RESUMEN

Mammary gland quarters have usually been considered to be anatomically and physiologically independent, but some recent research has indicated more interdependence than previously reported. The objective of this study was to compare milk composition (fat, total protein, lactose, solids-not-fat, and chloride) and health status (somatic cell count, differential leukocyte count, and lactate dehydrogenase) of milk samples from unaffected mammary glands of an udder with a single clinically inflamed quarter to results of milk samples from healthy mammary glands of healthy cows. The study was designed as a prospective case control study with case and control cows matched by parity and days in milk. Cases were defined as cows (n = 59) experiencing clinical mastitis in a single mammary gland, and controls (n = 59) were defined as cows that had not experienced clinical mastitis during the current lactation. Quarter milk samples were collected from all mammary glands adjacent to clinically affected quarters of cases and from the same mammary glands of controls. Samples were used to assess concentration of chloride and lactate dehydrogenase, fat, total protein, solids-not-fat, somatic cell count, and differential leukocyte count. Microbiological analysis was also performed on milk samples obtained from clinically affected mammary glands (n = 59). Logistic regression models were used to assess possible associations among quarter somatic cell count (≥150,000 cells/mL) and quarter type (adjacent to case or control). Multivariate linear models were used to compare milk composition and health status between quarter types. A total of 170 quarters were enrolled per group. Milk obtained from adjacent quarters of cases contained a lesser concentration of total protein, lactose, and solids-not-fat, but had a greater concentration of fat and chloride. The somatic cell count, total leukocyte count, and absolute numbers of neutrophils, lymphocytes, and macrophages were all increased in milk obtained from adjacent quarters of case cows compared with milk obtained from quarters of control cows. The relative proportion of neutrophils was increased, whereas the proportion of macrophages was decreased in milk obtained from cases. Approximately 30% of milk samples obtained from adjacent quarters of cases had a somatic cell count ≥150,000 cells/mL compared with 12% of milk samples obtained from quarters of control cows. The position of the mammary gland was not associated with any outcomes. In conclusion, our results support previous research that indicates the immune response to intramammary infection in a single mammary gland quarter alters milk composition and health status throughout the udder.

BACKGROUND: The objective of this study was to identify associations between the concentration of Mycobacterium avium subsp. paratuberculosis (MAP) antibodies in bulk milk and potential risk factors in herd management and herd characteristics, explaining high MAP antibody titers in milk. An extensive questionnaire was administered to 292 organic and conventional dairy farms from New York, Wisconsin and Oregon. Bulk milk samples were taken from each farm for MAP enzyme-linked immunosorbent assay (ELISA). A general linear model was constructed with MAP ELISA value as the outcome variable and the management factors and herd characteristics as independent variables, while at the same time controlling for the study design variables of state, herd size, and production system (organic or conventional). High bulk tank MAP ELISA value may be due to either a high prevalence of MAP in a herd with many cows contributing to the antibody titer or due to a few infected cows that produce large quantities of antibodies. RESULTS: Results of the regression models indicated that bulk milk ELISA value was associated with season of sampling and the presence or absence of protocols for managing MAP-positive cows. The concentration of MAP antibodies in bulk milk varied seasonally with a peak in the summer and low concentrations in the winter months. When compared to farms that had never observed clinical Johne's disease, keeping MAP-positive cows or only culling them after a period of delay was associated with an increase in optical density. CONCLUSIONS: The seasonal variation in MAP antibody titers, with a peak in the summer, may be due to a seasonal increase in MAP-bacterial load. Additionally, seasonal calving practices may contribute to seasonal fluctuations in MAP antibody titers in bulk tank milk. Keeping MAP-positive cows increases the antibody titer in bulk milk, likely due to direct antibody production in the infected cow and indirect triggering of antibody production in herdmates.

OBJECTIVE: To identify management factors associated with veterinary usage by organic and conventional dairy farms. DESIGN: Prospective case-control study. SAMPLE: 292 farms. PROCEDURES: Organic farms in New York, Oregon, and Wisconsin were matched to conventional farms on the basis of location and herd size. During a single herd visit, a questionnaire was administered, information about animal disease incidence and number of veterinarian visits in the preceding 60 days was collected, and forms to record similar information during the 60 days after the visit were left for the herd manager to complete. For analysis, conventional herds were classified as either grazing or nongrazing. Multiple correspondence analysis was used to assess relationships among management factors and selected outcomes for frequency of veterinary usage. RESULTS: Intensive management practices were closely associated with frequent veterinary usage. Generally, organic management practices were associated with less frequent veterinary usage than were conventional management practices. Conventional grazing practices were associated with intermediate veterinary usage (more than organic practices but less than intensive practices), whereas conventional nongrazing practices were associated with frequent veterinary usage. Cost of routinely scheduled veterinarian visits/45 kg (100 lb) of milk produced/y was greater for small farms than that for large farms. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that management intensiveness was more closely associated with frequency of veterinary usage than was organic status; therefore, veterinarians should characterize farms by factors other than organic status when investigating which farms are most likely to use their services. Economic factors substantially affected routine veterinary usage on small farms.
