RESUMEN

Hen diuresis syndrome has emerged over the past 5 yr as a significant cause of mortality in the U.S. broiler breeder industry. The condition affects hens in production and is characterized by transient muscle weakness in the vent region, transient diuresis, and often urate deposits on the skin below the vent. Affected hens are often seen straining to lay an egg, which suggests oviduct contraction is also impaired. Related hen mortality, often reaching 1% or more a week, is believed to be primarily the result of male aggression of the vent region (Turner et al., "Investigating Causes of Excessive Urate Production in Broiler Breeder Hens Associated with Peritonitis and Cannibalism Mortality," Oral Presentation at The American Association of Avian Pathologists Annual Meeting, p. 139, 2010). The exact association between the cause of mortality and this syndrome is unknown, but it may be the consequence of transient partial to full oviduct prolapse, which predisposes or stimulates cannibalism and aggression. Based on unpublished work done prior to this study (Turner et al., ibid.), the evidence suggests the underlying problem is metabolic. We feel that urine collection and analysis is an essential component to understanding this condition. This study serves as a pilot study for future investigations that attempt to identify the nature and cause of the metabolic disturbance through paired urine and serum collection and analysis. For the purpose of this study, a small sample of 10 affected and 10 unaffected birds was used for sample collection. In order to collect pure urine, the birds were surgically colostomized. Colostomy did prove to be a useful means of collecting urine free of feces, and for the purposes of our study it yielded adequate urine samples for analysis. There were statistically relevant urine values observed. Affected birds had a higher presence of blood in the urine, a lower uric acid excretion rate (mg/hr), higher concentration (mEq/L) of urine Na+, and a lower concentration (mEq/L) of urine K+ than unaffected birds. This pilot study helps to address some of the pitfalls previously associated with colostomy and to determine when collection can begin postoperatively so that we can better understand when and how to begin our sampling in future trials to address the etiology of this condition.

To obtain information about Salmonella from commercial birds and poultry environments within Mississippi, 50 Salmonella enterica isolates were collected and characterized by intergenic sequence ribotyping (ISR) serotyping and by determining antimicrobial resistance. ISR assigned serotype to all 50 Salmonella enterica isolates whereas the Kauffman-White-LeMinor antibody-based scheme assigned serotype to 48. Agreement between both methods was K = 89.58. Within the set, 12 serotypes were detected. The antimicrobial resistance patterns (ARP) of 12 serotypes, namely Enteritidis, Typhimurium, Kentucky, Bredeney, Mbandaka, Saintpaul, Montevideo, Cubana, Lille, Senftenberg, Johannesburg, and one serotype UN0094, were determined using minimum inhibitory concentration values. The antibiograms demonstrated differences between Salmonella serotypes and among isolates of the same serotype. All isolates were 100% susceptible to enrofloxacin and trimethoprim/sulfamethoxazole. The number of antimicrobials to which the isolates were resistant ranged from two to nine. Twenty-two different ARPs were identified and ARP1, with resistance to spectinomycin and sulfadimethoxine, was most frequently observed. Forty isolates (80%) were resistant to three or more antimicrobials and were thus designated multidrug resistant. Detection of a unique serotype, and variation in antibiograms within the set, demonstrates that it is important to survey isolates periodically from a region to follow epidemiologic trends.

This pilot analysis was conducted with data from 52 conventional grow-out broiler flocks in a prospective field observational study in the southeastern United States during 2003-2006. Each flock was sampled for Salmonella 1 wk before the end of grow-out, upon arrival at the processing plant, and during processing (prior to and immediately after carcass chilling). The broiler litter was sampled on the day of bird harvest. The grow-out feeding programs, including the medications delivered in feed, were surveyed with questionnaires completed by the broiler managers and feedmill managers. Each detail of the feeding program was tested for statistical association with the frequency of Salmonella in the flock at each sampling point, after accounting for variation in Salmonella frequency between the farms, broiler complexes, and companies. Significant associations were found between Salmonella frequency in the broiler flock pre- and postharvest and the inclusion of feeds containing individual coccidiostats and other antimicrobial growth promoters, days on feed, and total consumption of feeds containing these products, as well as with practices such as a mash feed and a nonmedicated withdrawal feed. The analysis provided testable hypotheses for how broiler feed medications impact the frequency of Salmonella in the flocks.

We analyzed factors involved in the introduction of infectious laryngotracheitis (ILT) virus (ILTV) onto broiler farms during a localized outbreak in an immunologically naive broiler population. The outbreak occurred in the state of Mississippi, United States in 2002-2003. From the responses to a retrospective survey questionnaire administered via personal interviews, 181 farm-level risk factors were defined and analyzed for their association with ILTV introduction using logistic regression. There were 27 case farms (93% of all the infected broiler farms) and two sets of controls: farms matched to the cases by location and those randomly selected among the broiler farms in Mississippi. We found that farm suppliers such as gas company representatives, who are likely to visit other farms, and farm-workers who visit other chicken farms, are likely vehicles of ILTV introduction onto broiler farms. These risks can be greatly reduced by following biosecurity procedures, in particular if farm workers bathe and change footwear prior to entering broiler houses on their own farm. Footbaths for farm visitors can provide a false sense of security during an ILT outbreak when, indeed, other practices such as plastic boots or changing boots are more effective in preventing ILTV transmission. Sharing of equipment used for removal of caked broiler litter between subsequent flocks may also serve as an important vehicle of ILTV transmission. During the 2002-2003 outbreak, shared litter removal equipment was associated with ILTV transmission despite a requirement being put in place for litter decontamination. We also found that tunnel-ventilated broiler houses with inlets toward a neighboring poultry farm are more likely to get infected with ILTV. In addition to this analysis, the data collected provide a good overview of the actual practices and deficiencies of biosecurity undertaken on broiler farms in this part of the United States.

During the routine histologic evaluation of an outbreak of inclusion body hepatitis (IBH) in Mississippi broilers, a high incidence of renal enlargement and glomerulonephropathy was observed in the birds presenting classic hepatic pathology. Characteristic intranuclear adenoviral inclusion bodies were demonstrated in the livers of these birds, and fowl adenovirus was identified by viral isolation and by PCR. The glomerular lesions were consistent with proliferative or membranoproliferative forms of glomerulonephritis. Histomorphometric evaluations were performed to generate a more quantitative analysis of altered glomerular size and cellularity, to detect statistically significant borderline changes, and to get a clearer insight into the incidence of the glomerular alterations. Marked increases in both the average glomerular size (area) and the total glomerular cellularity were observed for the affected glomeruli relative to normal controls. The average glomerular area values for normal glomeruli in the peripheral subcapsular cortical and central cortical kidney regions were 1791 microm2 and 5302 microm2, respectively. In contrast, glomerular measurements for kidneys exhibiting glomerulonephritis by routine histopathology, had average values for the two regions of 4429 microm2 and 11,063 microm2. The average glomerular cell counts for the two regions in controls were 44 and 107 cells/ glomeruli, while averages for birds with glomerulonephritis were 85 and 193 cells/glomeruli. The proportion of IBH-associated glomeruli greater than two standard deviations above the mean glomerular size of the normal controls was 52% for the central region and 62% for the peripheral region.

In 2007, an inclusion body hepatitis (IBH) outbreak affected several broiler farms in Mississippi. Results of logistic regression analyses showed significant associations between IBH occurrence and high enzyme-linked immunosorbent assay geometric mean titers for infectious bursal disease virus. However, there was no association between IBH occurrence and chicken infectious anemia virus status. Results of linear regression model analyses showed significant associations between IBH occurrence with average weight and with cost deviation. Broiler meat production cost was $0.0058/kg more expensive to produce when IBH occurred. Although feed conversion was higher with IBH occurrence, the association was not significant. IBH onset in the first farms affected occurred between 19 and 30 days of age, whereas in the last farms affected, IBH onset occurred as early as 10 days.

Routine and quantitative histologic studies on femoral head separation (FHS) associated with coxofemoral joint disarticulation at necropsy were conducted on 125 femoral heads collected from 21- to 50-day-old clinically normal broilers. The study compared groups demonstrating grossly detached femoral heads (DFHs) with those having attached femoral heads (AFHs). Marked microscopic lesions compatible with osteochondrosis (OCD) consistently occurred along the separation surface in the DFH population. The histologic changes consisted of cartilage degeneration and necrosis sometimes forming small clefts or microfractures. Hemorrhage and less frequent inflammatory cells were often present along the separation surfaces. Small foci of OCD in the femur occurred in the AFH group with lesser frequency and severity. The histologic changes were mainly found within the proximal proliferative zone of the physis near the epiphyseal junction. Histomorphometry disclosed significant quantitative reductions in chondrocyte density with increased pyknosis occurring adjacent to the separation site and to a lesser extent in deeper regions of the growth plate for the DFH compared with AFH. Measurements made along the separation surface of the percentage length occupied by osteochondrotic defects and actual separated cartilage disclosed significant differences between evaluation groups. However, determinations of vascular canal areas present within two or more regions of the growth plate revealed a slight and significant increased area for DFH compared with AFH. Severity scores for the occurrence of microthrombi within the growth plate showed no difference between the groups. The pathogenesis of FHS in broilers is related to defective cartilage production or degeneration resulting in increased fragility. This contrasts with the proposed pathogenesis of OCD in mammals, which involves ischemic necrosis due to underlying vascular defects. The results for the FHS-disarticulation model also differ from those reported for glucorticoid-induced femoral head necrosis in broilers. The FHS-associated lesions occurred without histologic evidence of bacterial chondritis or osteomyelitis.

Testicular histomorphometric methods were developed for the investigation of fertility and hatchability problems in commercial broiler-breeder roosters. Sertoli cell (SC) evaluations were emphasized in view of their role in influencing embryonic testicular organogenesis and spermatogenesis in adults. H&E stained sections prepared from formalized tissues were examined. SC evaluations were mostly made from computer generated images using image analysis software, but were also compared to direct counting with ocular grid reticle. Other measurements were germinal epithelial and total seminiferous tubule widths, quantification of individual spermatozoa and sperm clusters, and determinations of stromal area. Age-associated changes in SC occurred between 15 to 54 wk of age. A significant reduction in SC concentration occurred at 29 wk of age (P < 0.001) that corresponded with the histologic initiation of spermatogenesis. Expanded morphometric evaluations were performed on 59-week-old roosters comparing a flock having good body condition and footpad scores with normal testicular and body weights (optimal profile or OP group) to a second flock demonstrating relatively poor expression of these parameters and decreased fertility (sub-optimal profile or SOP group). Significant reductions (P < 0.05) in SC concentration, spermatozoa, and tubular widths, but an increased percentage of stroma area occurred in SOP compared to OP flocks. SC results also correlated well with testes weight. A unique finding was a significant relationship between decreasing SC concentration and increasing severity scores for footpad lesions. Differences in all histomorphometric parameters occurred between flocks demonstrating good hatchability compared to flocks exhibiting either low or decreasing hatchability. Significant reductions in SC and other morphometric parameters were observed for the poor relative to good hatchability group. A strong positive association was observed for SC results using computer generated images and direct reticle ocular counting (R = 0.80). Replicate counts on control sections demonstrated generally good reproducibility of methods between experiments. The findings are supportive of the value of testicular histomorphometrics for evaluations of hatchability issues in commercial roosters.

Previous studies documented the common occurrence of transitory cyanosis and echocardiographic aortic insufficiency in mature commercial broiler breeder roosters. During further investigations, we observed a high prevalence of hearts exhibiting extensive dilation of the left ventricle chamber compatible with dilated left ventricular cardiomyopathy present in both cyanotic and normal subpopulations. We conducted quantitative studies focused on documentation of cardiac ventricle parameters by using simple gross morphometric methods performed on formalin-fixed hearts obtained from both clinically normal roosters and those exhibiting variable transitory cyanosis, echocardiographic aortic insufficiency, or both. A high prevalence of often dramatic left ventricular dilation reflected in enlarged left ventricular chamber areas and elevated left ventricle-to-total ventricle area ratios was morphometrically documented. However, no statistically significant differences in the occurrence of ventricular abnormalities were observed between normal and cyanotic roosters. Age-associated changes were also demonstrated by comparative morphometric studies on hearts from normal market-age broilers (average age of 7 wk) and those of mature roosters (average age of 42 wk). Elevation in both left and right ventricular weight-to-total heart weight ratios dramatically increased with aging. In addition, values (average ± SD) for the left ventricle chamber area-to-total ventricle area ratios increased from 3.2 ± 2.0% in broilers up to 10.0 ± 8.8% in roosters. None of the normal broilers studied demonstrated left ventricular volume ratios above 10%, whereas 33% of the roosters had left ventricular volume ratios above 10%, including 13% with ratios of 20% or higher. However, the left ventricle wall area-to-body weight ratios were much closer for the two age groups (0.85 ± 0.18 cm(2)/kg in broilers and 0.79 ± 0.13 cm(2)/kg in roosters). Also, the standard right ventricle-to-total ventricle weight ratio (RV/TV) went from 0.18 ± 0.04 in broilers to 0.25 ± 0.12 in roosters, whereas the left ventricle-to-total ventricle weight ratios were similar for the two age groups (0.74 ± 0.12 and 0.75 ± 0.08 in broilers and roosters, respectively). Our results for RV/TV in normal broilers were similar to the reported values for normal market-age broilers. In contrast, 36% of the roosters had RV/TV above values reported for broilers considered reflective of right ventricular hypertrophy due to pulmonary hypertension, whereas 4% had values above the reported threshold for broilers dying with ascites (ratios greater than 0.0249 and 0.299, respectively). However, ascites was not observed for any of the roosters. Although essentially all cardiac morphometric parameters demonstrated statistically significant correlations with the age-class group comparisons, significance could not be documented for comparisons between cardiomorphometrics and the subjective occurrences of transitory cyanosis or echocardiographic aortic insufficiency.

BACKGROUND: Lighting is used during conventional broiler grow-out to modify bird behaviour to reach the goals of production and improve bird welfare. The protocols for lighting intensity vary. In a field study, we evaluated if the lighting practices impact the burden of Salmonella in broiler flocks. METHODS: Conventional grow-out flocks reared in the states of Alabama, Mississippi and Texas, USA in 2003 to 2006 were sampled 1 week before harvest (n = 58) and upon arrival for processing (n = 56) by collecting feathered carcass rinsate, crop and one cecum from each of 30 birds, and during processing by collecting rinsate of 30 carcasses at pre-chilling (n = 56) and post-chilling points (n = 54). Litter samples and drag swabs of litter were collected from the grow-out houses after bird harvest (n = 56). Lighting practices for these flocks were obtained with a questionnaire completed by the growers. Associations between the lighting practices and the burden of Salmonella in the flocks were tested while accounting for variation between the grow-out farms, their production complexes and companies. RESULTS: Longer relative duration of reduced lights during the grow-out period was associated with reduced detection of Salmonella on the exterior of birds 1 week before harvest and on the broiler carcasses at the post-chilling point of processing. In addition, starting reduced lights for > or = 18 hours per day later in the grow-out period was associated with decreased detection of Salmonella on the exterior of broilers arriving for processing and in the post-harvest drag swabs of litter from the grow-out house. CONCLUSIONS: The results of this field study show that lighting practices implemented during broiler rearing can impact the burden of Salmonella in the flock. The underlying mechanisms are likely to be interactive.
