RESUMEN

The aim of the study was to evaluate the effects of varying concentrations of ethylenediaminetetraacetic acid (EDTA) on blood samples from White Fulani breed of cattle and West African Dwarf goat from Nigeria. Sample sizes of 20 animals were used for both species. Different concentrations of EDTA (2, 4, 8 and 16 mg/ml) were used. The packed cell volume (PCV), red blood cell (RBC) and haemoglobin (Hb) concentration of blood samples collected from White Fulani breed of cattle and West African Dwarf goat into bottles containing 16 mg/ml of EDTA were significantly lower (P < 0.05) than those samples collected from the same animals into bottle containing 2 mg/ml (control). Similarly, the PCV, RBC and Hb values of the West African Dwarf goats in bottles containing 8 mg/ml of EDTA were significantly lower than those of the samples in the control (2 mg/ml). This study has shown that high concentration of EDTA as an anticoagulant can lead to a false erythrocytic index especially the PCV. In collecting blood samples for evaluation of haematological parameters, therefore, the blood volume/anticoagulant ratio must be strictly adhered to prevent error in the evaluated parameters in cattle and goats. Taken together, there is tendency for haemolytic anaemia to occur in blood sampled at higher concentration of anticoagulants in West African Dwarf goat than in White Fulani breed of cattle.

BACKGROUND: Concerted efforts to identify the pathogenesis and mechanism(s) involved in pansteatitis, (a generalized inflammation of the adipose tissue), that was attributed to the recent crocodile die off in the Olifants River and Loskop Dam in Kruger National Park, Mpumalanga, South Africa have been in the forefront of research in recent time. As part of the efforts, molecular characterization of healthy and pansteatitis adipose tissue was carried out by RNA sequencing (RNA-Seq) using Next Generation Sequencing (NGS) and de novo assembly of the adipose transcriptome, followed by differential gene expression analysis. METHODOLOGY: Healthy adipose tissue consisting of fifty samples was collected from the subcutaneous, visceral, intermuscular adipose tissues and the abdominal fat body of ten 4 years old juvenile crocodiles from a local crocodile farm in Pretoria, South Africa. Ten pansteatitis samples were collected from visceral and intermuscular adipose tissues of five crocodiles that were dying of pansteatitis. RESULTS: Forty-two thousand, two hundred and one (42,201) transcripts were assembled, out of which 37, 835 had previously been characterized. The de novo assembled transcriptome had an N50 (average sequence) of 436 bp, percentage GC content of 43.92, which compared well with previously assembled transcripts in the saltwater crocodile. Seventy genes were differentially expressed and upregulated in pansteatitis. These included genes coding for extracellular matrix (ECM) signaling ligands, inflammatory cytokines and tumour necrosis factor alpha (TNFα) receptors, fatty acid synthase and fatty acid binding proteins, peroxisome proliferator-activated receptor gamma (PPARγ), nuclear factor and apoptosis signaling ligands, and mitogen activated protein kinase enzymes among others. Majority (88.6%) of the upregulated genes were found to be involved in hypoxia inducible pathways for activation of NFkß and inflammation, apoptosis, Toll-like receptor pathway and PPARγ. Bicaudal homologous 2 Drosophila gene (BICD2) associated with spinal and lower extremity muscle atrophy was also upregulated in pansteatitis while Sphingosine -1-phosphate phosphatase 2 (SGPP2) involved in Sphingosine -1- phosphate metabolism was downregulated. Futhermore, Doublesex-mab-related transcription factor 1 (DMRT1) responsible for sex gonad development and germ cell differentiation was also downregulated. CONCLUSION: Thus, from the present study, based on differentially expressed genes in pansteatitis, affected Nile crocodiles might have died partly due to their inability to utilize stored triglycerides as a result of inflammation induced insulin resistance, leading to starvation in the midst of plenty. Affected animals may have also suffered muscular atrophy of the lower extremities and poor fertility.

RESUMEN

A case of polymelia with a rudimentary wing is described in an eight-week-old Nera black chicken (Gallus domesticus). It is a rare disorder with chromosomal aberrations, which are associated with congenital limb malformations. The condition was observed during routine physical examination of a flock of 2000 poultry birds in the month of February 2008 in Ibadan, Nigeria. After physical examination, the bird was found to have two extra well-developed legs, which were shorter than the normal legs. These extra legs were also found to be non-functional. Similarly, a rudimentary wing, which was highly vestigial, was found on the left lateral side of the bird very close to the cloaca. The bird died at the age of eight weeks. This is the first reported case of polymelia with a rudimentary wing in a domestic chicken in Nigeria.

This study was designed to investigate the effects of prolonged oral administration of calcium hypochlorite in the drinking water of commercial cockerels. It was carried out in order to ascertain probable toxicity associated with prolonged exposure to calcium hypochlorite. Thirty-two healthy birds were used; they were grouped into four groups of eight. Group 1, which served as the control, received 10 mL/kg body weight of physiological saline. Groups 2, 3 and 4 received 0.0375 g, 0.375 g and 0.75 g of calcium hypochlorite per 10 litres of drinking water for six weeks respectively. Six weeks after the administration of calcium hypochlorite, blood was collected from the jugular vein to assess liver function, lipid profiles and for markers of oxidative stress. The results revealed a significant (p < 0.05) increase in alanine aminotransferase activity in a dose-dependent manner when compared with the control. Also, there was a significant (p < 0.05) increase in aspartate aminotransferase and alkaline phosphatase activity. Similarly, there was a significant (p < 0.05) increase in total cholesterol, triglycerides, high-density lipoprotein and low-density lipoprotein levels compared with the control. There was a significant increase in malondialdehyde and hydrogen peroxide generation with a concomitant significant (p < 0.05) decrease in serum glutathione level in a dose-dependent manner when compared with the control. In this study, calcium hypochloriteinduced hepatic damage via oxidative stress and decrease in antioxidant defense system was found. Therefore, prolonged exposure of chickens to calcium hypochlorite is potentially harmful.

RESUMEN

The study was designed to carry out the phytochemical screening and the proximate analysis of Cnidoscolus aconitifolius leaves. The results obtained showed the presence of tannins, saponin, alkaloids, and flavonoids with the absence of glycosides. The proximate analysis and mineral composition of C. aconitifolius leaves showed high levels of crude protein, ash, and fiber, in that order, and low fat content with concomitant presence of minerals such as sodium, manganese, magnesium, potassium, calcium, iron, phosphate, and zinc. The leaves of C. aconitifolius have high nutrient potentials and could be used as nutraceuticals in complementary foods, especially in sub-Saharan Africa.

A wide variety of phenolic compounds derived from spices possess potent antioxidant, anti-inflammatory, antimutagenic, and anticarcinogenic activities. [6]-gingerol (1-[4'-hydroxy-3'-methoxyphenyl]-5-hydroxy-3-decanone) is the major pungent principle of ginger, with numerous pharmacological properties including antioxidant, anti-inflammation, and antitumor promoting properties. It could decrease inducible nitric oxide synthase (iNOS) and tumor necrosis factor alpha (TNF-alpha) expression through suppression of I-kappaB alpha (IkappaBalpha) phosphorylation, nuclear factor kappa B (NF-kappaB) nuclear translocation. Other antiproliferative mechanisms of [6]-gingerol include the release of Cytochrome c, Caspases activation, and increase in apoptotic protease-activating factor-1 (Apaf-1) as mechanism of apoptosis induction. Taken together, the chemopreventive potentials of [6]-gingerol present a promising future alternative to therapeutic agents that are expensive, toxic, and might even be carcinogenic.
