BACKGROUND: Insomnia is a prevalent sleep disorder that affects up to 15% of the population worldwide and is the second most common mental health issue. There is increasing interest in the effects of long-term insomnia on cognitive function. Electroacupuncture can effectively improve cognitive function and sleep quality, yet the underlying brain network mechanisms remain unclear. This study aims to explore the network regulatory mechanisms associated with enhanced cognitive function and sleep quality, providing theoretical support for the use of electroacupuncture in the clinical treatment of chronic insomnia. METHODS: This study is divided into two parts. Sixteen individuals with chronic insomnia and 16 healthy controls of similar age and gender will be recruited in Study 1 to examine the brain network topology of individuals with chronic insomnia. Study 2 will be a randomized controlled trial with 120 chronic insomnia patients divided into three groups: Group A (electroacupuncture plus placebo drug), Group B (drug plus placebo electroacupuncture), and Group C (placebo electroacupuncture plus placebo drug). Participants will be exposed to 24 treatments over an 8-week period (3 times per week) and monitored for 12 additional weeks. The primary outcome measure will be changes in brainwave data from before to after the treatment. In addition, the Wisconsin Card Sorting Test and the Pittsburgh Sleep Quality Index will be utilized as secondary outcomes to measure from before to after treatment and during the follow-up. A correlation analysis will be conducted to explore links among modifications in brainwave patterns, Wisconsin Card Sorting Test scores, and Pittsburgh Sleep Quality Index scores. Additionally, any adverse events will be strictly monitored. DISCUSSION: Electroacupuncture may represent an alternative treatment for chronic insomnia, and this trial is expected to reveal the brain mechanism by which electroacupuncture improves cognitive function and sleep quality in chronic insomnia patients. Intestinal microbiota is necessary for the guarantee of intestinal mucosal barrier. However, the detailed effect of probiotics on porcine intestinal development, especially in the early life, is still unclear. In this study, we treated 3-day-old newborn piglets with Lactobacillus reuteri (L. reuteri) D8 and observed its beneficial effect on piglets. The body weights, villus height, and crypt depth of jejunum were all significantly increased after L. reuteri treatment in piglets. L. reuteri also significantly increased the proliferation index of PCNA+ cells in the crypt, as well as c-Myc and Tcf4 expressions. Furthermore, L. reuteri also enhanced intestinal mucosal barrier with the increase of goblet cells and antimicrobial peptides (AMPs) expressions of Muc2, Lyz1, and pBD1. The well development of Peyer's patches and increased number of CD3+ T cells, combined with increased expression of IL-4 and IFN-γ, also demonstrated the immune stimulation effect of L. reuteri D8. This study demonstrated that L. reuteri promotes the development of intestine mucosal system and maintains intestinal mucosal barrier in newborn piglets.

Probiotics, such as Lactobacillus, have been proven to be effective in maintaining intestinal homeostasis. The modulatory effect of Lactobacillus on intestinal epithelial development in early life is still unclear. In this study, Lactobacillus isolates with good probiotic abilities were screened and orally administered to detect their regulatory effect on intestinal development in chickens. L. reuteri 22 was isolated from chickens and chosen for subsequent chicken experiments due to its strong acid and bile salt resistance and ability to adhere to epithelial cells. The 3-day-old chickens were orally administrated with 108 CFU L. reuteri 22 for consecutive 7 days. L. reuteri 22 increased Lgr5 mRNA expression (3.23 ± 0.40, P = 0.001) and activated the Wnt/ß-catenin signaling pathway, with increasing expression of proliferating cell nuclear antigen (PCNA) (49.27 ± 9.81, P = 0.021) to support the proliferation of chicken intestinal epithelial cells. Moreover, L. reuteri 22 also inhibited the Notch signaling pathway to induce intestinal stem cell differentiation into goblet cells with increased mucin 2 (Muc-2) expression (1.72 ± 0.34, P = 0.047). L. reuteri 22 significantly enhanced lysozyme mRNA expression (2.32 ± 0.55, P = 0.019) to improve intestinal innate mucosal immunity. This study demonstrated that L. reuteri administration could regulate chicken intestinal epithelium development to ensure the function of the intestinal mucosal barrier, which is beneficial for newborn animals.

Fluorescence quenching was used to study the potential interaction mechanism of Bovine serum albumin (BSA) with either hydrophilic ferroferric oxide (Fe3O4) nanoparticles (NPs) or hydrophobic Fe3O4 NPs. The experimental results indicated the mechanism between BSA and hydrophilic Fe3O4 NPs was static quenching and the one between BSA and hydrophobic Fe3O4 NPs was dynamic process that was drove by Förster's resonance energy transfer (FRET). And the binding parameters for the interaction of BSA with either hydrophilic or hydrophobic Fe3O4 NPs were calculated by using the fluorescence quenching measurement. The binding constant (KA) values of hydrophilic Fe3O4 NPs were 8518.73±23.35 (at 298K), 1190.31±15.41 (at 306K) and 321.97±8.57 (at 313K), respectively. The thermodynamic analysis implied that the intermolecular forces between BSA and hydrophilic Fe3O4 NPs were Van der Waals interaction or hydrogen bond, because the values of ΔH and ΔS between them were negative. While the one of BSA and hydrophobic Fe3O4 NPs involved hydrophobic forces, owing to the positive ΔH and ΔS between them. But they were all enthalpy-driven and exothermic, since their ΔG values were all negative. Synchronous fluorescence spectroscopy suggested that the conformation of tryptophan residue of BSA was changed in the presence of hydrophilic Fe3O4 NPs or hydrophobic Fe3O4 NPs, because the position of the maximum emission wavelength had a discernible red shift.
