RESUMEN

Salmonella is the leading cause of zoonotic foodborne illnesses worldwide and a prevalent threat to the poultry industry. For controlling contamination, the use of chemical sanitizers in combination with biological compounds (e.g., enzymes) offers a solution to reduce the chemical residues. The current study investigated the biofilm reduction effects of a food-grade enzyme-ficin-and a common sanitizer-peroxyacetic acid (PAA)-against an emerging pathogen, Salmonella enterica ser. Thompson, on plastic, eggshell, and chicken skin surfaces. Results showed that PAA could kill S. Thompson, but ficin cannot. Maximum biofilm reduction was 3.7 log CFU/cm2 from plastic after individual treatment with PAA. However, sequential treatment of ficin and PAA led to biofilm reductions of 3.2, 5.0, and 6.5 log CFU/cm2 from chicken skin, eggshell, and plastic, respectively. Fourier-transform infrared spectroscopy and microscopic analysis confirmed that ficin increased PAA action, causing biofilm matrix destruction. Moreover, the quality of the food surfaces was only altered by 12.5 U/mL ficin and was not altered by PAA. This combined use of enzyme and sanitizer solved major safety issues and proved promising against S. Thompson-associated contaminations in poultry and poultry processing lines.

Cdc6 is the bifunctional AAA+ ATPase that assembles prereplicative complexes on origins of replication and activates p21(CIP1)- or p27(KIP1)-bound Cdk2. During the G(1)-S transition, the Cdc6 gene essential for chromosomal replication is activated by the E2F transcriptional factor. Paradoxically, Apaf-1 encoding the central component of the apoptosome is also activated at the same time and by E2F. Consequently, genes for antipodal life and death are regulated in the same manner by the same transcriptional factor. Here we report a striking solution to this paradox. Besides performing prereplicative complex assembly and Cdk2 activation, Cdc6 obstructed apoptosome assembly by forming stable complexes very likely with a monomer of cytochrome c-activated Apaf-1 molecules. This function depended on its own ATPase domain but not on the cyclin-binding motif. In proliferating rodent fibroblasts, Cdc6 continued to block apoptosome assembly induced by a non-cytochrome c or some other mechanism, suppressing seemingly unintended apoptosis when promoting cell proliferation. Thus, Cdc6 is an AAA+ ATPase with three functions, all working for life.

Discotic hydrazone molecules are of particular interest as they form discotic phases where the discs are rigidified by intramolecular hydrogen bonds. Here, we investigate the thermotropic behavior and solid-state organizations of three discotic hydrazone derivatives with dendritic groups attached to their outer peripheries, containing six, eight, and ten carbons of linear alkoxy chains. On the basis of two-dimensional wide angle X-ray scattering (2DWAXS), the elevated temperature liquid crystalline (LC) phases were assigned to a hexagonal columnar (Colh) organization with nontilted hydrazone discs for all three compounds. With WAXS, advanced solid-state nuclear magnetic resonance (SSNMR) techniques, and ab initio computations, the compounds with six and ten carbons of achiral alkoxy side chains were further subjected to studies at 25 °C, revealing complex crystalline phases with rigid columns and flexible side chains. This combined approach led to models of coexisting helical columnar stacking morphologies for both systems with two different tilt/pitch angles between successive hydrazone molecules. The differences in tilt/pitch angles between the two compounds illustrate that the columns with short alkoxy chains (six carbons) are more influenced by the presence of other stacks in their vicinity, while those with long side chains are less tilted due to a larger alkoxy (ten carbons) buffer zone. The formation of different packing morphologies in the crystalline phase of a columnar LC has rarely been reported so far, which suggests the possibility of complex stacking structures of similar organic LC systems, utilizing small molecules as potential materials for applications in organic electronics.

This study investigated the antimicrobial and antibiofilm efficacy of separate and combined treatments of Lactobacillus curvatus B67-produced postbiotic and the polyphenolic flavanol quercetin against Listeria monocytogenes and Salmonella enterica ser. Typhimurium. The antimicrobial potentiality of the postbiotic was chiefly associated with organic acids (e.g., lactic and acetic acids). At sub-minimum inhibitory concentration (1/2 MIC), the postbiotic and quercetin effectively reduced the pathogenic biofilm cells on processed pork sausage and meat-processing surfaces (e.g., stainless-steel and rubber). Moreover, the postbiotic exhibited strong residual antimicrobial efficacy over diverse pH and temperature ranges. In addition, the combination of postbiotic with quercetin increased the leakage of pathogenic intracellular metabolites (e.g., nucleic acids and protein) and inhibited pathogenic biofilm formation on both biotic and abiotic surfaces. Therefore, this study confirmed that lactic acid bacteria-derived postbiotic and plant-derived quercetin could be used as potential alternative bioprotective agents in the meat processing industry.

Foodborne pathogen-mediated biofilms in food processing environments are severe threats to human lives. In the interest of human and environmental safety, natural substances with antimicrobial properties and generally regarded as safe (GRAS) status are the futuristic disinfectants of the food industry. In this study, the efficacy of bioactive, soluble products (metabolic by-products) from lactic acid bacteria (LAB) and plant-derived essential oils (EO) were investigated as biocidal agents. The postbiotic produced by kimchi-derived Leuconostoc mesenteroides J.27 isolate was analyzed for its metabolic components to reveal its antimicrobial potential against three pathogenic microorganisms (Vibrio parahaemolyticus, Pseudomonas aeruginosa, and Escherichia coli). Additionally, the efficacy of food-grade EO (eugenol and thymol, respectively) was also assessed in our study. Determination of the minimum inhibitory concentration (MIC) of postbiotic and EO against three tested pathogens revealed that the sub-MIC (0.5 MIC) of postbiotic and EO could efficiently inhibit the biofilm formation on both seafood (squid) and seafood-processing surfaces (rubber and low-density polyethylene plastic). Moreover, the polymerase chain reaction (PCR) analysis confirmed that the LAB J.27 isolate possesses bacteriocin- and enzyme-coding genes. The residual antibacterial activity of the produced postbiotic was maintained over a diverse pH range (pH 1-6) but was entirely abolished at neutral or higher pH values. However, the activity was unaffected by exposure to high temperatures (100 and 121 °C) and storage (30 days). Notably, the leakage of intracellular metabolites, damage to DNA, and the down-regulation of biofilm-associated gene expression in the pathogens increased significantly (p > 0.05) following the combination treatment of postbiotic with thymol compared to postbiotic with eugenol. Nonetheless, all in vitro results indicated the prospective use of combining Leu. mesenteroides J.27-derived postbiotic with both EO as a "green preservative" in the seafood industry to inhibit the formation of pathogenic microbial biofilms.

We prepared 1H-imidazolecarboxamide amphiphiles as potential organogelators. Compounds A1-A3, in which an imidazole head was connected to a hydrophobic trialkyloxyphenyl group, showed an ability to gelate nonpolar solvents, including alkanes. The dry gels obtained from compounds A1-A3 had columnar hexagonal structures. Polycatenar 1H-imidazolecarboxamide amphiphile B2, consisting of a 1H-imidazole head connected through a benzene ring to a tridecyloxyphenyl tail, formed an organogel in DMSO. In a concentrated THF solution (30 wt %), compound B2 exhibited a lyotropic liquid-crystalline phase with a columnar hexagonal structure. X-ray diffraction (XRD) results suggested a molecular arrangement consisting of a disk, via hydrogen bonding between successive imidazole moieties, and an assembly of columnar structures.

[reaction: see text] 1,3,5-Trisacetoacetamidobenzene with three 1,3-diketo groups was synthesized by the reaction of 1,3,5-triaminobenzene with diketene. Discotic hydrazone compounds were prepared by the diazo coupling reaction between 1,3,5-trisacetoacetamidobenzene and diazonium salts of 4-alkyloxyphenylamines. The compounds existed in hydrazone forms exclusively, being stabilized by the intramolecular hydrogen bonds, and showed discotic nematic or columnar hexagonal mesophases.

OBJECTIVE: When dexmedetomidine is used in patients undergoing spinal anesthesia, high incidence of bradycardia in response to parasympathetic activation is reported. Therefore, we aimed to evaluate the effectiveness of atropine premedication for preventing the incidence of bradycardia and the hemodynamic effect on patients undergoing spinal anesthesia with sedation by dexmedetomidine. DESIGN: Randomized, double-blind, placebo-controlled study. SETTING: Operating room. PATIENTS: One hundred fourteen patients (age range, 2-65 years; American Society of Anesthesiology class I-II) participated in this study, willing to be sedated and to undergo spinal anesthesia. INTERVENTION: The patients were divided into 2 groups: group A and group C. After performing spinal anesthesia, dexmedetomidine was infused at a loading dose of 0.6 µg/kg for 10 minutes, followed by an infusion at 0.25 µg/(kg h). Simultaneously with the loading dose of dexmedetomidine, patients in group A received an intravenous bolus of 0.5 mg atropine, whereas patients in group C received an intravenous normal saline bolus. MEASUREMENT: Data on administration of atropine and ephedrine were collected. Hemodynamic data including heart rate, systolic blood pressure, diastolic blood pressure (DBP), and mean blood pressure (MBP) were also recorded. MAIN RESULTS: The incidence of bradycardia requiring atropine treatment was significantly higher in group C than group A (P=.035). However, the incidence of hypotension needing ephedrine treatment showed no significant difference between the 2 groups (P=.7). Systolic blood pressure and heart rate showed no significant differences between the 2 groups (P=.138 and .464, respectively). However, group A showed significant increases in DBP and MBP, and group C did not (P=.014 and .008, respectively). CONCLUSION: Prophylactic atropine reduces the incidence of bradycardia in patients undergoing spinal anesthesia with dexmedetomidine sedation. However, DBP and MBP showed significant increases in patients when prophylactic atropine was administrated. Therefore, atropine premedication should be administered cautiously.

A series of ortho or meta Lewis base functionalized unbridged zirconocenes, [{1-(E(n)-Ph)-3,4-Me(2)C(5)H(2)}(2)ZrCl(2)] (E = NMe(2), OMe; n = 1, 2), and a half-functionalized zirconocene, [{1-(p-Me(2)NC(6)H(4))-3,4-Me(2)C(5)H(2)}{1-(p-tolyl)-3,4-Me(2)C(5)H(2)}ZrCl(2)], were prepared. The crystal structures of these compounds determined by X-ray diffraction revealed the presence of only C(2)-symmetric rac-like isomers in the asymmetric units. In combination with methylaluminoxane (MAO) cocatalyst, the meta-functionalized complexes afforded mixtures of polymers that exhibit multimelting transition temperatures and broad molecular-weight distributions (MWDs) in propylene polymerization at atmospheric monomer pressure, whereas the ortho-functionalized complexes did not give rise to polymerization. Stepwise solvent extraction of the polymer mixtures showed that the polymers consist of amorphous, moderately isotactic, and highly isotactic portions, the weight ratio of which is dependent on the reaction temperature. (13)C NMR spectral analysis indicated that the [mmmm] methyl pentad value of the isotactic portion reached around 90%. Among the meta-functionalized zirconocenes, the di-OMe-substituted one afforded the largest amount of the isotactic portion at all temperatures, and the portion comprised 82 wt % of the crude polymer obtained at 25 degrees C. In contrast, propylene polymerization with the half-functionalized unbridged zirconocene resulted in the formation of nearly atactic polypropylene with a narrow MWD of around 2. These results corroborate the proposition that the rigid rac-like cation-anion ion pair of type [rac-L(2)ZrP](+)[Me-MAO](-) generated in situ, through Lewis acid-base interactions between the functional groups and [Me-MAO](-), is responsible for the isospecific propylene polymerization with the given class of functionalized unbridged zirconocenes and further indicate that the formation of such ion pairs can be favored by difunctionalization at the meta position of the phenyl ring with OMe groups.
