RESUMEN

A paradox in evolutionary biology is how supergenes can maintain high fitness despite reduced effective population size, the suppression of recombination, and the expected accumulation of mutational load. The ruff supergene involves 2 rare inversion haplotypes (satellite and faeder). These are recessive lethals but with dominant effects on male mating strategies, plumage, and body size. Sequence divergence to the wild-type (independent) haplotype indicates that the inversion could be as old as 4 million years. Here, we have constructed a highly contiguous genome assembly of the inversion region for both the independent and satellite haplotypes. Based on the new data, we estimate that the recombination event(s) creating the satellite haplotype occurred only about 70,000 yr ago. Contrary to expectations for supergenes, we find no substantial expansion of repeats and only a modest mutation load on the satellite and faeder haplotypes despite high sequence divergence to the non-inverted haplotype (1.46%). The essential centromere protein N (CENPN) gene is disrupted by the inversion and is as well conserved on the inversion haplotypes as on the noninversion haplotype. These results suggest that the inversion may be much younger than previously thought. The low mutation load, despite recessive lethality, may be explained by the introgression of the inversion from a now extinct lineage.

Cutaneous melanoma (cM) is a prevalent invasive cancer resulting from the malignant transformation of melanocytes. At present, the primary treatment for melanoma is surgical resection, which is not appropriate for patients with metastasis. Therefore, it is necessary to identify effective therapeutic targets for the early diagnosis and treatment of metastatic melanoma. Acyl-CoA thioesterase 7 (ACOT7) has been reported to be involved in the progression of multiple cancer, while its role in melanoma has not been extensively researched. Through gain-of-function and loss-of-function experiments, ACOT7 was identified as a tumor promoter that facilitates the progression of melanoma cells. Cell proliferation was promoted by overexpressing ACOT7 in M14 cells, and was suppressed by silencing ACOT7 in MeWo cells. Knockdown of ACOT7 induced cell cycle arrest by increasing the expressions of cyclin dependent kinase inhibitor 1B (P27) and cyclin dependent kinase inhibitor 1 A (P21), while simultaneously reducing proliferating cell nuclear antigen (PCNA) expression. Upregulation of ACOT7 promoted the cell cycle of melanoma cells. Additionally, apoptosis was induced by the absence of ACOT7 through activating caspase-3 and poly (ADP-ribose) polymerase (PARP). The metastatic and invasive capacity of melanoma cells was significantly enhanced by the overexpression of ACOT7 and inhibited by the downregulation of ACOT7. Moreover, the cAMP responsive element binding protein 1 (CREB1) positively regulates ACOT7 expression by binding to its promoter region. A decrease of cell proliferation, migration and invasion, as well as an increase of cell apoptosis induced by silencing CREB1 were obviously reversed by ACOT7. In summary, ACOT7 transcriptionally activated by CREB1 elevates the progression of cM.

Frost-free areas have suitable climate for wetland plant growth and constructed wetlands (CW) technology. Information on the quantification of plant biomass and uptake efficiency in field-scale CWs is limited in these climates. The removal efficiency of total nitrogen (TN), total phosphorus (TP), chemical oxygen demand (COD), and total suspended solids (TSS) in wastewater from sewage plants, domestic sewage, and an industrial park in 15 rural and urban CWs in Guangdong Province, China, with an average temperature of 30 °C was evaluated. The effects of influent concentration, hydraulic load, the wastewater's physicochemical properties, operating conditions, and plant uptake were analysed. The mean removal rates were 40.0%, 45.2%, 41.1%, and 71.7% for TN, TP, COD, and TSS, respectively, which were higher than the removal load of the field-scale CWs in temperate regions. Removal loads of TN, TP, COD, and TSS were highest in CWs that have been operating for 5-6 years, treating wastewater volumes of over 1 m3/m2·d. The removal efficiency was mainly related to the inflow concentration and less affected by the type of CWs. Nutrient accumulation trends were primarily linked to influent concentrations (TN: r2 = 0.89, P = 0.007; TP: r2 = 0.96, P = 0.001) and plant biomass (TN: r2 = 0.96, P = 0.001; TP: r2 = 0.92, P = 0.004). Plant biomass contributed 2%-29% and 2%-70%, respectively, to removing N and P in CWs. The average uptake concentration of N and P in aboveground plant organs (15.66 ± 4.44 mg N/g, 2.15 ± 1.18 mg P/g) was generally higher than that of other temperate plants. A strong relationship between TN and TP in the biomass was also observed; however, the relationship is only restricted by the influent TP concentration. Arundo donax is well-adapted for nutrient accumulation and adaptation and is an ideal wetland plant to purify wastewater in frost-free climates.
