RESUMEN

The output signal from a photoacoustic cell based on a symmetrical Helmholtz resonator structure can be substantially increased if a counterphase light stimulation is applied to the cell cavities. However even slight differences in the intensity of the light beams irradiating the cavities may affect the frequency response of the cell and the output signal level. This paper shows the influence of the imbalanced light irradiation on the properties of such a cell. It was found that even at relatively high irradiation mismatch, and even with the photoacoustic signal detection implemented with a single microphone, the influence of the irradiation imbalance on the frequency response of the cell around the resonance frequency is not critical. In the case of differential detection of the photoacoustic signal, the imbalance of the light irradiation does not affect the frequency response of the cell, but only the output signal level.

All the preamplifiers dedicated for Quartz Enhanced PhotoAcoustic Spectroscopy (QEPAS) applications that have so far been reported in the literature have been based on operational amplifiers working in transimpedance configurations. Taking into consideration that QEPAS sensors are based on quartz tuning forks, and that quartz has a relatively high voltage constant and relatively low charge constant, it seems that a transimpedance amplifier is not an optimal solution. This paper describes the design of a quartz QEPAS sensor preamplifier, implemented with voltage amplifier configuration. Discussion of an electrical model of the circuit and preliminary measurements are presented. Both theoretical analysis and experiments show that use of the voltage configuration allows for a substantial increase of the output signal in comparison to the transimpedance circuit with the same tuning fork working in identical conditions. Assuming that the sensitivity of the QEPAS technique depends directly on the properties of the preamplifier, use of the voltage amplifier configuration should result in an increase of QEPAS sensitivity by one to two orders of magnitude.

A detailed investigation of a set of custom quartz tuning forks (QTFs), operating in the fundamental and first overtone flexural modes is reported. Support losses are the dominant energy dissipation processes when the QTFs vibrate at the first overtone mode. These losses can be decreased by increasing the ratio between the prong length and its thickness. The QTFs were implemented in a quartz enhanced photoacoustic spectroscopy (QEPAS) based sensor operating in the near-IR spectral range and water vapor was selected as the gas target. QTF flexural modes having the highest quality factor exhibit the largest QEPAS signal, demonstrating that, by optimizing the QTF prongs sizes, overtone modes can provide a higher QEPAS sensor performance with respect to using the fundamental mode.

An effective therapy for advanced melanoma, a skin cancer with the highest mortality, has not yet been developed. The endocannabinoid system is considered to be an attractive target for cancer treatment. The use of endocannabinoids, such as anandamide (AEA), is considered to be much greater than as a palliative agent. Thus, we checked its influence on various signaling pathways in melanoma cells. Our investigation was performed on four commercial cell lines derived from different progression stages (radial WM35 and vertical WM115 growth phases, lymph node WM266-4 metastasis, solid tumor A375-P metastasis). Cell viability, glucose uptake, quantification of reactive oxygen species production, expression of selected genes encoding glycosyltransferases, quantification of glycoproteins production and changes in the glycosylation profile and migration, as well as in cell elastic properties were analyzed. The cell glycosylation profile was investigated using the biophysical profiling method-the quartz crystal microbalance with dissipation monitoring (QCM-D). Anandamide treatment of only metastatic cells resulted in: an increase in the cell metabolism, a decrease in GFAT-1 and DPM1 expression, followed by a decrease in L1-CAM glycoprotein production, which further influenced the reduction in the cell glycosylation profile and migration. Considering our results, AEA usage is highly recommended in the combined therapy of advanced melanoma.

Modeling of small Helmholtz resonators based on electroacoustical analogies often results in significant disagreement with measurements, as existing models do not take into account some losses that are observed in practical implementations of such acoustical circuits, e.g., in photoacoustic Helmholtz cells. The paper presents a method which introduces loss corrections to the transmission line model, resulting in substantial improvement of simulations. Values of the loss corrections obtained from comparison of frequency responses of practically implemented resonators with computer simulations are presented in tabular and graphical form. A simple analytical function that can be used for interpolation or extrapolation of the loss corrections for other dimensions of the Helmholtz resonators is also given. Verification of such a modeling method against an open two-cavity Helmholtz structure shows very good agreement between measurements and simulations.

The paper presents the design of a low-noise preamplifier dedicated for condenser measurement microphones used in high sensitivity applications, in which amplifier noise is the main factor limiting sensitivity of the measurements. In measurement microphone preamplifiers, the dominant source of noise at lower frequencies is the bias resistance of the input stage. In the presented solution, resistors were connected to the input stage by means of switches. The switches are opened during measurements, which disconnects the resistors from the input stage and results in noise reduction. Closing the switches allows for fast charging of the microphone capacitance. At low frequencies the noise of the designed preamplifier is a few times lower in comparison to similar, commercially available instruments.
