Introduction: Understanding the interplay between cardiovascular parameters, cognitive stress induced by increasing load, and mental well-being is vital for the development of integrated health strategies today. By monitoring physiological signals like electrocardiogram (ECG) and photoplethysmogram (PPG) in real time, researchers can discover how cognitive tasks influence both cardiovascular and mental health. Cardiac biomarkers resulting from cognitive strain act as indicators of autonomic nervous system function, potentially reflecting conditions related to heart and mental health, including depression and anxiety. The purpose of this study is to investigate how cognitive load affects ECG and PPG measurements and whether these can signal early cardiovascular changes during depression and anxiety disorders. Methods: Ninety participants aged 18 to 45 years, ranging from symptom-free individuals to those with diverse psychological conditions, were assessed using psychological questionnaires and anamnesis. ECG and PPG monitoring were conducted as volunteers engaged in a cognitive 1-back task consisting of two separate blocks, each with six progressively challenging levels. The participants' responses were analyzed to correlate physiological and psychological data with cognitive stressors and outcomes. Results: The study confirmed a notable interdependence between anxiety and depression, and cardiovascular responses. Task accuracy decreased with increased task difficulty. A strong relationship between PPG-measured heart rate and markers of depression and trait anxiety was observed. Increasing task difficulty corresponded to an increase in heart rate, linked with elevated levels of depression and trait anxiety. A strong relationship between ECG-measured heart rate and anxiety attacks was observed. Increasing task difficulty corresponded to an increase in heart rate, linked with elevated levels of anxiety attacks, although this association decreased under more challenging conditions. Discussion: The findings underscore the predictive importance of ECG and PPG heart rate parameters in mental health assessment, particularly depression and anxiety under cognitive stress induced by increasing load. We discuss mechanisms of sympathetic activation explaining these differences. Our research outcomes have implications for clinical assessments and wearable device algorithms for more precise, personalized mental health diagnostics.

Objectives: Pupil dilation is controlled both by sympathetic and parasympathetic nervous system branches. We hypothesized that the dynamic of pupil size changes under cognitive load with additional false feedback can predict individual behavior along with heart rate variability (HRV) patterns and eye movements reflecting specific adaptability to cognitive stress. To test this, we employed an unsupervised machine learning approach to recognize groups of individuals distinguished by pupil dilation dynamics and then compared their autonomic nervous system (ANS) responses along with time, performance, and self-esteem indicators in cognitive tasks. Methods: Cohort of 70 participants were exposed to tasks with increasing cognitive load and deception, with measurements of pupillary dynamics, HRV, eye movements, and cognitive performance and behavioral data. Utilizing machine learning k-means clustering algorithm, pupillometry data were segmented to distinct responses to increasing cognitive load and deceit. Further analysis compared clusters, focusing on how physiological (HRV, eye movements) and cognitive metrics (time, mistakes, self-esteem) varied across two clusters of different pupillary response patterns, investigating the relationship between pupil dynamics and autonomic reactions. Results: Cluster analysis of pupillometry data identified two distinct groups with statistically significant varying physiological and behavioral responses. Cluster 0 showed elevated HRV, alongside larger initial pupil sizes. Cluster 1 participants presented lower HRV but demonstrated increased and pronounced oculomotor activity. Behavioral differences included reporting more errors and lower self-esteem in Cluster 0, and faster response times with more precise reactions to deception demonstrated by Cluster 1. Lifestyle variations such as smoking habits and differences in Epworth Sleepiness Scale scores were significant between the clusters. Conclusion: The differentiation in pupillary dynamics and related metrics between the clusters underlines the complex interplay between autonomic regulation, cognitive load, and behavioral responses to cognitive load and deceptive feedback. These findings underscore the potential of pupillometry combined with machine learning in identifying individual differences in stress resilience and cognitive performance. Our research on pupillary dynamics and ANS patterns can lead to the development of remote diagnostic tools for real-time cognitive stress monitoring and performance optimization, applicable in clinical, educational, and occupational settings.

Generalized anxiety disorder (GAD) is a prevalent disorder. The search for biomarkers may contribute to new knowledge about molecular pathogenesis and treatment. Since oxidative stress and micronutrient imbalance play a key role in the development of mental disorders, we aimed to study salivary antioxidant capacity and magnesium in patients with GAD in an anxiety model of solving problems with increasing complexity. The study subgroup consisted of 15 patients with GAD, and 17 healthy volunteers of the same age made up the control subgroup. Participants took a test with six levels of difficulty, which included false feedback. In this test, the participants were asked to remember the colors of balloons and react when the color changed. The reaction time, the number of correct answers, as well as biochemical parameters such as the antioxidant capacity of saliva and salivary magnesium, were assessed. There was no difference in the results of the quest between the subgroups; however, anxious participants spent more time at the moment of experimental frustration due to incorrect feedback and additional negative psycho-emotional load. Antioxidant capacity did not differ between the subgroups both before and after the experimental session. Average antioxidant capacity also did not change significantly at the endpoint of the experiment. However, the endpoint antioxidant capacity correlated negatively with the reaction time in anxious patients in the second block (where the false feedback as a frustrating factor appeared). Magnesium was initially significantly higher in the group of anxious participants and decreased at the experiment endpoint; in healthy patients, there were no changes in salivary magnesium at the endpoint. In conclusion, the compensatory potential of oxidative metabolism and magnesium in patients with GAD was spent with additional psycho-emotional stress, in contrast to healthy individuals, but it was sufficient to avoid exhaustion during experimental frustrating exposure.

Spelling errors are ubiquitous in all writing systems. Most studies exploring spelling errors focused on the phonological plausibility of errors. However, unlike typical pseudohomophones, spelling errors occur in naturally produced written language. We investigated the time course of recognition of the most frequent orthographic errors in Russian (error in an unstressed vowel in the root) and the effect of word frequency on this process. During event-related potentials (ERP) recording, 26 native Russian speakers silently read high-frequency correctly spelled words, low-frequency correctly spelled words, high-frequency words with errors, and low-frequency words with errors. The amplitude of P200 was more positive for correctly spelled words than for misspelled words and did not depend on the frequency of the words. In addition, in the 350-500-ms time window, we found a more negative response for misspelled words than for correctly spelled words in parietal-temporal-occipital regions regardless of word frequency. Considering our results in the context of a dual-route model, we concluded that recognizing misspelled high-frequency and low-frequency words involves common orthographic and phonological processes associated with P200 and N400 components such as whole word orthography processing and activation of phonological representations correspondingly. However, at the 500-700 ms stage (associated with lexical-semantic access in our study), error recognition depends on the word frequency. One possible explanation for these differences could be that at the 500-700 ms stage recognition of high-frequency misspelled and correctly spelled words shifts from phonological to orthographic processes, while low-frequency misspelled words are accompanied by more prolonged phonological activation. We believe these processes may be associated with different ERP components P300 and N400, reflecting a temporal overlap between categorization processes based on orthographic properties for high-frequency words and phonological processes for low-frequency words. Therefore, our results complement existing reading models and demonstrate that the neuronal underpinnings of spelling error recognition during reading may depend on word frequency.

Emotional dysfunction, including flat affect and emotional perception deficits, is a specific symptom of schizophrenia disorder. We used a modified multimodal odd-ball paradigm with fearful facial expressions accompanied by congruent and non-congruent emotional vocalizations (sounds of women screaming and laughing) to investigate the impairment of emotional perception and reactions to other people's emotions in schizophrenia. We compared subjective ratings of emotional state and event-related potentials (EPPs) in response to congruent and non-congruent stimuli in patients with schizophrenia and healthy controls. The results showed the altered multimodal perception of fearful stimuli in patients with schizophrenia. The amplitude of N50 was significantly higher for non-congruent stimuli than congruent ones in the control group and did not differ in patients. The P100 and N200 amplitudes were higher in response to non-congruent stimuli in patients than in controls, implying impaired sensory gating in schizophrenia. The observed decrease of P3a and P3b amplitudes in patients could be associated with less attention, less emotional arousal, or incorrect interpretation of emotional valence, as patients differed from healthy controls in the emotion scores of non-congruent stimuli. The difficulties in identifying the incoherence of facial and audial components of emotional expression could be significant in understanding the psychopathology of schizophrenia.

Lateral asymmetry is one of the fundamental properties of the functional anatomy of the human brain. Amygdala (AMYG) asymmetry was also reported in clinical studies of resting-state functional connectivity (rsFC) but rarely in healthy groups. To explore this issue, we investigated the reproducibility of the data on rsFC of the left and right AMYG using functional MRI twice a week in 20 healthy volunteers with mild-to-moderate anxiety. We found a resting-state network of the AMYG, which included regions involved in emotional processing and several other brain areas associated with memory and motor inhibition. The AMYG network was stable in time and within subjects, but the right AMYG had more significant connections with anatomical brain regions. The rsFC values of the right AMYG were also more sustained across the week than the left AMYG rsFC. Subjective ratings of anxiety did not correlate significantly with the patterns of seed-based AMYG connectivity. Our findings indicate that, for healthy subjects, rsFC may differ for the right and left AMYG. Moreover, the AMYG functional connectivity is variable in short-term observations, which may also influence the results of longitude studies.

The neural underpinnings of subjective experience during resting state remain elusive. Dynamic features of EEG oscillations may provide more understanding of the relationship between the content of inner conscious experience and electrical brain activity. We tested a correlation of rating on the Amsterdam Resting-State Questionnaire (ARSQ) with dynamic parameters of EEG recorded in 49 healthy volunteers during the 10-min resting session. The participants filled ARSQ immediately after the rest. We investigated both linear (1 Hz-band power spectral density - PSD) and dynamic features (standard deviation and frequency of Hilbert envelope) of EEG averaged for the whole resting-state segment. Besides, we conducted a procedure of k-mean clustering based on PSD, localization of components retrieved by independent component analysis for 10-sec EEG epochs to assess spectral and temporal variability of EEG. The correlation analysis showed that the increase of PSD and cluster duration of the high-frequency alpha rhythm (12-13 Hz) in central and frontal areas was positively associated with the rating of experienced thoughts related to Planning (r = 0.44). The time of the presence of low amplitude delta oscillations correlated negatively with Planning (r = -0.52). The participants with higher ARSQ scores of Visual Thoughts had a higher standard deviation of the wideband (1-30 Hz) Hilbert envelope. Our data suggest that the dynamic properties of EEG reflect cognitive states assessed by ARSQ.

Clinical neurology is constantly searching for reliable indices of ischemic brain damage to prevent a possible development of stroke. We suggest that resting state electroencephalogram (rsEEG) with respect to other clinical data may provide important information about the severity of ischemia. We carried out correlation analysis of rsEEG, data of transcranial Doppler ultrasonography of head vessels, and clinical assessment scores collected from healthy volunteers and four groups of patients with mild chronic microvascular ischemia (CMI-1), moderate CMI (CMI-2), severe atrophy of the cerebral hemisphere, ischemic stroke in the left middle cerebral artery stroke, and ischemic stroke in the right middle cerebral artery stroke. Using independent component analysis and k-mean clustering of EEG data, we observed prominent changes in rsEEG reflected in specific distributions of spectral peaks in all groups of patients. We found a significant correlation of EEG spectral distribution and the blood flow velocity in coronal arteries, which was also affected by the severity of ischemia and the localization of stroke. Moreover, EEG spectral distribution was more indicative of early stages of ischemia than the blood flow velocity. Our data support the hypothesis that rsEEG may reflect altered neural activity caused by ischemic brain damage.
