RESUMEN

Melanoidin is the hazardous byproduct formed during the production of ethanol in distilleries. In the present study, a highly effective melanoidin decolorizing bacterial isolate, SAG1, was isolated from the effluent enriched soil of a distillery. This strain, identified as Paracoccus pantotrophus, was highly efficient to decolorize melanoidins up to 81.2 ± 2.43% in the presence of glucose and NH4NO3. The effects of autoclaved as well as living cells and inoculums size on decolorization activity were investigated. The results indicated that only living cell showed the decolorization activity i.e. 78.6 ± 2.62%, while, no activity has been observed using autoclaved cells. The inoculums size of 8% v/v, showed maximum activity of 62.9 ± 3.00%. The isolate SAG1 was found to be more efficient in decolorizing the melanoidins from distillery effluent as compared to the reference culture Pseudomonas putida.

A probiotic strain of lactobacilli was isolated from traditional soft Churpi cheese of Yak milk and found positive for biosurfactant production. Lactobacilli reduced the surface tension of phosphate buffer saline (PBS) from 72.0 to 39.5 mNm(-1) pH 7.2 and its critical micelle concentration (CMC) was found to be 2.5 mg mL(-1). Low cost production of Lactobacilli derived biosurfactant was carried out at lab scale fermenter which yields 0.8 mg mL(-1) biosurfactant. The biosurfactant was found least phytotoxic and cytotoxic as compared to the rhamnolipid and sodium dodecyl sulphate (SDS) at different concentration. Structural attributes of biosurfactant were determined by FTIR, NMR ((1)H and (13)C), UPLC-MS, and fatty acid analysis by GCMS which confirmed the presence of glycolipid type of biosurfactant closely similar to xylolipids. Biosurfactant is mainly constituted by lipid and sugar fractions. The present study outcomes provide valuable information on structural characterization of the biosurfactant produced by L. helveticus MRTL91. These findings are encouraging for the application of Lactobacilli derived biosurfactant as nontoxic surface active agents in the emerging field of biomedical applications.

Paddy straw is a versatile and valuable resource with multifaceted benefits for nutrient cycling, soil health, and climate mitigation. Its role as a rich nutrient source and organic matter significantly enhances soil vitality while improving soil structure and moisture retention. The impact of paddy straw extends beyond traditional agricultural benefits, encompassing the promotion of microbial activity, erosion control, and carbon sequestration, highlighting its crucial role in maintaining ecological balance. Furthermore, the potential of paddy straw in bioenergy is explored, encompassing its conversion into biogas, biofuels, and thermal energy. The inherent characteristics of paddy straw, including its high cellulose, hemicellulose, and lignin content, position it as a viable candidate for bioenergy production through innovative processes like pyrolysis, gasification, anaerobic digestion, and combustion. Recent research has uncovered state-of-the-art techniques and innovative technologies capable of converting paddy straw into valuable products, including sugar, ethanol, paper, and fiber, broadening its potential applications. This paper aims to underscore the possibilities for value creation through paddy straw, emphasizing its potential use in bioenergy, bio-products, and other environmental applications. Therefore, by recognizing and harnessing the value of paddy straw, we can advocate for sustainable farming practices, reduce waste, and pave the way for a resource-efficient circular economy. Incorporating paddy straw utilization into agricultural systems can pave the way for enhanced resource efficiency and a more sustainable circular economy.

The current study emphasizes fungi as an important tool against heavy metals and how isolated fungal species can be used to create a successful strategy for the bioremediation of chromium and arsenic-contaminated sites/soils. Globally, heavy metal pollution is a serious issue. In the current investigation, contaminated sites were chosen, and samples could be taken from various localities of Hisar (29.1492° N, 75.7217° E) and Panipat (29.3909° N, 76.9635° E), India. A total of 19 fungal isolates were obtained from the collected samples through the enrichment culture technique using PDA media supplemented with Cr as chromic chloride hexahydrate (50 mg/L) and As as sodium arsenate (10 mg/L) and the potential of fungal isolates to be used for the removal of heavy metals was examined. The isolates were screened for minimum inhibitory concentrations (MIC) exhibiting tolerance capabilities, and the four best isolates C1, C3, A2, and A6 with the highest MICs (>5000 mg/L), were chosen for further investigations. To use the chosen isolates in the remediation of heavy metals (Cr and As), the culture conditions were optimized. The fungal isolates C1 and C3 estimated the highest removal of 58.60% and 57.00% at 50 mg/L chromium concentration, while the isolates A6 and A2 recorded the highest removal efficiency of 80% and 56% at 10 mg/L arsenic concentration under optimal conditions. Finally, the chosen fungal isolates C1 and A6 were molecularly identified as Aspergillus tamarii and Aspergillus ustus, respectively.

In the age of industrialization, numerous non-biodegradable pollutants like plastics, HMs, polychlorinated biphenyls, and various agrochemicals are a serious concern. These harmful toxic compounds pose a serious threat to food security because they enter the food chain through agricultural land and water. Physical and chemical techniques are used to remove HMs from contaminated soil. Microbial-metal interaction, a novel but underutilized strategy, might be used to lessen the stress caused by metals on plants. For reclaiming areas with high levels of heavy metal contamination, bioremediation is effective and environmentally friendly. In this study, the mechanism of action of endophytic bacteria that promote plant growth and survival in polluted soils-known as heavy metal-tolerant plant growth-promoting (HMT-PGP) microorganisms-and their function in the control of plant metal stress are examined. Numerous bacterial species, such as Arthrobacter, Bacillus, Burkholderia, Pseudomonas, and Stenotrophomonas, as well as a few fungi, such as Mucor, Talaromyces, Trichoderma, and Archaea, such as Natrialba and Haloferax, have also been identified as potent bioresources for biological clean-up. In this study, we additionally emphasize the role of plant growth-promoting bacteria (PGPB) in supporting the economical and environmentally friendly bioremediation of heavy hazardous metals. This study also emphasizes future potential and constraints, integrated metabolomics approaches, and the use of nanoparticles in microbial bioremediation for HMs.

Chemical fungicides are the backbone of modern agriculture, but an alternative formulation is necessary for sustainable crop production to address human health issues and soil/water environmental pollution. So, a green chemistry approach was used to form guar gum nanoemulsions (NEs) of 186.5-394.1 nm containing the chemical fungicide mancozeb and was characterized using various physio-chemical techniques. An 84.5% inhibition was shown by 1.5 mg/mL mancozeb-loaded NEs (GG-1.5) against A. alternata, comparable to commercial mancozeb (86.5 ± 0.7%). The highest mycelial inhibition was exhibited against S. lycopersici and S. sclerotiorum. In tomatoes and potatoes, NEs showed superior antifungal efficacy in pot conditions besides plant growth parameters (germination percentage, root/shoot ratio and dry biomass). About 98% of the commercial mancozeb was released in just two h, while only about 43% of mancozeb was released from nanoemulsions (0.5, 1.0 and 1.5) for the same time. The most significant results for cell viability were seen at 1.0 mg/mL concentration of treatment, where wide gaps in cell viability were observed for commercial mancozeb (21.67%) and NEs treatments (63.83-71.88%). Thus, this study may help to combat the soil and water pollution menace of harmful chemical pesticides besides protecting vegetable crops.

Biofouling roots damage to membrane bioreactors (MBRs), such as physical, functional and organisational changes and even therefore clogging of the membrane pores and successive microbial degradation. Further, it blocks the pores, results into a biomass cake and in due course reduces the membrane flux and leads to an increase in the operational costs. MBR fouling contributed to the rise in transmembrane pressure (TMP) and decrease in permeate flux (in case of constant pressure operation mode). Chemical surfactants adopted for the cleaning of membrane surfaces have certain disadvantages such as toxicity manifestations, damage to the membranes and high CMC concentrations. Biosurfactant surfactants have attained increasing interest due to their low toxicity, biodegradability, stability to extreme environmental conditions such as temperatures, pH and tolerance to salinity. The biosurfactants trapped the foulants via micelle formation, which distresses hydrophobic interactions amongst bacteria and the surface. Rhamnolipids as an anionic biosurfactant pose a significant interfacial potential and have affinity to bind organic matter. The present review discusses the problem of biofouling in MBRs, type and interactions of foulants involved and also highlights the mechanisms of biosurfactant cleaning, effect of different parameters, effect of concentration, TMP, flux recovery, permeability, mitigation practices and challenges.

Pesticides are essential to contemporary agriculture and are required to safeguard plants from hazardous pests, diseases, and weeds. In addition to harming the environment, overusing these pesticides causes pests to become resistant over time. Alternative methods and agrochemicals are therefore required to combat resistance. A potential solution to pesticide resistance and other issues may be found in nanotechnology. Due to their small size, high surface-area-to-volume ratio, and ability to offer novel crop protection techniques, nanoformulations, primarily biopolymer-based ones, can address specific agricultural concerns. Several biopolymers can be employed to load pesticides, including starch, cellulose, chitosan, pectin, agar, and alginate. Other biopolymeric nanomaterials can load pesticides for targeted delivery, including gums, carrageenan, galactomannans, and tamarind seed polysaccharide (TSP). Aside from presenting other benefits, such as reduced toxicity, increased stability/shelf life, and improved pesticide solubility, biopolymeric systems are also cost-effective; readily available; biocompatible; biodegradable; and biosafe (i.e., releasing associated active compounds gradually, without endangering the environment) and have a low carbon footprint. Additionally, biopolymeric nanoformulations support plant growth while improving soil aeration and microbial activity, which may favor the environment. The present review provides a thorough analysis of the toxicity and release behavior of biopolymeric nanopesticides for targeted delivery in precision crop protection.

Agriculture production faces many abiotic stresses, mainly drought, salinity, low and high temperature. These abiotic stresses inhibit plants' genetic potential, which is the cause of huge reduction in crop productivity, decrease potent yields for important crop plants by more than 50% and imbalance agriculture's sustainability. They lead to changes in the physio-morphological, molecular, and biochemical nature of the plants and change plants' regular metabolism, which makes them a leading cause of losses in crop productivity. These changes in plant systems also help to mitigate abiotic stress conditions. To initiate the signal during stress conditions, sensor molecules of the plant perceive the stress signal from the outside and commence a signaling cascade to send a message and stimulate nuclear transcription factors to provoke specific gene expression. To mitigate the abiotic stress, plants contain several methods of avoidance, adaption, and acclimation. In addition to these, to manage stress conditions, plants possess several tolerance mechanisms which involve ion transporters, osmoprotectants, proteins, and other factors associated with transcriptional control, and signaling cascades are stimulated to offset abiotic stress-associated biochemical and molecular changes. Plant growth and survival depends on the ability to respond to the stress stimulus, produce the signal, and start suitable biochemical and physiological changes. Various important factors, such as the biochemical, physiological, and molecular mechanisms of plants, including the use of microbiomes and nanotechnology to combat abiotic stresses, are highlighted in this article.

BACKGROUND: Acyl hydrazones are an important class of heterocyclic compounds promising pharmacological characteristics. Malaria is a life-threatening mosquito-borne blood disease caused by a plasmodium parasite. In some places, malaria can be treated and controlled with early diagnosis. However, some countries lack the resources to do this effectively. RESULTS: The present work involves the design and synthesis of some novel acyl hydrazone based molecular hybrids of 1,4-dihydropyridine and pyrazole (5a-g). These molecular hybrids were synthesised by condensation of 1,4-dihydropyridin-4-yl-phenoxyacetohydrazides with differently substituted pyrazole carbaldehyde. The final compound (5) showed two conformations (the major, E, s-cis and the minor, E, s-trans) as revealed by NMR spectral data and further supported by the energy calculations (MOPAC2016 using PM7 method). All the synthesised compounds were screened for their in vitro antimalarial activities against chloroquine-sensitive malaria parasite Plasmodium falciparum (3D7) and antimicrobial activity against Gram positive bacteria i.e. Bacillus cereus, Gram negative bacteria i.e. Escherichia coli and antifungal activity against one fungus i.e. Aspergillus niger [corrected]. All these compounds were found more potent than chloroquine and clotrimazole, the standard drugs. CONCLUSIONS: In vitro antiplasmodial IC50 value of the most potent compound 5d was found to be 4.40 nM which is even less than all the three reference drugs chloroquine (18.7 nM), pyrimethamine (11 nM) and artimisinin (6 nM). In silico binding study of compound 5d with plasmodial cysteine protease falcipain-2 indicated the inhibition of falcipain-2 as the probable reason for the antimalarial potency of compound 5d. All the compounds had shown good to excellent antimicrobial and antifungal activities.

Various lactic acid bacteria (LAB) have been isolated and screened for biosurfactant production and their biomedical and food applications. Additionally, various different concentrations of the biosurfactant (0.625-25 mg ml-1) were used to evaluate its antimicrobial and antiadhesive potential against a range of pathogenic microorganisms. Biosurfactant was found to be stable to pH changes over a range of 4.0-12.0, being most effective at pH 7 and showed no apparent loss of surface tension and emulsification efficiency after heat treatment at 125 °C for 15 min. Present study demonstrated that biosurfactant obtained from Lactobacillus helveticus has the ability to counteract effectively the initial deposition of biofilm forming pathogens to silicone surfaces and to significantly sluggish biofilm growth.

The present investigation was undertaken to isolate, screen and evaluate a selected promising PGPR Brevibacillus brevis on cotton crop. Out of 156 bacterial isolates one of the most promising isolate was analyzed for the various PGP traits. A seed germination analysis was conducted with cotton seeds to evaluate the potential of the isolate to promote plant growth. The bacterial isolate was checked for its growth and survival at high temperatures. The isolate was also analyzed for the PGP traits exhibited after the heat treatment. To identify the isolate morphological, biochemical and molecular characterization was performed. The isolate was found positive for many of the PGP attributes like IAA, ARA, anti-fungal activity and ammonia production. Effect of seed bacterization on various plant growth parameters was used as an indicator. The isolate showed significant growth and exhibited various PGP traits at high temperature making it suitable as an inoculant for cotton crop. Isolate was identified as Brevibacillus brevis [SVC(II)14] based on phenotypic as well as genotypic attributes and after conducting this research we propose that the B. brevis which is reported for the first time for its PGP potential in cotton, exerts its beneficial effects on cotton crop through combined modes of actions.

The objective of the present study was to isolate the biosurfactant (BS) producing lactic acid bacteria (LAB) from traditional fermented food (buttermilk) and its functional and structural characterization. BS isolated from strain MRTL9 reduced surface tension from 72.0 to 40.2 mN m(-1). The critical micelle concentration (CMC) of BS was 2.25 mg ml(-1) with emulsification efficiency (E24) after 24 h of 64% against kerosene oil. The cell bound BS was partially purified by silica gel column chromatography and found as glycolipid. The gas chromatography and mass spectroscopy data revealed the fatty acid as hexadecanoic acid. Xylose was determined as hydrophilic moiety. The BS was found to be stable to pH changes over a range of 4.0-12.0, being most effective at pH 7 and showed no apparent loss of surface tension and emulsification efficiency after heat treatment at 120°C for 15 min. The outcomes of cellular toxicity showed lower toxicity of BS in comparison to SDS and rhamnolipids. Current study confirmed the preventive anti-adhesion activity of BS. These amphiphilic molecules, interferes with the microbial adhesion and found to be least cytotoxic with cellular compatibility with mouse fibroblasts cells.

Distillery effluent retains very dark brown color even after anaerobic treatment due to presence of various water soluble, recalcitrant and coloring compounds mainly melanoidins. In laboratory conditions, melanoidin decolorizing bacteria was isolated and optimized the cultural conditions at various incubation temperatures, pH, carbon sources, nitrogen sources and combined effect of both carbon and nitrogen sources. The optimum decolorization (72.6 ± 0.56%) of melanoidins was achieved at pH 7.5 and temperature 37 °C on 5th day of cultivation. The toxicity evaluation with mung bean (Vigna radiata) revealed that the raw distillery effluent was environmentally highly toxic as compared to biologically treated distillery effluent, which indicated that the effluent after bacterial treatment is environmentally safe. This proves to be novel biological treatment technique for biodegradation and detoxification of melanoidin from distillery effluent using the bacterial strain SAG(5).
