BACKGROUND: In recent years, anthropogenic activities have released heavy metals and polluted the aquatic environment. This study investigated the ability of the silica-stabilized magnetite (Si-M) nanocomposite materials to dispose of lead nitrate (Pb(NO3)2) toxicity in Nile tilapia and African catfish. RESULTS: Preliminary toxicity tests were conducted and determined the median lethal concentration (LC50) of lead nitrate (Pb(NO3)2) to Nile tilapia and African catfish to be 5 mg/l. The sublethal concentration, equivalent to 1/20 of the 96-hour LC50 Pb(NO3)2, was selected for our experiment. Fish of each species were divided into four duplicated groups. The first group served as the control negative group, while the second group (Pb group) was exposed to 0.25 mg/l Pb(NO3)2 (1/20 of the 96-hour LC50). The third group (Si-MNPs) was exposed to silica-stabilized magnetite nanoparticles at a concentration of 1 mg/l, and the fourth group (Pb + Si-MNPs) was exposed simultaneously to Pb(NO3)2 and Si-MNPs at the same concentrations as the second and third groups. Throughout the experimental period, no mortalities or abnormal clinical observations were recorded in any of the treated groups, except for melanosis and abnormal nervous behavior observed in some fish in the Pb group. After three weeks of sublethal exposure, we analyzed hepatorenal indices, oxidative stress parameters, and genotoxicity. Values of alkaline phosphatase (ALP), gamma-glutamyl transferase (GGT), urea, and creatinine were significantly higher in the Pb-intoxicated groups compared to the control and Pb + Si-MNPs groups in both fish species. Oxidative stress parameters showed a significant decrease in reduced glutathione (GSH) concentration, along with a significant increase in malondialdehyde (MDA) and protein carbonyl content (PCC) concentrations, as well as DNA fragmentation percentage in the Pb group. However, these values were nearly restored to control levels in the Pb + Si-MNPs groups. High lead accumulation was observed in the liver and gills of the Pb group, with the least accumulation in the muscles of tilapia and catfish in the Pb + Si-MNPs group. Histopathological analysis of tissue samples from Pb-exposed groups of tilapia and catfish revealed brain vacuolation, gill fusion, hyperplasia, and marked hepatocellular and renal necrosis, contrasting with Pb + Si-MNP group, which appeared to have an apparently normal tissue structure. CONCLUSIONS: Our results demonstrate that Si-MNPs are safe and effective aqueous additives in reducing the toxic effects of Pb (NO3)2 on fish tissue through the lead-chelating ability of Si-MNPs in water before being absorbed by fish.

Parasites pose significant challenges to aquaculture and fisheries industries. Our study focuses on the Polyonchobothrium magnum and African catfish to address a potential health issue in aquaculture, explore host-parasite interactions that can help develop effective management practices to ensure fish health and industry sustainability. P. magnum was isolated from the stomach of African catfish (Clarias gariepinus) as the primary site of infection, with a prevalence of 10%. Most affected fish were heavily infected (8 out of 10). Infection was confirmed by sequencing the PCR-targeted region of the nicotinamide adenine dinucleotide dehydrogenase subunit 1 (ND1) gene, along with light and scanning electron microscopes. The parasite had an elongated scolex with deep bothria, a prominent apical disc wider than the scolex itself, and a four-lobed appearance. The scolex contained a central rostellum divided into two semicircles, bearing 26-30 hooks, with an average of 28. The apical disc had large hooks arranged in four quadrants, with 6-8 hooks each, averaging 7 per quadrant. No neck was observed. Phylogenetic analysis of our sequence showed a 100% match with isolates from Guangzhou, China. In infected fish, the anterior kidney showed increased expression levels of nuclear factor kappa B and lysozyme, but decreased levels of in major histocompatibility complex antigen II. Plasma analysis revealed a significant drop in superoxide dismutase, a rise in interleukin-1 beta, and lower IgM levels compared to non-infected controls. Non-infected fish displayed greater gut microbiota diversity, with dominant families including Moraxellaceae, Enterobacteriaceae, Fusobacteriaceae, and Caulobacteraceae, and prevalent genera such as Acinetobacter, Cetobacterium, and Brevundimonas. In contrast, infected fish exhibited very low diversity, with significantly higher proportions of Enterobacteriaceae (45.99%) and Aeromonadaceae (41.79%) compared to non-infected fish, which had 13.76% and 3.64% respectively. Cetobacterium somerae was prevalent in non-infected fish, while infected fish harboured Aeromonas fluvialis, Plesiomonas shigelloides, and Gallaecimonas xiamenensis. Overall, P. magnum disrupted the immune status and gut microbiota of the host, thereby impacting its health.

Fish parasitology contributes to our understanding of the potential risks posed by diverse groups of parasitic organisms on fish stocks in either wild and culture systems. This study was conducted in May 2023 and aimed at assessing the diversity of endohelminths in the invasive North African catfish Clarias gariepinus (Burchell, 1822) obtained from two freshwater lakes, Naivasha and Ol'Bolossat, in Kenya. Parasitological examination of 66 and 35 fish samples collected from the two lakes respectively was achieved using light and scanning electron microscopy methods. Results revealed endohelminth diversity broadly classified as four digeneans, two nematodes, and one cestode. Seven taxa of endohelminths were found in C. gariepinus samples, but only four of these taxa could be identified up to the species level. Six of the taxa (Diplostomum sp., Tylodelphys mashonense, Plagiorchioidea sp., Paracamallanus cyathopharynx, Contracaecum sp., and Tetracampos ciliotheca) were common in samples from the two lakes. Glossidium pedatum only occurred in samples from Lake Ol'Bolossat. Parasite prevalence ranged from 8.6 (T. mashonense) to 100% (Diplostomum sp., T. ciliotheca, and Contracaecum sp.) and mean intensity from 1.4 (T. mashonense) to 16.9 (Diplostomum sp.). The diversity and richness indices were comparatively higher in fish samples from Lake Ol'Bolossat and attributed to the occurrence of G. pedatum in the Ol'Bolossat. However, parasitic infestation of fish samples from the two lakes depicted close similarity, both in diversity and prevalence. These findings form an important baseline data for further follow-up studies, and they suggest the need for further molecular analyses to fully describe three of the taxa only identified up to the genus level.

The effects of dietary Lagnaria breviflora leaves extract (LBLE) on the growth performance, feed utilisation and haematological parameters of juveniles African catfish, Clarias gariepinus (mean = 8.45 ± 0.6 g) raised in a flow water (mean temperature 26.7°C, mean pH 7.52, mean dissolved oxygen 6.05 mg/L) were assessed in this study. Diets (400 g/kg crude protein; 4631.5 kcal/kg gross energy) supplemented with LBLE at levels of 0 (control), 0.5, 1.0, 1.5 or 2.0 g/kg diet were served to fish in triplicates (each replicate has 20 fish) until they appeared satiated for 12 weeks. Then, they were challenged with Aeromonas hydrophila infection for further 2 weeks. Fish served dietary LBLE had considerable higher weight gain (64.76 g) and specific growth rate (765.5% g/day) than the fish group fed with the control diet with a reduced feed conversion ratio (1.45) (p < 0.05). Compared to fish fed with the control diet, the villi height (146.30 µm) and width (284.35 µm) of the intestines increased greatly in a dose (LBLE in diets) dependent order. Similarly, dietary LBLE increased (p < 0.05) the packed cell volume (46.11%) and haemoglobin (17.03 g/dL), whereas 1.5 g of LBLE increased the counts of white blood cells. Glutathione S-transferase (769.43 U/mg protein), glutathione peroxidase (84.14 U/mg protein) and superoxide dismutase (433.15 U/mg protein) activities were significantly higher (p < 0.05) in fish fed with diets supplemented with LBLE compared to the control one. Additionally, dietary LBLE increased phagocytic and lysozyme activities and protected C. gariepinus against bacterial infections where the lowest death was observed in the fish fed on the diet containing 1.5 g LBLE/kg feed. These findings showed that the fish fed with LBLE/kg diet improved their immune system, antioxidant and growth performance in addition to providing protection from A. hydrophila infection with the optimum dose of 1.80 g/kg diet.

This study was designed to evaluate the tolerance of Clarias gariepinus juveniles to a gradual and abrupt increase in salinity over time. To this effect, C. gariepinus juveniles were exposed to three salinity incremental protocols namely 1 g L-1 day-1, 5 g L-1 day-1, and 10 g L-1 day-1. Changes in the hematological parameters and the gill histology of fish were analyzed to determine the impact of osmotic stress on the health status of the fish and its osmoregulatory ability. The result obtained showed that juveniles of C. gariepinus can tolerate salinity stress up to 14 g L-1. At 15 g L-1 and beyond, all samples died regardless of gradual (i.e., 1 g L-1 day-1 administered for 15 days) or abrupt salinity exposure (i.e., 5 g L-1 day-1 administered for three days and 10 g L-1 day-1 administered for two days). Interestingly, more than 90% of the fish survived a direct 10 g L-1 exposure for 24 h without prior acclimation. The hematological parameters accessed in the fish exposed to 10 g L-1 (either gradually or abruptly) showed a significant increase in the white blood cells and a decrease in the red blood cells, packed cell volume, hemoglobin concentration, and all derived blood parameters. The results of the serum biochemistry show a lower total protein and albumin in the salinity-treated fish compared to the control group. However, the serum glucose and the plasma electrolytes (i.e., K+, Na+, and Cl-) were higher in the former group than in the latter. Aside from the stress response expressed in the blood parameters, severe gill degenerations were seen in the histological micrograph obtained for the salinity-treated fish, while the control had a near-normal gill architecture. It was concluded that C. gariepinus could tolerate salinity exposure of 10 g L-1 day-1 (administered gradually or abruptly) and below without killing the fish within 24 h.

Aeromonas veronii is a pathogenic bacterium associated with various diseases in aquaculture. However, few studies address the antibacterial activity using nanoparticles (NPs). Hence, the current study is innovative to evaluate the antibacterial efficacy of silica nanoparticles (SiNPs) against A. veronii infection in-vitro with a trial for treatment in-vivo. Primarily, we assessed the in-vitro antibacterial activity against A. veronii. Further, we investigated the hematological profile, immune-antioxidant response, and gene expression of African catfish (Clarias gariepinus) in response to SiNPs exposure and the A. veronii challenge. Fish (N = 120; weight: 90 ± 6.19 g) were distributed into four groups (30 fish/group) for a ten-days-treatment trial. The first (control) and second (SiNPs) groups were treated with 0 mg/L and 20 mg/L SiNPs in water, respectively. The third (A. veronii) and fourth (SiNPs + A. veronii) groups were treated with 0 mg/L and 20 mg/L SiNPs in water, respectively, and infected with A. veronii (1.5 × 107 CFU/mL). Results demonstrated that SiNPs displayed an in-vitro antibacterial activity against A. veronii with a 21 mm inhibitory zone. A. veronii infection caused a high mortality rate (56.67%) and substantial reductions in hematological indices and immune indicators [nitric oxide (NO) and immunoglobulin M (IgM)]. Additionally, marked decline in the level of antioxidants [superoxide dismutase (SOD), catalase (CAT), and reduced glutathione content (GSH)] as well as down-regulation in the immune-related genes [interleukins (IL-1ß and IL-8) and tumor necrosis factor-alpha (TNF-α)] and antioxidant-related genes [SOD1, glutathione peroxidase (GPx), and glutathione-S-transferase (GST)] were the consequences of A. veronii infection. Surprisingly, treatment of A. veronii-infected fish with SiNPs lessened the mortality rate, enhanced the blood picture, modulated the immune-antioxidant parameters, and resulted in gene up-regulation. Overall, this study encompasses the significant role of SiNPs, a new versatile tool for combating hematological, immuno-antioxidant alterations, and gene down-regulation induced by A. veronii infection and sustainable aquaculture production.

In applying bioanalytical approaches, the aim of this study was to determine the toxicity of contaminants derived from a solid waste dumpsite in Calabar (Nigeria), by investigating the alterations of lipid and estrogen signaling pathways in Poeciliopsis lucida hepatocellular carcinoma-1 (PLHC-1) cells and compared to in vivo African catfish (Clarias gariepinus), using polar, nonpolar and elutriate extraction methods. Cells were exposed for 48 hr period to different concentrations of the contaminant extracts. The PLHC-1 cells were evaluated for lipid responses as follows adipoRed assay, retinoid x receptor (rxr), peroxisome proliferator-activated receptor isoforms (ppar-α and γ), estrogen receptor (er-α) and vitellogenin (vtg) transcripts. The lipid signaling activation was also assessed in vivo using C. gariepinus, where hepatic levels of ppar-α were determined at both transcript and functional proteins levels. Data showed variable-, extract type and concentration-specific elevations in mRNA and protein levels for lipidomic and estrogenic effects. These effects were either biphasic at low and high concentrations, depending upon extract type, or concentration-dependent elevations. In general, these toxicological responses may be attributed to soil organic and inorganic contaminants burden previously derived from the dumpsite. Thus, our data demonstrate a unique lipid and endocrine-disruptive chemical (EDC) effects of each soil extract, suggesting multiple and complex contaminant interactions in the environment and biota. Analysis of numerous soil- or sediment-bound contaminants have numerous limitations and cost implications for developing countries. Our approach provides a bioanalytical protocol and endpoints for measuring the metabolic and EDC effects of complex environmental matrices for ecotoxicological assessment and monitoring.

Aquatic pollution may continue to deepen following the emergence of new class of toxicants. The present study investigated the effect of water-soluble fraction of burnt tire-ash on Clarias gariepinus. The fish were exposed to sublethal doses; 0.00 g/L, 2.24 g/L, 1.12 g/L and 0.56 g/L of tire-ash solution, representing 1/5, 1/10 and 1/20 of 11.2 g/L median lethal concentration (96 LC50), for 28 days, followed by 14 days recovery trial. Biological sampling was done on exposure day 1, 14 and 28, and on day14 recovery period for biochemical analysis such as the liver and gill Na+/K+ and Ca2+-ATPase, serum immunoglobulin (IgM) and brain acetylcholinesterase (AChE) of the experimental fish. Also, body biomass and behavior were evaluated. The behavioral responses exhibited by the fish to BTA exposure include reduced feeding, hypoactivity, air gulping and skin discoloration, which was observed to be concentration dependent. The body weight of 2.24 g/L and 1.12 g/L BTA-exposed fish decreased significantly than 0.56 g/L exposed fish and the control. Furthermore, findings revealed evident induction of Na+/K+ and Ca2 +-ATPase activities in both tissues, elevation of serum immunoglobulin content and inhibition of AChE activity in the brain of the exposed fish relative to the control. However, it was also observed that the biochemical parameters normalized after the recovery period. In conclusion, water-soluble fraction of burnt tire-ash produced toxicological effects in the experimental model, hence the present study provides the ecotoxicological insight of tire ash.

This study investigated the acute toxicity in Clarias gariepinus to insecticides currently used in Benin cotton fields, including Thalis 112 EC (Emamectin benzoate 48 g L-1, Acetamiprid 64 g L-1), Vizir C 92 EC (Cypermethrin 72 g L-1, Abamectin 20 g L-1), Pyrinex Quick 212 EC (Deltamethrin 12 g L-1, Chlorpyrifos 200 g L-1) and Pyro FTE 472 EC (Cypermethrin 72 g L-1, Chlorpyrifos 400 g L-1) with emphasis on liver histopathological effects. A set of 180 juveniles of C. gariepinus (5.21 ± 3.22 g) was exposed for 96 h to increasing concentrations of each pesticide. The values of 96-hLC50 were 4.778, 0.002, 0.004, and 0.012 µL L-1 for Thalis, Vizir, Pyrinex, and Pyro, respectively, indicating that Vizir, Pyrinex, and Pyro were very highly toxic to C. gariepinus juveniles. During the experiments, the morphological and behavioral responses (discoloration, hyperactivity, lethargy, etc.) were observed in exposed fish, hypothesizing the neurotoxicity of these pesticides. Histopathological alterations observed in liver of contaminated fish were regressive changes, such as necrosis, vacuolation, bleeding, nuclear degeneration, hepatocytes degeneration, sinusoids dilatation, etc. Vizir induced the highest histological alteration indices while the lowest were induced by Thalis, confirming the highest toxicity of Vizir. These results indicate that acute concentrations of these insecticidal molecules have destructive effects on the liver of C. gariepinus.

Bacterial infections have been associated with immune dysfunction and oxidative stress in cultured fish species while essential elements could boost immunity and exhibit antioxidant properties in fish. This study was therefore aimed at determining the effects of pre-treatment with waterborne selenium on humoral immunity and redox status of Clarias gariepinus experimentally challenged with Serratia marcescens. Juveniles C. gariepinus were pre-treated with 50 µg/L selenium for 14 days after which they were challenged with 5 × 103 CFU/mL of S. marcescens via oral gavage for 24 or 48 h. The control fish were not pre-treated with selenium and not challenged with bacteria. Thereafter, fish were sacrificed, blood collected into EDTA bottles for the determination of plasma nitric oxide levels and respiratory burst, and the liver excised for the determination of reduced glutathione, lipid peroxidation, and activities of catalase, superoxide dismutase, and glutathione peroxidase. Fish that were pre-treated with selenium prior to bacterial challenge (Sel + Bact) had decreased levels of nitric oxide and lipid peroxidation but a significant increase in the levels of reduced glutathione (at 48-h post-infection period only) compared to the fish challenged with bacteria without prior selenium pre-treatment (Bact). The respiratory burst and catalase activity decreased significantly in the Sel + Bact group especially at 48-h post-infection period while the activity of glutathione peroxidase increased significantly in the Sel + Bact group (at 24-h post-infection period only) compared to the Bact group. The results from this study showed that infection with S. marcescens is capable of disrupting the immune system and redox homeostasis in C. gariepinus, while pre-treatment with selenium has the ability to improve the physiological status of fish that were challenged with bacteria probably through its antioxidant properties. HIGHLIGHT: The pre-treatment of Clarias gariepinus to waterborne selenium for 14 days improved the redox homeostasis and innate immunity of fish that were experimentally challenged with the bacterium, Serratia marcescens.

Pituitary adenylate cyclase-activating polypeptide (PACAP) is a multifunctional neuropeptide that belongs to the secretin/glucagon/GHRH/VIP superfamily. Some of these molecules have antimicrobial activity and they are capable of stimulating the immune system. The present work studied the antibacterial and immunostimulatory activity of PACAP-38 from African catfish Clarias gariepinus against the Gram-negative bacterium Pseudomonas aeruginosa in an in vivo test. PACAP-38 improved antimicrobial activity of skin mucus molecules against P. aeruginosa. The peptide modulates the gene expression profile of TLR-1, TLR-5, MyD88, IL-1ß, TNF-ɑ, IL-8, pardaxin, hepcidin and G/C-type lysozymes in skin, spleen and head kidney. The influenced exerted depended on the time after infection and tissue analyzed. This study provides the first evidence of a link between PACAP and antimicrobial peptides hepcidin and pardaxin. Our results suggest further use of PACAP as antimicrobial agent that could potentially be used to control disease in aquaculture.

Selection of suitable promoters is crucial for the efficient expression of exogenous genes in transgenic animals. Although one of the most effective promoters, the ß-actin promoter, has been widely studied in fish species, it still remains unknown in the economical important African catfish (Clarias gariepinus). In this study, the ß-actin promoter of African catfish (cgß-actinP) was cloned and characterized. In addition, recombinant plasmid pcgß-actinP-EGFP with enhanced green fluorescent protein (GFP) gene as the reporter gene was constructed to verify the transcriptional activity. We obtained a cgß-actinP fragment length of 1405 bp, consisting 104 bp of the 5' proximal promoter, 96 bp of the first exon, and 1205 bp of the first intron. Similar to those of other fish species, cgß-actinP contains three key transcription regulatory elements (CAAT box, CArG motif, and TATA box). GFP-specific fluorescent signals were detected in chicken embryonic fibroblasts cells (DF-1 cells) transfected with pcgß-actinP-EGFP, which was approximately 1.11 times of the positive control. In addition, GFP was effectively expressed in zebrafish larvae microinjected with linearized cgß-actinP-EGFP, with expression rate reaching approximately 49.84%. Our data indicate that cgß-actinP could be a potential candidate promoter in the practice of constructing "all fish" transgenic fish.

Parasitic infection may cause massive losses in Clarias gariepinus fries and fingerlings. Therefore, this study aimed to characterize the digenetic trematodes species (two adults' flukes and one metacercariae) infecting African catfish Clarias gariepinus, as well as their histopathological impacts on infected fish. The intestinal flukes were identified as Orientocreadium batrachoides and Masenia bangweulensis based on their morphological and molecular characteristics. Sequencing of their 28S (LSU rRNA) and 18S rRNA (SSU rRNA) genes confirmed that these trematodes belong to the families Orientocreadiidae and Cephalogonimidae, respectively. The metacercariae trematode infecting skin and muscles were only morphologically identified as Cyanodiplostomum sp. The gene expression levels of MHC II increased in naturally infected fish either with O. batrachoides or Cyanodiplostomum sp. alone, compared with uninfected catfish. In addition, lysozyme levels in individual fish serum increased in catfish infected either with O. batrachoides or Cyanodiplostomum sp. alone. Histopathological examination of the skin revealed embedded parasitic cysts that displaced tissue in the dermis. Surrounding tissues were infiltrated with melanomacrophages and displayed dermal edema. Histopathological analysis showed O. batrachoides or M. bangweulensis between the gastric folds of the stomach of infected catfish, causing infiltration of mononuclear inflammatory cells in the lamina propria.

Abattoirs are positioned close to freshwaters in most developing countries where untreated wastewaters are discharged into with potential risk to aquatic life such as fish and ultimately human health. We assessed physicochemical parameters, antibiotics (oxytetracycline and diclofenac) and algal load of effluent collected from a major abattoir in Nigeria. Furthermore, liver function enzymes, genotoxic and haemotoxic effects of subacute concentration (10% of 96 hLC50 value) of the wastewater were evaluated over a period of 28 d in Clarias gariepinus (The African Sharptooth Catfish). The 96 hLC50 value of the abattoir wastewater against C. gariepinus was 154.14 mL/L (15.4%). Nitrates, phosphates, sulphates, chloride, ammonia, TDS, TSS, BOD5 and heavy metals (Fe and Pb) in the wastewater were above permissible limits while diclofenac and oxytetracycline were below detection limit (BDL). Microalgae in the wastewater were mostly Bacilliariophyta (Navicula spp.) (45.64%) and euglenoids (Euglena and Phacus spp.) (49.48%). Liver function enzymes (LDH, AST, ALT) level were higher in exposed fishes except for ALP which was lower at day 28 compared to control levels. Erythrocytic genotoxic indices (nuclear abnormalities) were significantly higher (p < 0.05) in the exposed fishes particularly at day 28 compared to control. Haematologic indices level such as WBC, MCV MCH, MCHC increased significantly (p < 0.05) while lymphocytes, HGB, RBC, HCT levels decreased significantly (p < 0.05) in the exposed C. gariepinus by day 28 compared to control. These results demonstrate potential adverse effects posed to aquatic fish species in the Ogun River by the discharge of the abattoir effluent. The microalgae species identified in the effluent may be explored for pre-treatment of the effluent before discharge in order to prevent eutrophication and increased pollutant load in the River. The study results will contribute to evidence-based environmental risk management of the River which is relevant to the UN SDGs 6 (clean water and sanitation), 11 (sustainable cities and communities) and 14 (sustaining life below water).

The optimum distance and duration of ultraviolet (UV) irradiation for the complete inactivation of African catfish Clarias gariepinus egg nucleus was investigated in this study. The UV light was suspended above the unfertilized eggs at four distances (5, 10, 20 and 30 cm) and for five durations (1, 2, 3, 4 and 5 min). Then, the irradiated eggs were activated with sperm from diploid C. gariepinus and cold shocked at 5°C for 5 min just moments before cell cleavage. Ploidy analysis was performed using karyotype chromosome counting. The results obtained suggested that the further the distance, the better the hatchability rate, however prolonged duration seemed to significantly reduced hatchability. All treatments with surviving progenies at the end of the study showed evidence of successfully diploid gynogen (2n = 56) induction at different percentages. However, the optimal protocol that gave a moderately high hatchability/survival rate and completely induced gynogens was exposure of the eggs to UV irradiation at 20 cm for 1 min. It was concluded that the distance and duration of UV irradiation affects gynogenetic induction in African catfish C. gariepinus.

The safety of aquatic ecosystems has been compromised by numerous anthropogenic activities, especially leachates from non-point source toxicants, leaching into aquatic systems. This study evaluated the toxicity of the water-soluble fractions (WSFs) of burnt tire ash (BTA) on Clarias gariepinus via a battery of integrated biomarkers. Juvenile C. gariepinus were exposed to sublethal (0.56, 1.12, and 2.24 g/L) concentrations of BTA, derived from 11.2 g/L median lethal concentration (96 LC50), at duration intervals of 1, 14, and 28 days, followed by a recovery trial that lasted for 14 days. Serum biochemical parameters, antioxidant enzyme activities of the gill and liver, lysozymes activity and erythron profile were assessed. The findings of the present study revealed that BTA-WSF induced prominent alterations on biochemical parameters, lysozymes activity and antioxidant enzymes activities in the exposed fish. Furthermore, toxicant exposure promoted oxidative stress, cellular damage and genotoxicity (erythrocytic nuclear and cellular abnormalities) in the exposed fish. In general, a post-exposure trial showed partial recovery from the exposure effects of the toxicant, following the evident modifications of serum enzymes and erythron pathopathology in the experimental model. Biomonitoring of BTA, using sentinel aquatic species such as C. gariepinus, provides insights into the ecotoxicological potency of this toxicant.

The applications of exogenous hormones in different species for the induction of oocyte production, final oocyte maturation (FOM), and spawning for their reproduction is getting more attention day by day. The current preliminary research work was carried out to induce breeding in Clarias gariepinus, commonly known as African catfish, imported from Thailand. Single doses of two hormones as Ovaprim and human menopausal gonadotropin (hMG) were used and the research work was carried out at Muzaffargarh Fish Hatchery Punjab, Pakistan. A total of twenty-four (n = 24) C. gariepinus were selected having body weight approximately 2 kg and divided into two main groups based on gender as male (n = 12) and female (n = 12). For milt collection, all males were treated with Ovaprim 0.5 mg/kg body weight (b.w.) and female fish were divided into three groups as A, B, and C with four (n = 4) fish in each group. Group A was injected with only normal saline (control group) while fish in group B and group C were treated with hMG at 0.5 mg/kg b.w. and Ovaprim 0.5 mg/kg b.w., respectively. Then, after 6 h of hormone injections until 48 h, spawned eggs, eggs' weight, fertilization rate, hatching rate, survival rate, fecundity, and deformed larvae were investigated. The results revealed that Ovaprim injection significantly (p < 0.05) modulate the reproductive parameters in group C while no breeding was induced in both control and hMG-treated groups. Hence, it could be concluded that Ovaprim has the potential to induce breeding in African catfish, while in the current study, hMG failed to induce breeding. However, trials at large scales are required to further explore the effect of different doses of both tested hormones by increasing the treated subjects particularly in Pakistani fish farms.

The objective of the present work was to obtain scientific information on the ecological health of three freshwater lakes (Awassa, Koka, and Ziway) situated in the Ethiopian Rift Valley by investigating possible trace element contamination accumulated in fish. Accordingly, fish liver and kidney samples were collected from three commercially important fish species (Barbus intermedius, Clarias gariepinus, and Oreochromis niloticus) in the lakes to determine the concentrations of chromium (Cr), manganese (Mn), cobalt (Co), nickel (Ni), copper (Cu), zinc (Zn), arsenic (As), selenium (Se), cadmium (Cd), and lead (Pb), using ICP-MS. Trace element concentrations were generally higher in O. niloticus compared with concentrations in B. intermedius and C. gariepinus. Compared to background values of most freshwater fish species, higher liver concentrations of Cu in C. gariepinus and O. niloticus, Mn in O. niloticus, Co in all except B. intermedius, and Zn in C. gariepinus from Lakes Ziway and Awassa were found. Cr, Co, Ni, Cd, and Pb were enriched in kidney, while Mn, Cu, Zn, As, and Se seems retained in the liver tissues. Assessment of transfer factors indicated that bioaccumulation from water and diet occurred, while uptake from sediments was low. Furthermore, the transfer factor values were generally higher for essential elements compared to the non-essential elements. Multivariate statistical analyses showed that the differences between the trace element levels were generally not significant among the lakes (p = 0.672), while significant differences were found between the fish species (p = 0.042), and between accumulation in kidney and liver (p = 0.002).

The nutritional value of Jatropha curcas kernel (JCK) can be improved through different processing methods; however, when using a thermal treatment, optimization of the process is needed to prevent denaturation of nutrients. In this study, JCK was toasted for varying durations (0, 10, 20, and 30 min) and nutritionally evaluated. The implication of feeding Clarias gariepinus with dietary inclusions (35% CP; 315 kcal g-1) of the toasted JCK was also reported. The results obtained suggest that prolonged duration of toasting improved the nutritional characteristics of the JCK until the 20th min. Beyond this time, the protein content and essential amino acids are reduced. However, the antinutrients continuously decreased with prolonged processing. The growth, carcass analysis, and haematology of the fish groups fed toasted JCK at varying duration also did better than those fed raw JCK. Importantly, the performance tends to reduce for those fed JCK toasted beyond 20 min. The estimated cost of producing 1 kg of the fish also substantially reduced with feeding the processed JCK than feeding raw JCK. Histological examination of the intestine and liver tissues further revealed fewer signs of histopathological degeneration for fish-fed processed JCK compared to the control. It was concluded that the processing of JCK by toasting should not exceed 20 min to improve the nutritional composition of the feed ingredients and their dietary utilization by fish.

PURPOSE: Plastic particulates and chemicals are emerging environmental pollutants with significant impact on aquatic ecosystems. In this study, the effects of oral uptake of melamine, melamine formaldehyde, and polyvinyl chloride on serum biochemical profiles, antioxidant enzymes activities, lipid peroxidation levels and brain acetyl cholinesterase activities in Clarias gariepinus juveniles were investigated. METHODS: Fish specimens were fed diets spiked with melamine, melamine formaldehyde and poly vinyl chloride at 0.3% (3.0 g Kg-1) dietary inclusion for 45 days. Toxicity effect of these plastic chemicals was estimated by assaying relevant biomarkers. RESULTS: After 45 days exposure, Serum glucose was significantly elevated, whereas plasma protein levels were substantially reduced in the exposed fish groups. Serum transaminases were significantly elevated in the exposed groups. Brain acetylcholinesterase and antioxidant enzyme activities declined significantly, while malondialdehyde levels were elevated in the exposed groups. CONCLUSION: C. gariepinus is an important bioindicator to monitor the ecotoxicological impact of plastic chemicals such as melamine, and polyvinyl chloride.
