The buffalo (Bubalus bubalis) is a species of worldwide importance, raised to produce milk, meat, and hides, and often used as a working animal in rural contexts with low access to hi-tech solutions. In the present study, 100 lactating buffaloes (50 primiparous and 50 pluriparous) of five popular breeds were recruited to characterize and compare teat morphology. In particular, the focus was put on the Nili Ravi, Mediterranean, Egyptian, Bulgarian Murrah, and Azeri buffaloes raised in Pakistan, Italy, Egypt, Bulgaria, and Iran, respectively. In all countries, a longitudinal cross-section ultrasound was obtained before the milking to measure teat parameters at individual level: overall, teat canal length (TCL) averaged 24.13 mm, teat diameter (TD) 30.46 mm, cisternal diameter (CD) 17.80 mm, and teat wall (TW) 7.12 mm. The most variable trait across breeds was TCL which was positively correlated with CD and TD and negatively with TW, regardless of the teat position (front/rear or left/right). A strong negative correlation was found between TW and CD (- 0.43). The analysis of variance revealed that the fixed effect of breed significantly affected all the traits except TD. In fact, Bulgarian Murrah, Azeri, and Egyptian buffaloes presented the greatest estimate of TCL, whereas NR the smallest (14.70 mm). The TW was maximum in Nili Ravi, Egyptian, and Mediterranean buffaloes, with estimates equal to 8.19, 7.59, and 8.74 mm, respectively. Nili Ravi also showed the greatest TL (82.39 mm). In terms of CD, the lowest least square mean was that of Mediterranean buffaloes (12.14 mm). Primiparous and pluriparous buffaloes differed in terms of TD, TW, and TL, with older animals presenting the highest least square mean. In terms of position, instead, significant differences were observed for TD, CD, and TL when comparing front and rear teats, as left and right teats did not differ. Teat anatomy includes a set of heritable morphological features and is therefore breed-dependent. Differences presented in this study could be attributed to the divergent breeding objective and selective pressure across the five breeds; e.g., in some cases such as Mediterranean buffalo, selection for decades was oriented to improve milk production and milkability and achieve optimal conformation for mechanical milking. A better understanding of the mammary gland anatomical descriptors can be informative of the history of a breed and could provide useful insights to guide possible selection.

In this study, high-throughput gene amplicon sequencing was used to investigate the effects of 6 treatments [2 levels of hemp seed oil (HSO) × 3 levels of cysteamine (CS)] on bacterial and fungal communities in the rumen of 30 crossbred dairy buffalo. Our results indicate that the total numbers of bacterial and fungal taxa were unaffected regardless of diet (p > 0.05), while the total number of archaea was affected (p < 0.05) by the interaction of HSO and CS. Compared with control treatment, microbial composition of archaea was strongly influenced by CS (p < 0.05), while the addition of HSO, CS or both had a weak effect on fungus and bacteria. In addition, there was a significant increase in the lactic acid content with the addition of HSO, and the addition of CS to the feed caused a significant decrease in the ratio of acetic acid to propionic acid, compared with control treatment (p < 0.05). Correlation analysis showed that Acetobacter was significantly positively correlated with the genera Pichia, Klebsiella and Acinetobacter. pH was found to have a significant effect on the methanogens, and total volatile fatty acids (VFA) had a strong correlation with Butyrivibrio. The strong influence of CS on some methanogens shows that it may have potential in the development of methane reduction interventions.

Around 60% dairy animals developed moderate to severe hepatic lipidosis at the time of parturition or during early lactation stage. Most of clinician suspect the hepatic lipidosis during above time window only. However, negative energy balance or feeding of high concentrate diet can lead to hepatic lipidosis at any phase of life. The aim of the present study was to evaluate the potential for diagnosis of hepatic lipidosis by means of hemato-biochemical parameters and ultrasonography of the liver at any stage of life. Here, ultrasonographic back fat thickness measurement was correlated with ultrasonographic features of hepatic lipidosis. A total 60 buffaloes were included under the study and sampled for hematological and biochemical parameters. Hematological parameters did not exhibit any significant difference between healthy and hepatic lipidosis-affected buffaloes. Biochemical parameters like beta hydroxy butyric acid, non esterified fatty acid, aspartate amino transferase, gamma glutamyl transferase and alkaline phosphatase revealed a significant increase, while triglyceride, cholesterol, and glucose declined significantly in hepatic lipidosis-affected buffaloes. Total protein, albumin, and total bilirubin levels did not exhibit any significant difference. Based on ultrasonographic findings, the hepatic lipidosis-affected buffaloes were further sub divided into mild, moderate, and severe groups. Portal vein diameter and depth of portal vein were also estimated in current study. Ultrasonographic examination could diagnose 53.33% hepatic lipidosis cases in buffaloes. Among it, 37.50% buffalo had mild hepatic lipidosis, 33.33% had moderate hepatic lipidosis, and 29.16% had severe hepatic lipidosis. Depth of portal vein significantly increased in hepatic lipidosis cases. However, portal vein diameter exhibited a non-significant difference in mild, moderate, and severe groups of hepatic lipidosis. Back fat thickness also revealed a non-significant difference in mild, moderate, and severe hepatic lipidosis. Above study indicate that B mode ultrasonography of the liver can be employed to differentiate various grades of hepatic lipidosis in buffaloes. Biochemical parameters like NEFA, BHBA, AST, GGT, ALP, TG, cholesterol, and glucose can be helpful to screen the hepatic lipidosis at farm level.

The objectives of this study were to provide the buffalo research community with an updated SNP map for the Axiom Buffalo Genotyping (ABG) array with genomic positions for SNP currently unmapped and to map all cattle QTL from the CattleQTLdb onto the buffalo reference assembly. To update the ABG array map, all SNP probe sequences from the ABG array were re-aligned against the UOA_WB_1 assembly. With the new map, the number of mapped markers increased by approximately 10% and went from 106 778 to 116 708, which reduced the average marker spacing by approximately 2 kb. A comparison of results between signatures of autozygosity study using the ABG and the new map showed that, when the additional markers were used there was an increase in the autozygosity peaks and additional peaks in BBU5 and BBU11 could be identified. After sequence alignment and quality control, 64 650 (UMD3.1) and 76 530 (ARS_UCD1.2) cattle QTL were mapped onto the buffalo genome. The mapping of the bovine QTL database onto the buffalo genome should be useful for genome-wide association studies in buffalo and, given the high homology between the two species, the positions of cattle QTL on the buffalo genome can serve as a stepping stone towards a water buffalo QTL database.

We aimed to evaluate the chemical composition, production yield, and economic viability of Minas Frescal cheese produced with milk from buffaloes supplemented with açai seed as a substitute for corn meal. The experiment used five Murrah crossbred buffaloes with a mean body weight of 577 ± 20.4 kg, maintained under Megathyrsus maximus cv. Mombaça grazing in a 5 × 5 Latin square design. The treatments comprised four concentrate supplements, in which açai seed replaced corn meal at 0, 33, 66, and 100% substitution, and a control with no concentrate supplementation. Replacement of corn meal with açai seed in buffalo diet had no effect on chemical composition of produced cheese (P > 0.05) nor was there any difference in chemical composition between the control group and animals supplemented with concentrate (P > 0.05). The adjusted cheese yield (kg cheese/cow/day) demonstrated a quadratic pattern with maximum production at 33% açai seed substitution (P < 0.05). Hence, the profitability index (percentage of gross revenue) was greatest at this replacement level. In conclusion, 33% açai seed replacement of corn meal in the supplement of buffalo under a grazing regime improved the individual yield of Minas Frescal cheese without changing its chemical composition.

The aim of this study was to determinate whether coagulase-negative staphylococci (CNS) from buffalo milk or the milking environment possess virulence factors that are associated with intramammary infections or antimicrobial resistance. Milk samples (n = 320) from 80 lactating buffalo were evaluated for clinical and subclinical mastitis by physical examination, the strip cup test, California Mastitis Test (CMT), and somatic cell count (SCC) over a 4-mo period. In addition, swabs were obtained from the hands of consenting milkers (16), liners (64), and from the mouths (15) and nostrils (15) of buffalo calves. No clinical cases of mastitis were observed; however, CMT together with SCC results indicated that 8 animals had subclinical mastitis. Eighty-four CNS isolates were identified by MALDI-TOF MS and cydB real-time PCR (qPCR) and then evaluated by qPCR for presence of the eta, etb, sea, sec, cna, seb, sei, seq, sem, seg, see, and tst toxin genes, adhesion- and biofilm-associated genes (eno, ebps, fib, fnbA, coa), and the methicillin resistance gene (mecA). Resistance to antibiotics commonly used for mastitis treatment in Brazil was determined using the Kirby-Bauer test. Two strains were positive for the see and eta toxin genes; and mecA (1), eno (27), ebps (10), fnbA (10), and coa (5) genes were also detected. A notable number of isolates were resistant to erythromycin (30), penicillin (26), and cotrimoxazole (18); importantly, 10 vancomycin-resistant isolates were also detected. A smaller number of isolates were resistant to rifampicin (8), oxacillin (7), clindamycin (5), cefepime (4), tetracycline (3), ciprofloxacin (2), and chloramphenicol (1), and none were resistant to gentamicin or ciprofloxacin. Isolates with resistance to 2 (13 isolates), 3 (3), 4 (3), 5 (1), and 6 (1) antibiotics were detected. Taken together, our findings suggest that CNS isolates may not be a significant cause of clinical or even subclinical mastitis in buffaloes, but they may be a reservoir of virulence and antibiotic resistance genes.

A study was carried out to evaluate the effects of an automatic milking system (AMS) on milk yield and composition of buffalo (Mediterranean-type Bubalus bubalis) cows. Performed from January 2015 to December 2015 in an organic buffalo dairy farm equipped with both a traditional tandem milking parlor and an AMS, the study involved 90 primiparous buffaloes randomly allotted to a tandem or AMS group from 5 to 10 d of lactation onward. Number of milkings per day and daily milk yield of each cow were recorded, and individual milk sampling was carried out twice a month. Compared with the tandem, the AMS group showed significantly higher daily milk yield and persistence of lactation. Use of the AMS resulted in higher protein and casein contents, and lower somatic cell and total bacterial counts, whereas fat, freezing point, and pH were unaffected by the system. We conclude that, in terms of milk yield and quality, automatic milking may be a suitable alternative to conventional milking for buffaloes.

The objectives of this study were to estimate the genetic parameters and genetic trends for different measures of persistency (PM) for milk yield in Iranian buffaloes using a random regression test day model. Test day records of the first three lactations of buffaloes within 715 herds comprising 43 014, 38 941 and 33 937 records, respectively, were obtained from the Animal Breeding Center of Iran during 1992 to 2012. Legendre polynomial functions were chosen to fit the lactation curves in the framework of a random regression test day model for estimating (co)variance components. Three measures of persistency were modified based on the lactation curve conditions of buffaloes: (1) The average of estimated breeding values (EBVs) for test day milk yields from day 226 to day 270 as a deviation from the average of EBVs from day 44 to day 62 (PM1), (2) A summation of contribution for each day from day 53 to day 247 as a deviation from day 248 (PM2), (3) The difference between EBVs for day 257 and day 80 (PM3). The highest heritability estimates were observed for PM2 over the first three lactations (0·29, 0·80 and 0·42, respectively). Estimates of genetic correlations between persistency measures and milk yield varied from -0·48 to 0·29 (first lactation), from -0·12 to 0·48 (second lactation), and from -0·62 to 0·63 (third lactation). Estimate of genetic trend for first lactation PM2 was negative (-0·0088 ± 0·0043; P < 0·05). It is essential to include milk yield persistency in the breeding goal of Iranian buffaloes along with other important traits such as milk yield or reproductive traits.

The aim of this study was to compare seven non-linear mathematical models (Brody, Wood, Dhanoa, Sikka, Nelder, Rook and Dijkstra) to examine their efficiency in describing the lactation curves for milk fat to protein ratio (FPR) in Iranian buffaloes. Data were 43 818 test-day records for FPR from the first three lactations of Iranian buffaloes which were collected on 523 dairy herds in the period from 1996 to 2012 by the Animal Breeding Center of Iran. Each model was fitted to monthly FPR records of buffaloes using the non-linear mixed model procedure (PROC NLMIXED) in SAS and the parameters were estimated. The models were tested for goodness of fit using Akaike's information criterion (AIC), Bayesian information criterion (BIC) and log maximum likelihood (-2 Log L). The Nelder and Sikka mixed models provided the best fit of lactation curve for FPR in the first and second lactations of Iranian buffaloes, respectively. However, Wood, Dhanoa and Sikka mixed models provided the best fit of lactation curve for FPR in the third parity buffaloes. Evaluation of first, second and third lactation features showed that all models, except for Dijkstra model in the third lactation, under-predicted test time at which daily FPR was minimum. On the other hand, minimum FPR was over-predicted by all equations. Evaluation of the different models used in this study indicated that non-linear mixed models were sufficient for fitting test-day FPR records of Iranian buffaloes.

Modern molecular interventions are dynamic gears for breeding animals with superior genetic make-up. These scientific efforts lead us toward sustainable dairy herds with improved milk production in terms of yield and quality. Many of candidate genes have been dissected at molecular level, and suitable genetic markers have been identified in cattle, but this work has not been validated in buffaloes so far. Stearoyl-coenzyme A desaturase (SCD) has been a potential candidate gene for fat content of milk. Genomic analysis of SCD revealed a total of six variations that were identified through DNA sequencing of animals with lower and higher butter fat %age. After statistical analysis, genotype AB of p.K158I could be associated (P value <0.0001) with higher milk fat %age (10.5 ± 0.5464). This SNP was validated on larger data set by cleaved amplified polymorphic sequences (CAPS) by using DdeI. To scrutinize the functional consequences of p.K158I, 3D protein structure of SCD was predicted by homology modeling and this variation was found located in the vicinity of functional domain and a part of transmembrane helix of this membrane integrated protein. This is a first report toward genetic screening of SCD gene at molecular level in buffalo. This report illustrates the implication of SCD gene and in particular p.K158I variation, in imparting its effect on milk fat %age, which can be targeted in selection of superior dairy buffaloes.

Within the general aim of developing a Welfare Quality system for monitoring dairy buffalo welfare, this study focused on prevalence and interobserver reliability of the animal-related variables to be included in the scheme. As most of the measures were developed for cattle, the study also aimed to verify their prevalence for buffaloes. Thirty animal-based measures (22 clinical and 8 behavioral measurements) and 20 terms used for qualitative behavior assessment were assessed in 42 loose-housed buffalo farms. All farms were located in central-southern Italy. Two assessors were used (1 male and 1 female). The time needed to record all measures (animal-, resource-, and management-based) was 5.47 ± 0.48 h (mean ± SD). Interobserver reliability of animal-based measures was evaluated using Spearman rank correlation coefficient test (rs). If 0.7 is considered as threshold for high interobserver reliability, all animal-based measures were above this level. In particular, most of the coefficients were above 0.85, with higher values observed for prevalence of animals that can be touched (rs = 0.99) and prevalence of animals with iatrogenic abscess (rs = 0.97), whereas lower coefficients were found for the prevalence of vulvar discharge (rs = 0.74) and dewlap edema (rs = 0.73). Twelve out of the 20 terms used for the qualitative behavior assessment reached a satisfactory interobserver reliability (rs = 0.65). Principal component analysis of qualitative behavior assessment scores was conducted for each assessor. Both principal component 1 and principal component 2 showed high interobserver reliability (rs = 0.80 and 0.79, respectively). In addition, relevant proportions of animals were affected by welfare issues specific to buffaloes, such as overgrown claws (median = 34.1%), withers hygroma (median = 13.3%), and vulvar or uterine prolapse (median = 9.3%). We concluded that most of the investigated measures could be reliably included in the final scheme, which can be used as such to monitor buffalo welfare. However, to inform consumers about the welfare status of the animals, the data should be integrated into a single overall assessment of animal welfare, as already performed in the Welfare Quality project for dairy cattle.

The objective of this study was to evaluate the response of buffaloes to automatic milking, examining the relationships between milking interval, milk production, and milking time for this species. A total of 7,550 milking records from an average of 40 buffaloes milked by an automatic milking system (AMS) were analyzed during a 3-mo experimental period at a commercial farm with Italian Mediterranean buffaloes in southern Italy. Date and time of animal identification, milk yield, milking duration, milking interval, and average milk flow rate were determined for each milking. The results were also used to predict the maximum number of milkings per day and the optimal number of buffaloes per AMS for different levels of milk production. The average interval period between 2 consecutive milkings was 10.3 h [standard deviation (SD) 3.3]. Overall, 3.4 and 25.7% of the milkings had an interval of ≤ 6 h or >12 h, respectively. Milking duration averaged 8.3 min per buffalo per milking (SD 2.7). The average milk flow rate was 1.3 kg/min (SD 0.5) at a milk yield of 2.8 kg per milking (SD 1.4). Assuming that the milking station is occupied 80% of the time, the number of milkings ranged from 136 to 152 per day and the optimal number of buffaloes per AMS ranged from 59 to 66 when the production level increased from 2 to 5 kg of milk per milking. Automatic milking systems are suitable for buffalo, opening new options for the management of dairy buffalo farms.

The transition period is a critical metabolic phase for dairy ruminants, especially those with high production levels. In spite of this, little is still known about dairy water buffalo. The aim of this study was to evaluate the effect of a commercial feed additive based on diatomaceous earth and hydrolyzed yeasts on health status, milk quality and immune response of buffalo cows during the transition period. Eighty healthy Water buffaloes (Bubalus bubalis) of Italian Mediterranean breed were included in the trial. They were subdivided in two groups: one group received the additive (n = 40) while the control group (n=40) received a placebo. The trial lasted 120 days, from 60 days before calving to 60 days in milk. Blood samples were collected from each buffalo at -60d (60 days from the expected calving), -30 d, 0 d (calving), +15 d, +30 d, and +60 d (respectively, i.e., 15, 30 and 60 days in milking). The biochemical as well as the oxidative profile, and the antioxidant power and enzymatic activity were evaluated in the samples obtained. Moreover, acute phase proteins, reactive proteins and Interleukine plasma levels were determined. Peripheral blood mononuclear cells (PBMC) and monocytes were isolated and viability, reactive oxygen species (ROS) and reactive nitrogen species (RNS) were measured on PMBC and monocytes. The introduction of additives enhanced the total antioxidant capacity and enzyme activity, while no differences were observed in oxidation products throughout the trial. Additionally, it significantly reduced the synthesis of ROS in polymorphonuclear cells, supporting a potential positive response in animals experiencing inflammation. The impact of oxidation on the products was not evident. Despite higher enzyme levels in plasma, this did not necessarily correspond to significantly increased enzymatic activity, but rather indicated a higher potential. From these results, it was evident that the transition period in buffaloes differs notably from what reported in literature for cows, probably due to the absence of common postpartum production diseases in dairy cows and lower metabolic challenges linked to lower milk production in buffaloes. Few parameters exhibited notable changes during the transition period in buffaloes, notably certain antioxidant enzymes, PBMC viability, PBMC ROS production, and Hp levels.

Increases in temperature and the greater incidence of extreme events are the consequences of the climate change that is taking place on planet Earth. High temperatures create severe discomfort to animal farms as they are unable to efficiently dissipate their body heat, and for this, they implement mechanisms to reduce the production of endogenous heat (reducing feed intake and production). In tropical and subtropical countries, where buffalo breeding is more widespread, there are strong negative consequences of heat stress (HS) on the production and quality of milk, reproduction, and health. The increase in ambient temperature is also affecting temperate countries in which buffalo farms are starting to highlight problems due to HS. To counteract HS, it is possible to improve buffalo thermotolerance by using a genetic approach, but even if it is essential, it is a long process. Two other mitigation approaches are nutritional strategies, such as the use of vitamins, minerals, and antioxidants and cooling strategies such as shade, fans, sprinklers, and pools. Among the cooling systems that have been evaluated, wallowing or a combination of fans and sprinklers, when wallowing is not available, are good strategies, even if wallowing was the best because it improved the production and reproduction performance and the level of general well-being of the animals.

Buffaloes are raised mainly to obtain milk that is nutritionally very rich. The technological characteristics of buffalo milk are optimal for processing into cheese, and it is mainly used to produce mozzarella cheese. Under stressful conditions, buffaloes, like other animals, produce milk qualitatively poorly. The stressors that can affect the quality of production are, in addition to other factors, deficiencies in nutrients such as vitamins, antioxidants, and minerals. In this study, we evaluated the effect of antioxidant supplementation on the quality of buffalo milk. Sixty-six buffaloes were enrolled and subdivided into two balanced groups of 33 each. The ZnSe group received 0.2 kg/head/day of Bufalo Plus® containing antioxidants and barley meal, CaCO3 and MgCO3 mix; the control group was supplemented with 0.2 kg/head/day of barley meal, CaCO3 and MgCO3 mix. The two groups were fed ad libitum with a total mixed ration (TMR). The amount of diet distributed was recorded daily, and the residue in the trough manger was recorded three times per week. TMR samples were taken every two weeks for each group. Daily milk yield was recorded twice a week. Milk samples were collected every four weeks and analysed for chemical and technological properties. Furthermore, milk total antioxidant capacity was determined. The results obtained showed that the antioxidant supplement had no effect on feed intake, feeding behaviour, and feed efficiency. The treatment positively influenced milk production while it did not affect the chemical characteristics of the milk. In addition, the supplement of antioxidants improved the milk clotting properties (MCP). The supplement did not affect the antioxidant activity of the milk.

The objective of this study was to compare the accuracies of genomic prediction for milk yield, fat yield, and protein yield from Philippine dairy buffaloes using genomic best linear unbiased prediction (GBLUP) and single-step GBLUP (ssGBLUP) with the accuracies based on pedigree BLUP (pBLUP). To also assess the bias of the prediction, the regression coefficient (slope) of the adjusted phenotypes on the predicted breeding values (BVs) was also calculated. Two data sets were analyzed. The GENO data consisting of all female buffaloes that have both phenotypes and genotypes (n = 904 with 1,773,305-days lactation records) were analyzed using pBLUP and GBLUP. The ALL data, consisting of the GENO data plus females with phenotypes but not genotyped (n = 1,975 with 3,821,305-days lactation records), were analyzed using pBLUP and ssGBLUP. Animals were genotyped with the Affymetrix 90k buffalo genotyping array. After quality control, 60,827 single-nucleotide polymorphisms were used for downward analysis. A pedigree file containing 2,642 animals was used for pBLUP and ssGBLUP. Accuracy of prediction was calculated as the correlation between the predicted BVs of the test set and adjusted phenotypes, which were corrected for fixed effects, divided by the square root of the heritability of the trait, corrected for the number of lactations used in the test set. To assess the bias of the prediction, the regression coefficient (slope) of the adjusted phenotypes on the predicted BVs was also calculated. Results showed that genomic methods (GBLUP and ssGBLUP) provide more accurate predictions compared to pBLUP. Average GBLUP and ssGBLUP accuracies were 0.24 and 0.29, respectively, whereas average pBLUP accuracies (for GENO and ALL data) were 0.21 and 0.22, respectively. Slopes of the two genomic methods were also closer to one, indicating lesser bias, compared to pBLUP. Average GBLUP and ssGBLUP slopes were 0.89 and 0.84, respectively, whereas the average pBLUP (for GENO and ALL data) slopes were 0.80 and 0.54, respectively.

This study aimed to estimate heritabilities and genetic trends for different persistency measures for milk fat yield and their genetic correlations with 270-day milk yield in Iranian buffaloes. The records of test-day milk fat yield belonging to the first three lactations of buffaloes within 523 herds consisting of 43,818 records were got from the Animal Breeding Center and Promotion of Animal Products of Iran from 1996 to 2012. To fit the lactation curves based on a random regression test-day model, different orders of Legendre polynomial (LP) functions were selected. Three persistency measures were altered according to the specific condition of the lactation curve in buffaloes: (1) The average of estimated breeding values (EBVs) for test day fat yield from day 226 to day 270 as a deviation from the average of EBVs from day 44 to day 62 (PM1), (2) A summation of contribution for each day from day 53 to day 247 as a deviation from day 248 (PM2), and (3) The difference between EBVs for day 257 and day 80 (PM3). The estimates of heritability for PM1, PM2, and PM3 ranged from 0.20 to 0.48, from 0.36 to 0.47, and from 0.19 to 0.35 over the first three lactations, respectively. The estimate of genetic trends for different persistency measures of milk fat yield was not significant over the lactations (P > 0.05). Genetic correlation estimates between various measures of persistency were generally high over the first three lactations. Also, genetic correlations estimates between persistency measures and 270-day milk yield were mostly low and varied from 0.00 to 0.24 (between PM1 and 270-day milk yield), from -0.19 to 0.13 (between PM2 and 270-day milk yield), and from -0.02 to 0.00 (between PM1 and 270-day milk yield) over the first three lactations, respectively. Persistency measures that showed low genetic correlations with milk fat yield were considered the most suitable measures in selection schemes. Besides, medium to high heritability estimates for different persistency measures for milk fat yield indicated that relevant genetic variations detected for these characters could be regarded in outlining later genetic improvement programs of Iranian buffaloes.

The domestic buffalo (Bubalus bubalis), also known as water buffalo or Asian buffalo to prevent confusion with the American bison (Bison bison), wrongly named buffalo in North America, comprises two subspecies: the river buffalo (B. bubalis bubalis) and the swamp buffalo (B. bubalis kerebau). The swamp buffalo has a consistent phenotype and is considered as one type, even if many breeds are recognized within it; conversely, the river buffalo subspecies has many breeds. We found limited information available regarding the worldwide distribution of buffaloes. The best estimate is that 208,098,759 buffalo head are distributed in 77 countries in five continents. In this review, we presented the basic aspects of the water buffalo and unraveled the buffalo path followed from the origin of the species to its current global distribution. We reviewed several data sources to provide a better estimate of the world buffalo count and distribution.

Dairy buffalo has an integral role in the sustenance of economics due to its substantial contribution in milk and meat industry, however, the reproduction in this species is challenging. During the last decade, our laboratory conducted a series of experiments to encapsulate the solutions of the problems through optimizing pre- and post-insemination interventions in dairy buffaloes. In an unique study, we proposed that timing of ovulation with reference to the onset of standing heat during spontaneous estrus is delayed, and subsequently re-framed the traditional AM-PM rule (AI after 12 h of standing heat) to AM-AM or PM-PM (AI after 24 h of standing heat) to achieve the optimum fertility using frozen thawed semen in dairy buffaloes. Pregnancy per AI (P/AI) varied substantially either via injecting single shot of prostaglandin (PG) F2α to perform AI at detected estrus or applying standard ovsynch protocol for timed AI (TAI) in buffaloes. However, estrus response, and P/AI remained similar either with used or new controlled internal drug release device in dairy buffaloes. Additionally, the incorporation of estradiol benzoate in progesterone (P4) based protocol resulted in controlled emergence of follicular wave and increased the estrus intensity in buffaloes. Thereafter, we fine-tuned P4-based protocols to optimize the ovulation window for TAI either using GnRH or human chorionic gonadotropin (hCG) or equine chorionic gonadotropin that ultimately improved the fertility in dairy buffaloes. Although, these hormonal interventions resulted in decent fertility, yet it was consistently being compromised due to early or late embryonic losses in dairy buffaloes. Administration of hCG or GnRH on d 7 or 23 or 25 post AI has been proved beneficial to enhance the embryonic survival in buffaloes. Recently, resynchronization program as an aggressive reproductive management approach has been tested that served as a dual-purpose tool to increase overall herd fertility and reduce embryonic losses at commercial buffalo farm operations. Taken together, we concluded that the solutions to the problems of reproductive function are now clearly available with acceptable fertility, however, their application to the small holder buffalo farming remains challenging.

Water buffaloes (Bubalus bubalis) are very sensitive to environmental stimulus before and during milking, and this explains why disrupted milk ejections due to blood oxytocin level instability are frequent in this species. According to the literature, the manual stimulation (MS) of teats before milking promotes oxytocin release and allows milk ejection to start within 2-3 min. However, the pre-milking stimulation of teats is not always part of the milking routine in Italian buffalo farms; moreover, buffaloes with unstable milk let-down are sometimes treated with exogenous oxytocin (OX). Different types of pre-milking stimulation can impact differently on the mammary gland epithelium and structures and, therefore, on milk yield. In this study, we observed the changes in teat morphological traits before and after the application of three types of stimuli, i.e., no manual stimulation (NS), MS, and OX, in buffaloes reared in an Italian dairy farm. In particular, measurements were available for 23 and 21 buffaloes for front and rear teats, respectively. Subsequently, the effect of the pre-milking stimulation type was estimated on teat morphological characteristics and on milk traits recorded after the application of stimuli. The results showed that the teat canal length was shorter (P < 0.05) after stimulation in the case of MS and OX compared to NS. Cistern diameter was overall greater for MS and lower for OX. On the contrary, teat wall thickness was greater and lower for OX and MS, respectively. Milk yield and quality (fat, protein, and somatic cell score) were similar across the three types of pre-milking stimulation. In perspective, the impact of these types of pre-milking stimulation should be evaluated on a large scale, and the focus might be put on mammary gland epithelium integrity, mastitis incidence, and other udder health indicators in milk, e.g., electrical conductivity, differential somatic cell count, lactose content, and sodium and chloride concentration.
