The aim of this study was to determine the ergogenic effects of metformin in high-intensity exercise, as well as its effects on anaerobic capacity, in healthy and physically active men. Ten subjects (mean (± standard deviation) maximal oxygen uptake (VËO2max ) 38.6 ± 4.5 mL/kg per min) performed the following tests in a cycle ergometer: (i) an incremental test; (ii) six submaximal constant workload tests at 40%-90% (VËO2max ); and (iii) two supramaximal tests (110% (VËO2max ). Metformin (500 mg) or placebo was ingested 60 min before the supramaximal test. There were no significant differences between the placebo and metformin groups in terms of maximum accumulated oxygen deficit (2.8 ± 0.6 vs 3.0 ± 0.8 L, respectively; P = 0.08), lactate concentrations (7.8 ± 2.6 vs 7.5 ± 3.0 mmol/L, respectively; P = 0.75) or O2 consumed in either the last 30 s of exercise (40.4 ± 4.4 vs 39.9 ± 4.0 mL/kg per min, respectively; P = 0.35) or the first 110 s of exercise (29.0 ± 2.5 vs 29.5 ± 3.0 mL/kg per min, respectively; P = 0.42). Time to exhaustion was significantly higher after metformin than placebo ingestion (191 ± 33 vs 167 ± 32 s, respectively; P = 0.001). The fast component of VËO2 recovery was higher in the metformin than placebo group (12.71 vs 12.18 mL/kg per min, respectively; P = 0.025). Metformin improved performance and anaerobic alactic contribution during high-intensity exercise, but had no effect on overall anaerobic capacity in healthy subjects.

The objectives of this study were study a practical method to characterize bovine spermatogenic cells and test the efficiency cells conservation by refrigeration at 4°C and cryopreservation in different solutions using two cooling curves. Cellular identification was performing by analysis of shape, size and morphology, associated with nucleus positioning and nuclear-cytoplasm ratio (NCR). Cellular samples were kept at 4°C for a period of 96 h in refrigeration solution and every 24h plasma membrane and DNA integrity were evaluated. Cryopreservation of cells was carried out using solutions containing 10% Dimethyl sulfoxide, 5% Dimethylformamide, 7% Glycerol and 7% Ethylene glycol, using a controlled and non-controlled cooling curve. Results of cellular characterization demonstrated that spermatocytes II presented a cylindrical shape, NCR of 1:1.5 and diameter ranging from 14.5 to 17.5 µm. Round spermatids presented diameter ranging from 7.6 to 13.4 µm, acrosomal cap and NCR of 1:2. Elongation and elongated spermatids showed to marked divergence in shape. There was a daily significant loss of viability of cooled cells until third day of storage, however they presented 72.77±5.16% viability after 4 days of storage at 4°C. There was no difference among the cryoprotectant solutions and cooling curves. In conclusion we demonstrated that association of microscopes and staining was a practical method to identify bovine spermatogenic cells. Furthermore, refrigeration at 4°C is an important strategy to preserve over 70% of viable cells after 4 days and cryopreservation, regardless of cryoprotectant solution or cooling curve used, can maintain over 50% of cells viable.

Polymorphisms in genes encoding P450 cytochrome enzymes may increase the risk of sporadic colorectal cancer (SCRC). Here we investigated the association between SCRC and CYP2E1 (PstI) and CYP1A1 (MspI) polymorphisms in a case-control study. Moreover, we sought to determine any possible associations between this disease and the sociodemographic factors. We included 273 individuals (74 patients and 199 controls); the gender, age, tobacco usage, and alcohol consumption of the included subjects, and the clinico-histopathological parameters of the tumors, were analyzed. Molecular analyses were performed using PCR-RFLP. The effect of polymorphisms on SCRC development, and the association between this disease and sociodemographic factors were determined by multiple-logistic regression analyses. The combined genotype was also evaluated. Statistically significant differences between the patients and controls regarding the male gender (odds ratio, OR = 0.19, 95% confidence interval, CI = 0.08-0.46; P ≤ 0.05) and age ≥44 years (median = 44; OR = 96.84, 95%CI = 21.78-430.49; P ≤ 0.05) were observed. The evaluated polymorphisms were not associated with SCRC (PstI-CYP2E1: OR = 0.93, 95%CI = 0.30-2.85; P = 0.897; MspI-CYP1A1: OR = 0.75, 95%CI = 0.35-1.61; P = 0.463); the combined genotypes were not associated with the risk of disease. Thus, individuals aged ≥44 years are more sensitive to SCRC, while men are less susceptible. Additionally, polymorphisms in CYP2E1 (PstI) and CYP1A1 (MspI) were not associated with SCRC in the evaluated Brazilian population.

We investigated the karyotypes of 13 species of six sections of the genus Inga (Leguminosae-Mimosoideae) from Brazil. We used conventional Giemsa staining to identify numerical chromosomal variations and looked for karyotypic evolutionary patterns. The karyotypes generally had small chromosomes, varying from metacentric to submetacentric, with a basic number x=13. Nine of the species showed 2n=2x=26 (I. thibaudiana, I. cayennensis, I. ingoides, I. edulis, I. vera, I. subnuda, I. striata, I. bollandii, and Inga sp), while 2n=4x=52 was seen in a population of Inga cylindrical and of I. capitata, and in five populations of I. laurina. Additionally, 2n=8x=104 was observed in a population of I. cayennensis. Eight of these counts were new, while the counts of 2n=52 for I. laurina and 2n=26 for I. marginata, I. vera, I. subnuda, and I. edulis confirmed previous studies. We did not find cytological stability among the sections studied, with occurrence of significant intra- and inter-specific numerical variations. We conclude that polyploidy has played a significant role in karyotypic evolution in this group and that it occurred independently in several sections of the genus.

The ability to perceive polarization-related entoptic phenomena arises from the dichroism of macular pigments held in Henle's fiber layer of the retina and can be inhibited by retinal diseases, such as age-related macular degeneration, which alters the structure of the macula. Structured light tools enable the direct probing of macular pigment density and retinal structure through the perception of polarization-dependent entoptic patterns. Here, we directly measure the visual angle of an entoptic pattern created through the illumination of the retina with a structured state of light and a perception task that is insensitive to corneal birefringence. The central region of the structured light stimuli was obstructed, with the size of the obstruction varying according to a psychophysical staircase. Two stimuli, one producing 11 azimuthal fringes and the other three azimuthal fringes, were presented to 24 healthy participants. The pattern with 11 azimuthal fringes produced an average visual angle threshold of 10° ± 1° and a 95% confidence interval (C.I.) of [6°, 14°]. For the pattern with three azimuthal fringes, a threshold extent of 3.6° ± 0.3° C.I. = [1.3°, 5.8°] was measured, a value similar to the published extent of Haidinger's brush (4°). The increase in apparent size and clarity of entoptic phenomena produced by the presented structured light stimuli offers the potential to detect the early signs of macular disease over perception tasks using uniform polarization stimuli.
RESUMEN
This review provides the current state of knowledge regarding the use of nutritional nanocompounds on exercise performance. The reviewed studies used the following nanocompounds: resveratrol-loaded lipid nanoparticles, folic acid into layered hydroxide nanoparticle, redox-active nanoparticles with nitroxide radicals, and iron into liposomes. Most of these nutritional nanocompounds seem to improve performance in endurance exercise compared to the active compound in the non-nanoencapsulated form and/or placebo. Nutritional nanocompounds also induced the following physiological and metabolic alterations: 1) improved antioxidant activity and reduced oxidative stress; 2) reduction in inflammation status; 3) maintenance of muscle integrity; 4) improvement in mitochondrial function and quality; 5) enhanced glucose levels during exercise; 6) higher muscle and hepatic glycogen levels; and 7) increased serum and liver iron content. However, all the reviewed studies were conducted in animals (mice and rats). In conclusion, nutritional nanocompounds are a promising approach to improving exercise performance. As the studies using nutritional nanocompounds were all conducted in animals, further studies in humans are necessary to better understand the application of nutritional nanocompounds in sport and exercise science.

The present study investigated the reliability and sensitivity of a wearable near-infrared spectroscopy (wNIRS) device in moderate and heavy exercise intensity domains. On three separate days, eleven males performed an incremental test to exhaustion, and in the following visits, four submaximal constant-load bouts (i.e., test and retest) were performed in the moderate-intensity domain (100 and 130 W) and heavy-intensity domain (160 and 190 W). The local tissue oxygen saturation index (SmO2) and pulmonary oxygen uptake (VÌO2) were measured continuously. The absolute SmO2 and VÌO2 values and the change (Δ) from the 3rd to 6th min of exercise were calculated. There was good reliability for SmO2 measurements, as indicated by the high intraclass correlation coefficient analysis (ICC ≥0.84 for all) and low coefficient of variation between the two trials (CV ≤4.1% for all). Steady-state responses were observed for SmO2 and VÌO2 from the 3rd to the 6th min in the two moderate-intensity bouts (P>0.05), whereas SmO2 decreased and VÌO2 increased from the 3rd to the 6th min in the two heavy-intensity bouts (P<0.05). Together, these findings suggested that the SmO2 measured with a wNIRS device is reliable and sensitive to track local metabolic changes provoked by slight increments in exercise intensity.

Sites of 5S and 45S rDNA are more commonly located on different chromosomes of most angiosperms. Previous investigations have shown that in the subfamily Aurantioideae these sites may appear closely linked (adjacent sites), as in Poncirustrifoliata, or completely isolated, as in some species of Citrus. In the present work, the distribution of rDNA sites was investigated in representatives of 9 genera of Aurantioideae by FISH and CMA banding, aiming to understand the evolution of adjacent sites in the subfamily. A total of 57 rDNA sites were observed, 40 of them being adjacent to each other. All adjacent sites displayed the 45S rDNA array more terminally located. Assuming that the linked 5S-45S rDNA arrangement was the ancestral condition in Aurantioideae, the isolated rDNA sites observed in Clausena excavata,Bergera koenigii, and Fortunella obovata, as well as the complete linkage loss in Citrus maxima and C. medica indicates that unlinked sites arose independently several times in the evolution of the group. The linkage loss may be due to independent dispersion of one or both rDNA sequence families followed by deletion of the corresponding array in the adjacent site. The possible mechanisms behind these events and their occurrence in other groups are discussed.

BACKGROUND: Acute exercise increases IL-6, IL-10 and TNF-α levels in healthy subjects. There is no study evaluating the effect of exercise on cytokines level in systemic lupus erythematosus (SLE) patients. OBJECTIVE: Our aim was to assess IL-10, IL-6 and TNF-α levels at baseline and after acute physical exercise in patients with SLE. METHODS: In total, 27 female SLE patients and 30 healthy controls were evaluated. Serum levels of IL-10, IL-6 and TNF-α at baseline and soon after the ergospirometric test were measured by ELISA test. Student's t-tests and Mann-Whitney test were used for intra- and inter-group comparisons; p values <0.05 were considered significant. RESULTS: Patients with SLE presented worse ergospirometric parameters compared with controls: VO2max (25.78 ± 5.51 vs. 32.74 ± 5.85 ml/kg/min, p < 0.001); maximum heart rate (174.18 ± 12.36 vs. 185.15 ± 2.07 bpm, p = 0.001); maximum ventilation (65.51 ± 15.68 vs. 80.48 ± 18.98 l/min, p = 0.001) and maximum speed (7.70 ± 1.24 vs. 9.40 ± 1.22 km/h, p < 0.001). At baseline, SLE patients presented higher levels of IL-6 (2.38 ± 1.70 vs. 1.71 ± 0.29 pg/ml, p = 0.035) and IL-10 (1.09 ± 1.55 vs. 0.30 ± 0.11 pg/ml, p = 0.037) than controls. Acute exercise in controls increased IL-6 level (1.71 ± 0.29 vs. 2.01 ± 0.27 pg/ml, p = 0.003) without change in IL-10 and TNF-α levels. However, no significant change in cytokine levels was observed in SLE patients after acute exercise. CONCLUSION: This is the first study evaluating the effect of acute exercise on cytokine levels in patients with SLE. In contrast to healthy controls, acute physical exercise did not increase the levels of IL-6 in patients with SLE, and seems to be safe in those patients with inactive or mild active disease.

A parallel group randomized trial was designed to analyze the impact of 6 weeks of strength training programs performed with or without whole-body vibration on muscular and endurance performance parameters in long-distance runners. 22 endurance runners were allocated into strength with whole-body vibration (n=8), without (n=8), and control (n=6) groups. Before and after the experimental period the subjects performed the following tests: a) maximum dynamic strength test, b) maximal incremental treadmill test, and c) time to exhaustion at velocity corresponding to maximal oxygen uptake. The fractions of the aerobic and anaerobic contribution in time to exhaustion test were also calculated. Both strength trained groups showed a similar increase in maximum dynamic strength (~18%). The aerobic contribution was enhanced for strength training group without whole-body vibration (~25%) after experimental period. No statistical differences were observed in any other variable. These results suggest that 6 weeks of strength training performed with or without whole-body vibration improve similarly the maximum dynamic strength in long-distance runners. In addition, both training modes studied had no deleterious effects on the traditional parameters of endurance performance, traditional strength training program results in increased aerobic contribution during high-intensity aerobic exercise.

We examined the pacing strategy and the magnitude of the end spurt during a 200-kJ cycling time trial performed 12-14 h after an exercise protocol designed to reduce muscle glycogen content. 9 physically-active men performed 5 familiarization sessions and 2 experimental 200-kJ time trials in either a control condition (CON) or after an exercise protocol performed the previous evening that was designed to induce muscle glycogen depletion (EP). Mean total time was faster and power output was higher in the CON than in the EP (P<0.01). A fast-start was maintained until the 50-kJ section in CON, but only the 25-kJ section for EP (P<0.05). The power outputs during the 50-, 150- and 200-kJ sections, and the magnitude of the end-spurt, were significantly higher for the CON than for the EP condition (P<0.05). There was no significant difference in the rating of perceived exertion (overall feeling and feeling in legs) between conditions. In conclusion, a protocol designed to decrease muscle glycogen stores reduced the duration of the fast-start and the magnitude of the end spurt during a 200-kJ cycling time trial, impairing the overall performance.

Ruminal tympany (bloat) has long been an issue for large and small livestock operations. Though improvements in feedlot management practices have reduced its occurrence, it is still highly prevalent and is known to detrimentally affect animal performance, welfare, and in many instances, lead to animal death. Current decision support systems and diet formulation software omit the inclusion of bloat prediction based on animal performance. Here, we aim to predict bloat incidence in implanted and non-implanted feedlot steers from performance data comparing linear (LDB) and non-linear decision boundaries. Eighteen crossbred Angus × Hereford steers: BW (491.13 ± 25.78 kg) and age (12 ± 1 mo) were randomly distributed into implanted and non-implanted treatments. All animals were randomly assigned to one of two pens fit with automated monitoring systems for BW, freshwater intake, and water intake behavior: water intake event visit, no water intake event visit (NWIE), and time spent drinking. DM intake (DMI) was individually recorded from all animals through the Calan Gate system for 135 d (30 d adaptation, 105 d experimental diet). Incidences of bloat were recorded as bloat instances regardless of severity to ensure that early onset detection of bloat was recorded and properly identified in predictive models. Logistic regression with a binomial distribution and a logit link function was utilized to predict the incidences of bloat through LDB. Feature selection and penalization of coefficients were explored through L1 (sum of absolute values) and L2 (sum of squares) penalization to avoid overfitting of models. Additional NLDB and a non-parametric LDB are examined for prediction. Accuracy, specificity, and sensitivity were high for the models reported. No significant differences were observed between LDB and NLDB, with the highest specificity (predicting bloat) value of 0.820 for stepwise feature selection algorithms, and a value of 0.832 for the artificial neural network. Highest accuracy was 0.829 for ridge regression, and 0.847 for the random forest with hyperparameter tuning. DM intake, BW, and NWIE were the three most important variables for the prediction of feedlot bloat showing clear drops in DMI and BW and increases in NWIE when animals bloated. The lack of difference in predictive performance between LDB and NLDB highlights the often-overlooked concept that machine learning algorithms are not always the only/best modeling technique. Additionally, the models reported herein carry acceptable predictive performance for inclusion into management decisions that reduce bloat incidences in feedlot cattle.

PURPOSE: Analyze the association between parental stress and dental caries experience in adolescents in southern Brazil using data from the Pelotas 2004 Birth Cohort. METHODS: Interviews and oral health examinations for the determination of the main exposure and outcome of the study were performed in the homes of the adolescents. The outcome was dental caries experience in the permanent dentition analyzed using the Decayed, Missing and Filling Surfaces (DMFS) index. The main exposure was parental stress measured using the Parenting Stress Index-Short Form administered to the parents of the adolescents. Demographic/socioeconomic characteristics, oral health characteristics and oral health-related quality of life were considered potential confounding factors. Negative binomial regressions estimated mean ratios (MR) and 95% confidence intervals (CI). RESULTS: Nine hundred ninety-six adolescents were evaluated at 12 and 13 years of age. The prevalence of dental caries experience in the adolescents was 36.9% (95% CI: 33.8-40.0) and 15.1% (95% CI: 12.8-17.3) of the parents had parental stress. After adjusting for confounding factors, parental stress was associated with a higher mean number of decayed, missing and filling surfaces in the adolescents (MR = 1.10; 95% CI: 1.01-1.26; p = 0.045). CONCLUSION: Adolescents of parents with parental stress have more dental caries experience compared to those whose parents do not have parental stress.

In cycling, there is a body of evidence that supports that an all-out start strategy is superior to an even-pacing strategy, but it is unknown whether an all-out start strategy is superior to a self-paced strategy. In the present study, we investigated the effects of three different pacing strategies on 4-km cycling time trial performance. After preliminary trials (familiarization trials and a baseline 4-km cycling time trial), in a randomized and counterbalanced order, twelve male cyclists (32.3±7.2 years old, maximum rate of O2 uptake (VÌO2peak) 4.3±0.4 L/min) completed: 1) a self-paced 4-km cycling time trial; 2) an all-out start (â¼10 s), followed by maintenance of the average baseline trial power for the first km and self-paced cycling for the remaining trial (all-out+mean); and 3) an all-out start (â¼10 s), followed by a power 5% above the average baseline trial power for the first km and self-paced cycling for the remaining trial (all-out+5%mean). Although there was a significant interaction between power and distance (P=0.001) with different power distribution profiles throughout the trial, there was no significant difference (P=0.99) between the three strategies for overall exercise performance (self-paced 379.8±13.9 s, all-out+mean 380.0±16.0 s, and all-out+5%mean 380.2±11.5 s). Oxygen uptake, rating of perceived effort, and heart rate were also similar across the pacing strategies. Different all-out start strategies did not confer additional benefits to performance compared to a self-paced strategy.

In this paper, we determine by fluorescent in situ hybridization the variability in the chromosomal location of 45S rDNA clusters in 38 species belonging to 7 genera of the Triatominae subfamily, using a triatomine-specific 18S rDNA probe. Our results show a striking variability at the inter- and intraspecific level, never reported so far in holocentric chromosomes, revealing the extraordinary genomic dynamics that occurred during the evolution in this group of insects. Our results also demonstrate that the chromosomal position of rDNA clusters is an important marker to disclose chromosomal differentiation in species karyotypically homogenous in their chromosome number.

We examined the effects of listening to music on attentional focus, rating of perceived exertion (RPE), pacing strategy and performance during a simulated 5-km running race. 15 participants performed 2 controlled trials to establish their best baseline time, followed by 2 counterbalanced experimental trials during which they listened to music during the first (M start) or the last (M finish) 1.5 km. The mean running velocity during the first 1.5 km was significantly higher in M start than in the fastest control condition (p<0.05), but there was no difference in velocity between conditions during the last 1.5 km (p>0.05). The faster first 1.5 m in M start was accompanied by a reduction in associative thoughts compared with the fastest control condition. There were no significant differences in RPE between conditions (p>0.05). These results suggest that listening to music at the beginning of a trial may draw the attentional focus away from internal sensations of fatigue to thoughts about the external environment. However, along with the reduction in associative thoughts and the increase in running velocity while listening to music, the RPE increased linearly and similarly under all conditions, suggesting that the change in velocity throughout the race may be to maintain the same rate of RPE increase.

The current study examined the relationships among temporal processing speed, spatial visual span and Chinese character reading speed in normal central and peripheral vision. Maximum reading speed (MRS) and critical print size (CPS) of 26 native Chinese readers (13 young and 13 older adults) were determined at three visual field locations: central vision, 10° left and 10° below fixation using a rapid serial visual presentation (RSVP) task. Temporal processing speed was measured using trigrams of randomly selected Chinese characters presented at a range of exposure durations, while spatial visual span was measured using trigrams presented at different spatial positions. It was found that shorter temporal processing speed and larger spatial visual span were associated with faster MRS at the central and inferior visual field locations, but not at the left of fixation location. As expected, reading and visual span metrics were better in central vision compared to both peripheral locations. In addition, reading, temporal processing, and spatial visual span metrics were better in the young than older subjects (except for similar temporal processing speed at two peripheral locations). The results for central and inferior presentation locations support the hypothesis that temporal processing speed and spatial visual span were associated with Chinese character reading speed. Surprisingly, no correlation was observed for the 10° left of the fixation location, suggesting that the factors affecting reading speed might differ for inferior and lateral peripheral viewing locations.

We tested the ability of human observers to discriminate distinct profiles of spatially dependant geometric phases when directly viewing stationary structured light beams. Participants viewed polarization coupled orbital angular momentum (OAM) states, or "spin-orbit" states, in which the OAM was induced through Pancharatnam-Berry phases. The coupling between polarization and OAM in these beams manifests as spatially dependant polarization. Regions of uniform polarization are perceived as specifically oriented Haidinger's brushes, and study participants discriminated between two spin-orbit states based on the rotational symmetry in the spatial orientations of these brushes. Participants used self-generated eye movements to prevent adaptation to the visual stimuli. After initial training, the participants were able to correctly discriminate between two spin-orbit states, differentiated by OAM [Formula: see text], with an average success probability of [Formula: see text] ([Formula: see text], [Formula: see text]). These results support our previous observation that human observers can directly perceive spin-orbit states, and extend this finding to non-rotating beams, OAM modes induced via Pancharatnam-Berry phases, and the discrimination of states that are differentiated by OAM.

The aim of this study was to describe the muscle function, architecture, and composition of long-distance master runners, and verify the association between age and these variables. Additionally, different clusters of runners were compared based on age and training variables. Forty male runners (≥50 years) reported their training routine and had their muscle function evaluated through maximum knee extensor isometric peak torque (PT) assessed with an isokinetic dynamometer. The cross-sectional area (CSA), pennation angle (PA), fascicle length (FL), muscle thickness (MT), and echo intensity (EI) were evaluated through ultrasound (muscle architecture and composition). The participants were 58.7±6.2 years old and had been training for 18.4±10.3 years, 4 sessions/week with 298.8±164.7 min/week of training. The absolute torque was 226.92±63.44 N·m, and the specific torque (PT/CSA) was 7.29±3.78 N·m/cm2. Regarding muscle architecture, the phase angle was 17.34±4°, the fascicle angle 6.78±1.04 cm, muscle thickness 2.93±0.56 cm, and the cross-sectional area 21.24±5.88 cm2. Concerning muscle composition, the master runners showed echo intensity values of 62.05±11.68 AU. The analysis demonstrated a weak and negative association between age and some muscle architecture variables (CSA and MT) and muscle function (PT). No association was verified between age and muscle composition (EI). Age partially explained CSA, MT, and muscle function changes (13, 11, and 14%, respectively). Participants' high level of physical training might have contributed to the low association between these variables and the lack of association with muscle composition.
