RESUMEN

Electrooxidation of biomass-derived glycerol which is regarded as a main byproduct of industrial biodiesel production, is an innovative strategy to produce value-added chemicals, but currently showcases slow kinetics, limited Faraday efficiency, and unclear catalytic mechanism. Herein, we report high-efficiency electrooxidation of glycerol into formate via a Cu doped NiCo alloy catalyst supported on nickel foam (Cu-NiCo/NF) in a coupled system paired with nitrate reduction. The designed Cu-NiCo/NF delivers only 1.23 V vs. RHE at 10 mA cm-2, and a record Faraday efficiency of formate of 93.8%. The superior performance is ascribed to the rapid generation of NiIII-OOH and CoIII-OOH and favorable coupling of surface *O with reactive intermediates. Using Cu-NiCo/NF as a bifunctional catalyst, the coupled system synchronously produces NH3 and formate, showing 290 mV lower than the coupling of hydrogen evolution reaction, together with excellent long-term stability for up to 144 h. This work lays out new guidelines and reliable strategies from catalyst design to system coupling for biomass-derived electrochemical refinery.

RESEARCH QUESTION: What is the genetic cause of multiple congenital disabilities in a girl with a maternal balanced X-autosome translocation [t(X-A)]? Is preimplantation genetic testing (PGT), to distinguish non-carrier from euploid/balanced embryos and prioritize transfer, an effective and applicable strategy for couples with t(X-A)? DESIGN: Karyotype analysis, whole-exome sequencing and X inactivation analysis were performed for a girl with congenital cardiac anomalies, language impairment and mild neurodevelopmental delay. PGT based on next-generation sequencing after microdissecting junction region (MicroSeq) to distinguish non-carrier and carrier embryos was used in three couples with a female t(X-A) carrier (cases 1-3). RESULTS: The girl carried a maternal balanced translocation 46,X,t(X;1)(q28;p31.1). Whole-exome sequencing revealed no monogenic mutation related to her phenotype, but she carried a rare skewed inactivation of the translocated X chromosome that spread to the adjacent interstitial 1p segment, contrary to her mother. All translocation breakpoints in cases 1-3 were successfully identified and each couple underwent one PGT cycle. Thirty oocytes were retrieved, and 13 blastocysts were eligible for biopsy, of which six embryos had a balanced translocation and only four were non-carriers. Three cryopreserved embryo transfers with non-carrier status embryos resulted in the birth of two healthy children (one girl and one boy), who were subsequently confirmed to have normal karyotypes. CONCLUSIONS: This study reported a girl with multiple congenital disabilities associated with a maternal balanced t(X-A) and verified that the distinction between non-carrier and carrier embryos is an effective and applicable strategy to avoid transferring genetic and reproductive risks to the offspring of t(X-A) carriers.

PURPOSE: The purpose of this study is to summarize the clinical outcomes of apparently balanced chromosome rearrangement (ABCR) carriers in preimplantation genetic testing (PGT) cycles by next-generation sequencing following microdissecting junction region (MicroSeq) to distinguish non-carrier embryos from balanced carriers. METHODS: A retrospective study of 762 ABCR carrier couples who requested PGT for structural rearrangements combined with MicroSeq at the Reproductive and Genetic Hospital of CITIC-Xiangya was conducted between October 2014 and October 2019. RESULTS: Trophectoderm biopsy was performed in 4122 blastocysts derived from 917 PGT-SR cycles and 3781 blastocysts were detected. Among the 3781 blastocysts diagnosed, 1433 (37.9%, 1433/3781) were balanced, of which 739 blastocysts were carriers (51.57%, 739/1433) and 694 blastocysts were normal (48.43%, 694/1433). Approximately 26.39% of cycles had both carrier and normal embryo transfer, and the average number of biopsied blastocysts was 6.7. In the cumulative 223 biopsied cycles with normal embryo transfer, all couples chose to transfer the normal embryos. In the 225 cycles with only carrier embryos, the couples chose to transfer the carrier embryos in 169/225 (75.11%) cycles. A total of 732 frozen embryo transfer cycles were performed, resulting in 502 clinical pregnancies. Cumulatively, 326 babies were born; all of these babies were healthy and free of any developmental issues. CONCLUSION: Our study provides the first evaluation of the clinical outcomes of a large sample with ABCR carrier couples undergoing the MicroSeq-PGT technique and reveals its powerful ability to distinguish between carrier and non-carrier balanced embryos.

The MXenes attract great interest in catalytic and energy applications but suffer from inferior redox activity. An efficient strategy is presented to boost the intrinsic electrochemical activity of MXenes for electrocatalysis in various chemical environments by utilizing their plasmonic response to electromagnetic waves. The resulting significant thermoplasmonic effect lowers the endothermic enthalpy and potential barrier to the hydrogen evolution reaction (HER) in the Vis/near-IR region. Simultaneous hot-electron injection in a prolonged sub-femtosecond to picosecond timescale also remarkably facilitates interfacial charge transfer and decreases the activation energy of the reaction. Both effects boost the HER activity of various types of MXenes over fivefold with improved kinetics and Faradaic efficiency over a broad pH range.

BACKGROUND: Although the neuroprotective role of propofol has been identified recently, the regulatory mechanism associated with microRNAs (miRNAs/miRs) in neuronal cells remains to be poorly understood. We aimed to explore the regulatory mechanism of propofol in hypoxia-injured rat pheochromocytoma (PC-12) cells. METHODS: PC-12 cells were exposed to hypoxia, and cell viability and apoptosis were assessed by CCK-8 assay and flow cytometry assay/Western blot analysis, respectively. Effects of propofol on hypoxia-injured cells were measured, and the expression of miR-153 was determined by stem-loop RT-PCR. After that, whether propofol affected PC-12 cells under hypoxia via miR-153 was verified, and the downstream protein of miR-153 as well as the involved signaling cascade was finally explored. RESULTS: Hypoxia-induced decrease of cell viability and increase of apoptosis were attenuated by propofol. Then, we found hypoxia exposure up-regulated miR-153 expression, and the level of miR-153 was further elevated by propofol in hypoxia-injured PC-12 cells. Following experiments showed miR-153 inhibition reversed the effects of propofol on hypoxia-treated PC-12 cells. Afterwards, we found BTG3 expression was negatively regulated by miR-153 expression, and BTG3 overexpression inhibited the mTOR pathway and AMPK activation. Besides, hypoxia inhibited the mTOR pathway and AMPK, and these inhibitory effects could be attenuated by propofol. CONCLUSION: Propofol protected hypoxia-injured PC-12 cells through miR-153-mediataed down-regulation of BTG3. BTG3 could inhibit the mTOR pathway and AMPK activation.

Objective: To investigate the prevalence of echinococcosis in Yushu Prefecture of Qinghai Province in 2012. Methods: Two to three towns were selected in each of Chengduo, Nangqian, Qu malai, Yushu, Zaduo and Zhiduo Counties from June to August in 2012. Ultrasound examination was conducted for residents aged over 1 year, and ELISA was performed to detect serum antibody against Echinococcus. Visceral dissection was performed to detect hydatid infection in rodents and livestock. ELISA was used to detect Echinococcus antigen in collected dog feces. Results: A total of 7 025 residents received ultrasound examination, of whom 319 showed hydatid cysts with a morbidity rate of 4.54%. ELISA showed a serum antibody positive rate of 16.38% (457/2 790). The mobidity of hydatid disease was highest in Chengduo County (7.41%, 181/2 444), and the rate of serum antibody was highest in Yushu County (23.18%, 127/548). The morbidity and serum antibody in males were 3.91% (118/3 018) and 13.93% (172/1 235) respectively, and those in females were 5.02% (201/4 007) and 18.33% (285/1 555). In terms of age distribution, the morbidity was relatively higher in residents of 60- (8.39%, 38/453) and 40- years (6.61%, 67/1 014); and the rate of serum antibody was highest in residents over 70 years (33.93%, 19/56). In terms of occupation, the morbidity was relatively higher in herdsmen (5.28%, 252/4 777), Herdsmen-peasants (6.52%, 24/368), and religious workers(3.37%, 11/326), while the rate of serum antibody was relatively higher in children(24%, 6/25), religious workers (18.79%, 31/165) and herdsmen(18.34%, 328/1 788). In terms of education level, the morbidity and the rate of serum antibody were both highest in the uneducated(5.04%, 41/4 779; 18.34%, 359/1 958, respectively). In terms of residential pattern, the morbidity and the rate of serum antibody were both highest in those who were settled in winter and nomadic in summer (8.25%, 227/2 753; 19.48%, 158/811, respectively). There were significant differences in the morbidity and the rate of serum antibody in aspects of residential region, sex, age, occupation, education level and residential pattern (P<0.05 or P<0.01). In 872 rodents detected, the Echinococcus hydatid rate was 0.46% (4/872), while in 809 cattle and sheep detected, the Echinococcus hydatid rate was 10.14% (82/809). The fecal antigen positive rate in 838 samples of dog feces was 10.74%(90/838). Conclusion: It shows a high morbidity of hydatid diesease and serum antibody positive rate in residents, a high Echinococcus hydatid rate in cattle and sheep, and a high fecal antigen positive rate in dogs in Yushu Prefecture.

RESUMEN

MXenes are promising materials for electrocatalysis due to their excellent metallic conductivity, hydrophilicity, high specific surface area, and excellent electrochemical properties. Herein, we summarize the recent advancement of MXene-based materials for electrocatalysis and highlight their key challenges and opportunities. In particular, this review emphasizes on the major design principles of MXene-based electrocatalysts, including (1) coupling MXene with active materials or heteroatomic doping to create highly active synergistic catalyst sites; (2) construction of 3D MXene structure or introducing interlayer spacers to increase active areas and form fast mass-charge transfer channel; and (3) protecting edge of MXene or in situ transforming the surface of MXene to stable active substance that inhibits the oxidation of MXene and then enhances the stability. Consequently, MXene-based materials exhibit outstanding performance for a variety of electrocatalytic reactions. Finally, the key challenges and promising prospects of the practical applications of MXene-based electrocatalysts are briefly proposed.

Background: The effects of heart failure (HF) on cortical brain structure remain unclear. Therefore, the present study aimed to investigate the causal effects of heart failure on cortical structures in the brain using Mendelian randomization (MR) analysis. Methods: We conducted a two-sample MR analysis utilizing genetically-predicted HF trait, left ventricular ejection fraction (LVEF), and N-terminal prohormone brain natriuretic peptide (NT-proBNP) levels to examine their effects on the cortical surface area (SA) and thickness (TH) across 34 cortical brain regions. Genome-wide association study summary data were extracted from studies by Rasooly (1,266,315 participants) for HF trait, Schmidt (36,548 participants) for LVEF, the SCALLOP consortium (21,758 participants) for NT-proBNP, and the ENIGMA Consortium (51,665 participants) for cortical SA and TH. A series of MR analyses were employed to exclude heterogeneity and pleiotropy, ensuring the stability of the results. Given the exploratory nature of the study, p-values between 1.22E-04 and 0.05 were considered suggestive of association, and p-values below 1.22E-04 were defined as statistically significant. Results: In this study, we found no significant association between HF and cortical TH or SA (all p > 1.22E-04). We found that the HF trait and elevated NT-proBNP levels were not associated with cortical SA, but were suggested to decrease cortical TH in the pars orbitalis, lateral orbitofrontal cortex, temporal pole, lingual gyrus, precuneus, and supramarginal gyrus. Reduced LVEF was primarily suggested to decrease cortical SA in the isthmus cingulate gyrus, frontal pole, postcentral gyrus, cuneus, and rostral middle frontal gyrus, as well as TH in the postcentral gyrus. However, it was suggested to causally increase in the SA of the posterior cingulate gyrus and medial orbitofrontal cortex and the TH of the entorhinal cortex and superior temporal gyrus. Conclusion: We found 15 brain regions potentially affected by HF, which may lead to impairments in cognition, emotion, perception, memory, language, sensory processing, vision, and executive control in HF patients.

Mechanical thrombectomy is the gold standard in treating acute ischemic stroke complicated by large vessel occlusion. However, there are limited studies on repeated mechanical thrombectomy in acute ischemic stroke. In this case, we report a 68-year-old male with atrial fibrillation who developed sudden left limb weakness and motor aphasia for 6 hours, and his National Institutes of Health Stroke Scale (NIHSS) score was 10. Computed tomography angiography (CTA) showed occlusion of the right internal carotid artery, and mechanical thrombectomy was performed immediately. The patient's neurologic disability was utterly relieved, and the NIHSS score returned to 0. At 30 hours postoperatively, he again developed left limb weakness with motor aphasia. The NHISS Score was 11, and the CTA showed that the right internal carotid artery was re-occluded. After computed tomography perfusion evaluation, the patient underwent mechanical thrombectomy again, and the etiological examination confirmed Cardioembolism. Anticoagulation therapy was commenced 1 week post-thrombectomy. The prognosis of the patients was good.

Seawater electrolysis represents a potential solution to grid-scale production of carbon-neutral hydrogen energy without reliance on freshwater. However, it is challenged by high energy costs and detrimental chlorine chemistry in complex chemical environments. Here we demonstrate chlorine-free hydrogen production by hybrid seawater splitting coupling hydrazine degradation. It yields hydrogen at a rate of 9.2 mol h-1 gcat-1 on NiCo/MXene-based electrodes with a low electricity expense of 2.75 kWh per m3 H2 at 500 mA cm-2 and 48% lower energy equivalent input relative to commercial alkaline water electrolysis. Chlorine electrochemistry is avoided by low cell voltages without anode protection regardless Cl- crossover. This electrolyzer meanwhile enables fast hydrazine degradation to ~3 ppb residual. Self-powered hybrid seawater electrolysis is realized by integrating low-voltage direct hydrazine fuel cells or solar cells. These findings enable further opportunities for efficient conversion of ocean resources to hydrogen fuel while removing harmful pollutants.

An epidemiological investigation on echinococcosis was made in Jiuzhi County of Qinghai Province, western China. Ultrasonography and an indirect hemagglutination test revealed a morbidity of 8.0% (124/1,549) and a seroprevalence of 25.8% (287/1,113), respectively, in the Tibetan population. The morbidity in herdsmen (16.6%) and Buddhist priests (15%) was significantly higher than that in other occupation groups (3.2%), and it was higher in females (9.8%) than in males (6.2%). The ultrasound images showed a coexistence of cystic echinococcosis (CE) and alveolar echinococcosis (AE), occupying 69 and 31% of the cases, respectively. An Echinococcus Western blot assay was performed as a serological backup test for differentiating CE and AE. The assay revealed that serum samples from most cases with a positive AE image showed a specific antibody against antigen bands at 16/18 kDa. Autopsy proved that 9 out of 12 stray dogs were infected with Echinococcus granulosus (n = 8) and E. multilocularis (n = 1). Inspection at the abattoirs demonstrated a hydatid rate of 78.5% in yaks and 82.6% in sheep. The data indicate that Jiuzhi County is an important endemic area for both CE and AE, in both human and animal populations.

Local inhabitants in 8 towns/townships of the counties Yushu, Zhiduo and Chengduo were examined with serology and ultrasound in 2006. Among 2 251 people tested by indirect hemagglutination (IHA), 207 showed anti-hydatid IgG positive (9.2%). Ultrasound examination found 106 cases out of 2581 people, with a morbidity of 4.1%. Females showed higher sero-positive rate (11.3%) and morbidity rate (5.0%) than males (6.6% and 3.0% respectively). The highest sero-positive rate was in the group of 40-49 years old (16.4%). The morbidity rate increased with age, with the highest rate (15.5%) in the group of 60 years old and above. Occupationally, those involved in semi-agriculture and semi-animal husbandry showed highest sero-positive rate and morbidity rate, 21.5% and 11.4% respectively. The survey demonstrated that the prevalence of hydatid disease in human population is at a high level in Yushu Tibetan Autonomous Prefecture.

The MXene combining high conductivity, hydrophilic surface, and wide chemical variety has been recognized as a rapidly rising star on the horizon of two-dimensional (2D) material science. However, strong tendency to intersheet aggregate via van der Waals force represents a major problem limiting the functionalities, processability, and performance of MXene-based material/devices. We report a capillary-forced assembling strategy for processing MXene to hierarchical 3D architecture with geometry-based high resistance to aggregation. Aggregate-resistant properties of 3D MXene not only double the surface area without loss of the intrinsic properties of MXene but also render the characteristics such as kinetics-favorable framework, high robustness, and excellent processability in both solution and solid state. Synergistically coupling the 3D MXene with electrochemically active phases such as metal oxide/phosphides, noble metals, or sulfur yields the hybrid systems with greatly boosted active surface area, charge-transfer kinetics, and mass diffusion rate. Specifically, the CoP-3D MXene hybrids exhibit high electrocatalytic activity toward oxygen and hydrogen evolution in alkaline electrolyte. As a bifunctional electrocatalyst, they exhibit superior cell voltage and durability to combined RuO2/Pt catalysts for overall water splitting in basic solution, highlighting the great promise of aggregation-resistant 3D MXene in the development of high-performance electrocatalysts.

Powerful yet thinner lithium-ion batteries (LIBs) are eagerly desired to meet the practical demands of electric vehicles and portable electronic devices. However, the use of soft carbon materials in current electrode design to improve the electrode conductivity and stability does not afford high volumetric capacity due to their low density and capacity for lithium storage. Herein, we report a strategy leveraging the MXene with superior conductivity and density to soft carbon as matrix and additive material for comprehensively enhancing the power capability, lifespan, and volumetric capacity of conversion-type anode. A kinetics favorable 2D nanohybrid with high conductivity, compact density, accumulated pseudocapacitance, and diffusion-controlled behavior is fabricated by coupling Ti3C2 MXene with high-density molybdenum carbide for fast lithium storage over 300 cycles with high capacities. By replacing the carbonaceous conductive agent with Ti3C2 MXene, the electrodes with better conductivity and dramatically reduced thickens could be further manufactured to achieve 37-40% improvement in capacity retention and ultra-long life of 5500 cycles with extremely slow capacity loss of 0.002% per cycle at high current rates. Ultrahigh volumetric capacity of 2460 mAh cm-3 could be attained by such MXene-based electrodes, highlighting the great promise of MXene in the development of high-performance LIBs.

The survey was carried out in July, 2006 in Zhiduo County. The IHA and ELISA positive rate in human population was 4.5% (42/933) and 8.2% (76/931) respectively. Ultrasonography revealed a morbidity of 3.4% (33/979) with 3.2% Echinococcus granulosus and 0.2% of E. multilocularis respectively. Animal dissection showed an infection rate of 15.1% (14/93) in pikas with one infected by E. shiquicus proved by molecular biology. Coproantigen rate by ELISA was 62% (12/193) in dogs and 35.7% (5/14) in wolves. The results indicated that Zhiduo County is a mixed endemic area for echinococcosis.

The MXenes combining hydrophilic surface, metallic conductivity and rich surface chemistries represent a new family of 2D materials with widespread applications. However, their poor oxygen resistance causes a great loss of electronic properties and surface reactivity, which significantly inhibits the fabrication, the understanding of the chemical nature and full exploitation of the potential of MXene-based materials. Herein we report a facile carbon nanoplating strategy for efficiently stabilizing the MXenes against structural degradation caused by spontaneous oxidation, which provides a material platform for developing MXene-based materials with attractive structure and properties. Hierarchical MoS2 /Ti3 C2 -MXene@C nanohybrids with excellent structural stability, electrical properties and strong interfacial coupling are fabricated by assembling carbon coated few-layered MoS2 nanoplates on carbon-stabilized Ti3 C2 MXene, exhibiting exceptional performance for Li storage and hydrogen evolution reaction (HER). Remarkably, ultra-long cycle life of 3000 cycles with high capacities but extremely slow capacity loss of 0.0016% per cycle is achieved for Li storage at a very high rate of 20 A g-1 . They are also highly active HER electrocatalyst with very positive onset potential, low overpotential and long-term stability in acidic solution. Superb properties highlight the great promise of MXene-based materials in cornerstone applications of energy storage and conversion.

Preimplantation genetic diagnosis (PGD) is widely applied in reciprocal translocation carriers to increase the chance for a successful live birth. However, reciprocal translocation carrier embryos were seldom discriminated from the normal ones mainly due to the technique restriction. Here we established a clinical applicable approach to identify precise breakpoint of reciprocal translocation and to further distinguish normal embryos in PGD. In the preclinical phase, rearrangement breakpoints and adjacent single nucleotide polymorphisms (SNPs) were characterized by next-generation sequencing following microdissecting junction region (MicroSeq) from 8 reciprocal translocation carriers. Junction-spanning PCR and sequencing further discovered precise breakpoints. The precise breakpoints were identified in 7/8 patients and we revealed that translocations in 6 patients caused 9 gene disruptions. In the clinical phase of embryo analysis, informative SNPs were chosen for linkage analyses combined with PCR analysis of the breakpoints to identify the carrier embryos. From 15 blastocysts diagnosed to be chromosomal balanced, 13 blastocysts were identified to be carriers and 2 to be normal. Late prenatal diagnoses for five carriers and one normal fetus confirmed the carrier diagnosis results. Our results suggest that MicroSeq can accurately evaluate the genetic risk of translocation carriers and carrier screen is possible in later PGD treatment.
