RESUMEN

Dermatophytes are the most common causative agents of cutaneous mycosis and remain a major public health problem in spite of the availability of an increasing number of antifungal drugs. It was, therefore considered necessary to pursue the screening of different extracts (compounds) of selected traditional medicinal plants reportedly having antidermatophyte potential. The aim of this study was to isolate and identify specific compound from the most active extract (free flavonoid) of stem of Terminalia chebula of the selected plants to treat dermatophytosis induced on experimental mice. Mice which were experimentally induced with Trichophyton mentagrophytes were grouped in six of five animals each. To treat the lesions on infected mice, two concentrations of isolated apigenin ointment, i.e. 2.5 mg g(-1) (Api I) and 5 mg g(-1) (Api II), and terbinafine (standard) of concentration 5 mg g(-1) were used. Complete recovery from the infection was recorded on 12th day of treatment for reference drug Terbinafine and Api II (5 mg g(-1) ) concentration of ointment, whereas Api I (2.5 mg g(-1) ) ointment showed complete cure on 16th day of treatment. Fungal burden was also calculated by culturing skin scraping from infected mice's of different groups. Apigenin has shown potency as the infected animals recover completely by Api II comparable to the standard drug in 12th day. So Apigenin can be explored as an antifungal agent in the clinical treatment of dermatophytosis in future.

BACKGROUND & OBJECTIVES: Development of insect resistance to synthetic pesticides, high operational cost and environmental pollution have created the need for developing alternative approaches to control vector-borne diseases. In the present study, larvicidal activity of flavonoid extracts of different parts of Vitex negundo (Linnaeus) and Andrographis paniculata (Nees) have been studied against the late III or early IV instar larvae of Aedes aegypti and Anopheles stephensi (Liston). METHODS: Flavonoids were extracted from different parts of the selected plants using standard method. Bioassay test was carried out by WHO method for determination of larvicidal activity against mosquitoes. Different compounds of the most active extract were identified by the gas chromatography-mass spectrometry (GC-MS) analysis. RESULTS: Flavonoid extract of whole aerial part of A. paniculata was found to be inactive against the selected larvae of Ae. aegypti even at the concentration of 600 ppm, whereas it caused 70% mortality in An. stephensi at the concentration of 200 ppm. Flavonoid extract of flower-buds produced highest mortality (100%) at the concentration of 600 ppm for the late III or early IV instar larvae of Ae. aegypti and at the concentration of 200 ppm for the larvae of An. stephensi. GC-MS analysis of the most active flavonoid extract from flower-buds of Vitex showed 81 peaks. Phenol (26.83% area), naphthalene (4.95% area), 2,3-dihydrobenzofuran (6.79% area), Phenol-2,4-Bis (1,1-dimethyl) (4.49% area), flavones 4'-OH,5-OH,7-di-O-glucoside (0.25% area) and 5-hydroxy- 3,6,7,3',4'-pentamethoxy flavones (0.80% area) were present in major amount. CONCLUSION: Flavonoid extracts from different parts of two selected plants possess larvicidal activity against two selected mosquito species, hence, could be utilized for developing flavonoid-based, eco-friendly insecticide as an alternative to synthetic insecticides.

During antifungal evaluation of various plant extracts, free and bound flavonoids of Piper betle were found to be most effective as an antidermatophytic against human pathogenic dermatophytes Trichophyton rubrum, Trichophyton mentagrophytes, Microsporum gypseum and Candida albicans. Dermatophytic fungi cause both superficial and internal mycoses. These mycoses, although normally not lethal, are unpleasant and difficult to cure and cause considerable financial losses. Earlier workers prove that allopathic drugs are still found effective against dermatomycoses, but these drugs could not be accepted as a routine treatment for every case, because they are expensive and require long treatment. It is almost unaffordable by middle and lower class people. In view of such prospects and constraints, our aim was to explore more new compounds of plant origin for controlling dermatophytic infections. Author explored water, methanolic and flavonoid extracts for screening as antidermatophytic agent. Plant extracts that showed good results in vitro were selected for clinical studies. The study may give cheaper treatment for medium and lower class patients suffering with tinea and may provide them much relief. Well-established paper disc method was used for the screening of different extracts of their antidermatophytic activity. Moreover, it did not exhibit any adverse side effect on mammalian skin. Flavonoids in the form of ointment Pi be I and Pi be II were subjected to topical testing on patients attending out patients department of S.M.S. Hospital, Jaipur, India. Patients were diagnosed as tinea corporis, tinea capitis, tinea manum or tinea pedis. All patients showed positive potassium hydroxide (KOH) results at the beginning of trial. Patients between the ages of 3 months to 58 years were enrolled. At the end of treatment, while 64% of patients cured completely, 24% showed significant improvement and 12% showed little improvement from the disease. Allopathic treatment took 4-8 weeks when compared with only 1-5 weeks taken by Pi be I and Pi be II ointments. The ointment, thus not only showed maximum affectivity but was also found to be a better and cost effective alternative to allopathic drug for tinea infections. The acceptability of alternative medicines, particularly herbal medicines, has now become a critical need of the times.

Gadolinium as gadolinium nitrate is used as neutron poison in the moderator system for regulating and controlling the power generation of Pressurized Heavy Water Reactors (PHWR) and proposed to be used in Advanced Heavy Water Reactors (AHWR) owing to its high neutron absorption cross section. Removal of the added gadolinium nitrate (Gd3+ and NO3-) from the system after its intended use is done using ion exchange resins. In the present investigation, attempts have been made to optimize the ion exchange process for generation of low radioactive waste and maximize utilization of the ion exchange resins by employing different types of resins and different modes of operation. The investigations revealed that use of mixed bed (MB) resin column consisting of Strong Acid Cation (SAC) resin and Strong Base Anion (SBA) resin followed by SAC resin column is efficient in removing the Gd3+ and NO3- from the system besides maintaining the pH of the moderator system in the desirable regime, where gadolinium does not get precipitated as its hydroxide.

BACKGROUND: Epicardial adipose tissue thickness (EATT) measured by echocardiography is a reliable indicator of visceral adipose tissue in the body and metabolic syndrome. The objective of this work was to study the effect of 12-week aerobic exercise training on echocardiographic epicardial adipose tissue thickness (EEATT), body composition and other metabolic parameters in asymptomatic overweight adults. METHODS: A total of 170 overweight and mild obese individuals were randomly allocated to 12-week aerobic exercise program (intervention group) or no supervised exercise (control group). The body fat percentage and visceral fat levels were measured by bioelectric impedance analyzer, and blood tests were conducted to measure lipids, fasting blood sugar (FBS) and high-sensitivity C-reactive protein (Hs-CRP). The participants also underwent transthoracic echocardiography to measure EATT. All the outcomes were measured after 12 weeks. Repeated measures were analyzed using ANOVA and Mann-Whitney U-test were used to analyze the data. RESULTS: EEATT values significantly reduced by -16.24% (3.12±1.18 to 2.70±1.04, p<0.05) following 12 weeks of aerobic exercise along with a reduction in weight by 2.96% (80.66±12.12 to 78.27±12.31, p<0.05), body mass index by 3.11% (29.20±2.71 to 28.29±2.81, p<0.05) and waist circumference by 2.29% (94.82±7.23 to 92.64±7.33, p<0.05) and improved Hs-CRP values. CONCLUSION: The 12-week aerobic exercise was found to be effective in reducing EEATT in overweight and mild obese individuals. There was significant improvement in body composition, blood lipids, FBS, Hs-CRP and exercise capacity in intervention group compared to control group.

BACKGROUND: Terminalia chebula is called the "king of medicines" in Tibet and is always listed first in the Ayurvedic meteria medica because of its extraordinary powers of healing. OBJECTIVE: Identification, isolation and screening of pyrogallol which are responsible for antimicrobial property of fruits of Terminalia chebula. MATERIALS AND METHODS: Ethyl acetate fraction of fruits of Terminalia chebula was subjected to Gas chromatography-mass spectrometry (GC-MS) for the components present in the extract. RESULTS: Sixty four constituents were identified out of which kaempferol-3-O-rutinoside flavonoid and Vitamin E has been detected for the first time in fruits of this plant. Pyrogallol (46.26%) which was the major component of the extract in GC-MS analysis was isolated and screened for antimicrobial activity against selected test pathogens by Disc Diffusion Assay. Crude ethyl acetate fraction of the fruits was showing the same activity potential as was observed for pure pyrogallol which was the major component as per GC-MS analysis. The most sensitive species among the bacteria was Enterobacter aerogenes with highest inhibition zone (IZ = 31 mm; AI = 1.409 ± 0.046) even at minimum inhibitory concentration (0.039 mg/ml). CONCLUSION: Hence activity shown by crude ethyl acetate fraction might be due to pyrogallol present in the extract. On the basis of results it can be advocate that achieved crude ethyl acetate fraction can be explored for preparing antimicrobial drugs in future for the infectious caused by the pathogens tested in the study.

OBJECTIVE: This study aims to phytochemical and antimicrobial study of Euphorbia hirta Euphorbiaceae). MATERIALS AND METHODS: Antimicrobial activity of flavonoids (free and bound) of Euphorbia hirta L. was determined by disc diffusion assay against four bacteria (Escherichia coli, Pseudomonas aeruginosa, Proteus mirabilis, and Staphylococcus aureus) and four fungi (Aspergillus flavus, Aspergillus niger, Trichophyton mentagrophytes, and Candida albicans). Minimum inhibitory concentration (MIC) of the extract was evaluated through micro broth dilution method, while minimum bactericidal/fungicidal concentration was determined by subculturing the relevant samples. Total activity (TA) of extracts against each sensitive pathogen was also evaluated. RESULTS: Out of fungi; A. flavus, A. niger, and T. mentagrophytes were found to be resistant, against which none of the tested extracts showed activity. Bound flavonoids extract of root showed best activity against C. albicans (inhibition zone (IZ) 27.66, MIC 0.039, minimum fungicidal concentration (MFC) 0.039). TA of free flavonoid extract of root was found to be the same for P. mirabilis and S. aureus (192.30 ml/g). Two flavonoids quercetin and kaempferol were identified in the bound flavonoids of stem extract which showed activity against all the microorganisms. CONCLUSION: Results of the present investigation indicate that E. hirta has good antimicrobial activity with low range of MIC, hence can be exploited for future plant-based antimicrobial drugs.

Callus culture of Tridax procumbens has been established on Murashige and Skoog's medium supplemented with NAA and BAP from nodal segments. Free and bound flavonoids were extracted from 2, 4, 6 and 8 weeks old calli by a well-established method. These free flavonoids were then screened against Staphylococcus aureus (bacteria) and Candida albicans (yeast) for their antimicrobial potential. Minimum inhibitory concentration, minimum bactericidal/fungicidal concentrations and total activity were also evaluated. Apigenin, quercetin and kaempferol were identified from free flavonoids of 4 weeks old callus (most active) through, thin layer chromatography, (TLC) preparative TLC, MP and IR spectral studies.

The aim of present study was to investigate the antimicrobial activity of Cenchrus ciliaris and Cenchrus setigerus extracts in order to use it as a possible source for new antimicrobial substances against important human pathogens. Crude extracts of the stem of Cenchrus ciliaris and Cenchrus setigerus were evaluated against some medically important pathogens viz. Escherichia coli, Raoultella planticola, Staphylococcus aureus, Pseudomonas aeruginosa, Bacillus subtilis, Enterobacter aerogenes, Candida albicans and Aspergillus flavus. The dried and powdered stems were successively extracted with hexane, toluene, isopropyl alcohol, acetone and ethanol using soxhlet assembly. The antimicrobial activity assay was done by both disc diffusion and serial dilution methods. Isopropyl alcohol extract of Cenchrus setigerus showed highest activity against Escherichia coli. The test pathogens were more sensitive to the isopropyl alcohol, acetone and ethanol extracts than to the hexane and toluene extracts except against Bacillus subtilis. Result reveals that the most bioactive compound was cycloleucolenol-9,19-cycloergost-24 (28)-en-3-ol, 4, 14-dimethyl acetate in both the species of Cenchrus grass, (19.15%) in isopropanol extract of Cenchrus setigerus whereas, (14.03%) in acetone extract of Cenchrus ciliaris.

Antidermatophytic potential of three weed plants viz. Tridax procumbens L., Capparis decidua (forsk) Edgew and Lantana camara L. were explored and experimentally induced dermatophytic lesion was topically treated in mice. Microbroth dilution method was carried out for determination of MIC and MFC of different extracts of selected plants. In animal studies, mice were experimentally inoculated with Trichophyton mentagrophytes and infected animals were topically treated with 5 mg/g terbinafine and two concentrations, i.e., 5 and 10 mg/g of test extract ointment. Complete recovery from the infection was observed on 12th day of treatment for reference drug terbinafine (5 mg/g) and 10 mg/g concentration of test extract ointment whereas 5 mg/g concentration of test extract ointment showed complete cure on 16th day of treatment. Fungal burden was also calculated by culturing skin scrapings from infected animals of different groups. Test extract ointment successfully treated induced dermatophytosis in mice without any disease recurrence incidences, thereby indicating efficacy of test extract as an excellent topical antifungal agent for the cure of dermatophytosis.

Alkaloids are well known for their antimicrobial activity. Though all natural alkaloids come from plants, not all plants produce alkaloids. Plants of the Solanaceae family are known for their high alkaloid content. Alkaloids are found in all plant parts like roots, stems, leaves, flowers, fruits and seeds. In the present study, those plant parts of Solanum dulcamara were selected which have been reported to produce a high content of a specific alkaloid: solanine (from unripe fruits), solasodine (from flowers) and beta-solamarine (from roots). These alkaloids were extracted from various parts of S. dulcamara by well-established methods and were screened for their antibacterial activity. Human pathogenic bacteria, viz., Enterobacter aerogenes, Escherichia coli, Staphylococcus aureus, were selected for the study. All three alkaloids inhibited the growth of E. coli and S. aureus. However, no significant activity was observed against E. aerogenes. Minimum inhibitory concentration and minimum bactericidal concentration were also evaluated.

The present study was carried out to determine the possible bioactive components (steroids) of Cenchrus setigerus using GC-MS analysis and in vivo estimation of metabolites (total soluble sugar, soluble protein, proline and total phenolics), photosynthetic pigments (chlorophyll-a, chlorophyll-b and carotenoids) of seedlings and antimicrobial activity of extracts in various polar solvents from the leaves of C. setigerus. Antimicrobial activity was evaluated against three Gram-negative bacteria, including Proteusmirabilis, Klebsiella pneumonia and Agrobacterium tumefaciens andone fungus Aspergillus niger using 'disc diffusion' method, followed by the determination of minimum inhibitory concentrations (MIC) by broth dilution method. Results revealed the presence of some steroids in the isopropyl alcohol extract of C. setigerus:which are (22E)-stigmasta-4,22-dien-3-one(4.93%), δ4-sitosterol-3-one (stigmast-4-en-3-one) (4.31%), fagarsterol (lupeol) (1.25%) and ethyl iso-allocholate (0.32%). Total soluble sugars and chlorophyll-a were also recorded to be highest. The highest activity was exhibited by the isopropyl alcohol and ethyl acetate extract against P. mirabilis.
