RESUMEN

Rapid and selective separation, enrichment and detection of trace residue all-in-one in complex samples is a major challenge. Hydrogels with molecular sieve properties can selectively separate and enrich target analytes, and the combination with high sensitivity detection of surface-enhanced Raman scattering (SERS) is expected to achieve the above all-in-one detection. Herein, the core-shell structured Au@poly(N-isopropylacrylamide)-phenylboronic acid hydrogel (Au@PNIP-VBA) with boronic acid ester groups was prepared by thermally initiated polymerization. The boronic acid ester groups in hydrogel are selectively hydrolyzed by hydrogen peroxide (H2O2) to hydroxyl structures, leading to a reduction in SERS signals. The Au@PNIP-VBA hydrogel has molecular sieve properties and high SERS activity, making it suitable for separation, enrichment, hydrolysis and detection of H2O2 all-in-one. A rapid SERS method was developed for analysis of H2O2 based on the Au@PNIP-VBA hydrogel with the linear range of 8.5 × 10-2-6.8 mg L-1 and the detection limit of 33 µg L-1. The method was successfully applied to the determination of H2O2 residue in fresh milk, pure milk, yogurt and camel milk, with the recoveries were in the range of 82.2%-109.3% and the relative standard deviations were 2.8%-8.3%. This efficient all-in-one strategy has the advantages of simple sample pre-treatment, rapid analysis (30 min) and high sensitivity, making it highly promising for food quality and safety analysis.

Triazine herbicides have been widely detected in water resources and food, which poses a potential hazard to both ecosystem and human health. Due to their high polarity, conventional adsorbents have limitations for their extractions. Herein, for the effective magnetic extraction of triazine herbicides, a novel and effective magnetic adsorbent was prepared with a satisfactory extraction performance. In the experiments, five porous organic frameworks (POFs) with hydroxyl functional groups were synthesized by diazo-coupling reactions in aqueous solution with ß-cyclodextrin (ß-CD) as a green monomer. After evaluation of the five POFs, the DDM-CD-POF, which was synthesized with 4'4-diaminodiphenylmethane (DDM) and ß-CD, showed the largest specific surface area and the best adsorption capacity for the five triazine herbicides. Then, it was magnetized by introducing Fe3O4@SiO2 into it to prepare a magnetic adsorbent (M-DDM-CD-POF) to facilitate separation and recycling. Finally, the M-DDM-CD-POF-based magnetic solid-phase extraction in combination with high performance liquid chromatographic detection method was established for the quantitative determination of the triazine herbicides in environmental water and watermelon juice samples. The current strategy showed low limits of detection of 0.03-0.11 ng mL-1 for environmental water and 0.07-0.22 ng mL-1 for watermelon juice sample. The method recoveries for spiked samples ranged from 84.0% to 113.0% with the relative standard deviations ≤8.8%. This work provides a new approach for the detection of the triazine herbicides with good application prospect.

Three magnetic covalent organic frameworks (named M-TpPa-SO3Na, M-TpPa-SO3H and M-TpPa) were prepared by the solvothermal synthesis method with 1,3,5-trimethylphenol (TP) and either 2-sulfo-1,4-phenylenediamine (Pa-SO3H) or p-phenylenediamine (Pa) as monomers. Among them, the M-TpPa-SO3Na possessed relatively high hydrophilicity, good magnetic responsiveness, and high affinity for the benzoylureas (BUs) insecticides. It was then explored as the magnetic solid-phase extraction adsorbent for the extraction of six BUs (diflubenzuron, triflumuron, hexaflumuron, teflubenzuron, flufenoxuron and chlorfluazuron) from water, pear juice and honey samples prior to high-performance liquid chromatography with ultraviolet detection. Under the optimized experimental conditions, a good linearity was achieved within the concentration range of 0.27-40.0 ng mL-1 for water sample, 0.47-30.0 ng mL-1 for pear juice sample, and 2.70-200.0 ng g-1 for honey sample. The limits of detection for the analytes were 0.08-0.11 ng mL-1 for water sample, 0.14-0.19 ng mL-1 for pear juice sample and 0.80-1.00 ng g-1 for honey sample. The method recoveries for spiked samples were in the range of 85.0%-111.0% with the relative standard deviations less than 8.8%. The developed method was successfully used for the determination of the BUs in water, pear juice and honey samples.

As plant regulators, auxins can promote plant growth. However, they have toxicity and may cause harm to humans. Due to their low concentrations in food sample matrices, the enrichment and analysis of trace auxins in food samples is a challenging work. In this work, a series of hyper crosslinked polymers (HCPs) were synthesized by Friedel-Crafts acylation to extract four auxins (indole-3-acetic acid, indole-3-propionic acid, indole-3-butyric acid and 1-naphthylacetic acid). Among these HCPs, the QP-TC-HCP, synthesized from p-quaterphenyl (QP) and terephthaloyl chloride (TC), showed the best adsorption performance for the auxins. It was then applied as the adsorbent for the solid-phase extraction of the auxins from environmental water and peach beverage samples, followed by high performance liquid chromatography-fluorescence detection. Under the optimized conditions, the limits of detection were 3.0-12.0 pg mL-1 for environmental water and 18.0-36.0 pg mL-1 for peach beverage sample. The method recoveries of the auxins for the spiked samples were in the range of 85.0-110.0%. The established method provided an alternative approach for the determination of auxins in food samples. In addition, different types of organic compounds were tested for the extraction by the QP-TC-HCP to assess its application potential and adsorption mechanism. It was concluded that the QP-TC-HCP had better extraction performance for the compounds with certain hydrophilicity and more hydrogen bonding sites.

The three hypercrosslinked polymers (HCP) materials, designated as OPD-HCP, MPD-HCP and PPD-HCP, were synthesized by using o-phenylenediamine (OPD), m-phenylenediamine (MPD) and p-phenylenediamine (PPD) as monomers. They were characterized by infrared spectroscopy, powder X-ray diffraction, nitrogen sorption isotherms, and scanning electron microscopy. Then, the HCPs were explored as solid-phase extraction (SPE) adsorbent for the extraction of five nitroimidazoles (NDZs) (metronidazole, ronidazole, secnidazole, dimetridazole and ornidazole). Among the three HCPs, the MPD-HCP has the best adsorption performance for the NDZs. With the help of high-performance liquid chromatography with ultraviolet detection (HPLC-UV), good linear response range (0.07-40.0 ng mL-1), high method recovery (86.8%-113.3%), low limits of detection (0.02-0.15 ng mL-1) and good precision with the relative standard deviations of less than 8.1% were achieved for the determination of the NDZs in water samples. The effective determination of the NDZs in peach juice, honey tea, and honey samples were also realized by the developed method with satisfactory results. Based on both the experimental results and density functional theory calculation, the adsorption mechanism can be attributed to multiple interactions between the MPD-HCP and the NDZs, including hydrogen bonding, hydrophilic, and electrostatic interactions. The method provides a new alternative of choice for the determination of some NDZs in real samples.

RESUMEN

To promote the development of Traditional Chinese Medicine (TCM), it is necessary to innovate the traditional prescription. It is feasible to use one or several components to substitute TCM, which can be regarded as a process of discarding the dregs and preserving the essential components. In this way, traditional prescription can be converted into various combinations of pharmacological ingredients deriving from several TCMs. Furthermore, some of pharmacological ingredients should be modified to increase their efficacy. It is practical to select the main structural unit with specific substituents having strong pharmacological activity. After the innovation mentioned above, the prescription will evolve into a variety of modified components having distinct pharmacological activity, and this is the novel integration of active ingredients.

OBJECTIVE: To collect visualized proof of Tianmagouteng particles (TMGTP) in alleviating cognitive dysfunction and to explore its effects on brain activity in spontaneously hypertensive rats (SHRs) with hyperactivity of liver-yang (Gan Yang Shang Kang, GYSK). METHODS: Sixteen SHRs were randomized into treatment group and non-treatment. The SHR with GYSK was induced by gavaging aconite decoction (10mL/kg at 0.2g/mL). After the SHR models were prepared, the rats in the treatment group were administered TMGTP (10mL/kg) once a day for 14days.The rats in the non-treatment group or normal rats (control group) received an equivalent volume of saline. Morris water maze test was conducted before and after the treatment to observe cognitive function. Fluorine 18-deoxy glucose [F-18]FDG micro-PET brain imaging scans was performed after treatment. Data were analyzed with two-sample t-test (P<0. 001) using SPM2 image analysis software. RESULTS: Compared with the non-treatment group, the escape latency significantly decreased but the frequency of entrance into the target zone significantly increased in the treatment group. Consistent with the alteration of cognitive functions, TMGTP induced strong brain activity in the following sites: right dorsolateral nucleus and ventrolateral nucleus of thalamus, amygdala, left met thalamus, cerebellum leaflets, original crack, front cone crack, loop-shaped leaflets; but deactivation of right medial frontal gyrus, bilateral corpus callosum, hippocampus, and left dentate gyrus. CONCLUSION: TMGTP could alleviate cognitive dysfunction in SHRs with GYSK, which was possibly by inducing alteration of glucose metabolism in different brain regions with corresponding functions.

ETHNOPHARMACOLOGICAL RELEVANCE: Tong Luo Jiu Nao injection (TLJN), a Chinese medicine preparation, was extracted from the Chinese herbs Panax notoginseng and Gardenia jasminoides. Its pharmacological effect on cerebral ischemia was observed in the study. We previously reported that paracrine signaling of brain microvascular endothelial cells (BMECs) had a direct impact on the survival of neurons in mimicked cerebral ischemia in vitro. The current study was designed to investigate whether paracrine signaling of BMECs could be regulated by drug to achieve neuroprotection. MATERIALS AND METHODS: First, an in vitro model of cerebral ischemia in BMECs or neurons was established by oxygen-glucose-deprivation (OGD). TLJN was used as a medicine of intervention. Injured neurons were cultured in the conditioned media from normal and injured BMECs treated with TLJN. The changes in neurons, including the expression of N-methyl-D-aspartate receptor 1(NMDAR1), Ca(2+) concentration, cytochrome c release, the mitochondrial membrane potential, were determined by the immunofluorescence staining and molecular Probes. The content of Vascular endothelial growth factor (VEGF) and platelet activating factor (PAF) in various BMECs were also examined by Western blotting and Elisa. RESULTS: The results showed the activity of injured neurons was significantly increased when they were grown in conditioned media of normal or injured BMECs treated with TLJN, compared with that of normal or injured BMECs. These changes include a decrease of Ca(2+) concentration, of NMDAR1 expression, and of cytochrome c release, also an increase of the mitochondrial membrane potential. Moreover, the VEGF expression was up-regulated and the PAF expression was down-regulated by TLJN in BMECs. CONCLUSION: The results suggest that a neuroprotective effect of TLJN could be achieved by regulating paracrine signaling of BMECs, which could in part be explained by a TLJN-induced up-regulation of VEGF and a down-regulation of PAF in BMECs. Therefore, regulating the paracrine of BMECs could be the important target of the drug action on injured-neurons, which may be a novel path for therapeutic intervention in ischemic injury.
