The removal of protons (H+) produced during intense exercise is important for skeletal muscle function, yet it remains unclear how best to structure exercise training to improve muscle pH regulation. We investigated whether 4 wk of work-matched sprint-interval trining (SIT), performed 3 days/wk, with either 1 (Rest-1; n = 7) or 5 (Rest-5; n = 7) min of rest between sprints, influenced adaptations in acid/base transport protein content, nonbicarbonate muscle buffer capacity (ßmin vitro), and exercise capacity in active women. Following 1 wk of posttesting, comprising a biopsy, a repeated-sprint ability (RSA) test, and a graded-exercise test, maintenance of adaptations was then studied by reducing SIT volume to 1 day/wk for a further 5 wk. After 4 wk of SIT, there was increased protein abundance of monocarboxylate transporter (MCT)-1, sodium/hydrogen exchanger (NHE)-1, and carbonic anhydrase (CA) XIV for both groups, but rest interval duration did not influence the adaptive response. In contrast, greater improvements in total work performed during the RSA test after 4 wk of SIT were evident for Rest-5 compared with Rest-1 (effect size: 0.51; 90% confidence limits ±0.37), whereas both groups had similarly modest improvements in VÌo2peak When training volume was reduced to 1 day/wk, enhanced acid/base transport protein abundance was maintained, although NHE1 content increased further for Rest-5 only. Finally, our data support intracellular lactate as a signaling molecule for inducing MCT1 expression, but neither lactate nor H+ accumulation appears to be important signaling factors in MCT4 regulation.

NEW FINDINGS: What is the central question of this study? Following a training intervention, how is the interpretation of adaptations in skeletal muscle H+ transporters influenced by biopsy timing in the context of individual protein and mRNA kinetics after the final exercise bout? What is the main finding and its importance? We show that distinct postexercise protein and mRNA kinetics for monocarboxylate transporter 1/4 and sodium-hydrogen exchanger 1 indicate that timing of a single end-point biopsy after a training intervention can influence the inferences made. Furthermore, we found the intrasubject, intersample variability of the muscle buffer capacity titration assay to be greater than the typical training effect. In order to gain a better understanding of training-induced adaptations in skeletal muscle pH regulation, in this study we measured protein and mRNA kinetics of proton (H+ ) transporters for 72 h following a bout of high-intensity interval exercise (HIIE), conducted after 4 weeks of similar training. We also assayed muscle buffer capacity (ßm) by a titration technique (ßmin vitro ) over the same period. Sixteen active men cycled for seven bouts of 2 min at â¼80% of peak aerobic power, interspersed with 1 min rest. Compared with the first 9 h postexercise, monocarboxylate transporter (MCT) 1 protein content was â¼1.3-fold greater 24-72 h post-HIIE, whereas there was no such change in MCT4 protein content. Conversely, MCT1 and MCT4 mRNA expression progressively decreased 9-72 h post-HIIE. Sodium-hydrogen exchanger 1 (NHE1) protein content was lower 9 h post-HIIE (â¼0.8-fold) compared with every other postexercise time point, but NHE1 mRNA expression was 2.2 to 2.9-fold greater 24-72 h post-HIIE, compared with the first 24 h post-HIIE. Furthermore, we determined the intrasubject, intersample variability (11.5%) of ßmin vitro for resting samples taken on consecutive days to be greater than the typical training effect (mean 6%; 95% confidence limits ±4%). In conclusion, the delay in steady-state protein turnover should inform biopsy timing in studies investigating the response to training of the H+ transport proteins, whereas the temporal resolution provided by single time points has been shown to be of limited epistemological value for their corresponding mRNA expression. Finally, our data cast doubt on the ecological validity of the ßmin vitro assay for measuring true changes in ßm.

BACKGROUND: Exercise elicits a range of adaptive responses in skeletal muscle, which include changes in mRNA expression. To better understand the health benefits of exercise training, it is important to investigate the underlying molecular mechanisms of skeletal muscle adaptation to exercise. However, most studies have assessed the molecular events at only a few time-points within a short time frame post-exercise, and the variations of gene expression kinetics have not been addressed systematically. METHODS: We assessed the mRNA expression of 23 gene isoforms implicated in the adaptive response to exercise at six time-points (0, 3, 9, 24, 48, and 72 h post exercise) over a 3-day period following a single session of high-intensity interval exercise. RESULTS: The temporal patterns of target gene expression were highly variable and the expression of mRNA transcripts detected was largely dependent on the timing of muscle sampling. The largest fold change in mRNA expression of each tested target gene was observed between 3 and 72 h post-exercise. DISCUSSION AND CONCLUSIONS: Our findings highlight an important gap in knowledge regarding the molecular response to exercise, where the use of limited time-points within a short period post-exercise has led to an incomplete understanding of the molecular response to exercise. Muscle sampling timing for individual studies needs to be carefully chosen based on existing literature and preliminary analysis of the molecular targets of interest. We propose that a comprehensive time-course analysis on the exercise-induced transcriptional response in humans will significantly benefit the field of exercise molecular biology.

First published October 14, 2016; doi:10.1152/japplphysiol.00630.2016-This study measured the adaptive response to exercise training for each of the acid-base transport protein families, including providing isoform-specific evidence for the monocarboxylate transporter (MCT)1/4 chaperone protein basigin and for the electrogenic sodium-bicarbonate cotransporter (NBCe)1. We investigated whether 4 wk of work-matched, high-intensity interval training (HIIT), performed either just above the lactate threshold (HIITΔ20; n = 8), or close to peak aerobic power (HIITΔ90; n = 8), influenced adaptations in acid-base transport protein abundance, nonbicarbonate muscle buffer capacity (ßmin vitro), and exercise capacity in active men. Training intensity did not discriminate between adaptations for most proteins measured, with abundance of MCT1, sodium/hydrogen exchanger (NHE) 1, NBCe1, carbonic anhydrase (CA) II, and CAXIV increasing after 4 wk, whereas there was little change in CAIII and CAIV abundance. ßmin vitro also did not change. However, MCT4 protein content only increased for HIITΔ20 [effect size (ES): 1.06, 90% confidence limits × / ÷ 0.77], whereas basigin protein content only increased for HIITΔ90 (ES: 1.49, × / ÷ 1.42). Repeated-sprint ability (5 × 6-s sprints; 24 s passive rest) improved similarly for both groups. Power at the lactate threshold only improved for HIITΔ20 (ES: 0.49; 90% confidence limits ± 0.38), whereas peak O2 uptake did not change for either group. Detraining was characterized by the loss of adaptations for all of the proteins measured and for repeated-sprint ability 6 wk after removing the stimulus of HIIT. In conclusion, 4 wk of HIIT induced improvements in each of the acid-base transport protein families, but, remarkably, a 40% difference in training intensity did not discriminate between most adaptations.

UNLABELLED: The combined supplementation of caffeine (CAF) and sodium bicarbonate (NaHCO3) may have a potential ergogenic effect during intermittent-exercise tasks such as judo; however, its effect in this sport has not been tested. PURPOSE: To investigate the isolated and combined effects of CAF and NaHCO3 on judo performance. METHODS: Ten judokas performed 4 supplementation protocols-NaHCO3, CAF, NaHCO3 + CAF, and placebo (PLA) (cellulose)-followed by 3 Special Judo Fitness Tests (SJFTs) interspaced with 5 min rest. RESULTS: In the first SJFT, the combined supplement (NaHCO3 + CAF) resulted in a higher number of throws than with PLA (24.4 ± 0.9 and 23.2 ± 1.5 throws, respectively, P = .02). There was no significant difference between conditions for the 2nd SJFT (P = .11). In the 3rd SJFT, NaHCO3 and NaHCO3 + CAF resulted in more throws than with PLA (23.7 ± 1.6, 24.4 ± 1.0, and 22.0 ± 1.6 throws, P = .001 and P = .03, respectively). When the total throws performed in the 3 SJFTs were summed, they were higher than PLA only for NaHCO3 + CAF (68.8 ± 4.4 and 72.7 ± 3.1 throws, respectively, P = .003). Postexercise plasma lactate after each SJFT was higher in all experimental conditions than with PLA (P = .001). There was no significant difference in rating of perceived exertion across the conditions (P = .18). CONCLUSION: The results of the current study show that the combined supplementation of NaHCO3 + CAF increases judo performance compared with PLA.

The high forces undergone during repetitive eccentric, or lengthening, contractions place skeletal muscle under considerable stress, in particular if unaccustomed. Although muscle is highly adaptive, the responses to stress may not be optimally regulated by the body. Reactive oxygen species (ROS) are one component of the stress response that may contribute to muscle damage after eccentric exercise. Antioxidants may in turn scavenge ROS, thereby preventing or attenuating muscle damage. The antioxidant vitamins C (ascorbic acid) and E (tocopherol) are among the most commonly used sport supplements, and are often taken in large doses by athletes and other sportspersons because of their potential protective effect against muscle damage. This review assesses studies that have investigated the effects of these two antioxidants, alone or in combination, on muscle damage and oxidative stress. Studies have used a variety of supplementation strategies, with variations in dosage, timing and duration of supplementation. Although there is some evidence to show that both antioxidants can reduce indices of oxidative stress, there is little evidence to support a role for vitamin C and/or vitamin E in protecting against muscle damage. Indeed, antioxidant supplementation may actually interfere with the cellular signalling functions of ROS, thereby adversely affecting muscle performance. Furthermore, recent studies have cast doubt on the benign effects of long-term, high-dosage antioxidant supplementation. High doses of vitamin E, in particular, may increase all-cause mortality. Although some equivocation remains in the extant literature regarding the beneficial effects of antioxidant vitamin supplementation on muscle damage, there is little evidence to support such a role. Since the potential for long-term harm does exist, the casual use of high doses of antioxidants by athletes and others should perhaps be curtailed.

The purpose of this study was to examine the early-phase adaptations of traditional dynamic constant external resistance (DCER) training vs. a portable upper-body training device (Fortex). The Fortex is a concentric training device based on air resistance. Contractions using this device are slow (1.5-3 s) and have a limited range of motion. The exercises potentially allow maximal muscle action during each contraction. Healthy, sedentary men (n = 30) were assigned to begin either 8 weeks of weight training (W, n = 12) or 8 weeks of Fortex training (F, n = 9), and were compared with a control group (C, n = 9). Exercises were chosen for the W group that would train similar muscle groups and contain a similar volume of repetitions as the F group. However, movement patterns and force curves were not identical. Increases in the upper-arm cross-sectional area were not detected in any of the groups. Both training groups showed strength gains in the various strength tests that were distinct from each other. Our results indicate that both Fortex and DCER training proved effective in eliciting strength gains in sedentary men over an 8-week training period. There are, however, limitations with the Fortex in terms of progression needs and training asymmetry that indicate it should be used as a complement to other training.
