RESUMEN

A series of new studies reveal how the red/far-red light photoreceptors called phytochromes act. Phytochrome A and phytochrome B each move to the nucleus when activated by light, and phytochrome A is a kinase. Phytochrome-interacting proteins provide candidate signal transduction components and a recent physiological study suggests how phyA may mediate responses to far-red light. Regulation of phytochrome nuclear localization and kinase activities creates multiple phytochrome species, which may each have different regulatory activities.

RESUMEN

Auxin induces various distinct developmental responses, partly by regulating gene expression. The Aux/IAA genes are a large gene family, many of which are induced by auxin. Work on Arabidopsis Aux/IAA genes has begun to reveal that they can regulate development and auxin-induced gene expression. Furthermore, auxin responses require Aux/IAA protein turnover. Finally, recent evidence suggests that Aux/IAA proteins can mediate light responses. Work in the near future should test whether Aux/IAA proteins are antennae that connect auxin and light signals to endogenous developmental responses.

RESUMEN

Ambient light controls the development and physiology of plants. The Arabidopsis thaliana photoreceptor phytochrome B (PHYB) regulates developmental light responses at both seedling and adult stages. To identify genes that mediate control of development by light, we screened for suppressors of the long hypocotyl phenotype caused by a phyB mutation. Genetic analyses show that the shy (short hypocotyl) mutations we have isolated fall in several loci. Phenotypes of the mutants suggest that some of the genes identified have functions in control of light responses. Other loci specifically affect cell elongation or expansion.

Phytochrome, a red/far-red-light photoreceptor protein of plants, is encoded by a small gene family. Phytochrome A (PHYA), the product of the PHYA gene, is the predominant molecular species of phytochrome in etiolated tissue and has been best characterized biochemically. To define a role for PHYA, we isolated new mutants, designated fre1 (far-red elongated), in Arabidopsis thaliana that were specifically deficient in PHYA spectral activity and protein accumulation. These mutants were identified on the basis of their long hypocotyl phenotype under continuous far-red light. Although the fre1 mutants lacked the hypocotyl response to continuous far-red light, their responses to continuous white light and to end-of-day far-red-light treatments were normal. Thus, PHYA appears to play only a minor role in the regulation of hypocotyl elongation under natural conditions. In contrast, the fre1 mutation affected greening a fre1 mutant was less able than the wild type to deetiolate after growth in the dark. However, the potentiation effect of a red-light pulse on accumulation of chlorophyll was not changed significantly in the fre1 mutants. Thus, the function of PHYA might be highly specialized and restricted to certain phases of Arabidopsis development.

Plant responses to red and far-red light are mediated by a family of photoreceptors called phytochromes. In Arabidopsis thaliana, there are genes encoding at least five phytochromes, and it is of interest to learn if the different phytochromes have overlapping or distinct functions. To address this question for two of the phytochromes in Arabidopsis, we have compared light responses of the wild type with those of a phyA null mutant, a phyB null mutant, and a phyA phyB double mutant. We have found that both phyA and phyB mutants have a deficiency in germination, the phyA mutant in far-red light and the phyB mutant in the dark. Furthermore, the germination defect caused by the phyA mutation in far- red light could be suppressed by a phyB mutation, suggesting that phytochrome B (PHYB) can have an inhibitory as well as a stimulatory effect on germination. In red light, the phyA phyB double mutant, but neither single mutant, had poorly developed cotyledons, as well as reduced red-light induction of CAB gene expression and potentiation of chlorophyll induction. The phyA mutant was deficient in sensing a flowering response inductive photoperiod, suggesting that PHYA participates in sensing daylength. In contrast, the phyB mutant flowered earlier than the wild type (and the phyA mutant) under all photoperiods tested, but responded to an inductive photoperiod. Thus, PHYA and PHYB appear to have complementary functions in controlling germination, seedling development, and flowering. We discuss the implications of these results for possible mechanisms of PHYA and PHYB signal transduction.

BACKGROUND: Hypertension is a prevalent disease among African Americans, and successful treatment rates are low. Since calcium channel blockers are well-tolerated and efficacious in African Americans, we undertook this study to compare the efficacy, safety, and tolerability of 3 commonly prescribed calcium channel blockers: amlodipine besylate (Norvasc), nifedipine coat core (CC) (Adalat CC), and nifedipine gastrointestinal therapeutic system (GITS) (Procardia XL). METHODS: One hundred ninety-two hypertensive patients across 10 study centers were randomly assigned to double-blind monotherapy with amlodipine besylate (5 mg/d), nifedipine CC (30 mg/d), or nifedipine GITS (30 mg/d) for 8 weeks. Patients not achieving therapeutic response after 4 weeks had their dose doubled for the next 4 weeks. The primary end point was a comparison of the average reduction (week 8 minus baseline) in 24-hour ambulatory diastolic blood pressure (DBP). Secondary end points included a comparison of average 24-hour ambulatory systolic blood pressure (SBP), office SBP or DBP reduction, responder rates, safety, and tolerability. RESULTS: One hundred sixty-three patients were evaluable for efficacy after 8 weeks. There was no significant difference in the average 24-hour ambulatory DBP (-8.5, -9.0, and -6.1 mm Hg, respectively) or SBP (-14.3, -15.7, and -11.8 mm Hg, respectively) reduction. Average office SBP and DBP were reduced to a comparable degree (19-22 mm Hg [P =.50] and 12-14 mm Hg [P =.51], respectively). Responder rates (DBP <90 or reduced by > or = 10 mm Hg) were similar (P = .38). Discontinuation rates and adverse event frequency were distributed similarly across the 3 treatment groups. CONCLUSION: The efficacy, safety, and tolerability of the 3 dihydropyridine calcium channel blockers are equivalent in African Americans with stages 1 and 2 hypertension.

The 8-h double pulse oral glucose test is proposed as an alternative screening procedure for determining the adequacy of growth hormone (GH) release. The second pulse of glucose is timely in suppressing GH release and delaying it for a more predictable controlled elevation. Peak GH (mean plus or minus SD) values following the double pulse glucose test were 17 plus or minus 10.2, 16.5 plus or minus 2.2, and 1.3 plus or minus 0.5 ng/ml in normal controls, short stature patients and GH deficient patients, respectively. Peak GH values following insulin hypoglycemia were 36.9 plus or minus 13.8, 21.9 plus or minus 23.0 and 1.8 plus or minus 1.1 ng/ml in normal controls, short stature patients and GH-deficient patients. Peak GH values during a 5-h oral glucose tolerance test were 16.4 plus or minus 6.0 and 10.2 plus or minus 3.3 ng/ml in normal controls and short patients, respectively. These differences in peak GH values between the various clinical and control groups were not significantly different except for the greater GH peak values reached in short patients in the double pulse glucose test compared to the 5-h oral glucose tolerance test. The rise in GH following double pulse oral glucose is more timely predictab-e than after the 5-h oral glucose test and involves less professional time than the insulin tolerance test as it does not require close medical supervision.

RESUMEN

Twenty patients with benign thyroid cysts were studied in a prospective double-blind fashion to determine the effect of thyroid therapy on the recurrence of these cysts after aspiration. When the 10 patients receiving placebo medication were compared with the 10 patients ingesting thyroid hormone, no significant difference was found in the time either group was free of cyst recurrence. We conclude that thyroid therapy is not effective in preventing the recurrence of benign thyroid cysts after initial aspiration.

RESUMEN

Following radiotherapy for an extensive intraepithelial carcinoma of the conjunctiva and eyelid, a patient developed opacification of soft contact lenses used on the same side. Calcilm was demonstrated within the contact lenses by cytochemical methods and by energy-dispersive x-ray microanalysis. The phenomenon was associated with unilateral tear insufficiency and an elevated tear calcium level as determined by atomic absorption spectrophotometry.

RESUMEN

Rehabilitation can be readily carried out in an extended care facility. A technique is presented for accomplishing this objective by organizing existing personnel within the facility into a rehabilitation team which is goal-oriented. The Barthel Index serves as an objective measurement of the patient's progress. By use of this approach, significant numbers of patients with stroke, orthopedic disorders and other neuromusculoskeletal impairments can be returned to their previous living situations in a relatively short period, and at minimal cost.

RESUMEN

Vascularization was induced in rabbit corneas by the application of silver nitrate. Argon laser photocoagulation was applied to the entire length of these corneal vessels with relatively long pulses of energy. Untreated areas of vascularization were used as controls. There was a decrease in the corneal vascularization in the treated areas that exceeded any natural tendency toward remission. The results suggest the efficacy of inducing a remission of corneal vascularization with argon laser photocoagulation.

Currently used methods for calculating whole blood CO2 content from calculated plasma content, measured blood pH, hemoglobin concentration ([Hb]), and O2 saturation yield materially different results. In this study the constants of the fundamental equations relating blood CO2 content to plasma content have been reevaluated. An iterative computer technique was used to empirically derive appropriate constants from data obtained from nine healthy male subjects at rest and at several exercise work loads. A calculation was derived that fitted the data well [difference 0.02 +/- 1.19 ml/100 (SD) ml, r = 0.98] blood CCO2 = plasma CCO2 (Formula: see text) where plasma CCO2 = 2.226.s.plasma PCO2.(1 + 10pH-pK'), CCO2 is CO2 content, SO2 is O2 saturation, s is the plasma CO2 solubility coefficient, and pK' is the apparent pK [s and pK' are from the equations of Kelman (Respir. Physiol. 3: 111-115, 1967)].

Pulmonary function was measured before and after a 28-day saturation dive to a pressure of 0.25 MPa in eight subjects. PO2 was 40 kPa, with periods of 75 kPa for 2 h every 2nd day during the first 14 days, 50 kPa for the next 12 days, and a gradual fall to 21 kPa over the last 2 days in decompression. A 28-day saturation dive with six subjects to a pressure of 0.15 MPa and a PO2 of 21 kPa was used as control. The measurements included static and dynamic lung volumes and flows, transfer factor for carbon monoxide (TLCO), and a cycle ergometer exercise test. There was a significant reduction in TLCO of 9.8 +/- 6.0% (P < 0.001) after the dive when values were corrected for hemoglobin concentration changes. Effective alveolar volume was unchanged. There was a reduction in forced midexpiratory flow rate of 9.8 +/- 7.0% (P < 0.01), but forced vital capacity and forced expired volume in 1 s were unchanged. Peak oxygen uptake was reduced by 10.1 +/- 5.3% (P < 0.001). There were no significant changes in any of the lung function variables after the control dive. Exposure to raised PO2 contributes significantly to the changes in pulmonary function that have been reported after deep saturation dives to pressures of 3.1-4.6 MPa with a similar profile of oxygen exposure. TLCO is apparently a more sensitive index than vital capacity for oxygen toxicity.

RESUMEN

Nine males with varying degrees of subfertility with left varicocele underwent bilateral testicular biopsy for determination of testicular testosterone (T) and dihydrotestosterone (DHT) content. Testicular tissue T and DHT levels varied greatly, and there was no significant difference when the group T and DHT content of the left testis was compared with that of the right testis. When the respective mean preoperative sperm count was compared with the bilateral tissue T and DHT content, no consistent relationship was found.

We reviewed retrospectively the records of 12 patients with herpes zoster keratopathy who had undergone penetrating keratoplasty. Preoperatively, seven patients (58%) had noninflamed eyes with visually significant corneal scarring or edema. Five patients (42%) had progressive neurotrophic corneal ulceration, and four of those had corneal perforation. Tarsorrhaphies were placed in ten patients and appeared to be beneficial in preventing postoperative breakdown of the corneal surface. At an average follow-up time of 36 months, ten of the 12 grafts (83%) remained clear, with nine patients (75%) having a visual acuity of 20/80 or better.

A 46-year old woman and a 59-year-old man had corneal crystals, multiple myeloma, and IgG kappa hypergammaglobulinemia. In one case, crystalline deposits were also present in the lens. In both patients there was a marked decrease in the amount of crystals during chemotherapy. The crystals within the cornea of one case were identified by light and electron microscopy in material obtained during a lamellar keratoplasty. The crystalline deposits were located only in the corneal epithelium, and their regular repeating internal arrangement was confirmed by monochromatic optical diffraction of electron micrographs of sections through them. By immunofluorescent and immunoperoxidase techniques, the crystals reacted positively for immunoglobulin and particularly IgG kappa chains.

Solutions of the Ni(IV) complex of the dianion of 2,6-diacetylpyridine dioxime (chelate II in text) are reduced very slowly by 2-aminoethanethiol at pH 2.3-3.0, but this reaction is catalyzed dramatically and specifically by dissolved copper, with Cu(I) the active reductant. When the [thiol]/[Ni(IV)] ratio exceeds 1.6, each Ni(IV) oxidizes two molecules of thiol, forming Ni(II) and R(2)S(2). At low concentrations of catalyst and reductant, reaction profiles are almost exponential, but at higher concentrations of either, curves become progressively more nearly linear. Reactions are sharply retarded by increases in acidity. Profiles for 14 runs, carried out with [H(+)] = 0.001-0.0040 M, [Ni(IV)] = (0.94-1.2) x 10(-)(5) M, [thiol] = (2.0-32) x 10(-)(4) M, and [Cu(2+)] = (2.5-80) x 10(-)(6)M, are consistent with a reaction sequence (eqs 2-10 in text) in which Cu(I) is generated in competing homolyses of the complexes Cu(II)(SRH) and Cu(II)(SRH)(2). Reduction of Ni(IV) appears then to proceed through a Ni(IV)Cu(I) adduct, which can undergo electron transfer (yielding Ni(III) and Cu(II)), either in a unimolecular fashion or, alternatively, as a result of attack by a second Cu(I) species. The Ni(IV)Cu(I) + Cu(I) process is reflected in approach to second-order dependences on [Cu(II)] and [thiol] (which generate Cu(I)) at high concentrations of these reagents. Reductions of the Ni(III) intermediate are taken to be much more rapid than those of Ni(IV). Kinetic trends in the present system stand in contrast to the more familiar catalytic patterns such as those seen when the same combination of thiol and catalyst is used to reduce superoxo complexes of cobalt(III). With the latter reactions, decay profiles for the oxidant tend to be exponential at high reagent concentrations but approach linearity at low.

RESUMEN

The traumatic loss of iris is often associated with symptoms of glare, which can be debilitating. This symptom may be benefitted by wearing a contact lens with a clear, central optical zone and opacification of the remainder. The patient who is contact lens-intolerant may be helped by tattooing of all of the cornea except the central visual axis. Four patients who had traumatic iris loss and who were treated by this technique reported reduced glare as well as improvement in the cosmetic appearance of the eye. Although corneal tattooing is an ancient procedure, it continues to be of benefit for selected patients.
