RESUMEN

Extracellular concentration of heat shock protein (Hsp) with a molecular weight of 70 kDa (Hsp70) rapidly increases in the serum in response to stress and returns to the basal level during recovery. Further regulation of its blood concentration is unclear. A possible regulator is HspBP1, a protein binding Hsp70. Binding to ATPase domain of Hsp70, HspBP1 inactivates it, thus acting as a factor of nucleotide exchange. Blood sera from athletes were examined at the beginning and end of the last mesocycle of the training period by two-staged immunoaffinity test system. The concentration of HspBP1 increased with decreasing Hsp70 concentration under conditions of long-term training. Presumably, the dynamics of Hsp70 and HspBP1 concentrations can serve as the test for evaluating the adaptation potential.

RESUMEN

The mechanisms of early activation of natural killers were studied in volunteers of different athletic qualification. The initial levels of expression of KIR2DL2, PRF1, and GZMB genes regulating the functions of natural killer differ in athletes and untrained subjects. Moderate exercise stimulates transcription activities of these genes. In athletes, the expression increases more intensely than in controls. Stimulation of inhibitory (KIR2DL3) and activation (KIR2DS2) receptors was revealed. This indicated nonspecific stimulation of natural killers, probably mediated by an increase in serum concentration of heat shock protein with a molecular weight of 70 kDa.

RESUMEN

Latrophilin 1 (presynaptic receptor) binds α-latrotoxin from black widow spider venom and regulates neurotransmitter release from nerve endings. The study of the mechanism of action of this receptor is impeded by the existence of closely related latrophilins 2 and 3. A profile of differences detecting the most differing and identical sites in several proteins was developed in order to obtain highly specific antibodies for differentiation between isoforms of related proteins. In addition, we used an algorithm for prediction of immunogenic sites of the protein, based on the basic vector method. The peptides selected using this algorithm were used for immunization of animals. The resultant sera exhibited the estimated specificity and high affinity for the corresponding receptor forms.

RESUMEN

We developed a method of rapid assay for biochemical properties of the saliva based on attenuated total reflection infrared spectroscopy. This method allows evaluating saliva composition within 5 min without sample preparation; 10 microl sample is enough for the analysis. The concentration of total protein, glucose, secretory immunoglobulin A, urea, amylase, cortisol, inorganic phosphate in the saliva can also be measured. Precision and reproducibility of the evaluated parameters are comparable to those obtained by routine clinical analysis.

RESUMEN

Studying the copy number of ribosomal protein L7/L12 was performed using monoclonal antibody 3G9 to the linear epitope on the C-terminal domain of this protein from Escherichia coli. Immunohistochemical study showed that Agrobacterium tumefaciens ribosomes include 6 copies of protein L7/L12. Our results suggest that the copy number of this protein has an evolutionary role.

RESUMEN

The effects of 6-h marathon ultra-race (long aerobic work below the lactate threshold level) on the levels of IL-6, leukemia inhibiting factor (LIF), and stem cell growth factor (SCF) were studied. The athletes participating in the study had different endurance levels evaluated by the distance covered over 6 h. The level of IL-6 sharply increased after exercise. The degree of IL-6 increase correlated with the length of the distance covered (r=0.83, p=0.042). The concentration of LIF after exercise inversely correlated with the distance covered (r=-0.75), but this correlation was statistically insignificant. The IL-6/LIF proportion exhibited the highest correlation with the result in the marathon ultra-race. This parameter most fully characterized athlete endurance (r=0.92, p=0.009). Hence, the relationship of LIF with physical endurance was demonstrated. Involvement of LIF in antibody production processes can be responsible for it.
