlands while using different techniques.

It is well known that what we eat can influence our physical wellbeing, but interest is also increasing in the relationship between our diet and cognitive health. In recent years, blueberries have risen from relative obscurity to superfood status following a number of published epidemiological studies, rodent trials, and human RCTs, that suggest blueberries may convey benefits to cognition and mood. This commentary explores some of the evidence in humans, particularly during periods of cognitive development in the young and cognitive decline in the elderly. Evidence for possible mechanisms of action are also described. There is little doubt that blueberries convey a small, but tangible, benefit to cognitive function. Effects are seen following dose sizes easily achievable within a normal diet. Nevertheless, further research is needed on the cognitive domains influenced, additional benefits of longer-term supplementation, mechanisms of action responsible, and the real-world relevance of the cognitive benefits attained.

Adolescence is an important period for cognitive maturation and emotional regulation, and this age group is particularly vulnerable to developing depression. Diets rich in fruits and vegetables have been associated with decreased risk of developing depressive disorders across the lifespan, maybe due to the high flavonoid content of these foods. Previously, we have shown increases in transient positive affect (PA) in both children and young adults 2 h after administration of a wild blueberry (WBB) intervention. Here, using a randomised double-blind, placebo-controlled trial, we investigated the effects of 4 weeks, daily WBB supplementation (containing about 253 mg anthocyanins) on transient and chronic mood in adolescents. Healthy 12-17-year old (n 64, thirty-five females) participants were randomly assigned to receive either a WBB or matched placebo supplementation. Depression and anxiety symptoms were assessed before and after the intervention period using the Mood and Feeling Questionnaire and Revised Child Anxiety and Depression Scale. Transient affect was assessed before, 2 weeks and at 4 weeks using PA and negative affect. Following the intervention period, there were significantly fewer self-reported depression symptoms in participants who were supplemented with WBB compared with placebo (P = 0·02, 95 % CI -6·71, -5·35). There was no between-group effect on anxiety symptoms or on transient affect. Further investigation is required to identify specific mechanisms that link flavonoids consumption and mood. If replicated, the observed effects of WBB supplementation may be a potential prevention strategy for adolescent depression and may have benefits for public mental health.

The present study investigates the effect of anthocyanin (ACN), phenolic acid (PA) fractions, and their combination (ACNs:PAs) from wild blueberry powder (Vaccinum angustifolium) on the speed of endothelial cell migration, gene expression, and protein levels of RAC1 and RHOA associated with acute exposure to different concentrations of ACNs and PAs. Time-lapse videos were analyzed and endothelial cell speed was calculated. Treatment with ACNs at 60 µg/mL inhibited endothelial cell migration rate ( P ≤ 0.05) while treatment with PAs at 0.002 µg/mL ( P ≤ 0.0001), 60 µg/mL ( P ≤ 0.0001), and 120 µg/mL ( P ≤ 0.01) significantly increased endothelial cell migration rate compared with control. Moreover, exposure of HUVECs to ACNs:PAs at 8:8 µg/mL ( P ≤ 0.05) and 60:60 µg/mL increased ( P ≤ 0.001) endothelial cell migration. Gene expression of RAC1 and RHOA significantly increased 2 hours after exposure with all treatments. No effect of the above fractions was observed on the protein levels of RAC1 and RHOA. Findings suggest that endothelial cell migration is differentially modulated based on the type of blueberry extract (ACN or PA fraction) and is concentration-dependent. Future studies should determine the mechanism of the differential action of the above fractions on endothelial cell migration.

PURPOSE: Previous evidence suggests consumption of flavonoids, a sub-class of polyphenols, is associated with improved cognitive function across the lifespan. In particular, acute intervention of a flavonoid-rich wild blueberry (WBB) drink has been shown to boost executive function (EF), short-term memory and mood 2-6 h post-consumption in 7-10-year-old children. However, confirmation of the aspects of EF and memory susceptible to WBB ingestion is required, particularly during childhood, a critical period of neurological development. In addition, the child literature on berry flavonoid supplementation and cognition highlights the potential for such interventions to elicit positive benefits to real-world educational scenarios, such as reading, a complex ability which relies upon aspects of cognition already known to improve following WBB. METHODS: Here we examined which aspects of EF and memory are susceptible to acute WBB, as well as investigating whether acute WBB could further benefit reading ability. Fifty-four healthy children, aged 7-10 years, consumed a 200 ml WBB drink (253 mg anthocyanins) or a matched placebo according to a randomised, single-blind, parallel-groups design. Verbal memory (Auditory Verbal Learning Task; AVLT), EF (Modified Attention Network Task; MANT), and reading efficiency (Test of Word Reading Efficiency-2; TOWRE-2) were assessed at baseline and 2 h post-consumption. RESULTS: For the MANT, significantly quicker RTs were observed for WBB participants when compared to placebo participants on 120 ms trials, without cost to accuracy. Furthermore, WBB participants showed enhanced verbal memory performance on the AVLT, recalling more words than placebo participants on short delay and memory acquisition measures post-consumption. Despite these significant improvements in cognitive performance, no significant effects were observed for reading measures. CONCLUSION: Consumption of WBB was found to significantly improve memory and attentional aspects of EF. This indicates that a flavonoid-rich blueberry product, equivalent to 240 g or 1½ cups of fresh blueberries can provide acute cognitive benefits in children. These findings support accumulating evidence that flavonoid-rich products are beneficial for healthy brain function, particularly during critical developmental periods. However, the lack of findings relating to reading ability suggested acute WBB may not be sufficient to elicit benefits to reading. Chronic supplementation and other more sensitive reading measures should be considered for examining the effects of WBB on such a complex skill in the future.

The objective of this study was to analyze the cardioprotective roles of 3 wild blueberry genotypes and one commercial blueberry genotype by measuring markers of oxidative stress and cell death in H9c2 cardiac cells exposed to doxorubicin. Ripe berries of the 3 wild blueberry genotypes were collected from a 10-year-old clearcut forest near Nipigon, Ontario, Canada (49°1'39″N, 87°52'21″W), whereas the commercial blueberries were purchased from a local grocery store. H9c2 cardiac cells were incubated with 15 µg gallic acid equivalent/mL blueberry extract for 4 h followed by 5 µM doxorubicin for 4 h, and oxidative stress and active caspase 3/7 were analyzed. The surface area as well as total phenolic content was significantly higher in all 3 wild blueberry genotypes compared with the commercial species. Increase in oxidative stress due to doxorubicin exposure was attenuated by pre-treatment with all 3 types of wild blueberries but not by commercial berries. Furthermore, increase in caspase 3/7 activity was also attenuated by all 3 wild genotypes as well. These data demonstrate that wild blueberry extracts can attenuate doxorubicin-induced damage to H9c2 cardiomyocytes through reduction in oxidative stress and apoptosis, whereas the commercial blueberry had little effect.

The viability of murine norovirus (MNV-1), bovine rotavirus (boRV), and hepatitis A virus (HAV) was evaluated at 21 °C, 4 °C, and -20 °C on stainless steel surfaces, in bottled water, and on blueberries for up to 21 days. After 14 days of incubation at 21 °C on stainless steel, a viability loss >4 log for MNV-1, >8 log for boRV, and >1 log for HAV was observed. Losses were observed for MNV-1 (>1 log) and HAV (>2 log) incubated in water at 21 °C for 21 days. No significant loss was detected for MNV-1 and HAV at 4 °C and -20 °C and for boRV at 21 °C, 4 °C, and -20 °C. On blueberries incubated at 4 °C and -20 °C, they all maintained their infectivity. After 7 days at 21 °C, a loss >2 log, a loss of 3 log, and no loss were observed for boRV, MNV-1, and HAV, respectively. After RNase pretreatment, the detection of extracted RNA from infectious and noninfectious samples suggested the protection of RNA inside the capsid. Even though they all are enteric viruses, their persistence varied with temperature and the nature of the commodity. It is therefore important to use more than one viral surrogate, during inactivation treatments or implementation of control measures.

BACKGROUND: Blueberries are appreciated by consumers for their rich natural antioxidants and their good nutritional and health functions. However, blueberries are very perishable due to microbial infection and metabolic aging after harvest. Ethanol has been shown to have the effect of controlling postharvest microorganisms and improving storage quality of fruits and vegetables. This study aimed to clarify the effects of ethanol on the appearance quality and flavor attributes of postharvest blueberries. Blueberries were treated with ethanol (250, 500, and 1000 µL L-1 ) and stored at 0 ± 0.5 °C, 90% relative humidity (RH), for 40 days. RESULTS: The results indicated that ethanol treatment could slow the decline of blueberry firmness and reduce the decay rate significantly in a dose-dependent manner. The soluble solids content (SSC) and titratable acidity (TA) of ethanol-treated blueberries increased significantly (P < 0.05), improving the taste of the blueberries. The activities of alcohol dehydrogenase (ADH) and pyruvate decarboxylase (PDC) were stimulated with the accumulation of ethanol in blueberries, which catalyzed the conversion of ethanol, acetaldehyde, and pyruvate, increasing their levels in blueberries. More volatiles, especially esters, were detected in ethanol-treated blueberries, e.g. methyl acetate, ethyl acetate, ethyl propanoate, ethyl isobutyrate, ethyl 2-methylbutanoate, ethyl isovalerate, ethyl 3-methyl-2-butenoate, diethyl sebacate, and isopropyl myristate. CONCLUSION: The preservative effect of ethanol on blueberry was significantly affected by ethanol concentration. In this study, the effect of 500 µL L-1 ethanol fumigation on blueberry was the best in terms of appearance quality (firmness and decay rate) and flavor attributes (SSC, TA, and volatiles). © 2019 Society of Chemical Industry.

Human telomerase reverse transcriptase (hTERT) encodes the catalytic subunit of telomerase, which has been shown to be upregulated in many cancers. Pterostilbene is a naturally occurring stilbenoid and phytoalexin found primarily in blueberries that exhibits antioxidant activity and inhibits the growth of various cancer cell types. Therefore, the aim of this study was to determine whether treatment with pterostilbene, at physiologically achievable concentrations, can inhibit the proliferation of breast cancer cells and down-regulate the expression of hTERT. We found that pterostilbene inhibits the cellular proliferation of MCF-7 and MDA-MB-231 breast cancer cells in both a time- and dose-dependent manner, without significant toxicity to the MCF10A control cells. Pterostilbene was also shown to increase apoptosis in both breast cancer cell lines. Dose-dependent cell cycle arrest in G1 and G2/M phase was observed after treatment with pterostilbene in MCF-7 and MDA-231 cells, respectively. hTERT expression was down-regulated after treatment in both a time- and dose-dependent manner. Pterostilbene also reduced telomerase levels in both cell lines in a dose-dependent manner. Moreover, cMyc, a proposed target of the pterostilbene-mediated inhibition of hTERT, was down-regulated both transcriptionally and posttranscriptionally after treatment. Collectively, these findings highlight a promising use of pterostilbene as a natural, preventive, and therapeutic agent against the development and progression of breast cancer.

The consumption of berries has been linked to decreased risk of degenerative disease. Berries are regularly processed into juices. It is largely unknown how the juicing process affects the bioavailability of metabolites. As metabolomics has shown to be a valuable nutritional tool to study global metabolite differences, the aim of this study was to investigate the effect of juicing on the relative appearance of blueberry metabolites in humans using metabolomics. Nine healthy subjects consumed 250 g of fresh blueberries either as the whole fruit or after juicing, and provided blood and urine samples before and 2 h after intake in a cross-over design. Samples underwent metabolite profiling using LCMS, and data were mined with multivariate analysis. Overall, <12 % of all ions detected were significantly influenced by blueberry treatment (P<0·05). Partial least-squared discriminant analysis models of post-treatment samples revealed good discrimination. In urinary samples, whole blueberry treatment resulted in 108 ions that were significantly higher compared with juiced treatment (positive and negative mode combined), whereas only eight were significantly higher after juiced treatment. Examples of putative annotations included metabolites of ferulic and caffeic acids, several phenolic metabolites conjugated to sulphate, glycoside or glucuronide and fatty acyl derivatives, which were of higher intensity after whole blueberry treatment. In conclusion, consumption of whole blueberries resulted in a higher range of phenolic and other metabolites in plasma and urine samples 2 h after consumption. Both whole and juiced blueberries resulted in very similar metabolite profiles at 2 h, although this was the only time point measured.

Blueberry, rich in antioxidant and anti-inflammatory phytochemicals, has been demonstrated to lower inflammatory status in adipose induced by high-fat diet (HFD) and obesity. The effect of blueberry on systemic immune functions has not been examined. C57BL/6 mice were randomised to one of three diets - low-fat diet (LFD), HFD and HFD plus 4 % (w/w) blueberry (HFD+B) - for 8 or 12 weeks. Ex vivo T-cell mitogens (concanavalin A (Con A); phytohaemagglutinin), T-cell antibody (anti-CD3; anti-CD3/CD28)-stimulated T-cell proliferation and cytokine production were assessed. After 8 weeks, both HFD groups weighed more (>4 g) than the LFD group; after 12 weeks, HFD+B-fed mice weighed more (>6 g) and had 41 % more adipose tissue than HFD-fed mice (P<0·05). After 12 weeks, T-cell proliferation was less in both HFD groups, compared with the LFD group. HFD-associated decrements in T-cell proliferation were partially (10-50 %) prevented by blueberry supplementation. At 12 weeks, splenocytes from HFD mice, but not from HFD+B mice, produced 51 % less IL-4 (CD3/CD28) and 57 % less interferon-γ (Con A) compared with splenocytes from LFD mice (P<0·05). In response to lipopolysaccharide challenge, splenocytes from both HFD groups produced 24-30 % less IL-6 and 27-33 % less TNF-α compared with splenocytes from LFD mice (P<0·05), indicating impaired acute innate immune response. By demonstrating deleterious impacts of HFD feeding on T-cell proliferation and splenocyte immune responses, our results provide insights into how HFD/obesity can disrupt systemic immune function. The protective effects of blueberry suggest that dietary blueberry can buttress T-cell and systemic immune function against HFD-obesity-associated insults.
