During virus infection, Argonaute (AGO) proteins bind to Dicer-produced virus small interfering RNAs and target viral RNA based on sequence complementarity, thereby limiting virus proliferation. The Arabidopsis AGO2 protein is important for resistance to multiple viruses, including potato virus X (PVX). In addition, AGO5 is important in systemic defense against PVX. Normally AGO5 is expressed only in reproductive tissues, and its induction by virus infection is thought to be important for its participation in antiviral defense. However, it is unclear what mechanisms induce AGO5 expression in response to virus infection. Here, we show that dde2-2, a mutant compromised in jasmonic acid (JA) biosynthesis, displays constitutive upregulation of AGO5. This mutant also showed increased resistance to PVX and this resistance was dependent on a functional AGO5 gene. Furthermore, methyl jasmonate treatment ablated AGO5 expression in leaves during virus infection and resulted in increased susceptibility to virus. Our results further support a role for AGO5 in antiviral RNA silencing and a negative regulation by JA, a plant hormone associated with defense against plant-feeding arthropods, which are often the vectors of plant viruses. This paper describes OpenSpyrit, an open access and open source ecosystem for reproducible research in hyperspectral single-pixel imaging, composed of SPAS (a Python single-pixel acquisition software), SPYRIT (a Python single-pixel reconstruction toolkit) and SPIHIM (a single-pixel hyperspectral image collection). The proposed OpenSpyrit ecosystem responds to the need for reproducibility and benchmarking in single-pixel imaging by providing open data and open software. The SPIHIM collection, which is the first open-access FAIR dataset for hyperspectral single-pixel imaging, currently includes 140 raw measurements acquired using SPAS and the corresponding hypercubes reconstructed using SPYRIT. The hypercubes are reconstructed by both inverse Hadamard transformation of the raw data and using the denoised completion network (DC-Net), a data-driven reconstruction algorithm. The hypercubes obtained by inverse Hadamard transformation have a native size of 64 × 64 × 2048 for a spectral resolution of 2.3 nm and a spatial resolution that is comprised between 182.4 µm and 15.2 µm depending on the digital zoom. The hypercubes obtained using the DC-Net are reconstructed at an increased resolution of 128 × 128 × 2048. The OpenSpyrit ecosystem should constitute a reference to support benchmarking for future developments in single-pixel imaging.

C-H bond functionalization generates molecular complexity in single-step transformation. However, the activation of C-H bonds requires expensive metals or stoichiometric amounts of oxidizing/reducing species. In many cases, they often require pre-functionalization of starting molecules. Such pre-activating measures cause waste generation and their separation from the final product is also troublesome. In such a scenario, reactions activating elements generating from renewable energy resources such as electricity and light would be more efficient, green, and cost-effective. Further, incorporation of growing flow technology in chemical transformation processes will accelerate the safer accesses of valuable products. Arenes & heteroarenes are ubiquitous in pharmaceuticals, natural products, medicinal compounds, and other biologically important molecules. Herein, we discussed enabling tools and technologies used for the recent C-H bonds functionalization of arenes and heteroarenes.

Chronic diseases such as cystic fibrosis, inflammatory bowel diseases, rheumatoid arthritis, and cardiovascular illness have been linked to a decrease in selenium levels and an increase in oxidative stress. Selenium is an essential trace element that exhibits antioxidant properties, with selenocysteine enzymes like glutathione peroxidase being particularly effective at reducing peroxides. In this study, a series of synthetic organoselenium compounds were synthesized and evaluated for their potential antioxidant activities. The new selenohydantoin molecules were inspired by selenoneine and synthesized using straightforward methods. Their antioxidant potential was evaluated and proven using classical radical scavenging and metal-reducing methods. The selenohydantoin derivatives exhibited glutathione peroxidase-like activity, reducing hydroperoxides. Theoretical calculations using Density Functional Theory (DFT) revealed the selenone isomer to be the only one occurring in solution, with selenolate as a possible tautomeric form in the presence of a basic species. Cytocompatibility assays indicated that the selenohydantoin derivatives were non-toxic to primary human aortic smooth muscle cells, paving the way for further biological evaluations of their antioxidant activity. The results suggest that selenohydantoin derivatives with trifluoro-methyl (-CF3) and chlorine (-Cl) substituents have significant activities and could be potential candidates for further biological trials. These compounds may contribute to the development of effective therapies for chronic diseases such cardiovascular diseases.

Land use and management changes affect the composition and diversity of soil bacteria and fungi, which in turn may alter soil health and the provision of key ecological functions, such as pesticide degradation and soil detoxification. However, the extent to which these changes affect such services is still poorly understood in tropical agroecosystems. Our main goal was to evaluate how land-use (tilled versus no-tilled soil), soil management (N-fertilization), and microbial diversity depletion [tenfold (D1 = 10-1) and thousandfold (D3 = 10-3) dilutions] impacted soil enzyme activities (ß-glycosidase and acid phosphatase) involved in nutrient cycles and glyphosate mineralization. Soils were collected from a long-term experimental area (35 years) and compared to its native forest soil (NF). Glyphosate was selected due to its intensive use in agriculture worldwide and in the study area, as well as its recalcitrance in the environment by forming inner sphere complexes. Bacterial communities played a more important role than the fungi in glyphosate degradation. For this function, the role of microbial diversity was more critical than land use and soil management. Our study also revealed that conservation tillage systems, such as no-tillage, regardless of nitrogen fertilizer use, mitigates the negative effects of microbial diversity depletion, being more efficient and resilient regarding glyphosate degradation than conventional tillage systems. No-tilled soils also presented much higher ß-glycosidase and acid phosphatase activities as well as higher bacterial diversity indexes than those under conventional tillage. Consequently, conservation tillage is a key component for sustaining soil health and its functionality, providing critical ecosystem functions, such as soil detoxification in tropical agroecosystems.

The objective was to isolate lactic acid bacteria (LAB) from southern Brazil's wines and investigate their potential as starter cultures for malolactic fermentation (MLF) in Merlot (ME) and Cabernet Sauvignon (CS) wines through the fermentative capacity. The LAB were isolated from CS, ME, and Pinot Noir (PN) wines in the 2016 and 2017 harvests and evaluated for morphological (color and shape of the colonies), genetic, fermentative (increase in pH, acidity reduction, preservation of anthocyanins, decarboxylation of L-malic acid, yield of L-lactic acid, and content of reduced sugars), and sensory characteristics. Four strains were identified as Oenococcus oeni [CS(16)3B1, ME(16)1A1, ME(17)26, and PN(17)65], one as Lactiplantibacillus plantarum [PN(17)75], and one as Paucilactobacillus suebicus [CS(17)5]. Isolates were evaluated in the MLF and compared to a commercial strain (O. oeni), as well as a control (without inoculation and spontaneous MLF), and standard (without MLF). CS(16)3B1 and ME(17)26 isolates finished the MLF for CS and ME wines, respectively, after 35 days, similar to the commercial strain, and CS(17)5 and ME(16)1A1 isolates ended the MLF in 45 days. In the sensory analysis, ME wines with isolated strains received better scores for flavor and overall quality than the control. Compared to the commercial strain, CS(16)3B1 isolate obtained the highest scores for buttery flavor and taste persistence. CS(17)5 isolate received the higher scores for a fruity flavor and overall quality and the lowest for a buttery flavor. The native LAB displayed MLF potential, regardless of the year and grape species from which they were isolated.

Measuring the duration of cognitive processing with reaction time is fundamental to several subfields of psychology. Many methods exist for estimating movement initiation when measuring reaction time, but there is an incomplete understanding of their relative performance. The purpose of the present study was to identify and compare the tradeoffs of 19 estimates of movement initiation across two experiments. We focused our investigation on estimating movement initiation on each trial with filtered kinematic and kinetic data. Nine of the estimates involved absolute thresholds (e.g., acceleration 1000 back to 200 mm/s2, micro push-button switch), and the remaining ten estimates used relative thresholds (e.g., force extrapolation, 5% of maximum velocity). The criteria were the duration of reaction time, immunity to the movement amplitude, responsiveness to visual feedback during movement execution, reliability, and the number of manually corrected trials (efficacy). The three best overall estimates, in descending order, were yank extrapolation, force extrapolation, and acceleration 1000 to 200 mm/s2. The sensitive micro push-button switch, which was the simplest estimate, had a decent overall score, but it was a late estimate of movement initiation. The relative thresholds based on kinematics had the six worst overall scores. An issue with the relative kinematic thresholds was that they were biased by the movement amplitude. In summary, we recommend measuring reaction time on each trial with one of the three best overall estimates of movement initiation. Future research should continue to refine existing estimates while also exploring new ones.

The treatment planning for patients with hepatocellular carcinoma (HCC) relies predominantly on tumor burden, clinical performance, and liver function test results. Curative treatments such as resection, liver transplantation, and ablative therapies of small lesions should be considered for all patients with HCC. However, many patients are ineligible for these treatments owing to advanced disease stage and comorbidities. Despite efforts to increase screening, early-stage HCC remains difficult to diagnose, which decreases the possibility of curative therapies. In this context, local-regional treatment of HCC is accepted as a form of curative therapy in selected patients with early-stage disease, as a therapeutic option in patients who are not eligible to undergo curative therapies, as a downstaging approach to decrease tumor size toward meeting the criteria for liver transplantation, and as a bridging therapy to avoid tumor growth while the patient is on the waiting list for liver transplantation. The authors review the indications, types, mechanism of action, and possible complications of local-regional treatment, as well as the expected postprocedural imaging features of HCC. Furthermore, they discuss the role of imaging in pre- and postprocedural settings, provide guidance on how to assess treatment response, and review the current limitations of imaging assessment. Finally, the authors summarize the potential future directions with imaging tools that may add value to contemporary practice at response assessment and imaging biomarkers for patient selection, treatment response, and prognosis. ©RSNA, 2022.

BACKGROUND: Canine atopic dermatitis (cAD) is a chronic disease characterised by hypersensitivity to environmental allergens. Oclacitinib maleate selectively inhibits pro-inflammatory mediators associated with cAD. However, the impact of chronic oclacitinib use on immunocompetence requires further investigation. OBJECTIVES: Herein, we examined the potential immunomodulatory effects of prolonged oclacitinib treatment in dogs. ANIMALS: Thirteen privately owned dogs with cAD, treated with 0.4-0.6 mg/kg oclacitinib for 12 months. METHODS AND MATERIALS: Pruritus level was evaluated using a pruritus Visual Analog Scale (pVAS) and the canine atopic dermatitis extent and severity index, 4th iteration (CADESI IV). Peripheral blood samples were collected for routine laboratory assays and lymphocyte subtypes were analysed using flow cytometry. Antigen-specific intracellular cytokine production from CD4+ and CD8+ T lymphocytes was analysed following in vitro stimulation by Dermatophagoides farinae antigens. RESULTS: Oclacitinib treatment significantly reduced pVAS and CADESI-04 scores, by 51% and 86.7%, respectively. Flow cytometric analysis revealed increased CD4+ and CD14+ lymphocyte populations. The cytokine profile at 360 days after treatment initiation was similar to that before treatment and was not associated with clinical relapse. CONCLUSION: Oclacitinib, when administered at the currently labelled dose for one year, is associated with a significant increase in circulating CD4+ T cells, but does not alter cytokine production from antigen-stimulated T cells. The results reported do not support evidence for immunosuppression mediated by the mechanisms evaluated in this study.

OBJECTIVE: To evaluate the local tumour progression-free survival (LTPFS), metastasis-free survival (MFS), cancer-specific survival (CSS) and overall survival (OS) of healthy surgical candidates who underwent percutaneous thermoablation (TA) as a first-line therapy for small renal masses (T1a). METHODS: The institutional review board approved this bi-institutional retrospective study of 85 consecutive surgical candidates with 97 biopsy-proven malignant renal masses (T1a) treated with percutaneous TA from 2008 to 2016. The LTPFS, MFS, CSS and OS rates were calculated using the Kaplan-Meier method. Descriptive analysis was also performed. RESULTS: The median tumour size was 2.3 cm (range, 0.7-3.9 cm). The minimal and mean follow-up periods were 24 and 56 months, respectively. Local recurrence was detected in four patients (4.7%) at 8.5, 13.8, 58.0 and 64.0 months of follow-up and retreated successfully with percutaneous TA. No patient developed metastatic renal cell carcinoma, and none died due to renal oncologic complications. One patient died of heart attack. The 5-year LTPFS, OS, MFS and CSS rates were 93.0%, 98.4%, 100% and 100%, respectively. Only two patients (2.3%) had major complications (Clavien-Dindo grade > II), including ureteropelvic junction stenosis and urinary obstruction due to ureteral blood clots. CONCLUSIONS: Our study demonstrates that percutaneous TA is a feasible and effective first-line therapy for healthy surgical candidates with small renal masses (T1a). The 5-year LTPFS, OS, CSS and MFS rates were 93.0%, 98.4%, 100% and 100%, respectively, with a major complication rate of only 2.3%. KEY POINTS: • Image-guided percutaneous thermoablation of small renal malignancies was effective in 95.3% of the healthy surgical candidates. • Major complications were detected in 2.3% of the patients. • The local tumour progression-free survival rate was 97.6% and 93.0% at 3 and 5 years, respectively.

Herein, we present an overview of recent advances in the electrochemical halogenation of organic compounds. Halogen-containing compounds are essential building blocks for post-functionalization in academic research and in different industrial sectors, notably pharmaceutical companies. One of the main advantages of using electrosynthesis is its environmentally-friendly characteristics. Electrosynthesis is recognized as a methodology that can fulfill different criteria of the green chemistry concept and constitutes a promising tool in the development of new procedures in organic synthesis. In this review we target sp, sp2 and sp3 carbon fluorination, chlorination, bromination and iodination reactions using electrolysis as a redox medium. Mechanistic insights and substrate reactivity are also discussed.

Flowering time is a finely tuned process in plants, in part controlled by the age-regulated microRNA156 (miR156), which functions by suppressing the transcripts of SQUAMOSA-PROMOTER BINDING LIKE (SPL) transcription factors. ARGONAUTE (AGO) proteins are essential effectors of miRNA-mediated gene regulation. However, which AGO(s) mediate(s) the control of flowering time remains unclear. Here, we demonstrate a role of AGO5 in controlling flowering time by modulating the expression of SPL transcription factors. We show that AGO5 interacts physically and functionally with miR156 and that ago5 mutants present an early flowering phenotype in Arabidopsis. Furthermore, in ago5 mutants, the repression of flowering caused by miR156 overexpression is largely reversed, whereas leaf morphology remains unaffected. Our results thus indicate a specific role for AGO5 in mediating miR156 activity in meristematic, but not vegetative, tissue. As such, our data suggest a spatiotemporal regulation of the miR156 aging pathway mediated through different AGO proteins in different tissues.

RESUMEN

RNA silencing is a major mechanism of constitutive antiviral defense in plants, mediated by a number of proteins, including the Dicer-like (DCL) and Argonaute (AGO) endoribonucleases. Both DCL and AGO protein families comprise multiple members. In particular, the AGO protein family has expanded considerably in different plant lineages, with different family members having specialized functions. Although the general mode of action of AGO proteins is well established, the properties that make different AGO proteins more or less efficient at targeting viruses are less well understood. In this report, we review methodologies used to study AGO antiviral activity and current knowledge about which AGO family members are involved in antiviral defense. In addition, we discuss what is known about the different properties of AGO proteins thought to be associated with this function.

Organophosphorus chemistry is a broad field with multi-dimensional applications in research area of organic, biology, drug design and agrochemicals. Conventional methods have been adopted extensively to access phosphorylated compounds that rely on the use of toxic, moisture sensitive phosphorylating agents and occur in the presence of oxidants, catalysts, as well as high temperatures and harsh conditions are required for complete transformations. However, recent progress has been made for phosphorylation reactions using electricity to introduce green and sustainable synthetic procedures. These reactions can be performed at mild conditions and proceed with excellent atom economy. Herein, we targeted electrochemical phosphorylation reactions with generation of new bonds such as C(sp3 ) -P, C(sp2 ) -P, O-P, N-P, S-P and Se-P. This review is aimed to offer an overview of recent developments in the synthetic methodology to easy access of organophosphorus compounds using electrochemistry.

Herein, we report an eco-friendly, electrosynthetic approach for the intramolecular oxyselenylation of allyl-naphthol/phenol derivatives. This reaction proceeds with 0.2 equiv. of nBu4NClO4 as an electrolyte and Pt working electrodes in an undivided cell, resulting in the selenyl-dihydrofurans in good to excellent yields. Furthermore, several of the synthesized products presented a high percentage of acetylcholinesterase (AChE) inhibition, highlighting their potential anti-Alzheimer activity.

J Strength Cond Res 34(3): 808-817, 2020-The purpose of this study was to investigate the independent and interactive effects of situational variables, opposition team formation, and playing position on running performance and network analysis in Brazilian professional soccer players (n = 22). Global positioning system technology was used to determine total distance covered, mean speed, maximum running speed, and distance covered in 6 speed ranges. Social network analysis was used to assess interpersonal coordination (team interactions characterized as successful passes [n = 3,033] between teammates). Observations of match running performance (n = 129) and network analysis (n = 108) were obtained. The main results were: (a) no interactive effects between team formation and playing position were observed for running and network variables (unclear to possibly); (b) matches played at home or against "weaker" opponents presented greater running demands and individual/global metrics of network analysis (likely to almost certain); (c) match outcome demonstrated influence only for running performance; matches in which the reference team won resulted in higher values than in matches lost; (d) when the reference team competed in 1-4-4-2 formation, this resulted in greater running demands than 1-4-2-3-1 formation (likely to almost certain); (e) reduced values of running performance variables were reported in central defenders compared with other positions. Central/external midfielders reported greater closeness/betweenness centrality, outdegree, and eigenvector compared with central/external defenders and forwards (likely to almost certain). The results from this study provide practical information to potentially impact on physical, tactical, and technical training.

A novel and efficient SeCl2-mediated chalcogenative cyclization strategy toward 3-selenophen-3-yl-1 H-indoles from readily available and conveniently substituted propargyl indoles is described. It entails an unprecedented selenirenium-induced 1,2-indolyl shift prompted by the electrophilic addition of SeCl2 to the triple bond of the propargyl indole, followed by cyclization through the intermediacy of a 1-seleno-1,3-diene. The reaction takes place at room temperature and shows excellent selectivity, broad substrate scope, and wide functional group tolerance.

J Strength Cond Res 32(2): 313-325, 2018-The purpose of the study was to examine the influence of playing congested fixtures (2 matches a week) on running performance of Brazilian professional football players, pooled (all performance files regardless players' rotation between games) or individualized data treatment (including only files of athletes' who played pair(s) of noncongested-congested matches which occurred on the same week). Forty participants were observed 1-37 times during 59 official matches over 3 years. Global positioning system devices were used to compute total distance covered, maximal sprinting speed mean speed (VAVERAGE), and high-intensity activities (HIA: ≥15 km·h). Independent variables included match status (win, draw, and lose), location (home and away), opponent quality (weak and strong), competitive standard (national and state), team formation (4-4-2 and 4-3-3), and position (defender, midfielder, and forward). Student's t-test, effect sizes (d), and magnitude-based inferences were performed to detect differences between congested and noncongested fixtures for each independent factor. Overall, pooled and individualized data treatment revealed contrasting results. The first showed chances of positive outcomes playing congested matches (possibly to very likely; d = 0.09-0.55), for all variables, as using 4-4-2 and state tournament. Conversely, individualized data analysis revealed that during national leagues (possibly; d = 0.24) or forward position (likely; d = 0.70), the players presented reduced HIA when played congested than noncongested periods (p ≤ 0.05). Using 4-3-3 (d = 0.26) or when draw (d = 0.41), possibly lower HIA was also identified. Coaching staff should be aware of performance oscillations such as detrimental effects on HIA during congested periods.

The ability to coordinate the simultaneous movements of our arms is limited by a coalition of constraints. Some of these constraints can be overcome when the task conceptualisation is improved. The present study investigated how the movement preparation of bimanual reaching movements was affected by integrated visual feedback of the responses. Previous research has shown that the preparation of bimanual asymmetric movements takes longer than bimanual symmetric movements. The goal of the present study was to determine whether integrated, Lissajous feedback could eliminate this bimanual asymmetric cost. Fifteen participants made unimanual and bimanual symmetric and asymmetric reaches with separate feedback, where there was a cursor and a target for each hand. Participants also made bimanual symmetric and asymmetric movements with integrated feedback; a single cursor and a single target represented the locations and goals of both arms in this condition. The results showed a bimanual asymmetric cost with separate feedback, and that this cost persisted with integrated feedback. We suggest that integrated feedback improved continuous and discrete bimanual movements in other experiments by facilitating error detection and correction processes. We hypothesise that the bimanual asymmetric cost persisted in the present experiment because the uncertainty associated with choice reaction time prevented the facilitated error processing from improving the preparation of the next trial.

J Strength Cond Res 31(8): 2155-2161, 2017-The aim of this study was to examine the independent and interactive effects of match location, quality of opponents, and match status on the movement patterns in a professional Brazilian football team. Sixteen matches of the fourth division Brazilian Championship of 2015 were analyzed during the competitive stages (classifier, 8 matches; octave-finals, 2 matches; quarterfinals, 2 matches; semifinals, 2 matches; and finals, 2 matches). A 5-Hz Global Positioning System Sports QSTARZ was used to record the total distance (TD), maximum speed (VMAX), average speed (VAVERAGE), and frequency of high-intensity activities (HIA). The Student's t-test for independent samples showed significantly higher values (p ≤ 0.05) of VMAX, VAVERAGE, and HIA in home matches when compared with away matches. Comparing the quality of opponents, statistically higher values of TD, VMAX, and HIA were found when the team played against strong opponents. Regarding match status, 1-way analysis of variance demonstrated that when the team won presented significantly higher values of TD, VMAX, VAVERAGE, and HIA compared with matches when the team lost. There were no substantial interactive effects of match situational variables on movement patterns. Finally, multiple linear regression showed that the variable quality of opponents has a higher relative contribution to the variance in HIA (19%) than match status (16%) and match location (4%). In particular, the results indicate that physical performance in professional football is influenced by match situational variables, resulting in a change in the team's style of play.
