RESUMEN

Functional near-infrared spectroscopy (fNIRS) is a comparatively new noninvasive, portable, and easy-to-use brain imaging modality. However, complicated dexterous tasks such as individual finger-tapping, particularly using one hand, have been not investigated using fNIRS technology. Twenty-four healthy volunteers participated in the individual finger-tapping experiment. Data were acquired from the motor cortex using sixteen sources and sixteen detectors. In this preliminary study, we applied standard fNIRS data processing pipeline, i.e., optical densities conversation, signal processing, feature extraction, and classification algorithm implementation. Physiological and non-physiological noise is removed using 4th order band-pass Butter-worth and 3rd order Savitzky-Golay filters. Eight spatial statistical features were selected: signal-mean, peak, minimum, Skewness, Kurtosis, variance, median, and peak-to-peak form data of oxygenated haemoglobin changes. Sophisticated machine learning algorithms were applied, such as support vector machine (SVM), random forests (RF), decision trees (DT), AdaBoost, quadratic discriminant analysis (QDA), Artificial neural networks (ANN), k-nearest neighbors (kNN), and extreme gradient boosting (XGBoost). The average classification accuracies achieved were 0.75±0.04, 0.75±0.05, and 0.77±0.06 using k-nearest neighbors (kNN), Random forest (RF) and XGBoost, respectively. KNN, RF and XGBoost classifiers performed exceptionally well on such a high-class problem. The results need to be further investigated. In the future, a more in-depth analysis of the signal in both temporal and spatial domains will be conducted to investigate the underlying facts. The accuracies achieved are promising results and could open up a new research direction leading to enrichment of control commands generation for fNIRS-based brain-computer interface applications.

The rapid expansion of a country's economy is highly dependent on timely product distribution, which is hampered by terrible traffic congestion. Additional staff are also required to follow the delivery vehicle while it transports documents or records to another destination. This study proposes Delicar, a self-driving product delivery vehicle that can drive the vehicle on the road and report the current geographical location to the authority in real-time through a map. The equipped camera module captures the road image and transfers it to the computer via socket server programming. The raspberry pi sends the camera image and waits for the steering angle value. The image is fed to the pre-trained deep learning model that predicts the steering angle regarding that situation. Then the steering angle value is passed to the raspberry pi that directs the L298 motor driver which direction the wheel should follow. Based upon this direction, L298 decides either forward or left or right or backwards movement. The 3-cell 12V LiPo battery handles the power supply to the raspberry pi and L298 motor driver. A buck converter regulates a 5V 3A power supply to the raspberry pi to be working. Nvidia CNN architecture has been followed, containing nine layers including five convolution layers and three dense layers to develop the steering angle predictive model. Geoip2 (a python library) retrieves the longitude and latitude from the equipped system's IP address to report the live geographical position to the authorities. After that, Folium is used to depict the geographical location. Moreover, the system's infrastructure is far too low-cost and easy to install.

The ability to optimize power generation in sports is imperative, both for understanding and balancing training load correctly, and for optimizing competition performance. In this paper, we aim to estimate mechanical power output by employing a time-sequential information-based deep Long Short-Term Memory (LSTM) neural network from multiple inertial measurement units (IMUs). Thirteen athletes conducted roller ski skating trials on a treadmill with varying incline and speed. The acceleration and gyroscope data collected with the IMUs were run through statistical feature processing, before being used by the deep learning model to estimate power output. The model was thereafter used for prediction of power from test data using two approaches. First, a user-dependent case was explored, reaching a power estimation within 3.5% error. Second, a user-independent case was developed, reaching an error of 11.6% for the power estimation. Finally, the LSTM model was compared to two other machine learning models and was found to be superior. In conclusion, the user-dependent model allows for precise estimation of roller skiing power output after training the model on data from each athlete. The user-independent model provides less accurate estimation; however, the accuracy may be sufficient for providing valuable information for recreational skiers.

The current study attempted, for the first time, to qualitatively and quantitatively determine the phytochemical components of Elatostema papillosum methanol extract and their biological activities. The present study represents an effort to correlate our previously reported biological activities with a computational study, including molecular docking, and ADME/T (absorption, distribution, metabolism, and excretion/toxicity) analyses, to identify the phytochemicals that are potentially responsible for the antioxidant, antidepressant, anxiolytic, analgesic, and anti-inflammatory activities of this plant. In the gas chromatography-mass spectroscopy analysis, a total of 24 compounds were identified, seven of which were documented as being bioactive based on their binding affinities. These seven were subjected to molecular docking studies that were correlated with the pharmacological outcomes. Additionally, the ADME/T properties of these compounds were evaluated to determine their drug-like properties and toxicity levels. The seven selected, isolated compounds displayed favorable binding affinities to potassium channels, human serotonin receptor, cyclooxygenase-1 (COX-1), COX-2, nuclear factor (NF)-κB, and human peroxiredoxin 5 receptor proteins. Phytol acetate, and terpene compounds identified in E. papillosum displayed strong predictive binding affinities towards the human serotonin receptor. Furthermore, 3-trifluoroacetoxypentadecane showed a significant binding affinity for the KcsA potassium channel. Eicosanal showed the highest predicted binding affinity towards the human peroxiredoxin 5 receptor. All of these findings support the observed in vivo antidepressant and anxiolytic effects and the in vitro antioxidant effects observed for this extract. The identified compounds from E. papillosum showed the lowest binding affinities towards COX-1, COX-2, and NF-κB receptors, which indicated the inconsequential impacts of this extract against the activities of these three proteins. Overall, E. papillosum appears to be bioactive and could represent a potential source for the development of alternative medicines; however, further analytical experiments remain necessary.

A pandemic caused by the novel coronavirus (SARS-CoV-2 or COVID-19) began in December 2019 in Wuhan, China, and the number of newly reported cases continues to increase. More than 19.7 million cases have been reported globally and about 728,000 have died as of this writing (10 August 2020). Recently, it has been confirmed that the SARS-CoV-2 main protease (Mpro) enzyme is responsible not only for viral reproduction but also impedes host immune responses. The Mpro provides a highly favorable pharmacological target for the discovery and design of inhibitors. Currently, no specific therapies are available, and investigations into the treatment of COVID-19 are lacking. Therefore, herein, we analyzed the bioactive phytocompounds isolated by gas chromatography-mass spectroscopy (GC-MS) from Tinospora crispa as potential COVID-19 Mpro inhibitors, using molecular docking study. Our analyses unveiled that the top nine hits might serve as potential anti-SARS-CoV-2 lead molecules, with three of them exerting biological activity and warranting further optimization and drug development to combat COVID-19.

Elderly care at home is a matter of great concern if the elderly live alone, since unforeseen circumstances might occur that affect their well-being. Technologies that assist the elderly in independent living are essential for enhancing care in a cost-effective and reliable manner. Elderly care applications often demand real-time observation of the environment and the resident’s activities using an event-driven system. As an emerging area of research and development, it is necessary to explore the approaches of the elderly care system in the literature to identify current practices for future research directions. Therefore, this work is aimed at a comprehensive survey of non-wearable (i.e., ambient) sensors for various elderly care systems. This research work is an effort to obtain insight into different types of ambient-sensor-based elderly monitoring technologies in the home. With the aim of adopting these technologies, research works, and their outcomes are reported. Publications have been included in this survey if they reported mostly ambient sensor-based monitoring technologies that detect elderly events (e.g., activities of daily living and falls) with the aim of facilitating independent living. Mostly, different types of non-contact sensor technologies were identified, such as motion, pressure, video, object contact, and sound sensors. Besides, multicomponent technologies (i.e., combinations of ambient sensors with wearable sensors) and smart technologies were identified. In addition to room-mounted ambient sensors, sensors in robot-based elderly care works are also reported. Research that is related to the use of elderly behavior monitoring technologies is widespread, but it is still in its infancy and consists mostly of limited-scale studies. Elderly behavior monitoring technology is a promising field, especially for long-term elderly care. However, monitoring technologies should be taken to the next level with more detailed studies that evaluate and demonstrate their potential to contribute to prolonging the independent living of elderly people.

In recent years, human activity recognition from body sensor data or wearable sensor data has become a considerable research attention from academia and health industry. This research can be useful for various e-health applications such as monitoring elderly and physical impaired people at Smart home to improve their rehabilitation processes. However, it is not easy to accurately and automatically recognize physical human activity through wearable sensors due to the complexity and variety of body activities. In this paper, we address the human activity recognition problem as a classification problem using wearable body sensor data. In particular, we propose to utilize a Deep Belief Network (DBN) model for successful human activity recognition. First, we extract the important initial features from the raw body sensor data. Then, a kernel principal component analysis (KPCA) and linear discriminant analysis (LDA) are performed to further process the features and make them more robust to be useful for fast activity recognition. Finally, the DBN is trained by these features. Various experiments were performed on a real-world wearable sensor dataset to verify the effectiveness of the deep learning algorithm. The results show that the proposed DBN outperformed other algorithms and achieves satisfactory activity recognition performance.

BACKGROUND: The increasingly high incidence of ischemic stroke caused by thrombosis of the arterial vessels is one of the major factors that threaten people's health and lives in the world. The present treatments for thrombosis are still unsatisfactory. Herbal preparations have been used since ancient times for the treatment of several diseases. The aim of this study was to investigate whether herbal preparations possess thrombolytic activity or not. METHODS: An in vitro thrombolytic model was used to check the clot lysis effect of the crude extracts and fractions of five Bangladeshi plant viz., Trema orientalis L., Bacopa monnieri L., Capsicum frutescens L., Brassica oleracea L. and Urena sinuata L. using streptokinase as a positive control and water as a negative control. Briefly, venous blood drawn from twenty healthy volunteers was allowed to form clots which were weighed and treated with the test plant materials to disrupt the clots. Weight of clot after and before treatment provided a percentage of clot lysis. RESULTS: Using an in vitro thrombolytic model, different fractions of five Bangladeshi medicinal plants namely T. orientalis, B. monnieri, C. frutescens, B. oleracea and U. sinuata showed various range of clot lysis activity. Chloroform fractions of T. orientalis, B. monnieri, C. frutescens, B. oleracea and U. sinuata showed highest significant (P < 0.05 and P < 0.001) clot lysis activity viz., 46.44 ± 2.44%, 48.39 ± 10.12%, 36.87 ± .27%, 30.24 ± 0.95% and 47.89 ± 6.83% respectively compared with positive control standard streptokinase (80.77 ± 1.12%) and negative control sterile distilled water (5.69 ± 3.09%). Other fractions showed moderate to low clot lysis activity. Order of clot lysis activity was found to be: Streptokinase > Chloroform fractions > Methanol (crude) extract > Hydro-methanol fractions > Ethyl acetate fractions > n-hexane fractions > Water. CONCLUSIONS: Our study suggests that thrombolytic activity of T. orientalis, B. monnieri and U. sinuata could be considered as very promising and beneficial for the Bangladeshi traditional medicine. Lower effects of other extracts might suggest the lack of bio-active components and/or insufficient quantities in the extract. In vivo clot dissolving property and active component(s) of T. orientalis and B. monnieri for clot lysis could lead the plants for their therapeutic uses. However, further work will establish whether or not, chloroform soluble phytochemicals from these plants could be incorporated as a thrombolytic agent for the improvement of the patients suffering from atherothrombotic diseases.

This paper introduces a refined and broadened version of decision-theoretic rough sets (DTRSs) named Generalized Sequential Decision-Theoretic Rough Set (GSeq-DTRS), which integrates the three-way decision (3WD) methodology. Operating through multiple levels, this iterative approach aims to comprehensively explore the boundary region. It introduces the concept of generalized granulation, departing from conventional equivalence-relation-based structures to incorporate similarity/tolerance relations. GSeq-DTRS addresses the limitations encountered by its predecessor, Seq-DTRS, particularly in managing sequential procedures and integrating new attributes. A notable advancement is its seamless handling of continuous-scale datasets through a defined Generalized Granular Structure (GGS), enabling classification across multiple levels without necessitating reduction of attributes. A refined version of conditional probability (CP), aligned with tolerance classes, enhances the approach, supported by a meticulously designed algorithm. Extensive experimental analysis conducted on two datasets sourced from https://www.kaggle.com demonstrates the efficacy of the procedure for both continuous and discrete datasets, effectively classifying probable elements into POS or NEG regions based on their membership. Unlike traditional Seq-DTRS, it does not require reduction of attributes at each new level. Additionally, the algorithm exhibits lower sensitivity to parametric values and yields results in fewer iterations. Thus, its potential impact on decision-making processes is readily apparent.

Diabetic Retinopathy (DR) is a serious eye condition that occurs due to high blood sugar levels in patients with Diabetes Mellitus. If left untreated, DR can potentially result in blindness. Using automated neural network-based methods to grade DR shows potential for early detection. However, the uneven and non-quadrilateral forms of DR lesions provide difficulties for traditional Convolutional Neural Network (CNN)-based architectures. To address this challenge and explore a novel algorithm architecture, this work delves into the usage of contrasting cluster assignments in retinal fundus images with the Swapping Assignments between multiple Views (SwAV) algorithm for DR grading. An ablation study was made where SwAV outperformed other CNN and Transformer-based models, independently and in ensemble configurations with an accuracy of 87.00% despite having fewer parameters and layers. The proposed approach outperforms existing state-of-the-art models regarding classification metrics, complexity, and prediction time. The findings offer great potential for medical practitioners, allowing for more accurate diagnosis of DR and earlier treatments to avoid visual loss.

The alarming growth of misinformation on social media has become a global concern as it influences public opinions and compromises social, political, and public health development. The proliferation of deceptive information has resulted in widespread confusion, societal disturbances, and significant consequences for matters pertaining to health. Throughout the COVID-19 pandemic, there was a substantial surge in the dissemination of inaccurate or deceptive information via social media platforms, particularly X (formerly known as Twitter), resulting in the phenomenon commonly referred to as an "Infodemic". This review paper examines a grand selection of 600 articles published in the past five years and focuses on conducting a thorough analysis of 87 studies that investigate the detection of fake news connected to COVID-19 on Twitter. In addition, this research explores the algorithmic techniques and methodologies used to investigate the individuals responsible for disseminating this type of fake news. A summary of common datasets, along with their fundamental qualities, for detecting fake news has been included as well. For the purpose of identifying fake news, the behavioral pattern of the misinformation spreaders, and their community analysis, we have performed an in-depth examination of the most recent literature that the researchers have worked with and recommended. Our key findings can be summarized in a few points: (a) around 80% of fake news detection-related papers have utilized Deep Neural Networks-based techniques for better performance achievement, although the proposed models suffer from overfitting, vanishing gradients, and higher prediction time problems, (b) around 60% of the disseminator related analysis papers focus on identifying dominant spreaders and their communities utilizing graph modeling although there is not much work done in this domain, and finally, (c) we conclude by pointing out a wide range of research gaps, for example, the need of a large and robust training dataset and deeper investigation of the communities, etc., and suggesting potential solution strategies. Moreover, to facilitate the utilization of a large training dataset for detecting fake news, we have created a large database by compiling the training datasets from 17 different research works. The objective of this study is to shed light on exactly how COVID-19-related tweets are beginning to diverge, along with the dissemination of misinformation. Our work uncovers notable discoveries, including the ongoing rapid growth of the disseminator population, the presence of professional spreaders within the disseminator community, and a substantial level of collaboration among the fake news spreaders.

The coronavirus epidemic has spread to virtually every country on the globe, inflicting enormous health, financial, and emotional devastation, as well as the collapse of healthcare systems in some countries. Any automated COVID detection system that allows for fast detection of the COVID-19 infection might be highly beneficial to the healthcare service and people around the world. Molecular or antigen testing along with radiology X-ray imaging is now utilized in clinics to diagnose COVID-19. Nonetheless, due to a spike in coronavirus and hospital doctors' overwhelming workload, developing an AI-based auto-COVID detection system with high accuracy has become imperative. On X-ray images, the diagnosis of COVID-19, non-COVID-19 non-COVID viral pneumonia, and other lung opacity can be challenging. This research utilized artificial intelligence (AI) to deliver high-accuracy automated COVID-19 detection from normal chest X-ray images. Further, this study extended to differentiate COVID-19 from normal, lung opacity and non-COVID viral pneumonia images. We have employed three distinct pre-trained models that are Xception, VGG19, and ResNet50 on a benchmark dataset of 21,165 X-ray images. Initially, we formulated the COVID-19 detection problem as a binary classification problem to classify COVID-19 from normal X-ray images and gained 97.5%, 97.5%, and 93.3% accuracy for Xception, VGG19, and ResNet50 respectively. Later we focused on developing an efficient model for multi-class classification and gained an accuracy of 75% for ResNet50, 92% for VGG19, and finally 93% for Xception. Although Xception and VGG19's performances were identical, Xception proved to be more efficient with its higher precision, recall, and f-1 scores. Finally, we have employed Explainable AI on each of our utilized model which adds interpretability to our study. Furthermore, we have conducted a comprehensive comparison of the model's explanations and the study revealed that Xception is more precise in indicating the actual features that are responsible for a model's predictions.This addition of explainable AI will benefit the medical professionals greatly as they will get to visualize how a model makes its prediction and won't have to trust our developed machine-learning models blindly.

Ultrasound imaging is a valuable tool for assessing the development of the fetal during pregnancy. However, interpreting ultrasound images manually can be time-consuming and subject to variability. Automated image categorization using machine learning algorithms can streamline the interpretation process by identifying stages of fetal development present in ultrasound images. In particular, deep learning architectures have shown promise in medical image analysis, enabling accurate automated diagnosis. The objective of this research is to identify fetal planes from ultrasound images with higher precision. To achieve this, we trained several convolutional neural network (CNN) architectures on a dataset of 12400 images. Our study focuses on the impact of enhanced image quality by adopting Histogram Equalization and Fuzzy Logic-based contrast enhancement on fetal plane detection using the Evidential Dempster-Shafer Based CNN Architecture, PReLU-Net, SqueezeNET, and Swin Transformer. The results of each classifier were noteworthy, with PreLUNet achieving an accuracy of 91.03%, SqueezeNET reaching 91.03% accuracy, Swin Transformer reaching an accuracy of 88.90%, and the Evidential classifier achieving an accuracy of 83.54%. We evaluated the results in terms of both training and testing accuracies. Additionally, we used LIME and GradCam to examine the decision-making process of the classifiers, providing explainability for their outputs. Our findings demonstrate the potential for automated image categorization in large-scale retrospective assessments of fetal development using ultrasound imaging.

Recognizing emotional state of human using brain signal is an active research domain with several open challenges. In this research, we propose a signal spectrogram image based CNN-XGBoost fusion method for recognising three dimensions of emotion, namely arousal (calm or excitement), valence (positive or negative feeling) and dominance (without control or empowered). We used a benchmark dataset called DREAMER where the EEG signals were collected from multiple stimulus along with self-evaluation ratings. In our proposed method, we first calculate the Short-Time Fourier Transform (STFT) of the EEG signals and convert them into RGB images to obtain the spectrograms. Then we use a two dimensional Convolutional Neural Network (CNN) in order to train the model on the spectrogram images and retrieve the features from the trained layer of the CNN using a dense layer of the neural network. We apply Extreme Gradient Boosting (XGBoost) classifier on extracted CNN features to classify the signals into arousal, valence and dominance of human emotion. We compare our results with the feature fusion-based state-of-the-art approaches of emotion recognition. To do this, we applied various feature extraction techniques on the signals which include Fast Fourier Transformation, Discrete Cosine Transformation, Poincare, Power Spectral Density, Hjorth parameters and some statistical features. Additionally, we use Chi-square and Recursive Feature Elimination techniques to select the discriminative features. We form the feature vectors by applying feature level fusion, and apply Support Vector Machine (SVM) and Extreme Gradient Boosting (XGBoost) classifiers on the fused features to classify different emotion levels. The performance study shows that the proposed spectrogram image based CNN-XGBoost fusion method outperforms the feature fusion-based SVM and XGBoost methods. The proposed method obtained the accuracy of 99.712% for arousal, 99.770% for valence and 99.770% for dominance in human emotion detection.

Renal failure, a public health concern, and the scarcity of nephrologists around the globe have necessitated the development of an AI-based system to auto-diagnose kidney diseases. This research deals with the three major renal diseases categories: kidney stones, cysts, and tumors, and gathered and annotated a total of 12,446 CT whole abdomen and urogram images in order to construct an AI-based kidney diseases diagnostic system and contribute to the AI community's research scope e.g., modeling digital-twin of renal functions. The collected images were exposed to exploratory data analysis, which revealed that the images from all of the classes had the same type of mean color distribution. Furthermore, six machine learning models were built, three of which are based on the state-of-the-art variants of the Vision transformers EANet, CCT, and Swin transformers, while the other three are based on well-known deep learning models Resnet, VGG16, and Inception v3, which were adjusted in the last layers. While the VGG16 and CCT models performed admirably, the swin transformer outperformed all of them in terms of accuracy, with an accuracy of 99.30 percent. The F1 score and precision and recall comparison reveal that the Swin transformer outperforms all other models and that it is the quickest to train. The study also revealed the blackbox of the VGG16, Resnet50, and Inception models, demonstrating that VGG16 is superior than Resnet50 and Inceptionv3 in terms of monitoring the necessary anatomy abnormalities. We believe that the superior accuracy of our Swin transformer-based model and the VGG16-based model can both be useful in diagnosing kidney tumors, cysts, and stones.

Human Activity Recognition (HAR) systems are devised for continuously observing human behavior - primarily in the fields of environmental compatibility, sports injury detection, senior care, rehabilitation, entertainment, and the surveillance in intelligent home settings. Inertial sensors, e.g., accelerometers, linear acceleration, and gyroscopes are frequently employed for this purpose, which are now compacted into smart devices, e.g., smartphones. Since the use of smartphones is so widespread now-a-days, activity data acquisition for the HAR systems is a pressing need. In this article, we have conducted the smartphone sensor-based raw data collection, namely H-Activity, using an Android-OS-based application for accelerometer, gyroscope, and linear acceleration. Furthermore, a hybrid deep learning model is proposed, coupling convolutional neural network and long-short term memory network (CNN-LSTM), empowered by the self-attention algorithm to enhance the predictive capabilities of the system. In addition to our collected dataset (H-Activity), the model has been evaluated with some benchmark datasets, e.g., MHEALTH, and UCI-HAR to demonstrate the comparative performance of our model. When compared to other models, the proposed model has an accuracy of 99.93% using our collected H-Activity data, and 98.76% and 93.11% using data from MHEALTH and UCI-HAR databases respectively, indicating its efficacy in recognizing human activity recognition. We hope that our developed model could be applicable in the clinical settings and collected data could be useful for further research.

Coronavirus disease 2019 (COVID-19), which is a highly contagious viral illness caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has had a catastrophic effect on the world's demographics, resulting in more than 3.8 million deaths worldwide and establishing itself as the most serious global health crisis since the 1918 influenza pandemic. Several questions remain unanswered regarding the effects of COVID-19 disease during pregnancy. Although most infections are mild in high-risk populations, the severe disease frequently leads to intubation, intensive care unit admission, and, in some cases, death. Hormonal and physiological changes in the immune and respiratory systems, cardiovascular function, and coagulation may affect the progression of COVID-19 disease in pregnancy. However, the consequences of coronavirus infection on implantation, fetal growth and development, labor, and newborn health have yet to be determined, and, consequently, a coordinated global effort is needed in this respect. Principles of management concerning COVID-19 in pregnancy include early isolation, aggressive infection control procedures, oxygen therapy, avoidance of fluid overload, consideration of empiric antibiotics (secondary to bacterial infection risk), laboratory testing for the virus and co-infection, fetal and uterine contraction monitoring, prevention, and / or treatment of thromboembolism early mechanical ventilation for progressive respiratory failure, individualized delivery planning, and a team-based approach with multispecialty consultations. This review focuses on COVID-19 during pregnancy, its management, and the area where further investigations are needed to reduce the risk to mothers and their newborns.

Healthcare using body sensor data has been getting huge research attentions by a wide range of researchers because of its good practical applications such as smart health care systems. For instance, smart wearable sensor-based behavior recognition system can observe elderly people in a smart eldercare environment to improve their lifestyle and can also help them by warning about forthcoming unprecedented events such as falls or other health risk, to prolong their independent life. Although there are many ways of using distinguished sensors to observe behavior of people, wearable sensors mostly provide reliable data in this regard to monitor the individual's functionality and lifestyle. In this paper, we propose a body sensor-based activity modeling and recognition system using time-sequential information-based deep Neural Structured Learning (NSL), a promising deep learning algorithm. First, we obtain data from multiple wearable sensors while the subjects conduct several daily activities. Once the data is collected, the time-sequential information then go through some statistical feature processing. Furthermore, kernel-based discriminant analysis (KDA) is applied to see the better clustering of the features from different activity classes by minimizing inner-class scatterings while maximizing inter-class scatterings of the samples. The robust time-sequential features are then applied with Neural Structured Learning (NSL) based on Long Short-Term Memory (LSTM), for activity modeling. The proposed approach achieved around 99% recall rate on a public dataset. It is also compared to existing different conventional machine learning methods such as typical Deep Belief Network (DBN), Convolutional Neural Network (CNN), and Recurrent Neural Network (RNN) where they yielded the maximum recall rate of 94%. Furthermore, a fast and efficient explainable Artificial Intelligence (XAI) algorithm, Local Interpretable Model-Agnostic Explanations (LIME) is used to explain and check the machine learning decisions. The robust activity recognition system can be adopted for understanding peoples' behavior in their daily life in different environments such as homes, clinics, and offices.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) was first recognized in Wuhan in late 2019 and, since then, had spread globally, eventually culminating in the ongoing pandemic. As there is a lack of targeted therapeutics, there is certain opportunity for the scientific community to develop new drugs or vaccines against COVID-19 and so many synthetic bioactive compounds are undergoing clinical trials. In most of the countries, due to the broad therapeutic spectrum and minimal side effects, medicinal plants have been used widely throughout history as traditional healing remedy. Because of the unavailability of synthetic bioactive antiviral drugs, hence all possible efforts have been focused on the search for new drugs and alternative medicines from different herbal formulations. In recent times, it has been assured that the Mpro, also called 3CLpro, is the SARS-CoV-2 main protease enzyme responsible for viral reproduction and thereby impeding the host's immune response. As such, Mpro represents a highly specified target for drugs capable of inhibitory action against coronavirus disease 2019 (COVID-19). As there continue to be no clear options for the treatment of COVID-19, the identification of potential candidates has become a necessity. The present investigation focuses on the in silico pharmacological activity of Calotropis gigantea, a large shrub, as a potential option for COVID-19 Mpro inhibition and includes an ADME/T profile analysis of that ligand. For this study, with the help of gas chromatography-mass spectrometry analysis of C. gigantea methanolic leaf extract, a total of 30 bioactive compounds were selected. Our analyses unveiled the top four options that might turn out to be prospective anti-SARS-CoV-2 lead molecules; these warrant further exploration as well as possible application in processes of drug development to combat COVID-19.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), a contemporary coronavirus, has impacted global economic activity and has a high transmission rate. As a result of the virus's severe medical effects, developing effective vaccinations is vital. Plant-derived metabolites have been discovered as potential SARS-CoV-2 inhibitors. The SARS-CoV-2 main protease (Mpro) is a target for therapeutic research because of its highly conserved protein sequence. Gas chromatography-mass spectrometry (GC-MS) and molecular docking were used to screen 34 compounds identified from Leucas zeylanica for potential inhibitory activity against the SARS-CoV-2 Mpro. In addition, prime molecular mechanics-generalized Born surface area (MM-GBSA) was used to screen the compound dataset using a molecular dynamics simulation. From molecular docking analysis, 26 compounds were capable of interaction with the SARS-CoV-2 Mpro, while three compounds, namely 11-oxa-dispiro[4.0.4.1]undecan-1-ol (-5.755 kcal/mol), azetidin-2-one 3,3-dimethyl-4-(1-aminoethyl) (-5.39 kcal/mol), and lorazepam, 2TMS derivative (-5.246 kcal/mol), exhibited the highest docking scores. These three ligands were assessed by MM-GBSA, which revealed that they bind with the necessary Mpro amino acids in the catalytic groove to cause protein inhibition, including Ser144, Cys145, and His41. The molecular dynamics simulation confirmed the complex rigidity and stability of the docked ligand-Mpro complexes based on the analysis of mean radical variations, root-mean-square fluctuations, solvent-accessible surface area, radius of gyration, and hydrogen bond formation. The study of the postmolecular dynamics confirmation also confirmed that lorazepam, 11-oxa-dispiro[4.0.4.1]undecan-1-ol, and azetidin-2-one-3, 3-dimethyl-4-(1-aminoethyl) interact with similar Mpro binding pockets. The results of our computerized drug design approach may assist in the fight against SARS-CoV-2.
