RESUMEN

The main purposes of this work are designing new hybrid structures based on alumina nanoporous membranes with specific metallosupramolecular structure as well as studies of their usefulness in nonlinear optics (NLO). The NLO studies of the hybrid material is performed on the basis of two methods: the first by the Maker fringe technique, where the second harmonic generation (SHG) signal is recorded by rotating the sample; and the second by SHG imaging microscopy, where the SHG signal is collected point by point on the sample surface. The enhanced SHG signals were obtained without the use of the corona poling method needed during the experiment on thin films in our previous works and clearly shows the efficiency of hybrid materials based on nanoporous membranes as promising materials in devices developed based on NLO.

RESUMEN

The fabrication of one-dimensional (1D) nanostructures and microstructures inside the pores of porous templates is intensively investigated. The release of these structures is commonly accomplished by etching and destroying the templates. The 1D nanostructures and microstructures tend to condense because of the occurrence of capillary forces during drying of the specimens. It is shown that highly ordered arrays of polymer microfibers can be easily detached from silanized porous templates by mechanical lift-off. This procedure leaves the templates intact, thus allowing their recycling, and does not involve the use of solutions or solvents, thus circumventing condensation. Therefore, mechanical lift-off may enable the up-scaling of template-based approaches to the fabrication of highly ordered assemblies of 1D nanostructures and microstructures.

RESUMEN

This study describes a novel assay to visualize the macromolecular permeability of epithelial and endothelial cell layers with subcellular lateral resolution. Defects within the cell layer and details about the permeation route of the migrating solute are revealed. The assay is based on silicon chips with densely packed, highly ordered, dead-ended pores of µm-diameters on one side. The cells under study are grown on the porous side of the chip such that the pores in the growth surface serve as an array of femtolitre-sized cuvettes in which the permeating probe accumulates at the site of permeation. The pattern of pore filling reveals the permeability characteristics of the cell layer with a lateral resolution in the µm range. Coating of the chip surface with a thin layer of gold allows for impedance analysis of the adherent cells in order to measure their tightness for inorganic ions at the same time. The new assay provides an unprecedented look on epithelial and endothelial barrier function.

RESUMEN

We show that the crystal orientation in polymer nanotubes and nanorods inside porous templates is controlled by the kinetics of nucleation and growth under 2D confinement. Two clear limiting cases are identified: In separated nanostructures, any crystal orientation allowing the growth of lamellar crystals along the pores appears statistically. If a bulklike surface film connects the nanostructures, macroscopic arrays with uniform crystal orientation are obtained, in which the dominant growth direction of the crystals is aligned with the long axes of the pores of the template.

RESUMEN

Aligned liquid crystalline nanowires within ordered porous alumina templates show a pronounced texture on a macroscopic scale. We have investigated the influence of the geometric confinement and the nature of the pore walls on the mesophase formation by means of X-ray diffraction. The apparent texture is the result of a complex interplay of the pore geometry, interfacial phenomena, and the thermal history. Pores with a diameter of a few hundred nm guide the mesophase formation more efficiently than those with a diameter below 100 nm.

We report for the first time on pore-suspending lipid bilayers, which we call micro-black lipid membranes (micro-BLMs), based on a highly ordered macroporous silicon array. Micro-BLMs were established by first functionalizing the backside porous silicon surface with gold and then chemisorbing 1,2-dipalmitoyl-sn-glycero-3-phosphothioethanol followed by spreading 1,2-diphytanoyl-sn-glycero-3-phosphocholine dissolved in n-decane. Impedance spectroscopy revealed the formation of single lipid bilayers confirmed by a mean specific capacitance of 0.6 +/- 0.2 microF/cm2. Membrane resistances were in the G omega-regime and beyond. The potential of the system for single channel recordings was demonstrated by inserting the transmembrane domain of the HIV-1 accessory peptide Vpu(1-32), which forms helix bundles with characteristic opening states. We elucidated different amilorides as potential drugs to inhibit channel activity of Vpu.
