RESUMEN

The European eel Anguilla anguilla has shown decreased recruitment in recent decades. Despite increasing efforts to establish species recovery measures, it is unclear if the decline was caused by reduced numbers of reproductive-stage silver eels reaching the spawning area, low early larval survival, or increased larval mortality during migration to recruitment areas. To determine if larval abundances in the spawning area significantly changed over the past three decades, a plankton trawl sampling survey for anguillid leptocephali was conducted in March and April 2011 in the spawning area of the European eel that was designed to directly compare to collections made in the same way in 1983 and 1985. The catch rates of most anguilliform leptocephali were lower in 2011, possibly because of the slightly smaller plankton trawl used, but the relative abundances of European eel and American eel, Anguilla rostrata, leptocephali were much lower in 2011 than in 1983 and 1985 when compared to catches of other common leptocephali. The leptocephali assemblage was the same in 2011 as in previous years, but small larvae of mesopelagic snipe eels, Nemichthys scolopaceus, which spawn sympatrically with anguillid eels, were less abundant. Temperature fronts in the spawning area were also poorly defined compared to previous years. Although the causes for low anguillid larval abundances in 2011 are unclear, the fact that there are presently fewer European and American eel larvae in the spawning area than during previous time periods indicates that decreased larval abundance and lower eventual recruitment begin within the spawning area.

BACKGROUND, AIM AND SCOPE: Nonylphenol (NP) can be detected in the aquatic environment all over the world. It is applied as a technical mixture of isomers of which 353-NP is the most relevant both in terms of abundance (about 20% of total mass) and endocrine potential. 353-NP is metabolised in sewage sludge. The aims of the present study were to determine and to compare the acute toxicity of t-NP, 353-NP and its metabolites as well as to discuss if the toxicity of 353-NP changes during degradation. MATERIALS AND METHODS: 353-NP and two of its metabolites were synthesised. The zebrafish embryo test was performed according to standard protocols. Several lethal and non-lethal endpoints during embryonal development were reported. NOEL, LOEL and EC50 were calculated. RESULTS: All tested compounds caused lethal as well as non-lethal malformations during embryo development. 353-NP showed a higher toxicity (EC50 for lethal endpoints 6.7 mg/L) compared to its metabolites 4-(3.5-dimethyl-3-heptyl)-2-nitrophenol (EC50 13.3 mg/L) and 4-(3,5-dimethyl-3-heptyl)-2-bromophenol (EC50 27.1 mg/L). DISCUSSION: In surface water, concentrations of NP are far below the NOEC identified by the zebrafish embryo test. However, in soils and sewage sludge, concentrations may reach or even exceed these concentrations. Therefore, sludge-treated sites close to surface waters should be analysed for NP and its metabolites in order to detect an unduly high contamination due to runoff events. CONCLUSIONS: The results of the present study point out that the toxicity of 353-NP probably declines during metabolisation in water, sediment and soil, but does not vanish since the major metabolites exhibit a clear toxic potential for zebrafish embryos. RECOMMENDATIONS AND PERSPECTIVES: Metabolites of environmental pollutants should be included in the ecotoxicological test strategy for a proper risk assessment.

Toxaphene, also known as camphechlor, is a persistent organochlorine pesticide of complex composition. It is technically produced by photochlorination of camphene with elemental chlorine gas under ultraviolet irradiation. In the present work, a novel, laboratory-scale synthesis using sulfuryl chloride as a chlorinating reagent is described. This approach allowed the degree of chlorination of the resulting mixtures to be arbitrarily adjusted by varying the reaction conditions. Both the compositions and the chlorine contents of the low- and high-chlorinated mixtures acquired using this method were similar to those of environmentally altered toxaphene and technical toxaphene, respectively. For comparison of these mixtures regarding toxicity, they were subjected to the zebrafish (Danio rerio) embryo test. Median effective concentrations (EC50s) were calculated based on the presence of lethal and nonlethal embryonic malformations. Surprisingly, low-chlorinated toxaphene, comprising compounds that also are present in environmentally transformed toxaphene, exhibited a twofold-higher toxicity (according to the EC50 for nonlethal effects) toward the test organisms compared with high-chlorinated toxaphene, the composition of which resembled that of the technical product. Although the effective concentrations in the embryo test were much higher than those in aquatic ecosystems burdened with toxaphene, the present results lead to the assumption that toxaphene is becoming more toxic during transformation in the environment. A decrease in the total amount of toxaphene during environmental breakdown would then be compensated for, at least in part, by the higher toxicity of weathered toxaphene in sediments, soils, and biota of contaminated ecosystems.

GOAL, SCOPE AND BACKGROUND: Ethoxyresorufin-O-deethylase (EROD) is a well-accepted marker for biological effects in fish and is, therefore, part of numerous monitoring programmes. EROD activity was measured in livers of dab (Limanda limanda) from the German Bight (North Sea) from 1995 to 2003. The aim of the present long-term study was (a) time trend monitoring of EROD activity of dab from the German Bight and (b) to elucidate the needs for a successful application of EROD activity as an early warning system. METHODS: EROD activities were determined fluorimetrically in dab liver microsomes, using resorufin as an external standard. The limit of detection (LD) and the limit of quantification (LQ) were calculated. Results were referred to protein concentrations. RESULTS AND DISCUSSION: EROD activities of 610 female dab caught in different seasons between January 1995 and August 2003 were analyzed individually. Activities varied from < LD to 1768 pmol/(min mg protein) and showed an annual cycle as well as significant differences between the years. Highest EROD activities were observed in early summer and lowest activities during the winter period. In autumn 2002, significantly elevated EROD activities were detected, possibly related to effects of the River Elbe flood event. Two scenarios with different EROD baseline data are presented to discuss the prerequisites for the use of EROD as a monitoring tool. The comparison of these scenarios underlines the importance of appropriate season-specific baseline data. CONCLUSION: The use of EROD as an early warning tool for contaminant effects in dab in the German Bight has different prospects during the year, because, due to the high background variability, elevated EROD activities are less easy to detect in spring/ summer than during the remaining times of the year. RECOMMENDATION AND OUTLOOK: The availability of site-specific data on the EROD baseline level, its random variation and its annual cycle is a necessary prerequisite for monitoring. If monitoring is to be carried out only for a limited time period of the year, a season with low background variability in EROD activity (autumn) should be chosen to avoid the need for a compensation of the temperature-triggered shift in sexual cycles and the resulting changes in EROD activity.

Organic extracts of marine sediments from the North Sea and the Baltic Sea were investigated with two toxicity assays. The comet assay based on the fish cell line Epithelioma papulosum cyprini (EPC) was applied to determine the genotoxic potential; zebrafish embryos (Danio rerio) were used to quantify the teratogenic potential of the samples. EC(50) values were calculated from dose-response curves for both test systems. Highest teratogenic and genotoxic effects normalised to total organic carbon (TOC) content were detected in sediment samples of different origins. Polychlorinated biphenyls (PCBs) and polycyclic aromatic hydrocarbons (PAHs) are not likely to be the causes of the observed effects, as demonstrated by a two-step fractionation procedure of selected extracts. The toxic potential was more pronounced in fractions having polarity higher than those possessed by PAHs and PCBs. The suitability of the two in vitro test systems for assessing genotoxic and teratogenic effects of marine sediment extracts could be demonstrated.
