RESUMEN

Nacre plays an important role in bionic design due to its light weight, high strength, and structure-function integration. The key to elucidate its reinforcing and toughening mechanisms is to truly characterize its multi-layer structure and properties. In this work, the dynamic impact responses of graphene reinforced polymer nanocomposites with a unique brick-and-mortar structure are investigated using coarse-grained molecular dynamics simulations, in which the interfacial coarse-grained force field between graphene and the polymer matrix is derived by the energy matching approach. The influences of various geometrical parameters on dynamic impact responses of the nanocomposites are studied, including the interlayer distance, lateral distance, and number of graphene layers. The results demonstrate that the impact resistance of the nacre-like structure can be significantly improved by tuning the geometrical parameters of graphene layers. It is also found that the chain scission and interchain disentanglement of polymer chains are the main failure mechanisms during the perforation failure process as compared to the stretching and breaking of bonds. In addition, the microstructure analysis is performed to deeply interpret the deformation and damage mechanisms of the nanocomposites during impact. This study could be helpful for the rational design and preparation of graphene reinforced nacre-like nanocomposites with high impact resistance.

RESUMEN

Polyimide (PI), due to its exceptional performance, is commonly utilized in spacecraft. However, when such polymers are used in spacecraft navigating low Earth orbit, they are exposed to atomic oxygen (AO) that can cause the polymer to decompose. A protective coating method is a more effective way to safeguard the polymer from erosion caused by AO. This study employs the molecular dynamics simulation based on the reaction force field to investigate the protective effects of various coatings, including polydimethylsiloxane (PDMS), graphene (Gr), polytetrafluoroethylene (PTFE), and the (0 0 1), (0 1 1), and (1 1 1) surfaces of SiO2. The results indicate that the protective performance of the (0 1 1) surface is superior to that of the (0 0 1) and (1 1 1) surfaces. Moreover, protective coatings are classified into three categories based on different protective mechanisms: rebound, absorption, and sacrificial. The protective effectiveness of coatings depends on their anti-AO performance and ability to combine with the substrate. Gr displays exceptional anti-AO properties and can effectively shield the substrate from AO erosion. Silicone-based coatings have a superior ability to adhere to PI substrates, and PDMS is an excellent choice for protective coatings. This paper offers guidance for the protective coating method of PIs against AO erosion.

RESUMEN

Hepatocellular carcinoma (HCC) is a malignant tumor, which seriously threatens the life of patients. LncRNA SLC7A11-AS1 was reported to be abnormally expressed in HCC. Here, the functions and relative molecular regulatory mechanism of SLC7A11-AS1 in HCC were investigated. Nude mice and HCC cells were used as the experimental subjects. Knockdown or overexpression of exogenous genes was conducted in HCC cells. RT-qPCR, IHC, and western blot were employed to evaluate the abundance of genes and proteins. The malignant behaviors were evaluated using CCK-8, clone formation, wound-healing, and Transwell. The locations of SLC7A11-AS1 and KLF9 in cells were determined by FISH and IF assays. The total m6A level was evaluated by dot-blot assay. m6A modification of SLC7A11-AS1 was detected using RNA MeRIP. The interactions among molecules were validated by RIP, ChIP, dual luciferase reporter assay, and co-IP. SLC7A11-AS1 was elevated apparently in HCC cells and HCC tissues from mice. SLC7A11-AS1 silencing could suppress HCC progression, which was validated in in vivo and in vitro experiments. Furthermore, METTL3 mediated m6A modification of SLC7A11-AS1 to elevate its expression. In addition, SLC7A11-AS1 downregulated KLF9 expression by affecting STUB1-mediated ubiquitination degradation and KLF9 enhanced PHLPP2 expression to inactivate the AKT pathway. Eventually, rescue experiments revealed that KLF9 knockdown abolished SLC7A11-AS1 silencing-mediated suppression of HCC progression in vivo and in vitro. Our results unveiled that m6A-modified SLC7A11-AS1 promoted HCC progression by regulating the STUB1/KLF9/PHLPP2/AKT axis, indicating that targeting SLC7A11-AS1 might alleviate HCC progression.

RESUMEN

A visible-light-induced persulfate-promoted cascade phosphorylation/cyclization reaction to access various phosphorylated pyrrolo[1,2-a]indolediones under mild conditions was developed. Notably, the transformation was carried out with diethyl carbonate/H2O as a green medium at room temperature. More impressively, traditional metal catalysts and photocatalysts could be effectively avoided. The reactions are simple to operate, easy to scale up, and have good functional group tolerance.

RESUMEN

Poly(vinylidene fluoride) (PVDF) is a kind of important piezoelectric polymer used in spacecraft industry. But the atomic oxygen (AO) is the most abundant element in the low Earth orbit (LEO) environment. AO collision degradation is an important issue in the application of PVDF on spacecrafts. To investigate the erosion behaviors of PVDF during AO impacts and how to improve the stability of PVDF against AO impacts, the temperature evolution, mass loss, and erosion yields of neat PVDF, neat polyhedral oligomeric silsesquioxanes compound (3,3,3-trifluoropropyl)8Si8O12 (FP-POSS) and the PVDF/FP-POSS composite under AO impacts, as well as some key disintegrated structures and separated chemical compositions, were researched using the molecular dynamics (MD) simulations and the reactive ReaxFF force field. The simulation erosion yield result of PVDF is very close to the experiment results, which shows our simulations are reliable. The results of the temperature evolution, mass loss, and erosion yield of three materials show that the antierosion performance of PVDF is not outstanding. However, incorporating FP-POSS into PVDF matrix enhances the stability of PVDF against AO impact greatly and reduces the temperature rise, mass loss, and the erosion yield of PVDF rapidly. A detailed analysis on the flight chemical compositions and key snapshots of the structures reveals that the erosion process on PVDF and PVDF/FP-POSS is continuous and should be derived from the same PVDF matrix in two materials. In contrast, the erosion process on FP-POSS is stepped. The erosion will not take place until the number of AO reaches a specific value. There is a barrier for the erosion of high-energy AO because of the stable cagelike Si-O frame in FP-POSS molecules. This should be chiefly responsible for the high stability of FP-POSS and the reinforcement mechanism of FP-POSS on PVDF against AO impacts. This work is helpful for people to understand the erosion details of PVDF and POSS and provides valuable information to design effective protective structure for PVDF against AO impacts in LEO environment.

RESUMEN

This study aimed to analyze the polymorphism of the vasoactive intestinal peptide receptor-1 (VIPR-1) gene and its association with growth traits in quail using the PCR-RFLP and sequencing techniques. Genomic DNA was extracted from blood samples of 36 female Savimalt (SV) quails and 49 female French Giant (FG) quails. Growth traits were measured and used for VIPR-1 gene analysis, as body weight (BW), tibia length (TL), chest width (CW), chest depth (CD), sternum length (SL), body length (BL), and tibia circumference (TC). The results showed that 2 SNPs (BsrD I and HpyCH4 IV) were detected in exon 4 to 5 and exon 6 to 7 of the VIPR-1 gene, respectively. The results of association showed that the BsrD I site was not significantly associated with growth traits at 3 or 5 wk of age in the SV strain (P < 0.05), while the BsrD I site was significantly associated with BL at 3 or 5 wk of age in FG (P < 0.05). The HpyCH4 IV site was significantly associated with TL, CW, CD, SL, and BL at 3 wk of age in the SV strain (P < 0.05), while the HpyCH4 IV site was significantly correlated with BW, CW, SL, and BL at 5 wk of age in SV (P < 0.05). The HpyCH4 IV site was significantly associated with TL and TC at 3 wk of age in FG (P < 0.05), while the HpyCH4 IV site was significantly associated with TC at 5 wk of age in FG (P < 0.05). Four haplotype combinations based on 2 SNPs showed significantly association with BW, CW, CD, SL, BL, and TC at 3 or 5 wk of age in SV (P < 0.05). There was not significant association between 3 haplotype combinations with growth trait at 3 or 5 wk of age in FG (P > 0.05). In conclusion, the VIPR-1 gene could be used as a molecular genetic marker to improve growth traits in quail.

RESUMEN

Insulin-like growth factor 1 receptor (IGF-1R) gene is the main effector of insulin-like growth factor (IGF), which plays an important role in growth, development and reproduction of the animal organism. This study aimed to investigate the association of IGF-1R gene single nucleotide polymorphisms (SNPs) with egg quality and carcass traits of quail by direct sequencing. In this study, genomic DNA was extracted from quail blood samples of 46 Chinese yellow (CY) quail, 49 Beijing white (BW) quail and 48 Korean (KO) quail strains. Egg quality and carcass traits were measured and used for IGF-1R gene analysis in 3 quail strains. The results showed that 2 SNPs (A57G and A72T) of the IGF-1R gene were detected in 3 quail strains. The A57G was significantly associated with yolk width (YWI) in BW strain (P < 0.05). Whereas A72T was significantly associated with egg shell thickness (EST) in BW strain (P < 0.05), and significantly associated with egg weight (EW), egg long (EL), and egg short (ES) in KO strain (P < 0.05). Haplotypes based on 2 SNPs showed significant effect on EST in 3 quail strains (P < 0.05), it also has a significant effect on EW in KO strain (P < 0.05). Meanwhile, A72T was significantly associated with liver weight (LW) and dressing percentage (DP) in 3 strains (P < 0.05). Haplotypes showed significant effect on LW (P < 0.05). Therefore, the IGF-1R gene may be a molecular genetic marker to improve egg quality and carcass traits in quails.

RESUMEN

This study aimed to identify InDels from the FTO and PLIN1 genes and to analyze their association with morphometric traits in Hu sheep (HS), Dupor sheep (DS), and Small Tail Han sheep (STHS). The FTO and PLIN1 genes were genotyped using the insertion/deletion (InDel) method. A one-way ANOVA with SPSS 26.0 software (IBM Corp, Armonk, NY, USA) was used to assess the effect of the InDel FTO and PLIN1 genes on morphometric traits. The results revealed significant associations between certain InDels and the morphometric traits in different breeds of sheep. Specifically, FTO-2 was significantly associated with cannon circumference (CaC) in HS rams and body height (BoH) in HS ewes (p < 0.05). FTO-2 was also significantly associated with chest width (ChW), CaC, head length (HeL), and coccyx length (CoL) in the STHS breed (p < 0.05). FTO-3 showed significant associations with BoH in HS rams and BoH, back height (BaH), ChW, and chest depth (ChD) in HS ewes (p < 0.05). FTO-3 was also significantly associated with ChW in the DS and STHS breeds (p < 0.05). FTO-5 was significantly associated with body weight (BoW) in the DS breed and BoH in the STHS breed (p < 0.05). Furthermore, PLIN1 was significantly related to BoW in the DS breed and was significantly associated with CoL and forehead width (FoW) in the STHS breed (p < 0.05). In conclusion, the study suggested that InDels in the FTO and PLIN1 genes could provide practical information to improve morphometric traits in sheep breeding.

RESUMEN

OBJECTIVES: Paraneoplastic antigen Ma family (PNMA) is dysregulated in the pathological development of various cancers. However, the actions of PNMA member 5 (PNMA5) in cancers are still unknown. The aim of this study was to explore the biological actions of PNMA5 and its implication in epithelial-mesenchymal transition (EMT) during the progression of colorectal cancer (CRC). METHODS: Immunohistochemical staining, western blot and qPCR were used to explore PNMA5 expression in colorectal cancer tissues and cells. In addition, western blot, MTT assays, Colony formation assay, wound-healing, and transwell cell invasion assays were used to investigate the effects of PNMA5 on EMT in colorectal cancer. The lung metastasis models and xenografts in nude mice were established to explore the roles of PNMA5 in vivo. RESULTS: It was found that the expression level of PNMA5 in colorectal cancer tissues was significantly up regulated compared to that in the adjacent tissues. The overall survival rates of patients with a higher PNMA5 expression were markedly decreased. In addition, knockdown of PNMA5 expression decreased the proliferation, invasion and migration of both HCT-15 and HCT-116 cells. PNMA5 expression was found to be positively associated with the expression of C-myc, CyclinD1, Ki67, N-cadherin, zinc finger E-box binding homeobox 1 and vimentin, and negatively associated with E-cadherin. It was also found that PNMA5 knockdown attenuated TGF-ß-induced EMT in colorectal cancer cells. Finally, it was demonstrated that PNMA5 accelerated colorectal cancer cell proliferation, invasion and migration in vivo. CONCLUSION: The results revealed that PNMA5 increased cellular proliferation, invasion and migration in colorectal cancer. PNMA5 plays a key role in promoting CRC carcinogenesis and progression for patients with CRC.

RESUMEN

3-(2-(Ethynyl)phenyl)quinazolinones were designed and synthesized as a class of novel and efficient skeletons for phosphorylation/cyclization reactions. Under visible light irradiation, a series of phosphorylated quinolino[2,1-b]quinazolinones (35 examples, up to 87% yield) were first synthesized from 3-(2-(ethynyl)phenyl)quinazolinones and diarylphosphine oxides by using 4CzIPN as a photocatalyst under mild conditions. This reaction was also applicable under sunlight irradiation. Moreover, the reaction efficiency could be significantly improved under continuous-flow conditions.

RESUMEN

A general and metal-free visible-light-induced decarboxylative arylation procedure at room temperature was described for the construction of acylated heterocyclic derivatives, such as benzimidazo/indolo[2,1-a]isoquinolin-6(5H)-ones, aroylazaspiro[4.5]trienones, thioflavones, and so on. This practical arylation procedure was conducted by using 2,4,5,6-tetra(9H-carbazol-9-yl)isophthalonitrile (4CzIPN) as a photocatalyst under mild conditions, which avoided the use of an additional base, traditional heating, and metal reagents.
