In a society where people often communicate through digital technology, it is crucial to investigate whether the transmission mode influences destination memory performance (our capacity to remember to whom we transmitted certain information). In Experiment 1, we asked young adults (N = 31) to share of a set of familiar proverbs only by typing and the rest by both typing and saying them aloud. Better destination memory was observed when the information was transmitted by the two means (aloud and typing). Did this better performance occur because participants shared the information aloud or because the information was transmitted by two means? In Experiment 2, young adults (N = 34) shared familiar proverbs aloud, by typing, or simultaneously aloud and by typing. Results showed that transmission aloud led to a better destination memory than typing, and no further improvement occurred when the transmission was both aloud and by typing. Additionally, no differences were observed regarding item memory, supporting the idea that item and destination memories are two different types of memory.

Although effective in reducing virus transmission, face masks might compromise face recognition and trait judgments. With this study, we aimed to observe the influence of masks on face recognition and trait judgments-more specifically, in trustworthiness, dominance, and distinctiveness judgments. Also, we wanted to observe the possible influence of trait judgments on facial recognition for masked and unmasked faces, which has never been done before. For that, we conducted an online study where 140 participants observed and made trait judgments of masked and unmasked faces in a within-subjects design. After a distractive task, participants performed a recognition memory test. As expected, we observed a better recognition of faces shown without a mask during the study phase, which allowed the holistic processing of the faces. The worst performance was found for faces encoded with a mask but tested without it, occurring simultaneity disruption in holistic face processing and the violation of the encoding specificity principle. Regarding the trait judgments, unmasked faces were considered more distinctive, and masked faces were considered more trustworthy. More interestingly, we can conclude that facial distinctiveness predicts face recognition, regardless of mask use. In contrast, dominance judgments only predicted face recognition when faces were presented without a mask. When faces were exposed with masks, trustworthiness overrides dominance, becoming more critical to recognizing faces. We can interpret these results from an evolutionary perspective.

Triple-negative breast cancer has a poor prognosis and is non-responsive to first-line therapies; hence, new therapeutic strategies are needed. Enhanced store-operated Ca2+ entry (SOCE) has been widely described as a contributing factor to tumorigenic behavior in several tumor types, particularly in breast cancer cells. SOCE-associated regulatory factor (SARAF) acts as an inhibitor of the SOCE response and, therefore, can be a potential antitumor factor. Herein, we generated a C-terminal SARAF fragment to evaluate the effect of overexpression of this peptide on the malignancy of triple-negative breast cancer cell lines. Using both in vitro and in vivo approaches, we showed that overexpression of the C-terminal SARAF fragment reduced proliferation, cell migration, and the invasion of murine and human breast cancer cells by decreasing the SOCE response. Our data suggest that regulating the activity of the SOCE response via SARAF activity might constitute the basis for further alternative therapeutic strategies for triple-negative breast cancer.

Considering the global pandemic we currently experience, face masks have become standard in our daily routine. Even though surgical masks are established as a safety measure against the dissemination of COVID-19, previous research showed that their wearing compromises face recognition. Consequently, the capacity to remember to whom we transmit information-destination memory-could also be compromised. In our study, through a between-participants design (experiment 1) and a within-participants design (experiment 2), undergraduate students have to transmit Portuguese proverbs to masked and unmasked celebrity faces. Following our hypothesis, participants who shared information with masked faces had worse destination memory performance than those who shared information with unmasked faces. Also, we observed lower recognition for masked faces compared to unmasked faces. These results were expected since using a surgical mask affects facial recognition, thus making it harder to recognize a person to whom information was previously transmitted. More importantly, these results also support the idea that variables associated with the recipient's face are important for destination memory performance.

Destination memory involves remembering to whom we told information. Low accuracy of this memory is linked to higher self-focus and lower attentional resources allocated to the recipient of the information. The present paper aimed to investigate whether the existence of distinctive features (e.g., tattoos) of destination face would improve destination memory, in a within- (Experiment 1 and 2) and between-participants (Experiment 3) design. In a destination memory task, participants had to tell proverbs to faces that presented a distinctive feature and to other faces that did not. Results showed that a destination memory advantage only occurs when faces with different distinctive features are compared to faces without distinctive features (Experiment 1). These results are in accordance with the existing theoretical framework on destination memory and distinctiveness; highlighting the importance of personal attributes, namely, the distinctiveness of the destination face and the relativity of distinctiveness in destination memory.

Research on familiar faces has been conducted in different countries and resort to celebrities faces, stimuli that are highly constrained by geographic context and cultural peculiarities, since many celebrities are only famous in particular countries. Despite their relevance to psychological research, there are no normative studies of celebrities' facial recognition in Portugal. We developed a database with 160 black and white pictures of famous persons' faces in this work. The data collection took place in two different studies. In study 1, participants were asked to recognize and name celebrity faces; while in study 2, celebrity names were rated for AoA, familiarity, and distinctiveness. Data were gathered from two different samples of Portuguese young adults aged between 18 and 25 years old, and both procedures were performed online through a questionnaire created in Qualtrics software. This database provides ratings of AoA, familiarity, facial distinctiveness, recognition rate, and naming rate for each celebrity, which will allow further selection of celebrities, based on these five attributes, for studies using Portuguese samples. Also, possible relationships between these five variables were analyzed and presented, highlighting facial distinctiveness as a predictor for both naming and recognition rate of celebrity faces.

Chagas disease is a public health problem in America. Its parasite, Trypanosoma cruzi, presents different discrete typing units (DTUs), colonizes organs of mammalian hosts in chronic infections, and presents tropism for particular organs in experimental infections. We evaluated T. cruzi tropism towards organs on the naturally infected rodent Octodon degus, identifying the parasites' DTUs, by means of conventional PCR and hybridization. Almost all the analyzed organs presented T. cruzi. More than 42% of the tested oesophagus, skin, skeletal muscle, brain and intestine showed T. cruzi DNA. Other nine types of organs were infected in over 15%. These results suggest that there is some tropism by T. cruzi in chronically infected O. degus. DTU TcV was present in 92.5% of infected organs with identified DTUs; this DTU is frequently reported in human infections in the Southern Cone of South America. Few organs showed mixed DTU infections. This is one of the few reports on the outcome of chronic natural T. cruzi-infection in wild mammal hosts exposed to naturally infected vectors.

The dysregulation of the inflammatory response, including pro-inflammatory molecules, produces neuropsychiatric symptoms and depression-like behavior, including withdrawal from the physical and social environment. Genetic variants that enhance immune reactivity may thus increase inflammatory and withdrawn reactions to stress. Here we investigated a functional polymorphism of Interferon Gamma gene (IFNG +874 T > A, rs2430561) as moderator of the relationship between mothers' distress exposure and children's withdrawn behavior at preschool age. Participants were 198 Portuguese preschool children (mean age = 57.98 months). Exposure to mother's distress was assessed using the Brief Symptom Inventory, and withdrawn behavior with the Caregiver Teacher Report Form. All children provided saliva samples for genotyping. Contrary to expecations based on prior work, the rs2430561 AA genotype-not the T variant-interacted with (high levels of) mothers' distress exposure, to increase children's withdrawn behavior. No significant main effects were detected. The polymorphism in Interferon Gamma gene showed specific environmental stressor-dependent effects on withdrawn behavior during childhood, ones which are interpreted in light of the "behavioral immune system" hypothesis, and which proved inconsistent with diathesis-stress thinking.

BACKGROUND: Serological screening for celiac disease (CD) allows the identification of individuals genetically predisposed, as type 1 diabetes mellitus (T1DM). However, the diagnosis is confirmed by intestinal biopsy. The aim was to determine the prevalence of immunoglobulin-A anti-tissue transglutaminase antibodies (IgA-tTG) and CD in a large cohort of young T1DM patients. METHODS: Screening for CD was randomly conducted in 881 T1DM by IgA-tTG and total IgA. Individuals with positive antibodies were referred to endoscopy/duodenal biopsy. RESULTS: The age of the cohort at the screening was 14.3 ± 5.9 years and at T1DM onset was 7.9 ± 4.4 years. The prevalence of positive serology was 7.7%. Median IgA-tTG levels were 117.7 U/mL (interquartile range [IQR] 35.7-131.5 U/mL). Of the 62 duodenal biopsy, CD was diagnosed in 79.0%, yielding an overall prevalence of 5.6%. The mean age of CD patients was 15.6 ± 6.5 years and, at T1DM onset was 6.3 years (4.0-9.9 years). The modified Marsh-Oberhuber histological classification was 22.5% (3a), 36.7% (3b), and 40.8% (3c). In the biopsy-proven patients, T1DM onset occurred at slightly younger ages (6.3 vs 9.7 years, P = 0.1947), gastrointestinal (GI) manifestations, predominantly abdominal pain and distension, were more prevalent (71.4% vs 38.5%, P = 0.027) and higher IgA-tTG titers (128.0 vs 26.3 U/mL, P = 0.0003) were found than in those with negative-biopsies. CONCLUSION: Our results demonstrate the prevalence of 7.7% of IgA-tTG and 5.6% of CD in T1DM patients in South Brazil and, emphasize the importance of the screening in high-risk individuals. Furthermore, the presence of GI manifestations and higher IgA-tTG titers strongly suggest the diagnosis of CD.

BACKGROUND: The elimination of malaria depends on the blocking of transmission and of an effective treatment. In Brazil, artemisinin therapy was introduced in 1991, and here we present a performance overview during implementation outset years. METHODS: It is a retrospective cohort (1991 to 2002) of patients treated in a tertiary centre of Manaus, with positive microscopic diagnosis of Plasmodium falciparum malaria, under treatment with using injectable or rectal artemisinin derivatives, and followed over 35-days to evaluate parasite clearance, death and recurrence. FINDINGS: This cohort outcome resulted 97.6% (1554/1593) of patients who completed the 35-day follow-up, 0.6% (10/1593) of death and 1.8% (29/1593) of follow-up loss. All patients that died and those that presented parasitaemia recurrence had pure P. falciparum infections and received monotherapy. Considering patients who completed 35-day treatment, 98.2% (1527/1554) presented asexual parasitaemia clearance until D4 and 1.8% (27/1554) between D5-D10. It is important to highlight that had no correlation between the five treatment schemes and the sexual parasite clearance. Finally, it is noteworthy that we were able to observe also gametocytes carriage during all follow-up (D0-D35). MAIN CONCLUSIONS: Artemisinin derivatives remained effective in the treatment of falciparum malaria during first 12-years of use in north area of Brazil.

The biological responses of acidobiontic species to extremely acidic waters, as those related with Acid Mine Drainage (AMD) processes, are relevant not only to understand the toxicological degree of the waters as well as the mechanisms responsible for the survival and adaptation of such organisms. Therefore, the objective of this study was to evaluate the effect of acidic pH and metals in biofilms collected in two sites a lentic permanent acid pond (AP) and a temporary acid stream (AS) affected by Acid Mine Drainage (AMD), and taking as control (C), an uncontaminated site with circumneutral pH. The results showed typical conditions of AMD-contaminated waters in sites AS and AP (pH < 3 and high concentrations of Al, Cu, Fe, Mn, Pb and Zn), which are responsible for a substantial decrease in biomass and biodiversity of biofilms (with a dominance of acidobiontic species, such as Pinnularia aljustrelica and Eunotia exigua, comparing to the control site. Also expressed by low values of photosynthetic pigments and reduced amount of proteins. In addition, such extreme acidic conditions also induced biofilms' stress, increasing antioxidant [catalase (CAT), superoxide dismutase (SOD) and biotransformation glutathione S-transferases (GSTs)] enzymes activity and also membrane damage (peroxidized lipids). However, despite the quite similar physico-chemical conditions of both AMD-contaminated sites, differences in the toxicity status between them were indirectly detected through some of the biochemical parameters [GSTs, SOD and CAT], indicating site AS has the most toxic, which fact was attributed to the higher concentrations of Pb2+ in this water. This element is recognized to be highly toxic for biota, and in such acidic conditions remains in solution, easily available to the aquatic organisms. Since pH is similar in the acid sites, the different concentrations of heavy metals in the waters might be responsible for the observed changes in the biological community at these two sites. This fact was translated in a higher capacity of this site's biofilm to cope with the negative effects of the toxicants, evidenced in a higher positive correlation with GSTs, CAT and SOD when compared to biofilms of sites AP and C.

PURPOSE: This study aimed to study autobiographical memories in women with eating disorders regarding emotional verbal expression, according to age. Our hypotheses are threefold: due to the emotional avoidance that occurs in women with eating disorders, in the younger ages, it was hypothesized that younger participants with anorexia and bulimia nervosa will present a lower number of emotional expressions in the descriptions of their memories than women without eating disorders; that older participants with anorexia and bulimia nervosa will present a greater number of negative verbal expressions in the reports of their memories than women without eating disorders, given the development of negative bias that occurs with age in women with eating disorders; and that women with eating disorders will use more words in a description of their sad memories than women without eating disorders because of the existence of negative bias. METHODS: With a sample of 90 women and combining age and the presence or absence of eating disorders, we formed four groups. The task that they had to perform was to recall a sad and a happy life event. RESULTS: The younger women used more words than the older women to describe their memories, and women with eating disorders used more words in their descriptions of the sad memories. However, there were no differences in terms of the number of positive and negative expressions. CONCLUSIONS: Women with eating disorders exhibit higher levels of cognitive reappraisal and thus use more positive expressions and fewer negative expressions. To remember to whom we transmit information, we rely on destination memory, with worse performance occurring when participants share personal facts (e.g., my age is . . .) compared with interesting ones (e.g., a shrimp's heart is in its head). When reporting personal information, the internal attentional focus decreases the attentional resources available to associate that information with recipients, resulting in worse destination memory. Given that the poorer destination memory when participants transmitted personal facts was always compared with the transmission of interesting facts, in Experiment 1 (between-participants design: 41 participants) and Experiment 2 (within-participants design: 30 participants), we compared the generation and transmission of personal facts with the transmission of familiar proverbs. Again, the generation and transmission of personal facts hampered destination memory. Besides the type of information (personal vs. familiar proverbs), the conditions differed regarding the type of process (generation vs. transmission of information). To clarify the influence of generation on destination memory, in Experiment 3 (N = 31), participants (1) transmitted and (2) generated and transmitted familiar proverbs, and significant differences in destination memory between the conditions was not observed. In general, our experiments seem to support the assumption that transmitting personal information leads to worse destination memory not because participants generated the information but because personal facts drive the attentional focus to the self.

INTRODUCTION: Obesity is a chronic and multifactorial disease, and the COVID-19 pandemic and lockdown have led to changes in the lifestyle habits of those patients. This study aimed to compare compliance with dietary and lifestyle measures, physical activity, mental health status, and motivation to lose weight during the pandemic in these patients. METHODS: An observational study was conducted, which included 63 patients over 18 years old who were followed in the outpatient setting through obesity medical appointments and who agreed to answer a questionnaire. RESULTS: We found that the majority of patients lost weight during the pandemic (61.7%), associating it with a 68% change in dietary habits. Regarding physical activity, there was a 34.9% reduction in its practice among those who used to exercise before the pandemic. Moreover, 52.4% felt more anxious and 44.4% felt sadder during the lockdown. DISCUSSION: The weight loss reported in our study seems to be related to a decrease in the consumption of carbohydrates and snacks and a lower percentage of those who relied on takeaways. Additionally, we hypothesize a greater number of home-cooked meals. Regarding exercise, the closure of gyms and the limitations imposed on daily life appear to have contributed unfavorably to this matter. Home confinement, loneliness, and a lack of social activities had harmful effects on the mental health of our sample. CONCLUSION: Overall, the hostile influence of the pandemic on mental well-being and exercise habits was evident. Nevertheless, contrary to our expectations, we observed weight loss during the lockdown.

Pathogen genome sequencing during epidemics enhances our ability to identify and understand suspected clusters and investigate their relationships. Here, we combine genomic and epidemiological data of the 2022 mpox outbreak to better understand early viral spread, diversification and transmission dynamics. By sequencing 52% of the confirmed cases in Portugal, we identified the mpox virus sublineages with the highest impact on case numbers and fitted them into a global context, finding evidence that several international sublineages probably emerged or spread early in Portugal. We estimated a 62% infection reporting rate and that 1.3% of the population of men who have sex with men in Portugal were infected. We infer the critical role played by sexual networks and superspreader gatherings, such as sauna attendance, in the dissemination of mpox virus. Overall, our findings highlight genomic epidemiology as a tool for the real-time monitoring and control of mpox epidemics, and can guide future vaccine policy in a highly susceptible population.

Thyroid eye disease (TED), an autoimmune inflammatory disorder of the orbit, presents with a potential array of clinical sequelae. The pathophysiology behind TED has been partially characterized in the literature. There remain certain elusive mechanisms welcoming of research advances. Disease presentation can vary, but those that follow a characteristic course start mild and increase in severity before plateauing into an inactive phase. Diagnosis and evaluation include careful physical examination, targeted laboratory work up, appropriate imaging studies, and tailored treatment regimens. Special consideration may apply to certain populations, such as pediatric and pregnant patients.

Acute ischemic stroke (AIS) remains a leading cause of mortality, despite significant advances in therapy (endovascular thrombectomy). Failure in developing novel effective therapies is associated with unsuccessful translation from preclinical studies to clinical practice, associated to inconsistent and highly variable infarct areas and lack of relevant post-stroke functional evaluation in preclinical research. To outreach these limitations, we optimized the intraluminal transient middle cerebral occlusion, a widely used mouse stroke model, in two key parameters, selection of appropriate occlusion filaments and time of occlusion, which show a significant variation in the literature. We demonstrate that commercially available filaments with short coating length (1-2 mm), together with 45-min occlusion, results in a consistent affected brain region, similar to what is observed in most patients with AIS. Importantly, a dedicated post-stroke care protocol, based on clinical practice applied to patients who had stroke, resulted in lower mortality and improved mice welfare. Finally, a battery of tests covering relevant fine motor skills, sensory functions, and learning/memory behaviors revealed a significant effect of tMCAO brain infarction, which is parallel to patient symptomatology as measured by relevant clinical scales (NIH Stroke Scale, NIHSS and modified Rankin Scale, mRS). Thus, in order to enhance translation to clinical practice, future preclinical stroke research must consider the methodology described in this study, which includes improved reproducible surgical procedure, postoperative care, and the battery of functional tests. This will be a major step s closing the gap from bench to bedside, rendering the development of novel effective therapeutic approaches.

Freshwater quality has been changing due to the ever greater use of water resources and the contamination load resulting from human activities. Management of these systems, thus, requires constant diagnose of water quality with fast and efficient methodologies. The conventional methods adopted are, however, time-consuming, often very expensive, and require specialised expertise. Raman spectroscopy (RS) is a simple, fast and label-free technique that can be applied to environmental diagnosis using diatoms. Here, we developed a diagnostic method based on Raman spectroscopy applied to freshwater diatoms. For this, Raman spectra were recorded from diatoms of three lakes of a natural city park. The data acquired was analysed by chemometrics methods to describe the data (Partial Least Squares Regression), infer relationships in the dataset (Cluster Analysis) and produce classification models (Artificial Neural Network). The classification models developed diagnosed the lakes with excellent accuracy (89%) without requiring taxonomic information about the diatom species recorded. This study provides a proof-of-concept for the application of diatom Raman spectroscopy to diagnosing water quality, laying an important foundation for future environmental studies aiming at assessing freshwater systems, to be replicated at larger scales and to varied geographic settings.

Considering the current state of the worldwide pandemic, it is still common to encounter people wearing face protection masks. Although a safety measure against COVID-19, face masks might be compromising our capacity for face recognition. We conducted an online study where 140 participants observed masked and unmasked faces in a within-subjects design and then performed a recognition memory task. The best performance was found when there were no masks either at study and test phase, i.e., at the congruent unmasked condition. The worst performance was found for faces encoded with a mask but tested without it (i.e., masked-unmasked incongruent condition), which can be explained by the disruption in holistic face processing and the violation of the encoding specificity principle. Interestingly, considering the unmasked-masked incongruent condition, performance was probably affected by the violation of the encoding specificity principle but protected by holistic processing that occurred during encoding.

Ischemic stroke is a leading cause of death worldwide, mainly in western countries. So far, approved therapies rely on reperfusion of the affected brain area, by intravenous thrombolysis or mechanical thrombectomy. The last approach constitutes a breakthrough in the field, by extending the therapeutic window to 16-24 h after stroke onset and reducing stroke mortality. The combination of pharmacological brain-protective strategies with reperfusion is the future of stroke therapy, aiming to reduce brain cell death and decrease patients' disabilities. Recently, a brain-protective drug-nerinetide-reduced brain infarct and stroke mortality, and improved patients' functional outcomes in clinical trials. The success of new therapies relies on bringing preclinical studies and clinical practice close together, by including a functional outcome assessment similar to clinical reality. In this review, we focused on recent upgrades of in vitro and in vivo stroke models for more accurate and effective evaluation of therapeutic strategies: from spheroids to organoids, in vitro models that include all brain cell types and allow high throughput drug screening, to advancements in in vivo preclinical mouse stroke models to mimic the clinical reality in surgical procedures, postsurgical care, and functional assessment.
