RESUMEN

BACKGROUND: Monitoring of anticoagulation therapy is based on screening tests: prothrombin time (PT) and activated partial thromboplastin time (APTT). The accidental presence of a clot in the coagulation samples determines a false prolongation of PT by fibrinogen (FI) consumption and the false or delayed prolongation of APTT, depending on FI consumption or activation. The purpose of this study is to document from the present data re-garding procedures used to exclude the accidental presence of clot in the sample. METHODS: For a more efficient approach, we conducted a study based on research from the main databases that included original and peer-reviewed studies. RESULTS: We have reported studies in which pre-analytical procedures have been recommended and studies that have also presented post-analytical protocols. A correlation between the efficiency of the procedures in terms of additional laboratory costs has been performed, as well. CONCLUSIONS: Focusing on patient safety, it remains a continuous challenge for each laboratory to be able to establish its own pre-analytical and post-analytical procedure for highlighting accidental clot presence, thus ensuring provision of results with maximum confidence to the clinicians.

Enrofloxacin (ENR), a member of the fluoroquinolone class of antibiotics, is widely used in veterinary medicine to treat bacterial infections. Like many antibiotics, ENR has limited water solubility and low bioavailability. To address these challenges, drug formulations using solid dispersions, nanosuspensions, surfactants, cocrystal/salt formation, and inclusion complexes with cyclodextrins may be employed. The approach described herein proposes the development of ENR formulations by co-electrospinning ENR with custom-prepared cyclodextrin-oligolactide (CDLA) derivatives. This method benefits from the high solubility of these derivatives, enabling polymer-free electrospinning. The electrospinning parameters were optimized to incorporate significant amounts of ENR into the CDLA nanofibrous webs, reaching up to 15.6% by weight. The obtained formulations were characterized by FTIR and NMR spectroscopy methods and evaluated for their antibacterial activity against Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa. This study indicates that the presence of CDLA derivative does not inhibit the antibacterial activity of ENR, recommending these formulations for further development.

AIMS: This study reports a simple, cost-effective, and environmentally friendly method to obtain silver nanoparticles (AgNPs) using an aqueous extract of Quercus robur bark. METHODS: AgNPs synthesis conditions such as silver nitrate concentration, extract:AgNO3 volume ratio, pH, temperature, and reaction time have been examined. After optimizing the synthesis, the obtained AgNPs were characterized by different methods such as UV-Vis, TEM, EDX, and FTIR. The antioxidant activity was evaluated using lipoxygenase inhibition capacity and inhibition of erythrocyte hemolysis mediated by peroxyl free radicals tests. The antimicrobial potential of the samples was tested against Staphylococcus aureus, Pseudomonas aeruginosa, and Candida albicans. RESULTS: The AgNPs synthesis process is influenced by reaction conditions, the optimum established values being, in this case: concentration of 3 mM AgNO3, 1:9 extract: AgNO3 volume ratio, pH value of 6, 60 ºC temperature, and 90 minutes stirring time. The shape of the synthesized AgNPs was predominantly spherical, with an average size of 50 nm. The SPR band at 432 nm, the strong EDX signal at ~ 3 keV and the zeta potential of -13.88 mV revealed the formation of AgNPs and electrostatic stabilization of the colloidal solution. FTIR analysis confirmed the participation of molecules from the extract in the synthesis and stabilization of AgNPs. The obtained nanoparticles showed improved antioxidant, antifungal and antibacterial activities compared to the extract. CONCLUSION: The results open the possibility of exploring new applications of nanoparticles obtained via green synthesis.

Black-eyed Susan (Rudbeckia hirta L.), a flowering plant with various traditional medicinal uses, has recently garnered interest for its therapeutic properties. However, little is known about the potential therapeutic activities of the plant species. The current study focused on conducting a comprehensive investigation into the chemical composition and bioactivity of black-eyed Susan cultivated in Romania. Untargeted metabolite profiling and UHPLC-HR-MS phytochemical analysis of the studied extract revealed the presence of more than 250 compounds pertaining to different classes, including sesquiterpene lactones, polyphenolic acids, flavonoids, amino acids, and fatty acids. The tested extract exhibited inhibitory activity against Gram-positive bacteria and showed promising antifungal activity. It also demonstrated potent antioxidant properties through iron chelation and 15-LOX inhibition capacities, as well as inhibition of cell growth, particularly on the MCF-7 cell line, suggesting potential anticancer effects. Therefore, current research provides valuable information on the antioxidant, antimicrobial, and antitumor potential of Rudbeckia hirta flowers. Implicitly, the discovery of such a wide range of biosubstances, together with the biological activity observed for the studied extract in these preliminary in vitro studies, paves the way for future investigation of the potential application of the plant in the pharmaceutical and nutraceutical sectors.

Acute heart failure (AHF) is a life-threatening condition with high morbidity and mortality. Even though this pathology has been extensively researched, there are still challenges in establishing an accurate and early diagnosis, determining the long- and short-term prognosis and choosing a targeted therapeutic strategy. The use of reliable biomarkers to support clinical judgment has been shown to improve the management of AHF patients. Despite a large pool of interesting candidate biomarkers, endothelin-1 (ET-1) appears to be involved in multiple aspects of AHF pathogenesis that include neurohormonal activation, cardiac remodeling, endothelial dysfunction, inflammation, atherosclerosis and alteration of the renal function. Since its discovery, numerous studies have shown that the level of ET-1 is associated with the severity of symptoms and cardiac dysfunction in this pathology. The purpose of this paper is to review the existing information on ET-1 and answer the question of whether this neurohormone could be a promising biomarker in AHF.

This research describes an eco-friendly green route for the synthesis of AgNPs using an aqueous extract of Lythrum salicaria. Taguchi design was used to optimize the synthesis method, taking into account various working conditions. The optimum parameters were established using a 3 mM AgNO3 concentration, a 1:9 extract:AgNO3 volume ratio, a pH value of 8, 60 ℃ temperature, and 180 min reaction time. The synthesized AgNPs were characterized using UV-Vis and FTIR spectroscopy, and TEM and EDX analysis. The SPR band at 410 nm, as well as the functional groups of biomolecules identified by FTIR and the EDX signals at ~3 keV, confirmed the synthesis of spherical AgNPs. The average AgNPs size was determined to be 40 nm, through TEM, and the zeta potential was -19.62 mV. The antimicrobial assay showed inhibition against S. aureus and C. albicans. Moreover, the results regarding the inhibition of lipoxygenase and of peroxyl radical-mediated hemolysis assays were promising and justify further antioxidant studies.

Ochratoxins are mycotoxins that have been extensively studied lately due to the multiple toxic effects such as nephrotoxicity, hepatotoxicity, and carcinogenicity. These toxins contaminate plant and animal foods and after ingestion they reach into body fluids. The method of competitive direct enzyme immunoassay, in the solid phase, was validated through the determination of specific parameters (performance, linearity, recovery percentage, limit of detection, limit of quantification). The validated method was used to determine ochratoxin A in colostrum and cow's milk. The method applied for the determination of ochratoxin A was linear for the concentration range of 0.0-0.5 ng/mL, the value for the regression coefficient (r) was 0.9838. Ochratoxin A was present in 91.67% of the colostrum and in 93.33% of cow's milk samples. The linearity of the method, demonstrated for very low concentrations of analyte, the detection limit as well as the limit of quantification recommend the method for the determinations of micro-pollutants from foods, including biological fluids.

OBJECTIVE: This study was performed to determine whether a dual-biomarker approach using N-terminal prohormone of brain natriuretic peptide (NT-proBNP) and galectin-3 optimizes the diagnosis and risk stratification of acute cardiac dyspnea. Atypical clinical manifestations and overlapping pathologies require objective and effective diagnostic methods to avoid treatment delays. METHODS: This prospective observational study included 208 patients who presented to the emergency department for acute dyspnea. NT-proBNP and galectin-3 were measured upon admission. The patients were divided into two groups according to the etiology of their clinical manifestations: cardiac and non-cardiac dyspnea. The patients' New York Heart Association functional class, left ventricular ejection fraction, and discharge status were assessed. RESULTS: Diagnostic criteria for acute heart failure were fulfilled in 61.1% of the patients. NT-proBNP and galectin-3 were strongly and significantly correlated. Receiver operating characteristic analysis revealed similar areas under the curve for both markers in the entire group of patients as well as in the high-risk subsets of patients. CONCLUSIONS: The diagnostic performance of NT-proBNP and galectin-3 is comparable for both the total population and high-risk subsets. Galectin-3 adds diagnostic value to the conventional NT-proBNP in patients with acute cardiac dyspnea, and its utility is of major interest in uncertain clinical situations.
