BACKGROUND: The DNA mismatch repair (MMR) system serves as a sophisticated guardian of the precise functioning of the human genome. Dysregulation within this system is linked to the oncogenesis process. Reduced expression of MMR system proteins identified in salivary gland tumors (SGTs) suggests an increased risk of tumoral occurrence. This study aims to analyze the expression of MMR proteins in SGTs and discuss the relevance of this association to the development of these neoplasms. MATERIAL AND METHODS: This review was conducted following the PRISMA guidelines and was registered in PROSPERO (CRD42023465590). A comprehensive search of the PubMed/MEDLINE, Web of Science, Scopus, Embase, and ProQuest (non-peer reviewed platform) was performed to answer the question "Do DNA MMR system proteins exhibit expression in SGTs?". The methodological quality of the selected studies was assessed using the JBI's Critical Appraisal Tool. RESULTS: A total of 142 patients with benign SGTs and 84 with malignant SGTs were included in this review. The literature analysis showed a notable reduction in the expression of DNA MMR system proteins (hHMS2, hMLH1, hMSH3 and hMSH6) in the percentage of marked cells. CONCLUSIONS: The reduction in the expression of the DNA MMR system proteins suggests an interesting correlation with the development of malignant and benign SGTs. Nevertheless, further investigations are warranted to better clarify the precision of measuring biomarker protein expression.

Glioblastoma, which is highly invasive and has a poor patient prognosis, is the most common type of brain tumor. Flavonoids have known antiproliferative and antineoplastic effects, such as apoptosis induction and tumor growth inhibition. We investigated the effects of treatment with three flavonoids (BAS-1, BAS-4, and BAS-6) isolated from the Amazon plant Brosimum acutifolium on the proliferation and migration of the C6 glioma cell line. Cytotoxicity was evaluated by MTT assay, and morphological changes were evaluated by phase-contrast microscopy and by transmission electron microscopy. Apoptosis was determined using Annexin V-FITC-propidium iodide (PI) staining. A hemolysis assay was used to evaluate plasma membrane injury. Antiproliferative effects were assessed by wound migration and colony formation assays. Mitochondrial transmembrane potential (ΔΨm) was determined using JC-1 dye and flow cytometry. To identify the flavonoid targets, western blotting was performed. BAS-1 and BAS-4 reduced C6 cell proliferation in a dose-dependent manner. BAS-6 showed no effect. Due to its high toxicity toward primary glial cells and its high hemolytic index, BAS-1 was not used in the remaining experiments. BAS-4 treatment did not induce cytotoxicity in primary glial cells; however, in glioma cells, it suppressed migration and invasion and led to apoptosis through mitochondrial damage, ΔΨm loss, cell cycle arrest, and reduced AKT phosphorylation, which is a component of the main cell survival pathway. We conclude that BAS-4 showed potential activity against glioma by inducing apoptosis mediated by ΔΨm loss and AKT pathway disruption, and future studies should further evaluate BAS-4 as a promising antineoplastic agent against glioblastoma.

Normal aging is accompanied by renal functional and morphological deterioration and dietetic manipulation has been used to delay this age-related decline. We examined the effects of chronic administration of diets containing 5% lipid-enriched diet (LD, w/w) on renal function of rats at different ages. Three types of LD were tested: canola oil, fish oil and butter. Mean systemic tail-cuff blood pressure and glycemia remained within the normal range whatever the age and the diet of the animals. Proteinuria began to rise from the 8th month in the groups ingesting LD, while in the control group it increased significantly (above 10 mg/24 h) only after the 10th month. With age, a significant and progressive decline in glomerular filtration rate (GFR) and renal plasma flow was observed in the LD groups but after 6 months of lipid supplementation, the decline in these parameters was more marked in the butter and fish oil groups. By the 18th month, the lowest GFR level was observed in the group ingesting the butter diet (2.93 +/- 0.22 vs 5.01 +/- 0.21 ml min(-1) kg(-1) in control, P<0.05). Net acid excretion, evaluated in 9- and 18-month-old rats, was stimulated in the fish oil group when compared both to control and to the other two LD groups. These results suggest that even low levels of LD in a chronic nutritional regimen can modify the age-related changes in renal function and that the impact of different types of lipid-supplemented diets on renal function depends on the kind of lipid present in the diet.

This study evaluated the differences in composition, abundance and morphology of testate amoebae among different habitats of the same aquatic environment (plankton, aquatic macrophyte and sediment) in the Upper Paraná River floodplain. Triplicate samplings were undertaken monthly at each habitat from April 2007 to March 2008. The structure of the community of testate amoebae was different among the habitats. The species typical for each habitat, according to Indval, were classified by their shell morphology. Arcella species together with Difflugia gramen and Difflugia pseudogramem were more abundant for plankton. Trinema and Phryganella stood out by their abundance and frequency in aquatic macrophytes. Centropyxis was an indicator of sediment. The results indicated a higher frequency of hemispherical and spherical shells in plankton and spherical and elongated shells in aquatic macrophytes. In the sediment, there was a high frequency of elongated species. Our results support the hypothesis that the community of testate amoebae has different structures among the habitats, refuting the idea that the organization of this community in plankton is guided by random events like the resuspension of organisms from the sediment and their displacement from marginal vegetation.

We investigated the effect of interannual variation of hydrosedimentological regime and connectivity on the zooplankton biodiversity in the Upper Paraná River floodplain. Zooplankton samplings were undertaken between 2000 and 2007, in different environments of the floodplain, including connected and isolated floodplain lakes, backwaters, rivers and channels. The zooplankton included 541 species. Rotifers showed the highest species richness and abundance. Among the zooplankton species, 71 represent new occurrence records for the floodplain. The species accumulation curve showed a continuous increase in gamma diversity, demonstrating the importance of long-term research for accurate knowledge of biodiversity in heterogeneous and dynamic ecosystems, such as the floodplains. Interannual beta diversity among studied years indicated a lesser alteration in community composition in 2001, when a long limnophase period was observed. In most of the environments, the highest species richness values were related to the greatest flooding amplitudes. Flooding amplitude, which is associated with connectivity, favors faunal exchange amongst the environments and between the pelagic and littoral zones. This explains the occurrence of both planktonic and non-planktonic species within the community. On the other hand, mean zooplankton abundance values were higher when a long isolation period occurred. Differences between the potamophase and limnophase amplitude associated with connectivity among the environments were the most important factors for the structure and dynamics of the zooplankton community in the Upper Paraná River floodplain.

The glutathione S-transferases appear to form part of a protective mechanism against the development of cancer where environmental chemical carcinogens are involved. In humans one member of the mu class gene family (GSTM1) has been shown to be polymorphic and is only expressed in ~50% of individuals. Previous studies have shown a possible link between the null phenotype and susceptibility to cancer but have been equivocal regarding stomach cancer. To evaluate any association in Portuguese gastric cancer individuals with GSTM1 variability, we performed GST M 1 polymorphism by PCR amplification in 148 gastric cancer patients and in 84 healthy control individuals. We found no statistical differences between the gastric cancer and control populations (wild type phenotype: 52%, 48%; null phenotype: 48%, 52%, respectively). A subset analysis into site of tumour also revealed no significant differences between the groups, although we found a slight increase of the wild type phenotype in the samples of the antrum compared with the control population (57% vs 48%, respectively; 2= 1.18; p 0.28) and a slight increase of the null phenotype in the signet ring cells/mucocellular group (2= 1.05; p 0.3). However, in both cases it did not reach statistical significance. A subset analysis of the histological groups following the WHO criteria revealed a statistically significant difference (2= 3.704; p 0.05) between the moderately differentiated gastric adenocarcinoma and the presence of the wild type phenotype. These results do not support the hypothesis that the GSTM1 null phenotype predisposes to gastric cancer in the Portuguese population and the moderately differentiated gastric adenocarcinoma seems to be associated with the presence of the G STM 1 wild type phenotype.

O objetivo deste trabalho foi investigar o efeito da variação plurianual do regime hidrosedimentológico e da conectividade sobre a biodiversidade zooplanctônica da planície de inundação do Alto Rio Paraná. As amostragens do zooplâncton foram realizadas entre os anos de 2000 a 2007, em distintos ambientes dessa planície de inundação, incluindo lagoas abertas, lagoas fechadas, ressacos, rios e canais. A comunidade zooplanctônica foi constituída por 541 espécies, sendo os rotíferos o grupo mais especioso e abundante. Dentre essas espécies, 71 representam novas ocorrências para a planície de inundação. A curva de acumulação de espécies mostrou um incremento continuo da diversidade gama evidenciando a importância de estudos de longa duração para o conhecimento da real biodiversidade em ecossistemas heterogêneos e dinâmicos, como são as planícies de inundação. Os resultados da diversidade beta entre os anos estudados mostraram uma menor alteração da composição da comunidade em 2001, quando foi observado um longo período de limnofase. A maior riqueza média de espécies foi associada à grande amplitude de alagamento na maioria dos ambientes. A duração da inundação, associada à conectividade, favorece o intercâmbio de fauna entre os ambientes, bem como entre as regiões pelágica e litorânea, propiciando a ocorrência de espécies planctônicas e não planctônicas na comunidade. Por outro lado, a abundância da comunidade zooplanctônica foi maior quando ocorreu um extenso período de isolamento. Diferenças entre a amplitude de potamofase e a limnofase associada à conectividade entre os ambientes foram fatores preponderantes para estruturação e dinâmica da comunidade zooplanctônica na planície de Inundação do Alto Rio Paraná.
